Abstract:Clubroot disease caused by Plasmodiophora brassicae poses a significant threat to the yield of crucifers. However, there is no effective method to control clubroot. Therefore, an in vitro pot experiment was conducted to investigate the response of Pak choi at different concentrations of fluazinam and Trichoderma harzianum in clubroot suppression. T . harzianum was applied at different doses (1, 3, and 6 g/pot) by mixing with the substrate. The results showed that, as the concentration of T. harzianum increased (6 g/pot) it significantly improved plant total chlorophyll contents, shoot fresh weight by 56.11%, 44.60%, enzymatic activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) by 42.14%, 73.72%, and 94.01%, respectively as well as efficiently decreased clubroot disease index by 68.64% as compared to plants only supplemented with P. brassicae . Furthermore, 2 mL of P. brassicae and 20 mL of fluazinam from the diluted solution (0.1, 0.5, and 1 mL/L) were applied to the root zone of each plant. Increased concentration of fluazinam was observed toxic to the tested plants whereas, 0.5 mL/L of fluazinam efficiently suppressed clubroot up to 58.67%, enhanced total chlorophyll contents by 43.53%, shoot fresh weight up to 31.16%, enzymatic activities of SOD, POD, CAT by 37.68%; 64.43%; and 89.19%, respectively. Moreover, metabolomic profiling by liquid chromatography mass spectrometry (LC-MS) investigated the role of secondary metabolites in clubroot reduction which showed that the plants treated with T. harzianum upregulated 15 secondary metabolites including 6-Hydroxydelphinidin 3-glucoside, Jasmolone glucoside, Xanthotoxol glucoside, Chlorflavonin, 4'-Hydroxyanigorootin, Erosnin, 5-O-Methyleriodictyol 7-glucosyl-(1->4)-galactoside, Calomelanol G, Furano (2'',3'':6,7) aurone, Artelastochromene, Gliricidol, alpha,4,2'-Trihydroxy-4-O-geranyldihydrochalcone and 6,7-Dihydroxy-5,4'-dimethoxy-flavylium which might be the possible reason of clubroot suppression in T. harzianum treated plants. The results of this study will aid future research into the biological mechanisms of cruciferous clubroot management.
