A behavioural study on neuroprotective effect of noble gases

Abstract:Objective To evaluate the development,coordination exercise and memory function in a neonatal asphyxia model in rats,and to investigate the long-term neuroprotective effects of noble gases given in childhood.Methods Seven-day old postnatal Sprague-Dawley rats underwent right common carotid artery ligation.One hour later they were exposed to 0.08 volume fraction oxygen balanced with nitrogen for 90 minutes.Two hours later,they received 0.7 volume fraction argon,helium and xenon or 0.7 volume fraction nitrogen (control group) balanced with oxygen for 90 minutes,respectively.The body temperature was maintained by submerging the exposure chambers into a water bath of 37 ℃.The cohort pups without any intervention served as negative controls (negative group).There were 20 rats in each group.The rats were allowed to live up to 38 postnatal days for rotarod test,trace fear conditioning test and Morris water maze test.Results The body weight in the helium treated group was significantly lower than that in negative group and control group on the 38th postnatal day (both P<0.01).There was no significant difference in the body weight among argon,xenon,negative and control groups (all P>0.05).There was no statistical difference in the latency at the rotate speed of 30 r/min in the rotarod test among the five groups (all P>0.05).The latency of the control group at the rotate speed of 50 r/min was significantly shorter than that at the rotate speed of 30 r/min and that in the other four groups at 50 r/min (all P<0.01).There was no significant difference in the latency among argon,helium,xenon and negative groups (all P>0.05).In the trace fear conditioning test,the percent of freezing time of the control group was significantly less than that in the other four groups on day 2 context experiment (P<0.01,0.05).There was no statistical difference in the percent of freezing time among the five groups on day 3 tone experiment no matter during adaption period or after tone stimulation (all P>0.05).The latencies to find the platform of the five groups were similar on the first day of Morris water maze (all P>0.05).Except for the control group,the latency significantly reduced in the others in the next 4 days.The latency of the control group was significantly longer than that of the other four groups from day 3 to 5 (all P<0.05),but no significant difference was found in the latency among the other four groups (all P>0.05).The area under curve (AUC) showed that the cumulative time of the control group was significantly longer than that of the other four groups (P<0.01,0.05),and there was no statistical difference in the average swimming speed between groups (all P>0.05).Conclusion Argon,helium and xenon can provide certain degree of neuroprotection against hypoxia/ischemia induced brain injury in a rat mode of neonatal asphyxia,but helium has detrimental effects on physiological development.As a kind of widely available and cheap noble gas,argon may have a great potential to be a neuroprotectant for future use.
