Abstract:In the present study, a rapid and sensitive GC-MS method was developed for determination of essential oil from Aquilaria sinensis fossil. Forty-seven compounds were identified. The approach was effective in controlling the quality of the essential oil in the production process.

Abstract:Objective: To analyze the constituents of the essential oil extracted from the leaves of Aquilaria sinensis (Lour.) Gilg. and provide scientific basis for research & development and qualities control of the leaves of A. sinensis.Methods:The essential oil was extracted by water-steam distillation and separated by GC capillary column chromatography. The chemical components were quantitatively determined with normalization and were identified by GC-MS method. Results: GC-MS gave 154 peaks and 41 compounds have been identified, which were 89.92% in the total essential oil. Conclusion: In the total essential oil of leaves of A. sinensis, nonanoic acid (1.73%), tetradecanoic acid (7.22%), pentadecanoic acid (2.58%), hexadecanoic acid (48.86%), octadecanoic acid (1.31%), (E)-9-octadecenoic acid (6.04%), 6,10,14-trimethyl-2- pentadecanone (8.22%), 4,8,12,16-tetramethylheptadecan-4-olide (2.31%), phytol (1.91%) and isophytol (1.38%) were main components, which account for 81.56% of the total essential oil.

Abstract:We performed intraoperative arthrography of the knee in 12 children with congenital short femur, Blount's disease or Ollier's disease in whom the Ilizarov technique was used for correction of deformity, leg lengthening or both. In each case, arthrography revealed a joint surface considerably different from that assumed from plain radiographs, and resulted in a change in the placement of our reference wires before application of the frame. This gave significant improvement in the mechanical axis obtained at the time of removal of the frame. The technique is safe, cheap and easy to perform. It is a useful adjunct to the application of the Ilizarov frame when used for complex lengthening and correction of deformity in the leg.

Abstract:As a traditional medicinal herb and valuable natural spice in China, Aquilaria sinensis (Lour.) Gilg has many significant pharmacological effects. Agarwood is the resinous heartwood acquired from wounded A. sinensis trees, and is widely used in pharmaceuticals owing to its excellent medicinal value. In this study, the chemical composition of volatile components and alcohol extracts from different organs of A. sinensis and agarwoods grown in different regions were investigated using GC-MS. The results showed that Vietnam agarwood had the highest moisture content, which was attributed to the local climate, while the fruit and bark of A. sinensis had higher moisture contents than the other organs. The volatile components of A. sinensis organs included 3-ethyl-5-(2-ethylbutyl)-octadecane, oleic acid 3-(octadecyloxy) propyl ester, and docosanoic acid 1,2,3-propanetriyl ester, while the alcohol extracts of A. sinensis organs contained benzoic acid ethyl ester, hexadecanoic acid ethyl ester, oleic acid, and n-hexadecanoic acid. Furthermore, the main active ingredients in agarwood from different habitats were sesquiterpenoids, aromatic species, and chromone compounds. The role of chromone compound 2-phenylethyl-benzopyran as an elicitor and the mechanism of agarwood formation were also investigated. Antioxidant tests showed that essential oils from agarwood and A. sinensis had antioxidant capacities by comparison with butylated hydroxytoluene and vitamin E. An antibacterial activity test showed that the inhibition effect of the essential oil was better against Gram-positive bacteria than against Gram-negative bacteria.

Abstract:This work is focused on the characterization of the composition of a CO2 supercritical fluid extract of Aquilaria sinensis (Chinese agarwood) collected in the Dongguan area (China) and infected by mechanical methods. The constituents of this extract were analyzed by gas chromatography-mass spectrometry (GC-MS) and quantified accurately by gas chromatography with a flame ionization detector (GC-FID), using an internal reference and predicted response factors. Since a significant number of components of this extract remained non-identified after the initial GC-MS analysis of the whole extract, its fractionation by chromatography on silica gel helped to characterize several additional constituents by isolation and structural analysis by NMR spectroscopy. The main components are the classical agarwood chromones (Flindersia chromone and its mono-, di-, and trimethoxylated analogues (respectively, 11.01% and 0.11-4.02%) along with sesquiterpenic constituents typically found in agarwood essential oils, like baimuxinal (1.90%) and kusunol (1.24%), as well as less common selinane dialdehydes (1.58-2.27%) recently described in the literature. Moreover, the structure and stereochemistry of a new sesquiterpenic alcohol, 14β,15β-dimethyl-7αH-eremophila-9,11-dien-8β-ol (0.67%), was determined unambiguously by the combination of structural analysis (NMR, MS), hemisynthesis, and total synthesis, leading to dihydrokaranone and a neopetasane epimer.

Abstract:The resinous stem of Aquilaria sinensis (Lour.) Gilg is the sole legally authorized source of agarwood in China. However, whether other tissue parts can be potential substitutes for agarwood requires further investigation. In this study, we conducted metabolic analysis and transcriptome sequencing of six distinct tissues (root, stem, leaf, seed, husk, and callus) of A. sinensis to investigate the variations in metabolite distribution characteristics and transcriptome data across different tissues. A total of 331 differential metabolites were identified by chromatography–mass spectrometry (GC-MS), of which 22.96% were terpenoids. The differentially expressed genes (DEGs) in RNA sequencing were enriched in sesquiterpene synthesis via the mevalonate pathway. The present study establishes a solid foundation for exploring potential alternatives to agarwood.

Abstract:Angelica pubescens and Angelica sinensis belong to th e Umbelliferae family and both are used as traditional Chinese medicines. In the present study, headspace solid-phase microextraction (HS-SPME) with gas chromatography-mass spectrometry (GC-MS) was used for the analysis of the volatile constituents present in their roots. Eighty-seven compounds in Angelica pubescens and thirty-six compounds in Angelica sinensis were identified by GC-MS. Their relative contents were calculated by the peak area ratio. HS-SPME was compared to steam distillation (SD) by analyzing the volatile constituents of Angelica sinensis root. A good agreement between results obtained with both techniques was found. As a conclusion, HS-SPME is a powerful tool for determining the volatile constituents present in the TCMs.

Abstract:A comprehensive undergraduate experiment about extraction, separation, and identification of essential oils (EOs) from a natural plant is described. Acori tatarinowii Rhizoma (ATR) was used as the raw plant material. Two devices (Soxhlet and Clevenger) with different extraction principles were employed for EO extraction from ATR, separately. The chemical compounds of the extracted EOs were separated and identified by gas chromatography–mass spectrometry (GC–MS). The differences in the chemical components obtained by the two extraction devices were compared. The experiment can give students a deep insight into the difference between the extraction methods. Additionally, the experiment can enable students to learn how to use GC–MS to identify the chemical structures of the extracted complex samples. The grouped students who performed this experiment gained a deep understanding of extraction and detection techniques, as well as the benefits of teamwork.This article has not yet been cited by other publications.

Abstract:A simple, rapid, efficient and inexpensive method for the determination of essential oil in a traditional Chinese medicine (TCM), Acorus Tatarinowii Schott. was developed by using pressurized hot water extraction (PHWE) combined with solid-phase microextraction (SPME) and gas chromatography-mass spectrometry (GC-MS). Acorus Tatarinowii Schott. (0.050 g) was subjected to dynamic extraction with water at 50 bar, 150 degrees C and 1.0 ml/min for 5.0 min, and then the essential oil in the aqueous extract were extracted by SPME fibers at 60 degrees C for 10 min, finally the compounds on the fiber were desorbed and analyzed by GC-MS. The PHWE and SPME parameters were studied. The key active compound of alpha-asarone in the TCM samples from three different growing areas was quantitatively analyzed by external standard method. Compared to steam distillation (SD), the proposed method required little time (only 15 min) to prepare sample. Moreover, little sample mass and no organic solvent was needed. The present method provided good repeatability (R.S.D. less than 13.0%) and recovery (92% for alpha-asarone). It has been shown that PHWE-SPME-GC-MS is an alternative method for determination of essential oils in TCMs and a potential tool for TCM quality assessment.
Species Differentiation and Quality Evaluation for Atractylodes Medicinal Plants by GC/MS Coupled with Chemometric Analysis

Abstract:The utilization of rhizomes from the genus Atractylodes has been challenging due to their closely related origins. In this study, we developed an analytical strategy to differentiate Atractylodes lancea (A. lancea), Atractylodes chinensis (A. chinensis), Atractylodes japonica (A. japonica), and Atractylodes macrocephala (A. macrocephala), and compared their volatile compositions. Gas chromatography-mass spectrometry (GC/MS) was used to analyze the volatile profiles of essential oils extracted from 59 batches of samples. Chemometric methods enabled a better understanding of the differences in volatile oils between the four species and identified significant components affecting their classification and quality. A total of 50 volatile components were identified from the essential oils by GC/MS. Unsupervised and supervised chemometric analyses accurately distinguished A. lancea, A. chinensis, A. japonica, and A. macrocephala. Furthermore, five characteristic chemical markers, namely hinesol, β-eudesmol, atractylon, atractylodin and atractylenolide I, were obtained, and their respective percentage contents in individual species and samples were determined. This study provides a valuable reference for the quality evaluation of medicinal plants with essential oils and holds significance for species differentiation and the rational clinical application of Atractylodes herbs.

The structures were identified by spectral analyses of NMR, MS, et al.RESULT:Thirteen compounds were isolated and identified as 7-hyroxy-5, 4'-dimethoxy flovone (1), 5-hydroxy-7, 4'-dimethoxy flavone (2), luteolin-7-3',4'-trimethyl (3), isocorydine (4), 4-hydroxybenzoic acid (5), triacontenoic (6), hentriacontane (7), alpha-stigmasterol (8), epifriedelanol (9), friedelan (10), friedelin (11), genkwanin (12), 5, 4'-dihyroxy-7, 3'-dimethoxy flovone (13).CONCLUSION:Compound 4 was obtained from this genus for the first time, compounds 1, 6-11, 13 were obtained from this species for the first time.
Chemical Composition Analysis and Discrimination of Essential Oils of Artemisia Argyi Folium from Different Germplasm Resources Based on Electronic Nose and GC/MS Combined with Chemometrics

Abstract:In this study, the electronic nose and GC/MS were used to analyze the chemical components of essential oils from different germplasm resources of Artemisia argyi Folium (A. argyi), in order to quickly identify essential oils of A. argyi from different germplasm resources and clarify the differences among different A. argyi samples. The essential oils of A. argyi were extracted by steam distillation. This article describes for the first time that electronic nose combined with chemometrics can distinguish the essential oils of A. argyi from different germplasm, which proves the reliability and potential of this technology. GC/MS was used to identify 134 volatile components from the essential oil of A. argyi. The main bioactive components were cineole, thujarone, artemisia ketone, β-caryophyllene, (-)-4-terpinol, 3,3,6-trimethyl-1,5-heptadien-4-ol, (-)-α-thujone, camphor, borneol. In addition, the results of principal component analysis (PCA) and hierarchical cluster analysis (HCA) showed that there were significant differences in the essential oils of A. argyi from different germplasm resources, terpenes, alcohols and ketones played an important role in identifying the essential oils of A. argyi from different germplasm resources. This indicates that electronic nose and GC/MS combined with chemometrics can be used as reliable techniques to identify different germplasm resources of A. argyi, and provide certain reference value for quality evaluation, selection of high-quality varieties and rational development of resources of A. argyi.

Simultaneous determination of pesticides, mycotoxins and ferulic acid in Angelica sinensis by GC/LC-Q-TOF/MS

Abstract:In this study, a strong applicable method that could determine a total of 33 pesticides (54 compounds), 11 mycotoxins and functional components (ferulic acid) simultaneously in Angelica sinensis was developed. The compatibility of the sample pretreatment method for pesticides, mycotoxins, and functional components was improved by optimizing the acidity of extraction solvents, the sequence of ice bath and oscillation, the volumetric solution, and so on. The PRiME HLB SPE pretreatment method was chosen as the optimal one when comparing four pretreatment methods. Among the 65 contaminants, 38 of those determined by liquid chromatography and 41 of those by gas chromatography, which showed good linearity (R 2 > 0.9801), 97 % of them had a limit of quantitation (LOQ) of lower than 0.02 mg kg -1 . The recovery of all compounds were suited between 70 % to 120 % and the RSD were all lower than 20 % at the spiked levels of LOQ, 2 × LOQ, and 10 × LOQ. For ferulic acid, the LOQ was 50 ng/mL, and it showed good linearity (R 2 =0.9988) within the range of 0.5 to 10 μg/mL. The recovery and RSD were 98.1 %, and 3.2 % ( n = 6), respectively. The simultaneous determination of cross-category compounds in a single sample preparation was obtained by the combination of SPE and GC/LC-Q-TOF/MS. Therefore, this study could not only shorten the time for data acquisition and data analysis, but also improve the experimental efficiency.

Abstract:To comprehensively utilize the rich resource of Aquilaria sinensis,the ethanol extract of the leaves of A. sinensis was further chemically investigated, which led to the isolation of 12 compounds. By means of 'H- , "C-NMR, and ESI-MS data, and through comparison with those reported in literatures,their structures were identified as iriflophenone 2-(O-a-L)-rhamnoside(1) ,4'-hydroxy-5 methoxyflavone-7-O-glucoxyloside (2) ,7,3',5'-tri-O-methyltricetin(3) ,7-O-beta-D-glucopyranoside of 5-O-methylapigenin(4) ,2-phenyl-ethyl-D-glucopyranoside( 5), salidroside, (6) , benzyl alcohol O-beta-D-glucopyranoside (7) , 2, 6-dimethoxy-4-hydroxyphenol-1-O-beta-D-glucopyranoside(8) ,vanilloloside(9) , ( + ) -syringaresinol( 10) ,beta-tocopherol( 11) and stigmast-5-ene-3beta,7alpha-diol( 12). Among them, compounds 2,3,5-9,11 and 12 were isolated from this genus for the first time. This research hopefully provides valuable data for the further utilization and development of the leaves of A. sinensis.
[Simultaneous Determination of Benzophenones and Xanthone in Leaves of Aquilaria Sinensis by RP-HPLC-UV].

Abstract:This study is to develop a sensitive method by using reversed-phase high performance liquid chromatography coupled with UV detector (HPLC-UV) to simultaneously determine four bioactive compounds, iriflophenone 3-C-beta-D-glucoside, iriflophenone 3,5-C-beta-D-diglucoside, mangiferin, and iriflophenone 2-O-alpha-L-rhamnoside in the leaves of Aquilaria sinensis. An Agilent Zorbax SB-C, column (4, 6 mm x 250 mm, 5 microm) was used, and the gradient elution was performed with mobile phase of 0.1% aqueous phosphoric acid and acetonitrile at a flow rate of 1 mL x min(-1). The detection wavelength was 280 nm, and the column temperature was 25 degrees C. The four marker compounds were well separated with good linearity (R2 > 0.9990), precision, stability and repeatabili y. The-recovery rates were in the range of 98.80%-101.39%. For 15 branch of the leaves, the contents of iriflophenone 3-C-beta-D-gluoside, iriflophenone 3,5-C-beta-D-diglucoside, mangiferin, and iriflophenone 2-O-alpha-L-rhamnoside were between 0.41-14.48, 0.72-3.85, 4.30-29.07, 0.24-5.06 mg, respectivley. This method is precise, accurate and reliable, which provides an efficient way for the quality control of the leaves of A. sinensis. This will promote the comprehensive usage of this plant.

Molecular identification of Aquilaria species with distribution records in China using DNA barcode technology

Abstract:SAquilaria species is one of the main plant resources that produce agarwood, which containing black resin with important economic and medicinal values. There are about 15 species known to the genus around the world, but only two can be found in China, i.e. A. sinensis and A. yunnanensis. In this study, A. sinensis and A. yunnanensis that endemic respectively to Hainan and Yunnan were sampled, on the basis of the investigation and observation of their main morphological features in plantation. Five primers, i.e. ITS2, matK, trnL-trnF1, trnL-trnF2, and trnH-psbA, were eventually selected for DNA barcoding. The results showed that the seed surface of A. sinensis is smooth or sparsely pubescent, and the seed appendages were long. While the seed surface of A. yunnanensis is densely covered with yellow hairs and the seed appendages are short. The trnL-trnF1 sequence fragment has significant intraspecific and interspecific genetic distances. However, the species identification success rate of ITS2+matK combination was finally screened to be the highest, which was verified by the BBA method of TaxonDNA. The phylogenetic trees cluster analysis revealed that the classification of A. sinensis and A. yunnanensis is significant, and there is geographic isolation between the two species. Therefore, on the premise of accurate identification of plant morphological characters, ITS2+matK combination can be used to accurately identify the Aquilaria species in China.

Abstract:Introduction Atractylodes Macrocephala Rhizoma (AMR) is a traditional Chinese medicine containing several sesquiterpenoids with a series of effects. These bioactive compounds may be used as chemical markers for the quality control of AMR. It is necessary to optimise the extraction method and conditions in order to improve extraction productivity. Objective To develop a simple and effective method for the extraction of sesquiterpenoids from AMR and then to simultaneously determine four sesquiterpenoids, selina-4 (14), 7(11)-dien-8-one (SA), atractylenolide II (AII), atractylenolide III (AIII) and atractylenolide VII (AVII), in AMR. Methodology Ultrasound-assisted extraction (UAE) was optimised by central composite design (CCD) to obtain the maximum efficiency. The gas chromatography method was validated and applied for the quantification of four sesquiterpenoids. Results The optimum values of factors were: particle size (120?mesh), extraction time (26?min), extraction temperature (39 degrees C) and 31?mL of chloroform. The selectivity, linear range, limits of detection (LOD) and quantification (LOQ), accuracy, precision and repeatability of the method developed indicated its validity. The application of the method showed that the contents of four sesquiterpenoids in AMR were rather variable. Conclusion The results indicated that the described GC method could be used for the quality control of AMR and its related preparations. Meanwhile, this research revealed that UAE under optimum conditions could be considered as a powerful tool for the extraction of phytochemicals from plants. Copyright (C) 2011 John Wiley & Sons, Ltd.

Abstract:OBJECTIVE Essential oil from root of Angelica dahurica cv. Qibaizhi were studied. METHOD Essential oil was extracted by water-steam distillation and analyzed by GC-MS. RESULT Two hundreds and ninety chromatographic peaks were detected, among which 111 compounds have been identified. The contents of them made up 90.61% of the total essential oil. CONCLUSION The main compounds in the essential oil were 3-carene (12.70%), beta-elemene (6.20%), beta-terpinene (3.53%), beta-myrcene (1.97%), gamma-elemene (1.82%), beta-phellandrene (1.65%), and beta-maaliene (1.61%), et al. In addition, suberosin (0.16%), a coumarins compound, was also determined.
