Effects of Culture Conditions on Dehydrogenase Activity and Product Composition with Rhizopus Oryzae

Abstract:In order to improve the productivity of L-lactic acid in Rhizopus oryzae, the effects of culture conditions on key dehydrogenases (lactate dehydrogenase, alcohol dehydrogenase) and end products (L-lactic acid, ethanol) were examined. The results showed that 60 g/L glucose in the initial medium led to the highest specific activity of lactate dehydrogenase (LDH), further increase of glucose would reduce LDH's activity. The specific activity of alcohol dehydrogenase (ADH) decrease seriously with the addition of 20 g/L calcium carbonate, but the specific activity of LDH is less affected. The rate of LDH to ADH was improved greatly with the addition of calcium carbonate, and more L-lactic acid and less ethanol are determined in the fermentation broth. The supplement of Zn~(2+) improved the specific activity of ADH and decreased the specific activity of LDH, while the effect of Fe~(3+)as on the contrary. However, when calcium carbonate is added in the initial medium, Zn~(2+) would improve LDH's activity, and the stimulation of ADH's activity by Fe~(3+) is reduced significantly. The specific activities of LDH and ADH are reduced obviously with the increase of running rate, and the increased rate of LDH to ADH was contributed to the accumulation of L-lactic acid.
