Abstract:The monodisperse double emulsions obtained by microfluidic method can serve as ideal templates for preparing core-shell alginate microcapsules, which have attracted much attention in biological applications, such as drug delivery systems and cell encapsulation, tissue engineering. However, the formation behavior and dynamic analysis of double emulsion with an alginate shell is still unclear due to the complex rheological behavior of alginate solutions. Herein, we employ a dual-coaxial microfluidic device to generate the high-quality double emulsion droplets with alginate shell, focusing on the effects of the fluid properties of alginate solution in the middle phase (viscosity, μm) and the fluid flow rate on the droplet formation mechanism. As the viscosity of the middle fluid (μm) increased, the size of compound droplets (D2) increased and the size of inner droplets (D1) decreased, and the break-up regimes occurred a dripping-to-jetting transition when μm = 160 mPa s. The number of encapsulated inner droplets can be predicted and precisely controlled by regulating the generation frequency of inner (f1) and outer droplets (f2). The breakup dynamics of the alginate thread are also analyzed by using the volume-of-fluid/continuum-surface-force (VOF/CSF) method. The results show that the pressure and velocity in the neck of pinch-off is lower in the jetting than that in the dripping regime. This study will provide useful guidance for the rational design and controllable preparation of core-shell alginate microcapsules.

Abstract:Microfluidic devices can form monodisperse double emulsions, but the fabrication steps are complicated and require specialized equipment. Recently, a method to convert single emulsions into double emulsions using vortex mixers has been proposed. Using this method, we demonstrate the production of double emulsions using commercially available single-emulsion microfluidic chips. We characterize the effect of vortex speed, vortex duration and the number of vortex/flick cycles on the average diameter and coefficient of variation of the double emulsions. Using fluorescent nanoparticles as tracers, we show that droplet breakup occurs during the second emulsification (using the vortex), but did not observe any fusion between the cores of double emulsion droplets. We also found that some inverted double emulsion droplets containing the outer water phase in their core were produced during vortex emulsification. Finally, while commercial chips only exist with a finite range of channel size that sets the monodispersed emulsion droplet radius, we show that the double-emulsion droplet radius can be adjusted using osmotic pumping. Our method is simple, available and user-friendly for biomedical researchers.

Abstract:In this work, we developed a simple capillary microfluidic device to prepare double emulsions with two-phase cores. We can control the structure and size of double emulsions by changing four-phase flow rates. Based on the formation of these four-phase double emulsions, we investigated the structure evolution of the double emulsions for different systems, and successfully obtained different multiple emulsions with four typical inner structures, as a separately engulfing structure, triple emulsion structure, Janus-engulfing structure, and inner-Janus structure. The equilibrium morphologies of the double emulsions could be estimated by using the relationship of interfacial tension between different phase fluids. Remarkably, the physics of evolution of double emulsions with two-phase cores in a microfluidic device allows the synthesis of microparticles with new morphologies and more applications in encapsulation of multiple active substances.

Abstract:Microemulsions have found a wide range of applications exploiting their chemical and physical properties. Development of microfluidic-based approaches has allowed for the controlled production of highly monodispersed emulsions, including the formation of multiple and hierarchical emulsions. Conventional poly(dimethylsiloxane)-based microfluidic systems require tight spatial control over the surface chemistry when used for double emulsion generation, which can be challenging to achieve on the micrometer scale. Here, we present a two-dimensional device design, which can selectively be surface-treated in a straightforward manner and allows for the formation of uniform water/oil/water double emulsions by combining two distinct hydrophilic and hydrophobic surface properties. These surfaces are sufficiently separated in space to allow for imparting their functionalization without the requirement for lithographic approaches or complex flow control. We demonstrate that a mismatch between the wettability requirements of the continuous phase and the channel wall inherent in this approach can be tolerated over several hundreds of micrometers, opening up the possibility to use simple pressure-driven flows to achieve surface functionalization. The design architecture exhibits robust efficiency in emulsion generation while retaining simple device fabrication. We finally demonstrate the potential of this approach by generating water in oil in water emulsions with lipid molecules acting as surfactants.

Abstract:Double emulsions have great potential application in many fields. Although many companies and research institutions have developed various strategies for forming double emulsions, production and stability are not straightforward. Meanwhile, none of the well-organized simple device and scalable technique was available to form highly monodispersed double emulsions. Herein, we present a one-step strategy to produce highly monodisperse double emulsion via assembled micro-cross device. This device has a lot of advantages, such as easy assembly/disassembly and high repeatability. High-throughput is also easily achievable for its high-pressure capacity. Moreover, we systematically studied the effects of the three flow rates on the size and structure of the double emulsions. A mathematical model has been developed to predict the inner and outer droplets size of the double emulsion. We prepared the flame-retardant microcapsules with a dimethyl methylphosphonate (DMMP) inner core and an ultraviolet-curable polysiloxane shell. We hope that this assembled micro-cross device can be employed extensively in the area of microfluidics.

Abstract:This article presents a simple and novel approach to prepare monodispersed gas-in-oil-in-water (G/O/W) and gas-in-water-in-oil (G/W/O) double-emulsions in the same dual-coaxial microfluidic device. The effects of three phase flow rates on the sizes of microbubbles and droplets and the number of the encapsulated microbubbles were systematically studied. We successfully synthesized two different types of gas/liquid/liquid (G/L/L) double emulsions with different inner structures in the same geometry by adjusting the flow rates sequentially. Mathematical models were developed to predict the size and structures of the double emulsions. This simple approach gives a new idea for preparing hollow and porous microspheres with microbubbles as the direct core/pores templates.

Abstract:Double emulsions are useful geometries as templates for core-shell particles, hollow sphere capsules, and for the production of biomedical delivery vehicles. In microfluidics, two approaches are currently being pursued for the preparation of microfluidic double emulsion devices. The first approach utilizes soft lithography, where many identical double-flow-focusing channel geometries are produced in a hydrophobic silicone matrix. This technique requires selective surface modification of the respective channel sections to facilitate alternating wetting conditions of the channel walls to obtain monodisperse double emulsion droplets. The second technique relies on tapered glass capillaries, which are coaxially aligned, so that double emulsions are produced after flow focusing of two co-flowing streams. This technique does not require surface modification of the capillaries, as only the continuous phase is in contact with the emulsifying orifice; however, these devices cannot be fabricated in a reproducible manner, which results in polydisperse double emulsion droplets, if these capillary devices were to be parallelized. Here, we present 3D printing as a means to generate four identical and parallelized capillary device architectures, which produce monodisperse double emulsions with droplet diameters in the range of 500 μm. We demonstrate high throughput synthesis of W/O/W and O/W/O double emulsions, without the need for time-consuming surface treatment of the 3D printed microfluidic device architecture. Finally, we show that we can apply this device platform to generate hollow sphere microgels.

Abstract:Capillaries-based co-flowing microfluidic devices were designed and then fabricated for the controlled production of water-in-oil-in-water (W/O/W) double-emulsion droplets, where polystyrene dissolved in fluobenzene was employed as the oil phase. Two configurations of the co-flowing devices were employed in the experiments. In the so-called one-step device, the inner capillary tip locates at the same cross-section of the middle capillary tip. Thus, the core drop and the shell drop depart from their capillaries ends simultaneously, forming a double-emulsion drop in one step. While in the so-called two step device, the inner capillary tip locates upstream to the middle capillary tip. The core drop and the shell drop break off from their respective capillaries ends successively, forming a double-emulsion drop in two steps. The effect of the fluids flow rates on the double-emulsion droplets size as well as their formation mechanism was studied comprehensively in the proposed devices. Experimental results implied that slight difference in device configurations would lead to significant difference in droplet formation mechanisms and thus size-control laws. In the two-step device, the inner stream of fluid had little influence on the breakage and the outer diameter of the double-emulsion drops, while in the one-step device the effect was significant. With the produced double-emulsion droplets, millimeter-sized polymer capsules with single hollow cavity and ultra-thin shells were obtained by solidifying the oil layers of the compound drops using the solvent evaporation method.

Abstract:In this paper we present a comprehensive study of the step-emulsification process for high-throughput production of (sub-)$\mu$m-size monodisperse droplets. The microfluidic device combines a Hele-Shaw nanofluidic cell with a step-like outlet to a deep and wide reservoir. The proposed theory based on Hele-Shaw hydrodynamics provides the quasi-static shape of the free boundary between the disperse liquid phase engulfed by the co-flowing continuous phase prior to transition to oscillatory step-emulsification at low enough capillary number. At the transition the proposed theory anticipates a simple condition for critical capillary number as a function of the Hele-Shaw cell geometry. The transition threshold is in excellent agreement with experimental data. A simple closed-form expression for the size of the droplets generated in step-emulsification regime derived using simple geometric arguments also shows a very good agreement with the experimental results.

Abstract:In this work, we generated gas-in-water-in-oil (G/W/O) multi-cores double emulsions with a one-step microfluidic device and found a new stable flow pattern, where multi-cores double emulsions were easily fabricated. The mechanism of the particular flow pattern was discussed. Moreover, we systematically studied the effects of the three flow rates on the size and structure of the double emulsions. We then developed mathematical models to predict the size and structure of the double emulsions. On this basis, porous polyacrylamide hydrogel microspheres were successfully fabricated by in situ UV polymerization method. The structure of the double emulsions can be tuned easily by the three phase flow rates and the dimensional range of the encapsulated microbubbles can be expanded by integrating CO2 adsorption. Abstract:Double emulsions are of great importance for both science and engineering. However, the production of multicore double-emulsion droplets is challenging and normally requires sophisticated microfluidic devices, which limits their availability to broader communities. Here, we propose a simple, precise, and scalable batch method for producing double emulsions with monodispersed multicores at milliliter per minute rates, using the most common means in laboratory, temperature. By rapidly cooling liquid crystal emulsions, the introduced temperature gradient around the emulsion droplets leads to the injection of monodispersed guest droplets to form double-emulsion droplets. The number of injected water droplets can be precisely controlled by adjusting the thermally induced mechanical force through the temperature difference and the cooling rate. In contrast to conventional microfluidic fabrication, this method processes all emulsion droplets simultaneously in a noncontact and in situ manner. Therefore, it has great flexibility, allows multiple processing of double emulsions of arbitrary shape, has good capacity for mass production, and offers excellent compatibility with technologies such as microfluidics. Finally, we demonstrate that temperature changes can also be used to release the inner droplets from the double emulsion. The proposed method offers a reversible tool for processing double emulsions with minimal cost and expertise and is applicable to droplet-based microsystems in materials science, photonics, sensors, pharmaceuticals, and biotechnology.

Novel glass capillary microfluidic devices for the flexible and simple production of multi-cored double emulsions

Abstract:Hypothesis: Double emulsions with many monodispersed internal droplets are required for the fabrication of multicompartment microcapsules and tissue-like synthetic materials. These double emulsions can also help to optically resolve different coalescence mechanisms contributing to double emulsion destabilization. Up to date microfluidic double emulsions are limited to either core-shell droplets or droplets with eight or less inner droplets. By applying a two-step jet break-up within one setup, double emulsion droplets filled with up to several hundred monodispersed inner droplets can be achieved. Experiments: Modular interconnected CNC-milled Lego®-inspired blocks were used to create two separated droplet break-up points within coaxial glass capillaries. Inner droplets were formed by countercurrent flow focusing within a small inner capillary, while outer droplets were formed by co-flow in an outer capillary. The size of inner and outer droplets was independently controlled since the two droplet break-up processes were decoupled. Findings: With the developed setup W/O/W and O/W/O double emulsions were produced with different surfactants, oils, and viscosity modifiers to encapsulate 25-400 inner droplets in each outer drop with a volume percentage of inner phase between 7% and 50%. From these emulsions monodispersed multicompartment microcapsules were obtained. The report offers insights on the relationship between the coalescence of internal droplets and their release.

Abstract:The droplet-based microfluidic techniques have been applied widely in functional material synthesis and biomedical information measurements, wherein step emulsification as an integrated system combines the advantages of homogeneity and throughput in monodisperse droplet formation. This paper reviews the mechanisms and classical structures of step emulsification. In terms of droplet formation mechanisms, we describe the droplet size and detachment regimes related to the microchannel geometry. Distinguished by droplet formation, microfluidic step emulsification driven by interfacial tension and centrifugal step emulsification related to buoyancy are introduced respectively, including their improved structures for enhancing the droplet homogeneity and throughput. Finally, the perspectives about the developments of step emulsification in mechanisms, fabrications, and applications are discussed.

Abstract:This study includes an examination of the design, fabrication, and experimentation of a rudimentary droplet generator. The device has potential applications in on-demand double and higher-order emulsions as well as tailored emulsions with numerous cores. The phenomenon of a pendant double droplet creation is observed when an inner phase is transported through a capillary, while a middle phase envelops the external surface of the capillary. This leads to the occurrence of a pinching-off process at the tip of the pulled capillary. Following this, the double droplet is introduced into a container that is filled with the outer phase. The present study examines the force equilibrium throughout the droplet break-up process and aims to forecast the final morphology of the droplets within the container by considering the impact of interfacial tension ratios. The shell thickness in a core–shell formation can be calculated based on the inner and middle phase flow rates as well as the middle droplet formation period. The present platform, which enables the simple production of double and higher emulsions, exhibits promising prospects for the controlled manufacturing of complex emulsions. This technology holds potential for various applications, including the experimental exploration of collision behavior or electro-hydrodynamics in emulsions as well as millimeter-size engineered microparticle fabrication.

Abstract:Microfluidic devices are promising tools for the production of monodispersed tuneable complex emulsions. This review highlights the advantages of microfluidics for the fabrication of emulsions and presents an overview of the microfluidic emulsification methods including two-step and single-step methods for the fabrication of high-order multiple emulsions (double, triple, quadruple and quintuple) and emulsions with multiple and/or multi-distinct inner cores. The microfluidic methods for the formation of multiple emulsion drops with ultra-thin middle phase, multi-compartment jets, and Janus and ternary drops composed of two or three distinct surface regions are also presented. Different configurations of microfluidic drop makers are covered, such as co-flow, T-junctions and flow focusing (both planar and three-dimensional (3D)). Furthermore, surface modifications of microfluidic channels and different modes of droplet generation are summarized. Non-confined microfluidic geometries used for buoyancy-driven drop generation and membrane integrated microfluidics are also discussed. The review includes parallelization and drop splitting strategies for scaling up microfluidic emulsification. The productivity of a single drop maker is typically <1 mL/h; thus, more than 1000 drop makers are needed to achieve commercially relevant droplet throughputs of >1 L/h, which requires combining drop makers into twodimensional (2D) and 3D assemblies fed from a single set of inlet ports through a network of distribution and collection channels.

Abstract:To achieve high-throughput generation of monodisperse double emulsions via controllable symmetric splitting, the splitting dynamics of double emulsions in Y-shaped microchannels are systematically investigated by the combination of experimental study and numerical simulation. Four distinct breakup flow patterns of double emulsions splitting are initially identified, namely non-breakup, once uneven breakup, twice uneven breakup and twice even breakup, and the underlying hydrodynamic mechanisms of each flow pattern are further elucidated through the flow field structures and pressure distributions. The results show that the upstream pressure created by the flow obstruction of continuous phase governs the stable and symmetric splitting of double emulsions in Y-shaped microchannels. Effects of the flowrates of continuous phase, the outer and inner diameters of double emulsions, the physical properties of all the inner, middle and outer fluids as well as the angle of Y-shaped microchannel on the flow pattern transition laws are systematically clarified, and the corresponding universal flow pattern diagrams for predicting the critical boundary for the transition of flow patterns are established. Based on controllable symmetric splitting of double emulsions, three-level splitting microfluidic networks are rationally designed to achieve high-throughput generation of monodisperse double emulsions as well as monodisperse microcapsules via template synthesis. The results in this study not only advance the deep understanding of the breakup dynamics of double emulsions in Y-shaped microchannels, but also offer valuable guidance for the rational design of microfluidic devices for mass production of monodisperse double emulsions, thus providing a significant contribution to the fields of micro-chemical engineering and droplet microfluidics.
