Abstract:To investigate the damage of nano-titanium dioxide (Nano-TiO2) on brain cells of mice, so as to provide more research basis for further evaluation of the biological safety of Nano-TiO2 and its molecular mechanism of action. In this study, that observed the pathological changes of mice brain treated with nano-titanium dioxide at concentrations of 5,10,50,100,150,200 mg/kg, the content changes of titanium in the brain, the activities of antioxidant enzymes and the contents of important non-enzymatic substances, as well as the important neurochemistry of glutamate (Glu), acetylcholinesterase (AchE) and nitric oxide (NO). The changes of molecular content index showed that with the increase of nano-titanium dioxide dosage, the brain-body ratio showed a gradual downward trend, while the content of titanium in brain tissue continued to increase and had a significant dose-dependent effect. The content of non-enzymatic antioxidants in high-dose nano-titanium dioxide treatment group decreased significantly, the levels of NO increased significantly, Glu and AchE gradually decreased and the above data were significantly different from the control group . It was proved that nano-titanium dioxide administered intraperitoneally was absorbed after digestive tract and eventually transported to the brain, resulted in oxidative stress in brain tissue and unbalanced metabolism of neurochemical substances, even abnormal. It caused various brain injuries.
