Abstract:L-threo-4-methylsulfonylphenylserine ethyl ester [(2S, 3R)-1d], a key building block of florfenicol which synthesized by esterification using ethanol and L-threo-4-methylsulfonylphenylserine [(2S, 3R)-1b] as substrates. L-threonine aldolase (LTA) is a promising biocatalyst for producing (2S, 3R)-1b through a one-step process taking 4-Methylsulphonyl benzaldehyde (1a) and glycine as substrates under the mild condition. However, the moderate Cβ-stereoselective blocked the industrial application of LTA. To address this issue, rational design by combination with prereaction state molecular dynamics (MD) simulation and per-residue energy decomposition algorithm are employed to engineer LTA for enhancing Cβ-stereoselective. As a result, a triple mutant N16A/E98S/Y314R (Mu3) is screened out to produce (2S, 3R)-1b with 93.7% de and 90.2% conversion under the 300 mM 1a substrate loading. Furthermore, esterification is applied to synthesize (2S, 3R)-1d with over 99% chiral purity and 99% product purity. The success of this study provides new insights for the rational design of LTA with improved Cβ-stereoselectivity and proves a chemo-enzymatic route for green synthesis of (2S, 3R)-1d.

Abstract:Alkoxy-L-serines are useful for peptide syntheses. The demand for alkoxy-L-serines in the pharmaceutical industries continues to increase because of their multiple physiological effects. In this research, an improved method for alkoxy-L-serines synthesis is reported. A series of substrates, DL-β-alkoxy-α-amino propionamides, was used for the synthesis of alkoxy-serines catalyzed by Escherichia coli cells with peptidase B(PepB) activity. The results show that PepB has a high resolution activity with DL-β-alkoxy-α-amino propionamides as substrate. Reaction conditions were optimized, i.e., DL-β-methoxy-α-amino propionamide as substrate at pH=9.0, 40 °C and 14 h, and the optimal reaction concentration is 400 mmol/L. The results also show that divalent metal cations exhibit different effects on the PepB activity, for example, Zn2+ and Cu2+ can obviously inhibit the activity of PepB, whereas Co2+, Ca2+, Mn2+ and Mg2+ at low concentrations can activate PepB. This research provides access to enantiomerically enriched and valuable alkoxy-L-serines from a simple amino-amide racemate.

Abstract:Achieving the preparation of enantiomerically enriched S-stereogenic compounds is a long-standing objective in stereoselective synthesis, owing to the fundamental importance and broad applications of these chiral scaffolds in various fields. Despite recent significant advancements, catalyst-controlled stereoselective synthesis of S-stereogenic compounds remains to be a considerable challenge, particularly by means of small-molecule catalysts. Herein, we disclosed an organocatalytic strategy for highly practical and enantioselective sulfinylation of alcohols and amines through the activation of sulfinates by forming mixed sulfinic anhydrides. Tuning the structure of the reactive species with sterically congested moieties, a simple, naturally occurring quinine catalyst effectively controls the chemo- and enantioselectivity over the nucleophilic S–O and S–N bond constructions, affording a wide range of chiral sulfinyl derivatives with excellent optical purities. Notably, the protocol readily facilitates the coupling with various natural products and commercial drugs that could offer an attractive strategy for the late-stage diversification of important biologically intriguing molecules.

Abstract:Herein, multiple types of chiral Os(II) complexes have been designed to address the appealing yet challenging asymmetric C(sp3)-H functionalization, among which the Os(II)/Salox species is found to be the most efficient for precise stereocontrol in realizing the asymmetric C(sp3)-H amidation. As exemplified by the enantioenriched pyrrolidinone synthesis, such tailored Os(II)/Salox catalyst efficiently enables an intramolecular site-/enantioselective C(sp3)-H amidation in the γ-position of dioxazolone substrates, in which benzyl, propargyl and allyl groups bearing various substituted forms are well compatible, affording the corresponding chiral γ-lactam products with good er values (up to 99:1) and diverse functionality (> 35 examples). The unique performance advantage of the developed chiral Os(II)/Salox system in terms of the catalytic energy profile and the chiral induction has been further clarified by integrated experimental and computational studies.

Abstract:Enzymatic asymmetric sulfoxidation using molecular oxygen as the oxidant is a promising green chemistry approach to chiral sulfoxide production. Despite the broad substrate spectrum of cyclohexanone monooxygenases (CHMOs), some unnatural substrates with bulky functional groups, such as the pharmaceutically relevant omeprazole sulfide, cannot be effectively accepted by CHMOs. Herein, we describe a set of variants derived from an Acinetobacter calcoaceticus CHMO (AcCHMO), whose active sites adjacent to the substrate tunnel were altered to shift the substrate specificity from cyclohexanone monooxygenation toward omeprazole sulfide sulfoxidation. We performed homologous modeling and molecular docking to identify key residues that might affect the substrate specificity. Two libraries of residues lining the active center of AcCHMO were then constructed and screened by an effective halo-based selection method using the solubility difference between the substrate (omeprazole sulfide) and product (esomeprazole). Functional evaluation of the resultant variants showed that the substrate specificity of AcCHMO was markedly altered from the small natural substrate (cyclohexanone) toward the desired bulky substrate (omeprazole sulfide) despite the extremely poor activity detected even for the best variant, M2 (0.61 U/g(prot)). The crystal structure of M2 complexed with a flavin adenine dinucleotide (FAD) prosthetic group was determined, which provided insight into the altered substrate specificity. To improve the activity of enzyme M2 toward pharmaceutical precursor omeprazole sulfide, we performed both local and global protein engineering among the two CASTing libraries surrounding FAD(+) and NADP(+) prosthetic groups and an error-prone PCR library of the full-length AcCHMO. As a result, variant M6 was obtained, giving a 50-fold higher activity compared to M2. This structure-guided protein engineering of AcCHMO provided a promising candidate for converting omeprazole sulfide into (S)-omeprazole using a green biocatalytic method.

Abstract:An organocatalytic asymmetric enantioselective domino oxa-Michael–Mannich-[1,3]-amino rearrangement reaction of N-tosylsalicylimines with a wide range of α,β-unsaturated aldehydes utilizing diarylprolinol silyl ether catalysis is described. The catalytic reactions proceed with excellent enantioselectivity (up to 99% ee) to produce the corresponding chair N-tosylimines-chromenes with a yield of up to 99%, tolerating a range of functional groups. This methodology offers a new method with great potential to further extend the synthetic power and versatility of chiral aminocatalysis.

The method is mild in reaction condition, high in product purity and suitable for industrial production.

Abstract:Abstract Chiral sulfoxides are valuable building blocks in asymmetric synthesis. However, the biocatalytic synthesis of chiral sulfoxides is still challenged by low product titres. Herein, we report the use of peroxygenase as a catalyst for asymmetric sulfoxidation under non‐aqueous conditions. Upon covalent immobilisation, the peroxygenase showed stability and activity under neat reaction conditions. A large variety of sulfides was converted into chiral sulfoxides in very high product concentration with moderate to satisfactory optical purity (e. g. 626 mM of ( R )‐methyl phenyl sulfoxide in approx. 89 % ee in 48 h). Further polishing of the ee value via cascading methionine reductase A (MsrA) gave>99 % ee of the sulfoxide. The robustness of the enzymes and high product titer is superior to the state‐of‐the‐art methodologies. Gram‐scale synthesis has been demonstrated. Overall, we demonstrated a practical and facile catalytic method to synthesize chiral sulfoxides.

Abstract:A rhodium-catalyzed enantioselective construction of triorgano-substituted silicon-stereogenic siloxanes and alkoxysilanes is developed. This process undergoes a direct intermolecular dehydrogenative Si–O coupling between dihydrosilanes with silanols or alocohols, giving access to a variety of highly functionalized chiral siloxanes and alkoxysilanes in decent yields with excellent stereocontrol, that significantly expand the chemical space of the silicon-centered chiral molecules. Further utility of this process was illustrated by the construction of CPL-active (circularly polarized luminescence) silicon-stereogenic alkoxysilane small organic molecules. Abstract:In this paper, chiral aryl iodides were used for the first time to catalyze the formation of C-O and C-N bonds of inactive olefins to obtain optically active aminolactone derivatives. The reaction is compatible with various substituted alkenyl carboxylic acid substrates. This method usually shows high activity and enantioselectivity, with a yield of up to 83% and an ee of up to 99%. It is recovery and reuse of chiral aryl iodide catalyst CIC4 showed almost no loss in product yield and enantioselectivity after 3 runs.

Abstract:Baeyer–Villiger monooxygenases (BVMOs) are able to catalyse the asymmetric oxidation of sulfides. This property has made them attractive catalysts for the synthesis of chiral sulfoxide drugs. Here, we have designed and synthesised an exhaustive combinatorial mutant library of the previously identified lansoprazole sulfide monooxygenase Cb BVMO V1 . From this synthetic combinatorial mutant library, the best mutant, Cb BVMO V3 , was selected with a specific activity of approximately 1 U mg −1 for lansoprazole sulfoxides. We then optimised the reaction conditions of a two-phase system, achieving the enzymatic asymmetric synthesis of ( R )-lansoprazole in a space-time yield of 213 g L −1 d −1 and an enantiomeric excess of >99% ( R ) with no detectable by-products. In addition, Cb BVMO V3 showed higher activity towards other prazole sulfides. These results indicate the potential application of Cb BVMO in the chiral sulfoxide drug industry.

Abstract:(S)-2-chlorophenylglycine ((S)-CPG) is a key chiral intermediate for the synthesis of clopidogrel. Herein, a novel, efficient and environmentally friendly chemo-enzymatic route for the preparation of optically pure (S)-CPG was developed. A straightforward chemical synthesis of the corresponding prochiral keto acid substrate (2-chlorophenyl)glyoxylic acid (CPGA) was developed with 91.7% yield, which was enantioselectively aminated by leucine dehydrogenase (LeuDH) to (S)-CPG. Moreover, protein engineering of LeuDH was performed via directed evolution and semi-rational design. A beneficial variant EsLeuDH-F362L with enlarged substrate-binding pocket and increased hydrogen bond between K77 and substrate CPGA was constructed, which exhibited 2.1-fold enhanced specific activity but decreased thermal stability. Coupled with a glucose dehydrogenase from Bacillus megaterium (BmGDH) for NADH regeneration, EsLeuDH-F362L completely converted up to 0.5 M CPGA to (S)-CPG in 8 h at 40 °C.

Abstract:A simple and efficient approach to obtain optically pure 1,4-diols was established. The asymmetric oxidation of sulfides to sulfoxides with cumyl hydroperoxide in moderate yields and moderate to high enantioselectivities (up to 84%) catalyzed by chiral Ti/1,4-diols complexes has been achieved. A 76% ee value was obtained in the asymmetric synthesis of esomeprazole.

Abstract:We developed an efficient and enantioselective method for the synthesis of chiral alpha-silyl amines. In the presence of a copper(I)-chiral monodentate N-heterocyclic carbene complex (5 mol%), smooth nucleophilic silicon transfer from Me(2)PhSiBpin (pin = pinacolato) to tosyl-protected aldimines affords the corresponding alpha-silyl amines in 77-93% yields over 15 examples and satisfactory ee of 79-99%. The asymmetric silylation of ketimines was also explored.

Abstract:(S)-Arylpropionic acids are the pharmacologically active enantiomers of profen drugs that are challenging to synthesize chemically. Here, we report a chemoenzymatic synthesis of (S)-profens from alkenes involving styrene oxide isomerase (SOI)-catalyzed enantioretentive Meinwald-type isomerization of epoxides to aldehydes. This success relies on the engineering of SOI to accept larger substrates with enhanced activity for the rate-limiting isomerization step using a multi-Harmony analysis to guide directed evolution, which represents the first successful rational-based evolution of a membrane-associated enzyme without structural information. The engineered SOI-F35A/A131Y demonstrated 10.5-fold improvement in catalytic efficiency compared to wild-type SOI and was successfully combined in a cascade with aldehyde dehydrogenase to produce four (S)-profens from chemically derived (S)-epoxides in 83–89% yield and 90–94% ee. This work represents a simple and cleaner route to synthesize (S)-profens from alkenes, and the engineered SOI can be useful to produce other types of chiral molecules.

Abstract:Despite the growing demand for the enantioenriched silicon-stereogenic silanols in materials science, medicinal chemistry, and modern synthetic chemistry, the catalytic asymmetric synthesis of which remains a considerable challenge compared with their carbinol analogues. Herein, a copper-catalyzed desymmetrization of silanediols for the synthesis of various functionalized chiral silanols is demonstrated. The reaction features high atom economy, decent yield with excellent stereoselectivity, and H2 as the sole byproduct. Key to the success for discrimination of the gem-diol groups in silanediol relies on an enantioselective σ-bond metathesis process. Further straightforward elaboration of the enantioenriched silicon-stereogenic silanols delivers several interesting chiral silane scaffolds without the loss of enantiopurities.

Enantioselective high-performance liquid chromatography of aryl-substituted oxazolines as an efficient tool for determination of chiral purity of serine medicinal components

Abstract:A new approach for the evaluation of chiral purity of serine esterification products bearing long-chain alkyl substituents was developed. The compounds were simply converted to aryl-substituted oxazolines which: (i) facilitates effective chromatographic enantioseparation and (ii) enables direct detection using ultraviolet absorption. The method employs a polysaccharide-based chiral stationary phase and allows enantioseparation of highly stable oxazoline products in less than 6 min using a simple binary mobile phase. As opposed to the previously used normal phase method the developed method was performed in the reversed-phase mode. Aside from the benefits of switching to less hazardous solvents with regard to the principles of Green Chemistry, this has also led to a reduction in the analysis time. In comparison with known serine chromophores, the best enantioseparation of aryloxazoline rigid structure may be achieved only based on non-polar interactions with the chiral stationary phase. In contrast, the substitution of the chromophore moiety with hydroxyl substituent affected intra and intermolecular interactions that caused enantioseparation differences. Concurrently, we found high chirality retention of (R)- and (S)-configuration oxazoline standards (≥99% enantiomeric excess) during the introduction of the ultraviolet label. The method is suitable for rapid injection of the mixture containing the ultraviolet absorption marker without prior purification.
