Abstract:BackgroundNear-infrared II (NIR-II, 900-1700 nm) fluorescence bioimaging with advantages of good biosafety, excellent spatial resolution, high sensitivity and contrast, has attracted great attentions in biomedical research fields. However, most nanoprobes used for NIR-II fluorescence imaging have poor tumor-targeting ability and therapeutic efficiency. To overcome these limitations, a novel NIR-II-emissive theranostic nanoplatform for imaging and treatment of cervical cancer was designed and prepared. The NIR-II-emissive dye IR-783 and chemotherapy drug doxorubicin (DOX) were encapsulated into liposomes, and the tumor-targeting peptide TMTP1 was conjugated to the surface of the liposomes to form IR-783-DOX-TMTP1 nanoparticles (NPs) via self-assembly methods.ResultsThe IR-783-DOX-TMTP1 NPs showed strong NIR-II emission, excellent biocompatibility, a long lifetime, and low toxicity. Further, high-definition NIR-II fluorescence microscopy images of ear blood vessels and intratumor blood vessels were obtained from IR-783-DOX-TMTP1 NPs-stained mice with high spatial resolution under 808 nm laser excitation. Moreover, IR-783-DOX-TMTP1 NPs showed strong tumor targeting ability and high efficiently chemotherapeutic character towards cervical tumors. ConclusionsThe novel targeting and NIR-II-emissive IR-783-DOX-TMTP1 NPs have potential in diagnosis and therapy for cervical cancer.

Abstract:The future of personalized cancer treatments relies on the development of functional agents that have tumor-targeted anticancer activities and can be detected in tumors using imaging. However, application of these functional agents in the clinic has been limited due to inefficient drug delivery, low specificity for tumor imaging, development of drug resistance, low signal-to-noise ratio and safety concerns regarding potential toxicity. Currently, the most common strategy to develop these functional agents is to conjugate therapeutic agents with the appropriate fluorescent probe. The present study synthesized a novel mitochondria-targeted heptamethine cyanine (Cy) derivative Cy‑triphenylphosphonium. The newly developed compound exhibited stronger near infrared (NIR) fluorescence and reacted with bovine serum albumin. In addition, it preferentially accumulated in the mitochondria of cancer cells, as observed using confocal microscopy, and efficiently reduced cancer cell viability (IC50=3.04 µM). This novel multifunctional heptamethine Cy derivative, with cancer mitochondria targeting and NIR fluorescence imaging, may be promising as an alternative anticancer agent.

Abstract:Subcellular organelle mitochondria are becoming a key player and a driver of cancer. Mitochondrial targeting phototheranostics has attracted increasing attention for precise cancer therapy. However, those phototheranostic systems still face great challenges, including complex and multiple components, light scattering, and insufficient therapeutic efficacy. Herein, a molecular fluorophore IR-TPP-1100 was tactfully designed by molecular engineering for mitochondria-targeted fluorescence imaging-guided phototherapy in the second near-infrared window (NIR-II). IR-TPP-1100 not only exhibited prominent photophysical properties and high photothermal conversion efficiency but also achieved excellent mitochondria-targeting ability. The mitochondria-targeting IR-TPP-1100 enabled NIR-II fluorescence and photoacoustic dual-modality imaging of mitochondria at the organism level. Moreover, it integrated photothermal and photodynamic therapy, obtaining remarkable tumor therapeutic efficacy by inducing mitochondrial apoptosis. These results indicate that IR-TPP-1100 has great potential for precise cancer therapy and provides a promising strategy for developing mitochondria-targeting NIR-II phototheranostic agents.

Abstract:Both photothermal therapy (PTT) and sonodynamic therapy (SDT) are neoteric therapeutic method for tumor control and ablation without invasive. Herein, we report the synthesis of a newly designed near-infrared (NIR) trifluoromethyl-heptamethine cyanine (TFHC) dye, which could synergistically work as a mitochondrial-targeting sonosensitizer for SDT and also a photothermal therapy agent (PTA) for PTT. The structure and chemical components of TFHC were determined by 1H NMR and mass spectrum (MS). Furthermore, the introduce of trifluoromethyl was proved drastically improving the biocompatibility of TFHC. Moreover, TFHC was capable to kill cancer cells with a high efficiency (80%) under NIR and ultrasound (US) without causing any significant side effects. In general, the prepared mitochondria-targeting TFHC obtains the synergy effect for the PTT and SDT with excellent therapeutic effects and low side effects, which can be harnessed for the combinational cancer non-invasive therapy in the near future.

Abstract:Near-infrared (NIR) heptamethine cyanine (HCy) dyes are promising photothermal transducers for image-guided cancer treatment owing to their prominent photophysical properties and high photothermal conversion ability. However, HCy photothermal transducers usually have poor photostability due to degradation induced by the self-generated reactive oxygen species. Herein, a novel mitochondria-targeting dimeric HCy dye, named dimeric oBHCy, is rationally designed, exhibiting strong near-infrared II (NIR-II) fluorescence emission, high photothermal conversion efficiency (PCE), and excellent photostability. The large π-conjugation and drastic intramolecular motion of the diphenol rotor in the dimeric oBHCy enhance the nonradiative energy dissipation and suppress the intersystem crossing process, thereby achieving a high PCE (49.2%) and improved photostability. Impressively, dimeric oBHCy can precisely target mitochondria and induce mitochondrial damage upon NIR light irradiation. Under the guidance of in vivo NIR-II fluorescence imaging, efficient NIR light-activated photothermal therapy of 4T1 breast tumors is accomplished with a tumor inhibitory rate of 96% following a single injection of the dimeric oBHCy. This work offers an innovative strategy for designing cyanine photothermal transducers with integrated NIR-II fluorescence and photothermal properties for efficient cancer theranostics.

Abstract:Because mitochondria are the key regulators for many cellular behaviors and are susceptible to hyperthermia and reactive oxygen species, mitochondria-specific reagents for simultaneous targeting, imaging, and treatment are highly desirable in cancer theranostics. Herein, we developed a mitochondria targeted cyanine dye IR825-Cl, which possesses two separated excitation wavelength channels for both red fluorescence imaging and near-infrared (NIR) photothermal therapy (PTT). For imaging, IR825-Cl rapidly entered cells and selectively targeted mitochondria. Although IR825-Cl was completely quenched in water, interestingly, this dye had a turn-on response of red fluorescence (610 nm) in mitochondria under 552 nm excitation due to its polarity-responsive fluorescence emission. More interestingly, IR825-Cl realized the selective mitochondrial staining of cancer cells over normal cells and thus served as an ideal fluorescent probe for identifying cancer cells in normal tissues, which is extremely beneficial for cancer theranostics. For PTT, we demonstrated that under 808 nm NIR laser irradiation, this dye efficiently converted optical energy into heat, realizing mitochondria-targeted photothermal cancer therapy. Collectively, this molecule realized both high fluorescence emission (quantum yield > 43%) and effective light-to-heat conversion (17.4%), enabling its applications for wash free fluorescence imaging for mitochondria and highly efficient fluorescence imaging-guided PTT.

Abstract:The construction of intelligent near-infrared (NIR) fluorophores for high specificity to cancer cells and application in multiple therapeutic modalities is crucial for precise cancer diagnostic and therapy. In this study, an aggregation-induced emission-active NIR fluorophore (TACQ) with mitophagy-modulating activity was synthesized and developed for mitochondrial targeting multimodal cancer theranostics. The strengthened push-pull interaction extended the emission of TACQ into the NIR-II region (>1000 nm). Further, the rotor structure and twisted molecular conformation enables nanoaggregates of TACQ to balance the radiative and nonradiative decays to simultaneously exhibit bright NIR emission, high photothermal conversion efficiency (55%), and efficient generation of reactive oxygen species. The lipocationic property of TACQ allows it to selectively accumulate in the mitochondria of cancer cells. TACQ can induce mitophagy and block mitophagic flux facilitating cancer cell apoptosis. Both in vitro and in vivo evaluations revealed that TACQ is an efficient theranostic agent for NIR fluorescence and photothermal imaging-guided synergistic chemo-photothermal and photodynamic therapy.

Abstract:Photodynamic therapy has been developed as a prospective cancer treatment in recent years. Nevertheless, conventional photosensitizers suffer from lacking recognition and specificity to tumors, which causing severe side effects to normal tissues, while the enzyme-activated photosensitizers are capable of solving these conundrums due to high selectivity towards tumors. APN (Aminopeptidase N, APN/CD13), a tumor marker, has become a crucial targeting substance owing to its highly expressed on the cell membrane surface in various tumors, which has become a key point in the research of anti-tumor drug and fluorescence probe. Based on it, herein an APN-activated near-infrared (NIR) photosensitizer (APN-CyI) for tumor imaging and photodynamic therapy has been firstly developed and successfully applied in vitro and in vivo. Studies showed that APN-CyI could be activated by APN in tumor cells, hydrolyzed to fluorescent CyI-OH, which specifically located in mitochondria in cancer cells and exhibited a high singlet oxygen yield under NIR irradiation, and efficiently induced cancer cell apoptosis. Dramatically, the in vivo assays on Balb/c mice showed that APN-CyI could achieve NIR fluorescence imaging (λem = 717 nm) for endogenous APN in tumors and possessed an efficient tumor suppression effect under NIR irradiation.

Abstract:Multimodal imaging-guided phototherapy is an emerging treatment for tumor elimination that can provide more physiological and pathological information for precise theranostics with minimal side effects. Herein, we proposed a novel hypoxia-activated near infrared (NIR) probe (AzoCyS-N) based on a thioxanthene-hemicyanine scaffold for tumor multimodal imaging and photothermal/photodynamic therapy. The thioxanthenehemicyanine fluorophore (CyS-NH2) could be released from AzoCyS-N via reductive cleavage of the azo group under hypoxia, resulting in a dramatically enhanced NIR fluorescence signal at 760 nm and redshifted absorption at 730 nm. AzoCyS-N was further encapsulated into a folic acid modified amphiphilic polymer to form AzoCyS-N NPs to further improve the tumor selectivity. In addition, the released Cys-NH2 could generate singlet oxygen and exhibited decent photothermal effects under 690 nm light irradiation. The in vivo studies confirmed that the NIR fluorescence and PA signals in the tumor region were selectively lightened after intravenous injection of AzoCySN NPs. Finally, solid tumor growth was significantly suppressed by the synergistic effect of PDT and PTT. This work provides a powerful multimode imaging-guided synergistic phototherapeutic system for precise tumor theranostics.

Abstract:Organelle-targeted photosensitizers have been reported to be effective cell apoptosis agents. Mitochondria is recognized as an ideal target for cancer treatment due to its central role in oxidative metabolism and apoptosis. Meanwhile, two-photon (TP) fluorescence microscopy has become a powerful tool for fluorescence imaging in biological events based on its minimizing photodamage/photobleaching and intrinsic 3D resolution in deep tissues and in vivo. In this study, a series of novel mitochondrial-targeted TP fluorescent photosensitizers (TP-tracers) are designed, synthesized, and systematically investigated. These TP-tracers exhibit extraordinary anti-interference capability among different cations, anions, and amino acids as well as the insensitivity to the changes of pH and complex biological environments. TP-tracers are further used in fluorescence living cells, Drosophila brains, and zebrafish imaging with low cytotoxicity, excellent mitochondria-targeting, and TP properties. The results demonstrate efficient mitochondria-targeting cell selective apoptosis based on TP-activated cancer cells with highly single cell selectivity, and the pharmacokinetic study reveals that MitoY2 does not have accumulation in rats. It is believed that these molecules hold great potential in TP-related smart phototherapy.

Abstract:Photodynamic (PDT) and photothermal therapies (PTT) are emerging treatments for tumour ablation. Organic dyes such as porphyrin, chlorin, phthalocyanine, boron-dipyrromethene and cyanine are the clinically or preclinically used photosensitizer or photothermal agents. Development of structurally diverse near-infrared dyes with long absorption wavelength is of great significance for PDT and PTT. Herein, we report a novel near-infrared dye ML880 with naphthalimide modified cyanine skeleton. The introduction of naphthalimide moiety results in stronger electron delocalization and larger redshift in emission compared with IR820. Furthermore, ML880 is co-loaded with chemotherapeutic drug into ROS-responsive mesoporous organosilica (RMON) to construct nanomedicine NBD&ML@RMON, which exhibits remarkable tumor inhibition effects through PDT/PTT/chemotherapy in vivo.

Abstract:Mitochondria targeted phototherapy, including photodynamic therapy (PDT) and photothermal therapy (PTT), has excelled as an effective approach among other non-specific techniques for its high selectivity, non-invasiveness and low systemic toxicity. Derivatives of porphyrins, indocyanine dyes and rhodamine are widely utilized for cancer PDT or PTT. However, limitations, such as hypoxia and heat resistance of PDT and PTT, have restricted their efficacy in tumor treatment, making it urgent to develop highly efficient theranostic agents with synergistic effects. Aza-boron-dipyrromethene (aza-BODIPY) has shown promising prospects for synergistic phototherapy due to its outstanding reactive oxygen species (ROS) generation and photothermal effect. Herein, we designed and synthesized a near-infrared (NIR) aza-BODIPY derivative MeOABBr (ΦΔ = 84%). By encapsulating it with polyethylene glycol-folic acid (PEG-FA) and polyethylene glycol-triphenylphosphonium (PEG-TPP), tumor and mitochondria dual targeting nanoparticles (FMAB NPs) have been obtained. Triggered by NIR irradiation, FMAB NPs could generate ROS and hyperthermia (η = 40%) to cause mitochondrial dysfunction, resulting in cell apoptosis. Simultaneously, FMAB NPs, with unique optical properties, can be monitored precisely by photoacoustic, fluorescence and photothermal imaging in vivo. In particular, as proved by both in vitro and in vivo experiments, tumor-mitochondria dual targeted FMAB NPs exhibit high phototherapeutic efficacy without toxicity to normal tissues.

Abstract:Simultaneous targeted cancer imaging, therapy and real-time therapeutic monitoring can prevent over-or under-treatment. This work describes the design of a multifunctional nanomicelle for recognition and precise near-infrared (NIR) cancer therapy. The nanomicelle encapsulates a new pH-activatable fluorescent probe and a robust NIR photosensitizer, R16FP, and is functionalized with a newly screened cancer-specific aptamer for targeting viable cancer cells. The fluorescent probe can light up the lysosomes for real-time imaging. Upon NIR irradiation, R16FP-mediated generation of reactive oxygen species causes lysosomal destruction and subsequently trigger lysosomal cell death. Meanwhile the fluorescent probe can reflect the cellular status and in situ visualize the treatment process. This protocol can provide molecular information for precise therapy and therapeutic monitoring.

Abstract:Theranostic nanoparticles (NPs) capable of mitochondrial targeting/imaging, cancer/normal cell differentiation, early stage cancer diagnosis, and mitochondria-based photothermal therapy (PTT) were developed. The NPs were fabricated by physical encapsulation of near infrared (NIR) heptamethine cyanine dye me-IR82.5 into the inner core of the micelle-forming copolymer Pluronic F127 (PF127). The PF127/me-IR825 NPs exhibited two fluorescence emissions at similar to 610 nm (excited by 550 nm) and 845 nm (excited by 780 nm). The former was used for in vitro mitochondrial fluorescence imaging, cancer/normal cell differentiation, and early stage cancer detection with high fluorescence contrast. The latter was used for in vivo NIR fluorescence imaging. Besides, the NPs could also be used for in vivo photoacoustic imaging under 808 nm excitation. After irradiation by an 808 nm laser at an elevated power density, the NPs achieved excellent photothermal tumor ablation both in vitro and in vivo. Furthermore, me-IR825 inside the inner core of PF127/me-IR825 NPs could be degraded into biocompatible products after PTT treatment, which guaranteed the post-treatment biosafety of the NPs. Benefiting from their simple preparation, good colloidal dispersibility/stability, excellent cancer/normal cell differentiation ability, and superb in vivo dual-modal imaging-guided therapeutic outcome, the PF127/me-IR825 NPs may be used as a theranostic nanoplatform for applications in the biomedical field.

However, fluorescent probes that can be used for monitoring mitochondrial proteins have not yet been widely developed, thereby severely limiting the exploration of the functions of proteins in mitochondria. Towards this end, here we propose a near-infrared (NIR) fluorescence probe MPP to effectively illuminate the dynamic changes in mitochondrial proteins in live cells under oxidative stress, with excellent temporal and spatial resolution. Of particular importance, MPP extends the study of the pharmacology involved in apoptosis induced by anti-cancer drugs (hydroxycamptothecin (HCPT), epirubicin (Epi) and cyclophosphamide (CPA)) for the first time. Furthermore, employing a protein-activatable strategy, this probe could serve as an excellent phototherapeutic agent in photodynamic therapy (PDT). Finally, in vivo experiments suggest that this versatile probe can be used to image tumors in HeLa tumor-bearing mice for 24 h, which demonstrates that our probe could play a dual role as a robust phototherapeutic and imaging agent.
A Simple Strategy for Simultaneously Enhancing Photostability and Mitochondrial-Targeting Stability of Near-Infrared Fluorophores for Multimodal Imaging-Guided Photothermal Therapy.

Abstract:Near-infrared fluorophores are emerging as promising molecular tools for cancer theranostics because of their inherent biodegradability, low toxicity, and synthetic flexibility. However, they still suffer from several limitations, such as poor photostability and insufficient organelle-targeting stability during photothermal therapy. In this work, we introduce an "aldehyde functionalization" strategy for simultaneously enhancing photostability and mitochondria-immobilization of near-infrared fluorophores for the first time. Based on the proposed strategy, representative near-infrared organic molecules, namely AF-Cy, were rationally designed and synthesized. Upon aldehyde modification, the AF-Cy dyes displayed both remarkable photostability and mitochondrial-targeting stability. The strong absorption in the near-infrared region confers the AF-Cy dyes with outstanding fluorescent/photoacoustic imaging and photothermal therapy capabilities. Finally, in vitro and in vivo studies revealed the enhanced performance in inhibiting the growth of breast tumors under NIR laser radiation, and these results suggested the strong potential of AF-Cy dyes as efficient multimodal imaging-guided photothermal therapy agents, further highlighting the value of this simple strategy in the design high performance near-infrared fluorophores for tumor theranostics.
A Novel Heptamethine Cyanine Photosensitizer for FRET-amplified Photodynamic Therapy and Two-Photon Imaging in A-549 Cells.

Abstract:In this study, a new cyanine-based photosensitizer Cy-N-Rh was developed for photodynamic therapy. Based on fluorescence resonance energy transfer (FRET) mechanism, utilizing the absorption of the donor rhodamine (Rh), the acceptor heptamethine cyanine unit (Cy) was indirectly excited to produce singlet oxygen (O-1(2)). The efficiency of energy transfer from the donor Rh to the acceptor Cy was 78.5%. Meanwhile, the singlet oxygen yield of Cy-N-Rh (Phi(delta) = 12.00%) was much higher than that of the acceptor Cy (Phi(delta) = 4.35%) without FRET. Moreover, the dual cation gave Cy-N-Rh with excellent mitochondria targeting ability with Pearson's correlation coefficients of 0.90 and 0.91, respectively. In the MTT test, Cy-N-Rh had low dark cytotoxicity with cell survival rate above 90% and high photo cytotoxicity with cell survival rate below 40%. The cell apoptosis assay also demonstrated the role of the photosensitizer Cy-N-R visually. More importantly, Cy-N-Rh fulfilled two-photon excitation fluorescence imaging under the 800 nm femtosecond laser. All results indicate that this design strategy provides a new method for the development of higher-level cyanine photosensitizers. (C) 2022 Elsevier B.V. All rights reserved.
Enhanced porphyrin-based fluorescence imaging-guided photodynamic/photothermal synergistic cancer therapy by mitochondrial targeting

Abstract:Mitochondria are the power plants of the cell and play key roles in activating the apoptotic pathway in cancer cells, which are readily susceptible to cytotoxic reactive oxygen species and temperature elevations. Herein, we develop a "nanomissile" that targets mitochondria to enhance tumor treatment effects by facilitating mitochondrial dysfunction and releasing cytochrome C to activate the apoptotic pathway of cancer cells under 650-nm laser irradiation. Porphyrin-grafted polydopamine nanomaterial (PTPF-MitP) is designed as a nanomissile, with integrated O2-evolving photodynamic therapy and moderate photothermal therapy, which can selectively deliver to the mitochondria through a targeting unit, MitP. The cytotoxicity of PTPF-MitP to human lung tumor cells is twice as high as that of PTPF that does not have mitochondrial targeting units. In addition, it represents a realtime visualization and highly efficient treatment for tumor sites in vivo. This development represents a viable strategy for cancer therapy.
