Abstract:This paper describes the preparation of semiconducting polymer dots (Pdots) and their application for single-molecule imaging and biosensing. The Pdots possessed high fluorescence brightness, with a 16 nm Pdot being similar to 9 times brighter than 13 nm Quantum Dots (Qdots). The surface of Pdots was successfully conjugated with streptavidin, which made Pdots suitable for specific subcellular labeling and targeting. The interior composition of Pdots was also successfully modified, through which the Pdots obtained additional functionalities. We demonstrated the utility of gold nanoparticle embedded Pdots in dual-modality imaging. We also demonstrated that Rhodamine B embedded Pdots were able to function as ratiometric temperature sensor in live-cell imaging mode.

Abstract:This paper describes a method, based on co-precipitation, for generating small semiconducting polymer dot (Pdot) nanocomposites, which contain either gold or iron oxide nanoparticles within the Pdot matrix. We demonstrate the utility of Pdot-Au nanoparticles (Au-NP-Pdots) in dual-modality imaging in which co-localization of fluorescence from Pdot and scattering from Au was used to identify Au-NP-Pdot probes for downstream single-particle tracking and cellular imaging. We also demonstrate the potential of employing Pdot-FeOx nanoparticles (FeOx-NP-Pdots) for both sample preparation, where cells tagged with FeOx-NP-Pdots were isolated using an external magnet, and cellular imaging and detection, owing to the intense fluorescence from Pdots. The method we present here should be generalizable to the formation of other Pdot nanocomposites for creating the next generation of multifunctional Pdot probes.

Abstract:The search for emerging materials combining magnetic and semiconducting properties has attracted widespread interest in contemporary materials science. Chalcopyrite (CuFeS2), as an earth abundant and nontoxic chalcogenide compound in the I-III-VI2 family, is a promising class of such materials that exhibit unusual electrical, optical, and magnetic properties. However, its successful implementation largely depends on our ability to understand, control and manipulate their structural, transport and spin behavior. Here we show that solution processing monodispersed CuFeS2 quantum dots exhibit a strong coupling among optical, electronic, and magnetic degrees of freedom. The photoresponse and magnetoconductance of CuFeS2 quantum dots are realized under external stimuli. We further exploit a fast and efficient way to achieve an exceptionally large performance in a magneto-optoelectronic hybrid system consisting of magnetic semiconducting CuFeS2 and conducting polymer matrix. The results demonstrate a promising potential of magnetic semiconductor CuFeS2 in the field of spin electronics.

Abstract:Nanomaterials filled polymers system is a simple method to produce organic/inorganic hybrid with synergistic or complementary effects. The properties of nanocomposites strongly depend on the dispersion effects of nanomaterials in the polymer and their interfaces. The optimized interface of nanocomposites would decrease the barrier height between filler and polymer for charge transfer. To avoid aggregation of metal nanoparticles and improve interfacial charge transfer, Pt nanodots filled in the non-conjugated polymer was synthesized with an in situ method. The results exhibited that the absorbance of nanocomposite covered from the visible light region to NIR (near infrared). The photo-current responses to typical visible light and 808 nm NIR were studied based on Au gap electrodes on a flexible substrate. The results showed that the size of Pt nanoparticles was about 1-2 nm and had uniformly dispersed in the polymer matrix. The resulting nanocomposite exhibited photo-current switching behavior to weak visible light and NIR. Simultaneously, the nanocomposite also showed electrical switching responses to strain applied to a certain extent. Well-dispersion of Pt nanodots in the polymer is attributable to the in situ synthesis of metal nanodots, and photo-current switching behavior is due to interface optimization to decrease barrier height between metal filler and polymer. It provided a simple way to obtain organic/inorganic hybrid with external stimuli responses and multi-functionalities.

Abstract:The expansion applications of semiconducting polymer dots (Pdots) among optical nanomaterial field have long posed a challenge for researchers, promoting their intelligent application in multifunctional nano-imaging systems and integrated nanomedicine carriers for diagnosis and treatment. Despite notable progress, several inadequacies still persist in the field of Pdots, including the development of simplified near-infrared (NIR) optical nanoprobes, elucidation of their inherent biological behavior, and integration of information processing and nanotechnology into biomedical applications. This review aims to comprehensively elucidate the current status of Pdots as a classical nanophotonic material by discussing its advantages and limitations in terms of biocompatibility, adaptability to microenvironments in vivo, etc. Multifunctional integration and surface chemistry play crucial roles in realizing the intelligent application of Pdots. Information visualization based on their optical and physicochemical properties is pivotal for achieving detection, sensing, and labeling probes. Therefore, we have refined the underlying mechanisms and constructed multiple comprehensive original mechanism summaries to establish a benchmark. Additionally, we have explored the cross-linking interactions between Pdots and nanomedicine, potential yet complete biological metabolic pathways, future research directions, and innovative solutions for integrating diagnosis and treatment strategies. This review presents the possible expectations and valuable insights for advancing Pdots, specifically from chemical, medical, and photophysical practitioners' standpoints.

Abstract:Stable and highly fluorescent small semiconducting polymer dots (Pdots) are prepared by D. T. Chiu and co-workers using a covalent cross-linking strategy described on page 3498. In this confocal fluorescence microscopy image, microtubules in HeLa cells are specifically labeled by the cross-linked Pdots, and individual tubular structures can be seen under 488 nm laser illumination. Cross-linked Pdots are shown to have desirable characteristics for improved labeling efficiency and sensitivity in cellular assays.

Abstract:A facile cross-linking strategy covalently links functional molecules to semiconducting polymer dots (Pdots) while simultaneously providing functional groups for biomolecular conjugation. Abstract:Two types of small photoblinking semiconductor polymer dots (Pdots) with high brightness, strong photostability, and favorable biocompatibility are designed, by C. Wu, Y. Sun, and co-workers as described in article 1604850. Super-resolution optical fluctuation imaging is achieved using these superior small Pdots. Imaging of subcellular structures demonstrates that these small photoblinking Pdots are outstanding probes for fast, long-term, super-resolution fluorescence imaging.

Abstract:This paper describes a synthetic approach for photocrosslinkable polyfluorene (pc-PFO) semiconducting polymer dots, and demonstrates their superior ability to crosslink and form 3-D intermolecular polymer networks. The crosslinked pc-PFO Pdots are equipped with excellent encapsulating ability of functional small molecules. Optimum conditions of light irradiation on pc-PFO Pdots were investigated and clarified by using polymer thin films as a model. By employing the optimal light irradiation conditions, we successfully crosslinked pc-PFO Pdots and studied their particle sizes, photophysical, and colloidal properties. Single-particle imaging and dynamic-light-scattering measurements were conducted to understand the behaviors of photocrosslinked Pdots. Our results indicate pc-PFO Pdots can be easily photocrosslinked and the crosslinked species have excellent colloidal stability, physical and chemical stability, fluorescence brightness, and specific binding properties for cellular labeling. Considering that optical stimulus can work remotely, cleanly, and non-invasively, this study should pave the way for a promising approach to further develop stimuli-responsive ultrabright and versatile Pdot probes for biomedical imaging.

Abstract:Bright dots: Semiconducting polymer dots (Pdots) doped with europium complexes possess line-like fluorescence emission, high quantum yield, and long fluorescence lifetime. The Pdots successfully labeled receptors on cells. The long fluorescence lifetime of the Pdots was used to distinguish them from other red fluorescence emitting nanoparticles, and improve the signal-to-noise ratio for time-gated cellular imaging. Abstract:Semiconducting polymer dots (Pdots) have shown great promise in biomedical applications, including biosensing, drug delivery, and live imaging of cells and biomolecules. Insight into the mechanism and regulation of cellular uptake and intracellular metabolism of Pdots is important for the development of superior Pdots-based theranostic nanoconjugates. Herein, we performed real-time imaging of endocytosis and intracellular trafficking of a type of fluorescent Pdots that showed excellent biocompatibility in various types of cells. The endocytic routes and kinetics of Pdots were differently regulated in distinct cell types. Following endocytosis, Pdots were transported in vesicles along microtubule and destined for lysosomes. Furthermore, our results revealed exosome-mediated extracellular release of Pdots and have tracked the dynamic process at the single particle level. These results provide new insight into the design of more effective and selective imaging probes as well as drug carriers.

Abstract:This work describes the preparation and validation of single-chain semiconducting polymer dots (sPdots), which were generated using a method based on surface immobilization, washing, and cleavage. The sPdots have an ultrasmall size of ∼3.0 nm as determined by atomic force microscopy, a size that is consistent with the anticipated diameter calculated from the molecular weight of the single-chain semiconducting polymer. sPdots should find use in biology and medicine as a new class of fluorescent probes. The FRET assay this work presents is a simple and rapid test to ensure methods developed for preparing sPdot indeed produced single-chain Pdots as designed.

Abstract:Semiconducting polymer dots (Pdots) recently have emerged as a new class of ultrabright fluorescent probes with promising applications in biological detection and imaging. We developed photoswitchable Pdots by conjugating photochromic spiropyran molecules onto poly[9,9-dioctylfluorenyl-2,7-diyl)-co-1,4-benzo-{2,1'-3}-thiadiazole)] (PFBT). The modulation of fluorescence was achieved by ultraviolet irradiation, which converted spiropyran into its visible-absorbing merocyanine form. The merocyanine efficiently quenched the fluorescence of PFBT via Förster resonance energy transfer (FRET). We then reversed the quenching by subsequent irradiation with visible light to get back the fluorescence of PFBT. This FRET-based photomodulation of Pdot fluorescence could be repeated multiple times. We next conjugated biomolecules onto the surface of these photoswitchable Pdots and demonstrated their specific cellular and subcellular labeling to different types of cells without any noticeable nonspecific binding. We anticipate these photoswitchable and biocompatible Pdots will be useful in developing bioimaging techniques in the future.

Abstract:A simple and effective method was reported for the preparation from MEH-PPV of conjugated polymer nanoparticles (Pdots) that are water-soluble and well dispersed. The as-prepared Pdots show bright orange fluorescence at a quantum yield up to 32.37%. The fluorescence intensity of Pdots can be quenched with good selectively by the successive addition of Fe3+. Abstract:Fluorescent semiconducting polymer dots (Pdots) have attracted great interest because of their superior characteristics as fluorescent probes, such as high fluorescence brightness, fast radiative rates, and excellent photostability. However, currently available Pdots generally exhibit broad emission spectra, which significantly limit their usefulness in many biological applications Involving multiplex detections. Here, we describe the design and development of multicolor narrow emissive Pdots based on different boron dipyrromethene (BODIPY) units. BODIPY-containing semiconducting polymers emitting at multiple wavelengths were synthesized and used as precursors for preparing the Pdots, where intraparticle energy transfer led to highly bright, narrow emissions. The emission full width at half-maximum of the resulting Pdots varies from 40 to 55 nm, which is 15-2 times narrower than those of conventional semiconducting polymer dots. BODIPY 520 Pdots were about an order of magnitude brighter than commercial Qdot 525 under identical laser excitation conditions. Fluorescence imaging and flow cytometry experiments indicate that the narrow emissions from these bright Pdots are promising for multiplexed biological detections.

Abstract:Different from the most extensively used inorganic quantum dots (Qdots) for the current state-of-the-art photoelectrochemical (PEC) bioanalysis, this work reports the first demonstration of polymer dots (Pdots) for novel PEC bioanalysis. The semiconducting Pdots were prepared via the reprecipitation method and then immobilized onto the transparent indium tin oxide glass electrode for PEC biodetection of the model molecule L-cysteine. The experimental results revealed that the as-fabricated Pdots exhibited excellent and interesting PEC activity and good analytical performance of rapid response, high stability, wide linear range, and excellent selectivity. In particular, the PEC sensor could easily discriminate L-cysteine from reduced L-glutathione (L-GSH). This work manifested the great promise of Pdots in the field of PEC bioanalysis, and it is believed that our work could inspire the development of numerous functional Pdots with unique properties for innovative PEC bioanalytical purposes in the future.
