Abstract:Water-soluble upconversion luminescent (UCL) nanoparticles based on triplet-triplet annihilation (TTA) were successfully prepared by coloading sensitizer (octaethylporphyrin Pd complex) and annihilator (9,10-diphenylanthracene) into silica nanoparticles. The upconversion luminescence quantum yield of the nanoparticles can be as high as 4.5% in aqueous solution. As determined by continuous kinetic scan, the nanoparticles have excellent photostability. Such TTA-based upconversion nanoparticles show low cytotoxicity and were successfully used to label living cells with very high signal-to-noise ratio. UCL imaging with the nanoparticles as probe is capable of completely eliminating background fluorescence from either endogenous fluorophores of biological sample or the colabeled fluorescent probe. In particular, such blue-emissive upconversion nanoparticles were successfully applied in lymph node imaging in vivo of living mouse with excellent signal-to-noise ratio (>25), upon low-power density excitation of continuous-wave 532 laser (8.5 mW cm(-2)). Such high-contrast and low-power excited bioimaging in vivo with a blue-emissive upconversion nanoparticle as probe may extend the arsenal of currently available luminescent bioimaging in vitro and in vivo.

Abstract:Near-infrared (NIR) dye-sensitized upconversion nanoparticles (UCNPs) can broaden the absorption range and boost upconversion efficiency of UCNPs. Here, we achieved significantly enhanced upconversion luminescence in dye-sensitized core/active shell UCNPs via the doping of ytterbium ions (Yb(3+)) in the UCNP shell, which bridged the energy transfer from the dye to the UCNP core. As a result, we synergized the two most practical upconversion booster effectors (dye-sensitizing and core/shell enhancement) to amplify upconversion efficiency. We demonstrated two biomedical applications using these UCNPs. By using dye-sensitized core/active shell UCNP embedded poly(methyl methacrylate) polymer implantable systems, we successfully shifted the optogenetic neuron excitation window to a biocompatible and deep tissue penetrable 800 nm wavelength. Furthermore, UCNPs were water-solubilized with Pluronic F127 with high upconversion efficiency and can be imaged in a mouse model.

Near‐Infrared Upconversion Luminescence and Bioimaging in Vivo Based on Quantum Dots

Abstract:Recently, upconversion luminescence (UCL) has been widely applied in bioimaging due to its low autofluorescence and high contrast. However, a relatively high power density is still needed in conventional UCL bioimaging. In the present study, an ultralow power density light, as low as 0.06 mW cm−2, is applied as an excitation source for UCL bioimaging with PbS/CdS/ZnS quantum dots (UCL‐QDs) as probes. The speculated UCL mechanism is a phonon‐assisted single‐photon process, and the relative quantum yield is up to 4.6%. As determined by continuous irradiation with a 980 nm laser, the UCL‐QDs show excellent photostability. Furthermore, UCL‐QDs‐based probe is applied in tumor, blood vessel, and lymph node bioimaging excited with an eye‐safe low‐power light‐emitting diode light in a nude mouse with few heat effects.

Abstract:Upconversion optical imaging has great application prospects in biomedical imaging. However, research on molecular upconversion luminescence materials is very limited. In this study, a cyanine dye modified using polyethylene glycol is reported, which has frequency upconversion luminescence (FUCL) capability and can be applied for in vivo FUCL imaging after forming nanomicelles. The optical properties of PEGylated cyanine dye show that the light absorption property of cyanine does not be influenced and can still emit near-infrared fluorescence. However, under the condition that the illumination wavelength is larger than the maximal fluorescence wavelength, the FUCL of cyanine dye is generated at the same position as the down-converted luminescence. The nanomicelles exhibit good biocompatibility. When applied to in vivo imaging, the cyanine-based nanomicelles not only qualify for high-resolution subcutaneous blood vessel imaging but also exhibit good imaging ability for various organs and tumors. These results point out that cyanine-based nanomicelles with FUCL characteristics are expected to be further applied in the field of a basis for the construction of FUCL molecules.

Abstract:Upconversion nanophosphors are considered to be important components in advanced materials designed for in vivo bioapplications and benefit from their unique anti-Stokes upconversion luminescence properties. The near-infrared light excitation allows for a large penetration depth in biotissues during in vivo applications. This review presents recent advances in the optimization and functionalization of upconversion nanomaterials for bioimaging and in vivo therapy. The most effective protocols are introduced in detail. Current limitations and future prospects are also included to provide a general summary and suggest future research directions. Upconversion nanophosphors have unique ability to generate anti-Stokes luminescence. The rapid developments of nanotechnology in recent years have demonstrated the successful synthesis and optimization of such upconversion nanophosphors. This review summarizes the recent advances using upconversion nanophosphors as bioprobe for in vivo applications. Upconversion nanoparticles emit photons at shorter wavelengths than the wavelengths of the photons they absorb under excitation, and thus convert low-energy light into higher-energy light. With obvious potential for solar cells, these nanoparticles also show promise in bioimaging and therapeutic applications: they can be excited by near-infrared radiation and emit shorter near-infrared or visible light, leading to minimal scattering by biological tissues and good penetration depth. Fuyou Li and colleagues from Fudan University, China, review recent progress toward optimizing and functionalizing these nanoparticles for in vivo applications such as multi-modality imaging and near-infrared light-driven therapy. Challenges include insufficient luminescent efficiencies, which can be improved through core-shell structures, as well as the alleviation of overheating effects—achieved, for example, through incorporating additional ions that act as antennas and result in more efficient wavelength conversion. Additionally, the potential toxicity of these inorganic particles—including their distribution and effects in the body—needs to be investigated further.

Abstract:Lanthanide-doped luminescent nanoparticles with both emission and excitation in the near-infrared (NIR-to-NIR) region hold great promise for bioimaging. Herein, core@shell structured LiLuF4: Nd@LiLuF4 (named as Nd@Lu) nanoparticles (NPs) with highly efficient NIR emission were developed for high-performance in vivo bioimaging. Strikingly, the absolute quantum yield of Nd@Lu NPs reached as high as 32%. After coating with polyethylene glycol (PEG), the water-dispersible Nd@Lu NPs showed good bio-compatibility and low toxicity. With efficient NIR emission, the Nd@Lu NPs were clearly detectable in tissues at depths of up to 20 mm. In addition, long-term in vivo biodistribution with a high signal-to-noise ratio of 25.1 was distinctly tracked upon an ultralow-power-density excitation (10 mW cm(-2)) of 732 nmfor the first time.

Abstract:We designed and synthesized NaYF4:Yb,Tm@LiLuF4:Nd core-shell nanoparticles, which can emit at nm under 980 nm excitation and at similar to 1060 nm under 808 nm excitation, simultaneously having an upconversion and downshifting mechanism for near-infrared (NIR) emission. After surface modification with sodium citrate, the soluble nanoparticles were used in the in vitro NIR luminescence imaging to compare the penetration depth and the scattering of tissue. Furthermore, to determine the differences between the upconversion and downshifting fluorescence for biological imaging, the soluble nano particles also were operated on the aforementioned two modes for in vivo imaging.

Abstract:Organic fluorescent probes are widely used in bioimaging and bioassays, but the notorious photobleaching hampers their applications. Encapsulation of organic dyes into nanoparticles (NPs) is an effective strategy to minimize photobleaching, but classical organic dye molecules tend to have their fluorescence quenched in aggregate states, which is termed aggregation-caused quenching (ACQ). Here we demonstrate our attempt to tackle this problem through the aggregation-induced emission (AIE) strategy. 3,4: 9,10-Tetracarboxylic perylene bisimide (PBI) is a well-known organic dye with a serious ACQ problem. By attaching two tetraphenylethene (TPE) moieties to the 1,7-positions, the ACQ-characteristic PBI-derivative was converted to an AIE-characteristic molecule. The obtained PBI derivative (BTPEPBI) exhibits several advantages over classical PBI derivatives, including pronounced fluorescence enhancement in aggregate state, red to near infrared emission, and facile fabrication into uniform NPs. Studies on the staining of MCF-7 breast cancer cells and in vivo imaging of a tumor-bearing mouse model with BTPEPBI-containing NPs reveal that they are effective fluorescent probes for cancer cell and in vivo tumor diagnosis with high specificity, high photostability and good fluorescence contrast.

Abstract:Persistent luminescence bioimaging provides an optimal method of eliminating autofluorescence for a higher resolution and sensitivity due to the absence of excitation light. However, ultraviolet-light is still necessary in common energy charging processes, which limits its reactivation in vivo due to its low penetration depth. In the present study, we introduce a type of hybrid nanocluster (UCPL-NC) composed of upconversion nanoparticles, β-NaYbF4:Tm@NaYF4, and persistent nanoparticles, Zn1.1Ga1.8Ge0.1O4:0.5%Cr, which can be activated by a 980 nm laser and exhibits an afterglow at 700 nm to realize near-infrared to near-infrared upconverted persistent luminescence bioimaging. The persistent luminescence of the UCPL-NCs can be reactivated even when covered with a 10-mm pork. We demonstrate that these polyethylene glycol-modified phospholipid functionalized UCPL-NCs can be reactivated in vivo and applied in the persistent luminescence lymphatic imaging on small animals.

Abstract:Simultaneous deep macroscopic imaging and microscopic imaging is in urgent demand, but is challenging to achieve experimentally due to the lack of proper fluorescent probes. Herein, we have designed and successfully synthesized simplex Er(3+)-doped upconversion nanoparticles (UCNPs) with double excitation bands for simultaneous deep macroscopic and microscopic imaging. The material structure and the excitation wavelength of Er(3+)-singly doped UCNPs were further optimized to enhance the upconversion emission efficiency. After optimization, we found that NaYF4:30%Er(3+)@NaYF4:2%Er(3+) could simultaneously achieve efficient two-photon excitation (2PE) macroscopic tissue imaging and three-photon excitation (3PE) deep microscopic when excited by 808 nm continuous wave (CW) and 1480 nm CW lasers, respectively. In vitro cell imaging and in vivo imaging have also been implemented to demonstrate the feasibility and potential of the proposed simplex Er(3+)-doped UCNPs as bioprobe.

Abstract:Upconversion luminescence involves conversion of two or more low energy photons,usually near infrared(typically 980 nm),to higher energy visible emission through multiple photon absorption or energy transfer.Compared with the traditional ultraviolet excitation,upconversion luminescence exhibits deep tissue penetration depth,minimal photodamage to biological samples and non-autofluorescence of biological samples.Therefore,upconversion luminescence has great application prospects in biological imaging.Herein,on basis of the unique upconversion luminescence of rare earth materials,the novel upconversion luminescence imaging technologies including upconversion luminescence microscopy and upconversion luminescence whole-body imaging have been developed for application in targeted bioimaging in vitro and in vivo.

Abstract:In vivo optical imaging is an important application value in disease diagnosis. However, near-infrared nanoprobes with excellent luminescent properties are still scarce. Herein, two boron-dipyrromethene (BODIPY) molecules (BDP-A and BDP-B) were designed and synthesized. The BODIPY emission was tuned to the near-infrared (NIR) region by regulating the electron-donating ability of the substituents on its core structure. In addition, the introduction of polyethylene glycol (PEG) chains on BODIPY enabled the formation of self-assembled nanoparticles (NPs) to form optical nanoprobes. The self-assembled BODIPY NPs present several advantages, including NIR emission, large Stokes shifts, and high fluorescence quantum efficiency, which can increase water dispersibility and signal-to-noise ratio to decrease the interference by the biological background fluorescence. The in vitro studies revealed that these NPs can enter tumor cells and illuminate the cytoplasm through fluorescence imaging. Then, BDP-B NPs were selected for use in vivo imaging due to their unique NIR emission. BDP-B was enriched in the tumor and effectively illuminated it via an enhanced penetrability and retention effect (EPR) after being injected into the tail vein of mice. The organic nanoparticles were metabolized through the liver and kidney. Thus, the BODIPY-based nanomicelles with NIR fluorescence emission provide an effective research basis for the development of optical nanoprobes in vivo.

Abstract:Upconversion based on triplet-triplet annihilation (TTA-UC) has received much attention due to its unique ability to effectively harvest low-energy photons in the solar spectrum for widespread investigations involving energy, environment, and bioapplication. However, the utilization of conventional TTA-UC systems for low-power bioimaging in vivo is still a challenge due to their incompatibility and moderate efficiency in aqueous media. Herein, we pioneer the molecular modification strategy by introducing steric hindrance to the BODIPY annihilators to realize robust TTA-UC in biocompatible nanocapsule. The improved molecules could maintain excellent luminescence even at a very high concentration wherein the separation distance between neighbouring chromophores was reduced to facilitate the pivotal process of Dexter energy transfer. In this way, the encapsulated TTA-UC system became highly efficient in biological circumstance with quantum efficiency ФUC of 6.3%. Consequently, the constructed nanocapsule was capable of operating at low-power with high contrast for in vivo imaging.
Dual-Functional Alpha-Nayb(Mn)F-4:Er3+@Naluf4 Nanocrystals With Highly Enhanced Red Upconversion Luminescence

Abstract:Rare earth doped upconversion nanoparticles (UCNPs) used as bioprobes for in vivo upconversion luminescence (UCL) deep tissue imaging have been extensively studied. Red emission possesses high penetration length in biological tissue, so it's more convenient for naked eyed red emission to be performed in biological imaging and detection. However, improved in vivo imaging depth based on the NIR-to-red UCL is still challenging and desirable. Herein, novel alpha-NaYb(Mn)F-4:Er3+@NaLuF4 core-shell nanocrystals with enhanced red emission and improved imaging depth have been synthesized. Under 980 nm excitation, there is a 220% increase in red-to-green ratio and even a 400% increase in red-emission when the Yb3+ core doping concentration is increased from 18% to 68% in the core-shell structure. And the UCL signals detection of pork muscle tissues could be visualized even at a depth of 18 mm. By the tail vein injection of PEG modified alpha-NaYb(Mn)F-4:Er3+@NaLuF4 UCNPs, the in vivo and ex vivo NIR-to-red UCL deep tissue imaging present significant UCL signals in liver and spleen. Moreover, the UCNPs could also be used as in vitro and in vivo X-ray computed tomography (CT) contrast agent owing to the large X-ray absorption efficiency and high atomic number of ytterbium and lutetium elements. The synergistic combination of the in vivo NIR-to-red UCL deep tissue and CT imaging could provide comprehensive diagnosis information for cancer.

Abstract:Under 980 nm near-infrared (NIR) excitation, upconversion luminescent (UCL) emission of GdF(3):Yb,Er upconversion nanoparticles (UCNPs) synthesized by a simple and green hydrothermal process can be tuned from yellow to red by varying the concentration of dopant Li(+) ions. A possible mechanism for enhanced red upconverted radiation is proposed. A layer of silica was coated onto the surface of GdF(3):Yb,Er,Li UCNPs to improve their biocompatibility. The silica-coated GdF(3):Yb,Er,Li UCNPs show great advantages in cell labeling and in vivo optical imaging. Moreover, GdF(3) UCNPs also exhibited a positive contrast effect in T(1)-weighted magnetic resonance imaging (MRI). These results suggest that the GdF(3) UCNPs could act as dual-modality biolabels for optical imaging and MRI.

Abstract:Rare-earth (RE) upconversion nanophosphors (UCNPs) have attracted extensive attention in the field of bioimaging. Through a unique anti-Stokes multiphoton UC process, the UCNPs can convert the low-energy near-infrared (NIR) photons into high-energy ultraviolet (UV), visible (VIS) and shorter NIR emission. Such unique UC emission overcome the disadvantages faced by conventional downconversion (DC) fluorescent probes with the advantages including high depth of light penetration, the absence of photodamage to living organisms, ignorable autofluorescence and high detection sensitivity, thus making them an ideal type of fluorescent label for bioimaging. In this chapter, the UC emission mechanism of UCNPs excited by different wavelengths was introduced. In detail, recent reports regarding the UCNPs-based optical bioimaging are summarized, including the applications of UCNPs for single-modal upconversion luminescence (UCL) optical imaging, UCL-based multimodal imaging, nanoscopy imaging, multiplexed imaging, ratiometric imaging, hypoxia imaging, potassium imaging, myeloperoxidase imaging, mRNA imaging and neuronal activity imaging. Finally, we discuss the challenges and opportunities in the development of UCNPs for bioimaging, with an emphasis on how to use them to achieve imaging-guided disease therapy.

Abstract:BODIPY (4,4-difluoro-4-bora-3a,4a-diaza-s-indacene) is an emissive chromophore in solutions but suffers from fluorescence quenching when aggregated due to its flat molecular conformation and small Stokes shift. To create aggregate-state emissive BODIPY luminogens, tetraphenylethene (TPE), which is a popular luminogen with intriguing aggregation-induced emission (AIE) characteristic, is introduced as periphery to a methylated BODIPY core. Three TPE-BODIPY adducts are synthesized and characterized, and their photophysical properties and electronic structures are investigated. The incorporation of AIE-active TPE units alleviates aggregation-caused quenching of BODIPY core, furnishing emissive nanoparticles based on TPE-BODIPY adducts. Significantly, the two-photon absorption (TPA) and two-photon excited fluorescence (TPEF) properties are improved as more TPE units are attached. The luminogens with 3TPE units (3TPE-BODIPY) shows the strongest TPA and TPEF in the wavelength range of 750-830 nm, with cross-section values of 264 and 116 GM at 810 nm, respectively. Red emissive nanoparticles with a Stokes shift of 60 nm and a fluorescence quantum yield of 16% are attained by encapsulating 3TPE-BODIPY with 1,2-sistearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000]. The nanoparticles are biocompatible and function well in TPEF cellular imaging and mouse brain blood vascular visualization.

Abstract:Lanthanide-doped upconverting nanoparticles (UCNPs) possess uniqueanti-Stokes optical properties, in which low energy near-infrared (NIR) photons can beconverted into high energy UV, visible, shorter NIR emission via multiphoton upconversionprocesses. Due to the rapid development of synthesis chemistry, lanthanide-doped UCNPscan be fabricated with narrow distribution and tunable multi-color optical properties. Theseunique attributes grant them unique NIR-driven imaging/drug delivery/therapeuticapplications, especially in the cases of deep tissue environments. In this brief review, weintroduce both the basic concepts of and recent progress with UCNPs in material engineeringand theranostic applications in imaging, molecular delivery, and tumor therapeutics. The aimof this brief review is to address the most typical progress in basic mechanism, materialdesign as bioimaging tools.

Abstract:The red emission of upconversion nanoparticles (UCNPs) is located in an “optical window” range of the biological tissue, which has a promising prospect in biology application. In this work, novel red-emitting NaYF4:Yb3+/Er3+ UCNPs with CH3COO− modifications were synthesized by single-mode focused microwave method. The morphology, structure, and luminescence properties of as-prepared UCNPs were characterized. The results showed that single-mode focused microwave method was an effective approach to prepare hydrophilic and strongly red emission UCNPs within a drastically reduced reaction time (only 10 min) compared to the traditional methods. Importantly, we also demonstrated that the CH3COO− modified NaYF4:Yb3+/Er3+ UCNPs could be successfully applied for in vitro imaging of HeLa cells. This work not only provides a rapidly synthetic methodology for the preparation of red-emitting UCNPs but also offers an effective material for cancer cell imaging.
