Stability of the Probiotic Lactobacillus Plantarum P-8 after Long-Term Continuous Subculturing for 1000 Generations

Abstract:Long-term continuous cultivation is one of the classical methods to measure microbial stability.In this study,the probiotic Lactobacillus plantarum P8 was propagated separately in either normal or carbon source-limited MRS broth for a thousand generations,phenotypic characteristics,including the bacterial colony morphology,viable counts,growth(OD600) and activity,were monitored during the long-term continuous propagation.The results showed that after subculturing for a thousand generations,no change was observed in the bacterial colony morphology,growth and viable counts of L.plantarum P-8 grown in either culture medium.Furthermore,the bacterial activity of L.plantarun P-8 was stable after subculturing for the thousand generations.To sum up,our results suggest that L.plantarun P-8 is a probiotic with desirable stability in its phenotypic characteristics.
