Effect of chronic stress on the chronicity of acute postsurgical pain

Abstract:Objective To investigate the influence of chronic stress on the chronicity of acute postsurgical pain in mice.Methods Forty-eight male C57BL/6 mice were randomly divided into 4 groups (n =12):nonstress+ sham operation group,nonstress+ operation group,chronic stress+ sham operation group,and chronic stress+ operation group.Mice of operation group received a left plantar incision.The mice in stress groups were individually restrained for 8 hours per day for 10 consecutive days to establish chronic restraint stress models.Body weight of each mouse was measured before and after chronic restraint stress.Elevated plus maze test and forced swimming test were used to assess the anxiety and depression in mice with chronic restraint stress.The threshold of mechanical pain was tested before operation and 1,3,5,7,10,14,18,21,25 and 28 days after operation.Results Compared with nonstress groups,the body weight was significantly decreased in stress groups after chronic restraint stress (P<0.01),total immobility time in the forced swimming test was significantly elevated (P<0.05) and the open arm duration was declined significantly in the elevated plus maze test (P<0.05).There were no significant differences in baseline mechanical pain threshold between groups (all P>0.05).Compared to the baseline,the pain threshold of both chronic stress groups and nonstress groups declined significantly after left plantar incision (all P<0.01),and the pain threshold in the nonstress + operation group and chronic stress + operation group went back to the baseline on the day 7 and 21 after operation,respectively.Moreover,on the day 3,5,7,10,14,and 18 after operation,the pain threshold in the chronic stress group was significantly lower than that in the nonstress + operation group (P<0.05,P<0.01).Conclusion The chronic restraint stress can induce both anxiety and depressive behaviors,which makes acute postoperative pain change into chronic pain.
