Resting-State Brain Network Dysfunction Characteristic of Alzheimer’s Disease

Abstract:Alzheimer’ s disease ( AD) is a common neurodegenerative disease which seriously harms the health of the old.There is no effective way of early diagnosis yet.This paper aims to study interval brain functional connectivity ( FC) anomalies in patients with AD from a new perspective of the brain functional network to explore new ideas in early diagnosis to AD.The study collected the resting-state functional magnetic resonance imaging( fMRI) data of 27 cases of AD patients, 20 cases of mild cognitive impair-ment ( MCI) patients and 36 cases of normal control ( NC) , constituting 4 large-scale brain networks of the central executive network ( CEN) , salience network ( SN) , default mode network ( DMN) and dorsal attention network ( DAN) respectively, and calculated the network FC parameters based on seed point analysis method.In each brain network, pairwise comparisons of two-sample t-test statistical analysis was made to the data of AD, MCI patients and NC, finding significant differences of changing characteristics between groups within the network FC.The results show that the FC of AD patients reduced:in right dor-sal prefrontal cortex and the right precuneus of CEN;in bilateral medial frontal, bilateral angular gyrus, and bilateral posterior cingulate cortex of the DMN; and in bilateral parietal lobule of DAN.The FC of MCI patients reduced in right side of the parietal cortex in CEN.There were no significant changes of FC in SN.The results not only confirm that the brain network FC variability of changes is consistent with cog-nitive impairment in AD patients, but also are expected to explore the pathogenesis of MCI to AD conver-sion and provide reference to AD early diagnosis based on brain FC analysis.
