Abstract:This research explores the role of surface modifications and particle size in the development of microparticles (MPs) for targeted drug delivery. Eight distinct types of MPs were fabricated, including PLGA, PLGA-PEG and their glucan-modified counterparts (GLU-PLGA and GP-PLGA). Characterization showed that glucan modifications significantly enhanced zeta potential and drug encapsulation efficiency. In vitro release studies indicated that particle size influenced the release dynamics, with smaller particles exhibiting a greater burst release. Notably, glucan-modified MPs showed markedly increased uptake in antigen-presenting cells (APCs), with GLU-PLGA and GP-PLGA achieving uptake rates of 64.33% and 47.33%, respectively. Furthermore, confocal microscopy demonstrated superior endosomal escape capabilities in APCs compared to unmodified MPs. These findings underscore the importance of glucan receptors in mediating effective uptake and distribution of MPs in APCs. Overall, this study positions glucan-modified MPs as promising candidates for enhancing drug delivery and vaccine efficacy, suggesting a strategic approach for future immunotherapy applications. The uptake rates for GLU-PLGA MPs reached 64.33% in macrophages and 47.74% in DCs, compared to lower rates for unmodified MPs. Additionally, glucan-modified GLU-PLGA and GP-PLGA MPs demonstrated improved cytoplasmic distribution, with up to and 42.90% and 54.77% localization in macrophages. These findings indicate that glucan modification facilitates effective endosomal escape, which is crucial for enhancing immunological responses.
