Abstract:Shake flask culture was used and the yield of polysaccharide was taken as main index to ensure the optimum culture conditions of submerged fermentation for Ganoderma lucidum by corn mash medium. The optimum culture conditions were obtained as followed: the culture temperature 28 ℃; the rotation rate 180 r/min; the initial pH6.0; the culture time 144 h. Under the optimum culture conditions, the yield of polysaccharide increased by 26.98 % as compared with the original one.

Abstract:Objective: In order to raise the yield of Ganoderma lucidum polysaccharide,submerged fermentation conditions of Ganoderma lucidum were optimizated.Methods: Using medium of corn mash as main component,single factor experiment and orthogonal experiment were conducted to study submerged fermentation conditions such as fermentation temperature and shaking table rotation rate etc.which affecte the yield of Ganoderma lucidum polysaccharide.Results: By range analysis and variance analysis,the optimum submerged fermentatio conditions were obtained as followed: the fermentation temperature 27℃,the shaking table rotation rate 170r/min,the medium initial pH 6.0,the fermentation time 144h.Conclusion: By optimizating submerged fermentation conditions,the yield of Ganoderma lucidum polysaccharide could be raised remarkably.Under the optimum fermentation conditions,the yield of Ganoderma lucidum polysaccharid increased from 1.851g/L to 2.439g/L,that had been raised 31.0% as compared with the original yield.

Abstract:Compared shake culture,two-stage culture process by combining shake culture with static cultureand solid-state fermentationof Ganoderma lucidum,analyzed the influence of different cultivation methods on total ganoderic acid,ganoderic acid Mk,ganoderic acid S,ganoderic acid T and ganoderic acid Me synthesis.Corn was used as the main medium to produce ganoderic acids,Plackett-Burman(PB)design was applied to screen potential key factors associated with total ganoderic acid content.Then,the steepest ascent experiment,central composite design and response surface methodology were applied to optimize the significant factors.The final results were as follows:39.0 g solid substrate (moisture content 38.0％) shake flask,1.08％ silkworm chrysalis powder,0.86％ sucrose,0.08％tri-ammonium citrate,0.22％ KH2PO4,0.13％ MgSO4,0.02％ VB1,culture temperature 29.5 ℃,and fermentation time 10 days.Under this condition,the total ganoderic acids content was 24.93 mg every gram of the ganoderma lucidum mycelium and corn powder,and ganoderic acid Mk,ganoderic acid S and ganoderic acid content reached 0.86,0.69 and 3.69 mg/g,respestively.Production of three ganodericacids by Ganoderma lucidum solid-state fermentation has good application prospect.
Optimization of Shake-Flask Fermentation Conditions of Ganoderma Lucidum

Abstract:The inoculum,rotation speed and medium volume in flask affect the growth and polysaccharide production of Ganoderma lucidum to vari-ous extents. Orthogonal tests showed that the inoculum had significant impacts. The fermentation conditions of G. lucidum were optimized as fol-lows: inoculum 10%,rotation speed 150 r/min,media volume in flask 125ml/500ml. The study provides data for the scale-up fermentation of G. lu-cidum in fermentor.
Optimization of Cultural Condition of Ganoderma Lucidum in the Compound Culture Medium by Response Surface Methodology

Abstract:The purpose is to obtain optimal cultural conditions for Ganoderma lucidum in the solid medium mainly containing the residual of Pteridium aquilinum,and to provide theoretical foundation for the further use of this material from Pteridium aquilinum.Response surface methodology was applied to optimize the main culture conditions including proportion of the residual compound in medium,water content in medium and culture temperature.The results showed that proportion of residual of Pteridium aquilinum in medium,water content in medium and cultural condition had noticeably significant effects on daily average growth rate of mycelium of Ganoderma lucidum(p0.01).Obviously correlation was found to be existed between proportion of residual of Pteridium aquilinum and water content in medium,or water content in medium and the cultural temperature.The optimal proportion of residual of Pteridium aquilinum in medium was 85.0%.The optimal water content in medium was 62.4%,and the optimal cultural temperature was 27℃.Under these optimized conditions,daily average growth rate of mycelium of Ganoderma lucidum was 3.48mm/d.Multivariate regression analysis showed that the model for regressing proportion of residual of Pteridium aquilinum in medium,water content in medium,cultural temperature and daily average growth rate of mycelium of Ganoderma lucidum might be available for the culture of the fungi.Practical growth rate of mycelium of Ganoderma lucidum could be forecasted by applying the model.The application of residual of Pteridium aquilinum as a main additive in the culture of Ganoderma lucidum was firstly reported.It has laid a pave for the further investigation of residual of Pteridium aquilinum in medium.

Abstract:Effect of sources of carbon and nitrogen on mycelia dry weight and conditions optimization under submerged culture of Armillariella tabescens was studied.It is showed that the best carbon and nitrogen source were corn flour and soybean powder respectively.The optimum medium was: corn flour: 4%,soybean powder: 1%,yeast extract 0.3%,KH2PO4: 0.1%,MgSO4: 0.1%.The optimum technology was initial pH 5.0,started glucose concentration 4%,100 ml medium in 300 ml flask and calcium carbonate 0.5%.High yield of exopolysaccharide will reach under the optimum culturing conditions.

Abstract:The culture conditions of Helvella crispa Fr.in shake flask liquid culture were investigated in the experiment.Results showed that the optimal medium of liquid submerged culture was as follows:wheat flour 20 g/L,HN4SO4 2 g/L,MgSO4 0.5 g/L,KH2PO4 0.46 g/L,K2HPO4 1.0 g/L;when the medium capacity was 300-400 mL/L,and the seed(about 5 mL) was inoculated,the shake flask culture experiment was carried out at 26-28 ℃,pH 5.5-6.5 in a shake incubator with a rotation rate 100 r/min for 7 days.The maximum mycelial biomass was as high as 5.867 g/L.

Abstract:Fermentation medium and culture conditions to improve the spore concentration and antifungal activity of Brevibacillus brevis ch2-22 at the condition in shaking-flask were optimized. Firstly,based on single factor experiment the ferment medium for B. brevis ch-22 was optimized by response surface methodology,then confirmed the optimized ferment conditions with single factor experiment,obtained the optimized ferment medium was as followed,starch 35. 05 g/ L,soybean powder 26. 08 g/ L,sucrose 10 g/ L,fish meal 5 g/ L,NaCl 1 g/ L,MgSO40. 3g/ L, (NH4 )2SO43 g/ L,MnSO40. 1g/ L,K 2 HPO43 g/ L,yeast extract 1g/ L,CaCO32g/ L,incubated at 32 ℃,sha-king 180,filling at volume 100 mL in 500 mL,inoculum at 4％,initial pH 7. 0,culture for 45 h. The colony-forming units per milliliter were improved from 1. 6 × 109 to 8. 1 × 109 after optimization,and the spore concentration was in-creased by 3. 7 times. The broth valance reached as much as 3350 IU/ mL,increased by 109％,2000 IU/ mL higher than the strain of same kinds.

Abstract:The effect of culture medium compositions on gellan gum production produced by fermentation with a halobacterium Sphingomonas paucimobilis QHZJUJW CGMCC2428 was studied. In this work, a fractional factorial design was applied to investigate the main factors that affected gellan gum production by S. paucimobilis QHZJUJW CGMCC2428. Sucrose was the best carbon source for gellan gum and peptone displayed better inducing effect. Central composite design and response surface methodology were adopted to derive a statistical model for optimizing submerged culture medium composition. These experimental results showed that the optimum culture medium for producing gellan gum was composed of 40.00 (w/v) sucrose, 3.00% peptone (w/v), MgSO4 (w/v), 9.20% KH2PO4 (w/v), 7.50% Na2HPO4 (w/v), 4.30% K2SO4 (w/v), pH 6.8-7.0. The maximal gellan gum was 19.89 +/- 0.68 g/L, which was agreed closely with the predicated value (20.12 g/L). After incubated for 72 h under the optimized culture medium in 5-L bioreactor, the gellan gum fermentation reached about 19.90 +/- 0.68 g/L, which was higher than that in the initial cultivation medium. Abstract:Effects of different carbon and nitrogen sources and their concentration on submerged culture for Cordyceps militaris were investigated in shake flasks.Glucose and peptone were found to be most favorable to mycelia biomass and exopolysaccharides production.To determine the optimal culture conditions,response surface methodology was used to analyze the three major factors including glucose concentration,peptone concentration and fermentation time.The optimal submerged culture conditions achieved were glucose 33.30g/L,peptone 12.50g/L,fermentation time 8d.Under these conditions,maximum productions of mycelial biomass and exopolysaccharides reached 18.05 g/L and 1.13 g/L respectively.

Abstract:In this paper,culture conditions on liquid strain of the three white-rot fungi were studied for the optimization.The efficiency of study and application in white-rot fungi in the relevant fields can be improved in a large extant.The results showed that the optimum culture condition on liquid strain of Phanerochaete chrysosporium was 4 days culture period,the initial pH 6.0,liquid capacity 50 mL,agar recruitment 0.2%.The optimum culture condition on liquid strain of W3 was 5 days culture period,the initial pH 6.5,liquid capacity 50mL,agar recruitment 0.3%.The optimum culture condition on liquid strain of Trametes versicolor was 5 days culture period,the initial pH 6.5,liquid capacity 75 mL,agar recruitment 0.1%.

Abstract:Objective: To study the optimum culture media and liquid conditions in shaking flasks for laccase production by Ganoderma lucidum. Methods: Taking S3 stain of G. lucidum as test materials and laccase activity of G. lucidum as measurement index to optimize the culture media and liquid conditions through orthogonal test. Results: The optimum culture components of media were: Glucose 30 g/L, cotton 0.2%, (NH4)2HPO4 0.66 g/L, casein 0.5%, Tween-80 0.15 mL; the optimum conditions were: initial pH value of medium was 5.5, 75 mL medium was in 250 mL flask, inoculation was 12.5%, mycelium age was 5 × 24 h for 9 × 24 h culture. Conclusion: Laccase activity of G. lucidum has been improved remarkably in the optimum culture media and liquid conditions.

Abstract:The production capacities of polysaccharide (GLP) had been examined among the Gandoerma lucidum strains by submerged fermentation. The results showed that the highest yield of GLP was produced by Ganoderma lucidum G22. The single factor test and orthogonal design test for culture conditions of submerged fermentation for GLP production were obtained as follows: when fermenting with constant temperature and constantly offering oxygen,the culture temperature was 27℃; The equipment rotational speed was 170r/min; the beginning pH value of medium was 6.0; the culture period was 144h. Under the optimal culture conditions,the yield of GLP was 2.421g/L by submerged fermenting Ganoderma lucidum G22.

Abstract:Clostridium cellulolyticum, as one of obligate anaerobic bacteria capable of secreting cellulosome, has not been efficiently cultured due to its strict requirement of growing conditions. In this study, culture conditions of C. cellulolyticum were optimized using response surface methodology. Plackett-Burman design was first used to screen the dominant impact factors for the growth of C. cellulolyticum, which were determined as yeast extract concentration, cellobiose concentration and culture temperature. The steepest ascent path design was then applied to gain the suitable range close to the optimal culture conditions for obtaining high cell density. The central composite design and the response surface analysis were finally used to determine the optimal levels of the influential factors, which were 3 g/L for yeast extract concentration, 7 g/L cellobiose concentration and 34 degrees C for culture temperature. The optimized medium was used for flask culture, and OD600 of C. cellulolyticum was increased from 0.303 to 0.586. With a pH-controlled fermentor at batch mode, OD600 reached 3.432, which was 2.8 times higher than elsewhere reported. These results support further study on the high-density culture of C. cellulolyticum and its application.

Abstract:This study aimed at optimizing the medium of a new Ganoderma lucidum strain CAU5501 to enhance the yield of exopolysaccharides (EPS) and mycelial growth. Firstly, the suitable level of glucose, magnesium, phosphate and C/N ratio was determined by single factor experiment. Subsequently, the optimum concentrations of these medium components were investigated using the orthogonal matrix method. The results indicated that the higher levels of EPS were correlated with the level of cell growth when glucose concentration was studied (data no show). The optimum medium for EPS yield was found to be 70 g/l glucose, 5 C/N ratio, 2.5 g/l KH2PO4, 0.75 g/l MgSO4·7H2O, and for mycelial growth was 50 g/l glucose, 5 C/N ratio, 1.5 g/l KH2PO4, 0.5 g/l MgSO4·7H2O. When cultivated in the obtained optimal media in 3 L shake flask, compared to the basal medium, the EPS yield increased markedly from 1.003 to 1.723 g/l, and the mycelium formation was also markedly improved from 2.028 to 7.235 g/l. Results obtained in this study are beneficial to further study for enhancing the production of Ganoderma lucidum polysaccharides in large scale commercialized production.

Abstract:The solid culture medium of Cordyceps militaris was optimized with Plackett-Burman and Box-Behnken designs combined with desirability function.Desirability function was used to construct a comprehensive value D,which was an integration index of the growth cycle of fruiting body,the yield of fruiting body,and the contents of polysaccharide,protein,adenosine and mannitol,for the optimization of solid culture medium of C.militaris mutant N165.The optimal medium formula for C.militaris N165 consisted of 10.33 g/L yeast extract,27.24g/L sucrose,5.60g/L KH2PO4,5.00g/L peptone,1.00g/L MgSO4·7H2O,0.011g/L ZnCl2,0.05g/L VB1 and 21.00g/L(NH4)2SO4.
Optimization of solid-state fermentation medium of Ganoderma lucidum and nutrition component analysis

Abstract:The solid-state fermentation medium of Ganoderma lucidum was optimized by the response surface methodology.And the nutrient component of powder fermentated by Ganoderma lucidum was tested,so as to provide a reference for further development of functional food.The results showed that the optimal medium formulations of solid-state fermentation by Ganoderma lucidum were 24% wheat,12% soybean flour and 64% corn powder of 8 mesh.The result of validation test indicated that the polysaccharide content was 3.27% under the optimum conditions.The analysis of nutritional components of substrate before and after fermentation showed that the solid-state fermentation of Ganoderma lucidum could reduce the content of starch and crude fat in the substrate by 43.27% and 9.76%,respectively,and increase the content of reducing sugar,protein and vitamine B1 by 684%、35.94% and 167%,respectively.According to amino acid analysis,it was found that the amino acid content increased by 7% after Ganoderma lucidum fermentation,moreover the content of all the essential amino acids except lysine increased.
Statistical Optimization Of Cultivation Conditions For Saccharomyces Boulardh Via Central Composite Design

Abstract:Saccharomyces boulardii (S. boulardii) is used in many countries as both a preventive and therapeutic agent for diarrhea and other gastrointestinal disorders caused by the administration of antimicrobial agents. However, there are few reports on its culture conditions. Therefore, the aim of this study was to optimize the cultivation conditions of S. boulardii by single factor experiment and central composite design of Response Surface Methodology (RSM). The effects of temperature (28, 31, 34, 37, 41 degrees C), initial pH value (2, 4, 6, 8 and 10), liquid loading (25, 50, 75, 100 and 125 mL in 250 mL triangular flask), inoculum size (1, 3, 5, 7 and 9 %), and shaking speed (140, 160, 180, 200, 220 rot.min(-1)) were investigated. Among these variables, the combination of 37 degrees C temperature, 6.4 the initial pH value, 250 mL triangular flask with 52 mL medium volume, 3 % inoculum size, and 181 rot.min(-1) shaking speed were found to be the most suitable cultivation conditions. Under the optimized conditions, the maximal yeast count was (1.81 +/- 0.06)x10(8) CFU.mL(-1), which was in a good agreement with the predicted value of the model. This study has provided useful information on how to improve the viable cells of yeast cells for S. boulardii culture.
Cultural characteristics of mycelia of Ganoderma gibbosum

Abstract:To study the cultural characteristics of mycelia of Ganoderma gibbosum, a medicinal fungus used in China. The growth rate and biomass of G. gibbosum mycelia were measured under different temperature, lightning carbon and nitrogen sources conditions. It showed that the optimal growth temperature for mycelia was 25 degrees C. Darkness was beneficial for mycelium growth. The initial pH 5.5 was suitable. The sucrose was the best carbon source and yeast extract the best nitrogen source, the optimal carbon-nitrogen ratio 60:2. These conclusions will offer references for further artificial cultivation and liquid fermentation.
