Abstract:Physiological states of marine microalgal cells can influence photosynthesis efficiency, which affects approximately half of global carbon fixation. The detection of the algae physiological profiles is important for marine ecology and economy. In this paper, we propose a polarized light-scattering method to detect sensitive changes in the physiological states of the suspended marine microalgal cells. Our experimental setup is designed to measure the scattered polarization parameters of the cells suspended individually in the seawater. Two species of microalgal cells cultured in the laboratory were measured for several days. Experimental results showed that both species display distinctive changes in their polarized photon scattering features corresponding to changes in their physiological states. The changes are far more prominent than those displayed in unpolarized light scattering. Microscopy observations, simulations for microspheres of different diameters and refractive indices, or different shapes, indicated that the polarization features of the scattered photons are sensitive to the submicrometer microstructures of the cells. This study demonstrates the potential of the polarized light-scattering technique to characterize the physiological states of suspended marine microalgae. (C) 2020 Optical Society of America

Abstract:The polarization characteristic of oceanic lidar returns includes a great deal of seawater information, which plays an important role in subsurface layer detection and seawater optical property retrieval. Along with the microphysical properties of particles, the multiple scattering effect between laser light and particles in seawater is also a source of polarization, which obscures the original depolarization properties of the seawater. However, modeling multiple scattering in polarized oceanic lidar with analytical methods is a complex problem. In this paper, a semianalytic Monte Carlo (MC) polarization radiative transfer model is established to assess the impact of multiple scattering on the backscattering depolarization measurement of oceanic lidar. The MC-simulated lidar returns of both the parallel and perpendicular channels and the depolarization ratios coincide well with the experimental results from three stations in the Yellow Sea of China by shipborne polarized lidar. Further analyses show that the multiple scattering effect enhances the depolarization of seawater; specifically, the depolarization ratio of the lidar return increases with increasing penetration depth, receiver's field of view (FOV), single scattering albedo and beam attenuation coefficient of seawater. This semianalytic polarized MC model is useful for understanding the mechanism of polarized multiple scattering in seawater and for further improving the precision of optical property retrieval using oceanic polarized lidar. Abstract:In situ marine algae detection is very important for many global issues. Even though polarized-light scattering has the promising application in the biomedical characterization, its potential to detect marine algae is still not clear. The movements of the suspended marine algae due to their own migrations, arise challenges for the polarization measurement and information acquisition. In this paper, we response the challenges and show the potential of the polarized-light scattering to detect the marine algae. We measure the scattering signals of the aqueous suspension of the glass fibers by use of a 2-channel simultaneous polarization measurement setup and describe the properties of the signals. After comparing and analyzing of the polarizations of glass fibers with different orientations, we reveal the ability of polarized-light scattering to retrieve the orientation distribution of the cylindrical scatterers of the sample, and show the method's potential to detect the marine algae.

Abstract:Microalgae play a vital role in aquatic ecological research, but the fine classification of these tiny and various microalgae cells is still challenging for the community. In this paper, we propose a multimodality technique to simultaneously acquire the polarized light scattering, fluorescence and laser-induced acoustic wave signals originated from individual microalgae cells in water. Experiments of different species of Spirulina and different states of Microcystis have been conducted to test our experiment setup, and the results demonstrate that this method can well discriminate microalgae cells with pigment or microstructural differences. Moreover, with these modalities, the consumption of absorbed energy is evaluated quantitively, and a possible way to assess photosynthesis on a single-cell level is presented. This work is expected to be a powerful technique to probe the biophysical states of microalgae in the aquatic ecosystem.

Abstract:An inelastic hyperspectral Scheimpflug lidar system was developed for microalgae classification and quantification. The correction for the refraction at the air-glass-water interface was established, making our system suitable for aquatic environments. The fluorescence spectrum of microalgae was extracted by principal component analysis, and seven species of microalgae from different phyla have been classified. It was verified that when the cell density of Phaeocystis globosa was in the range of 10(4) similar to 10(6) cell mL(-1), the cell density had a linear relationship with the fluorescence intensity. The experimental results show our system can identify and quantify microalgae, with application prospects for microalgae monitoring in the field environment and early warning of red tides or algal blooms. Characterizing the Kinetics of Suspended Cylindrical Particles by Polarization Measurements

Abstract:Polarization has promising potential to retrieve the information of the steady samples, such as tissues. However, for the fast changing sample such as the suspended algae in the water, the kinetics of the particles also influence the scattered polarization. The present paper will show our recent results to extract the information about the kinetics of the suspended cylindrical particles by polarization measurements. The sample is the aqueous suspension of the glass fibers stirred by a magnetic stirrer. We measure the scattered polarization of the fibers by use of a simultaneous polarization measurement system and obtain the time series of two orthogonal polarization components. By use of correlation analysis, we obtain the time parameters from the auto-correlation functions of the polarization components, and observe the changes with the stirring speeds. Results show that these time parameters indicate the immigration of the fibers. After discussion, we find that they may further characterize the kinetics, including the translation and rotation, of the glass fibers in the fluid field.
Obtaining Shape Information of Marine Microorganisms Using Polarized-Light Scattering

Abstract:Polarized-light scattering methods have shown promising potential in characterizing the structural properties of complex biological tissues. Similar methods can also obtain the shape and intracellular information of aqueous suspended microorganisms. In this paper, we measured the Mueller matrix of algae and an alga phantom which consists of cylindrical and spherical scatterers. We also used a Monte Carlo (MC) model containing spheres and randomly oriented infinitely long cylinders to reconstruct the experimental results of cylindrical microorganisms, such as Synedra sp. and the phantom. Based on the analysis of the phantom's polarization properties, the relationship between the depolarization ratio and the ratio of scattering coefficient of sphere versus cylinders (RSCSC) is given, which proves the potential of polarized-light scattering methods to obtain shape information of the microorganisms.

Abstract:Probing suspended particles in seawater, such as microalgae, microplastics and silts, is very important for environmental monitoring and ecological research. We propose a method based on polarized light scattering to differentiate different suspended particles massively and rapidly. The optical path follows a similar design of a commonly used marine instrument, BB9, which records backscattering of non-polarized light at 120°. In addition, polarization elements are added to the incident and scattering path for taking polarization measurements. Experiments with polystyrene microspheres, porous polystyrene microspheres, silicon dioxide microspheres, and different marine microalgae show that by carefully choosing the incident polarization state and analyzing the polarization features of the scattered light at 120°, these particles can be effectively differentiated. Simulations based on the Mie scattering theory and discrete dipole approximation (DDA) have also been conducted for particles of different sizes, shapes and refractive indices, which help to understand the relationship between the polarization features and the physical properties of the particles. The laboratory system may serve as a prove-of-concept prototype of new instrumentations for applications on board or even with submersibles.
Machine Learning Powered Microalgae Classification by Use of Polarized Light Scattering Data

Abstract:Microalgae are widely distributed in the ocean, which greatly affects the ocean environment. In this work, a dataset is presented, including the polarized light scattering data of 35 categories of marine microalgae. To analyze the dataset, several machine learning algorithms are applied and compared, such as linear discrimination analysis (LDA) and two types of support vector machine (SVM). Results show that non-linear SVM performs the best among these algorithms. Then, two data preparation approaches for non-linear SVM are compared. Subsequently, more than 10 categories of microalgae out of the dataset can be identified with an accuracy greater than 0.80. The basis of the dataset is shown by finding the categories independent to each other. The discussions about the performance of different incident polarization of light gives some clues to design the optimal incident polarization of light for future instrumentation. With this proposed technique and the dataset, these microalgae can be well differentiated by polarized light scattering data.

Abstract:Microalgae classification is important for aquatic environment monitoring, but it is difficult in some in-situ cases. The Mueller matrix is known to encode the physical properties including the sophisticated microstructure of microalgae cells. In this paper, the concept of polarization fingerprint based on the Mueller matrix is presented to extract the parameters to classify microalgae. An experimental setup based on the particulate Mueller matrix polarimetry (PMP) is used to measure 25 kinds of microalgae covering seven phyla to form a Mueller matrix dataset. The proposed sixteen polarization parameters with explicit physical meanings and associations with the structural properties are selected to compose the polarization fingerprint. As a result, different microalgae can be effectively classified by the polarization fingerprint and machine learning algorithms. The contribution of the parameters in the polarization fingerprint is analyzed to provide the specific features for microalgae classification, which gives insights into microalgae's exclusive physical properties. The extensibility and flexibility of the polarization fingerprint are discussed. This work shows the polarization fingerprint's power to classify microalgae, which can help retrieve in-situ information on the community structures of microalgae, and further monitor the aquatic environment.

Abstract:Nanoplastics have been found in marine biological habitats. There is now an urgent need to effectively detect these nanoplastics. Due to the small size of these plastics, it is challenging to directly measure the total abundance while in a suspension. In this study, a backward 120° polarized light scattering device was used to measure the polarization parameters of Phaeodactylum tricornutum, before and after the adsorption of nanoplastics. The polarization parameters were observed to be affected by the adsorption of nanoplastics, with different concentrations and sizes. Moreover, the polarization parameter X changed with time after mixing the nanoplastics with the microalgae, which showed the details of the adsorption of nanoplastics by the microalgae, and was further confirmed by a scanning electron microscope image. The simulation results demonstrated how the polarization parameter X of the scatterers (e.g. microalgae) were affected by the adsorption of the nanoplastics. The results of this work indicate that the polarization scattering method can be used to probe the concentration and size of nanoplastics adsorbed by microalgae in water.

Abstract:The time-dependent optical properties of microalgae are crucial for light transfer in photobioreactor (PBR) designs. Here, the time-dependent optical properties were derived using electromagnetic scattering theory informed by the experimentally measured optical properties of Chlorella protothecoides. The temporal scaling functions (TSFs) of nearly wavelength-independent absorption and scattering cross-sections were demonstrated using the electromagnetic scattering theory, leading to the first concrete expression of the TSF. The TSF establishes the relationship between the time-dependent absorption/scattering cross-sections at the stationery and growth phases of microalgal development. The concrete expression of the TSF provides a new way to calculate the time-dependent optical properties of microalgae using electromagnetic scattering theory. The areas where the TSF of microalgae has exciting potential are in remote sensing and photobioreactor applications.

Abstract:We take low resolution Muller matrix images of microalgae using a Mueller matrix microscope based on time sequential polarization measurements and obtain a characteristic average Mueller matrix (AMM) which can be evaluated for each cell from the average values of the polarization component images. Using the 136 first and second order terms of the characteristic Mueller matrix elements and linear discriminant analysis (LDA), we can obtain a characteristic polarization parameter for each type of algae and use it to stain and identify these specific cells in a mixed sample containing three strains microalgae with an accuracy higher than 84%. The results demonstrate that AMM can be used for the high-throughput detection of marine microalgae. © 2018 Elsevier GmbH
