Optimization of Spore Production of Aspergillus niger by Response Surface Methodology (RSM) in Solid-State Fermentation

Abstract:In this study, effects of medium components and culture conditions on the spore production of Aspergillus niger were investigated by response surface methodology (RSM). Carbon, nitrogen sources and culture conditions were evaluated by a one-variable-at-a-timemethod; through Plackett-Burman design, stirring, illumination, and sucrose were found to be the important factors affecting A. niger spore production significantly; finally, a 2(3) full factorial central composite design and RSM were applied to determine the optimal concentration of each significant variable. A second-order polynomial was evaluated by the multiple regression analysis of the experimental data. The final optimal values for culture conditions and medium components are initial pH 7.2, inoculation volume 2 mL (1x10(5) spores mL(-1)), initial moisture level 8: 8 (m/v), NaNO3 1 g/L, sucrose 0.119 (131.2 g/L), illumination -0.225 (22.24 h), and stirring 0.017 (44.03 h), with a predicted value of maximum spore production of 7.139 x 10(9) +/- 2.17 x 10(7) spores.g(-1) IDM. Under the optimal conditions, the practical spore production was 7.16 x 10(9) spore.g(-1) IDM.
