Abstract:It is known that chemoresistance is a major cause of treatment failure in acute myeloid leukemia (AML). Substantial data indicate that the CD44 adhesion molecule is strongly expressed on AML blasts and that it can also inhibit apoptosis. Our study shows that drug resistance of the AML cell line HL60/ADM is due to overexpression of CD44. In an in vitro study, we knocked down CD44 in the HL60/ADM cell line using small interfering RNA (siRNA). Cell proliferation and the 50 % inhibitory concentrations (IC50) were determined by Cell Counting Kit-8 (CCK-8) assay. Cell apoptosis and intracellular ADM accumulation were detected by flow cytometry. Expression of CD44, Bcl-2, c-Myc were assayed by quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot. The results indicate that the expression of CD44 in HL60/ADM cell line was much higher than in HL60 cell, and siRNA targeted CD44 (siRNA/CD44) could silence its expression in both mRNA and protein levels effectively. siRNA/CD44 substantially induces cell apoptosis, inhibits cell proliferation, enhances susceptibility to ADM and Ara-C, and at the same time increases intracellular ADM accumulation even reverses chemoresistance to ADM and Ara-C. Furthermore, by qRT-PCR and Western blot, we found that siRNA/CD44 decreases Bcl-2 and c-Myc synthesis. Our study provides a novel clue that CD44 plays a significant role in the chemoresistance of AML cells to Ara-C and ADM. Moreover, this provides a new direction to the approaches that combination therapy including targeting CD44 may overcome drug resistance and improve treatment effects.
Mechanism underlying drug resistance of HepG2/ADM cells silently reversed by lentivirus-mediated BC047440 gene during chemotherapy

Abstract:Objective To observe the effect of RNA interference(RNAi)-silenced BC047440 gene on multidrug resistance of HepG2/ADM cells and its molecular mechanism.Methods BC047440 protein siRNA interference model was established in HepG2/ADM cells.Expression of BC047440 protein was detected by Western blotting.Experiment was conducted in BC047440-shRNA group,control-shRNA group and HepG2/ADM group.Effect of adriamycin on cell growth was analyzed with CCK-8 method.Cell cycle and apoptosis were assayed by flow cytometry.Expression of NF-κB protein,Survivin mRNA and CCNL1 mRNA was detected by Western blotting and RT-PCR,respectively,after BC047440 was silenced.Results The siRNA interference model was successfully established.The NF-κB protein was expressed about 71.5% in BC047440-shRNA group and HepG2/ADM group,and about 70.0% in control-shRNA group.The cytotoxicity of adriamycin at 24 and 48 h was significantly higher in BC047440-shRNA group.Flow cytometry showed that the number of apoptotic cells was significantly greater in BC047440-shRNA group,the cell cycle was maintained at the G1/G0 phase,and the number of HepG2/ADM cells was reduced at the S phase.RT-PCR showed that the Survivin mRNA was expressed about 37% in control-shRNA group and about 42% in HepG2/ADM group,while the expression of CCNL1 mRNA was about 3.5-fold higher in BC047440-shRNA group than in control-shRNA group and about 2.4-fold higher than in HepG2/ADM group.Conclusion Silencing BC047440 gene can reverse the multidrug resistance of HepG2/ADM cells though the NF-κB,and downstream survivin and CCNL1 signaling pathways,which indicates that BC047440 gene may be one of the important molecules for the multidrug resistance of liver cancer.

Abstract:CD44 is a ubiquitous leukocyte adhesion molecule involved in cell-cell interaction, cell adhesion, migration, homing and differentiation. CD44 can mediate the interaction between leukemic stem cells and the surrounding extracellular matrix, thereby inducing a cascade of signaling pathways to regulate their various behaviors. In this review, we focus on the impact of CD44s/CD44v as biomarkers in leukemia development and discuss the current research and prospects for CD44-related interventions in clinical application.

[Effect of CD44 Gene Silence on Multi-Drug Resistance Reversal and Biologic Activity in K562/A02 Cells].

Abstract:This study was aimed to investigate the effect of CD44 gene silence on the drug resistance and biologic activity of human multidrug resistant leukemia cell line K562/A02. The oligonucleotides of CD44 gene were designed according to related data of GenBank, double-stranded DNA was produced by annealing, and was inserted into pGCsilencerU6/Neo/GFP vector. The resultant recombinant plasmid pGCsiRNA-CD44 was transfected into K562/A02 cell line. Expressions of CD44, mdr-1 and blc-2 mRNA were assayed by real time RT-PCR. The 50% inhibitory concentration (IC50) of doxorubicin (ADM) for K562/A02 cell line was determined by MTT method. Cell cycle was determined by flow cytometry. The morphology of apoptotic cells was examined by Hochst 33258 staining. The results indicated that the siRNA eukaryotic plasmid directing at CD44 gene could effectively silence the CD44 gene of K562/A02 cells; as compared with control group, the CD44 expression in K562/A02 cells transfected with 4 pGCsiRNA-CD44 plasmids was obviously inhibited, while the inhibition of CD44 expression in cells transfected with siCD44-1 was strongest. After being transfected with pGCsiRNA-CD44, the expression of CD44 mRNA in K562/A02 cells reduced by 64.1% (p<0.05), at the same time the expression of mdr-1 and bcl-2 mRNA in pGCsiRNA-CD44-transfected K562/A02 cells reduced by 25.6% and 50.8% respectively. IC50 of K562/A02 cells after transfection decreased to (8.77+/-1.63) microg/ml and was obviously lower than that of control (17.97+/-1.61) microg/ml (p<0.01). After transfection for 48 hours, the ratio of K562/A02 cells in G0/G1 increased by 10.7%, and the cells displayed karyopyknosis, nuclear margination and apoptotic bodies. It is concluded that the siRNA plasmid specifically targeting CD44 gene can remarkably down-regulate the expression of CD44 gene, inhibit K562/A02 cell proliferation, induce its apoptosis and effectively reverse the multidrug resistance of K562/A02 cells.

Abstract:Objective CD44 is an adhesion molecule of cell surface,which is closely related with metastasis and prognosis of breast cancer,but the relationship between CD44 and drug resistance of breast cancer chemotherapy is not clear so far.We mainly study the relationship between CD44 and drug resistance of breast cancer chemotherapy.Methods The expression of CD44 was detected by SP immunohistochemical methods,the different expression of CD44 was investigated between drug-sensitive groups and drug-resistant groups of neoadjuvant chemotherapy,before and after chemotherapy.The different expressions of CD44 on the level of mRNA and protein in MCF-7 cell line and drug-resistant cell lines MCF-7/ADR was tested by reverse transcription polymerase chain reaction(RT-PCR) and flow cytometry.Results Expression of CD44 before chemotherapy in drug-resistant groups was much higher than drug-sensitive ones(χ2=4.22,P <0.05).This was also found after chemotherapy(χ2=9.954,P<0.01).The expression of CD44 after chemotherapy was higher than that before chem otherapy(t=3.43,P<0.01).Expression of mRNA and protein of CD44 in MCF-7/ADR cell lines was higher than drug-sensitive cell lines(MCF-7).Conclusion CD44 may be conferred as a predictive index of breast cancer chemotherapy drug resistance,and chemotherapy may enrich CD44 positive breast cancer cell.
Enhancement of Sensitivity of Multidrug-Resistant Leukemia K562/ADM Cell Line and Its Leukemia Stem Cells to Adriamycin by Interferon Alpha

Abstract:Aim To investigate the effect of interferon alpha(IFN-α) on the sensitivity of multidrug resistant leukemia K562/ADM cell population and its leukemia stem cells(LSC) to the cell-cycle specific agent adriamycin.Methods The drug-resistant leukemia K562/ADM cells were used as the target cells.The cell proliferating activity was assessed with an MTT colorimetric assay.Flow Cytometry(FCM) was employed to determine the distribution of cell cycle,and the cellular apoptosis was assayed with FITC-Annexin V and propidium iodide(PI) double staining.The proportion and apoptosis of LSC in K562/ADM cell population were determined with co-staining of anti-CD34,anti-C38,anti-CD123 antibodies and Annexin V.Results After the treatment with IFN-α,the K562/ADM cells exhibited obvious changes in the cell cycle distribution,the cells in G0/G1 phase decreased and those in S phase increased.Combination of IFN-α and adriamycin significantly augmented the proliferation-inhibiting and apoptotic effects of adriamycin to K562/ADM cells,and the apoptotic rate of the population cells signally increased.Furthermore,IFN-α could increase the proportion of LSC in K562/ADM cell population and improve the sensitivity of LSC to adriamycin,and the amount of apoptotic LSC(Annexin V+) significantly increased.Conclusion IFN-α possesses the potential to improve the sensitivity of drug-resistant K562/ADM cell population and its LSC to the cell-cycle specific agent adriamycin by driving the cells to enter the cell division cycle.
DEVELOPMENT OF A K562 MULTIDRUG-RESISTANT CELL LINE AND STUDY ON PROTEINS WITH ALTERED EXPRESSION

Abstract:In an attempt to study the whole protein expression alterations of tumer cells after becoming multidrug-resistant, which may provide useful information on new drug target identification, an adriamycin-resistant variant of the human leukemia cell line K562 (K562/ADR) was developed in vitro by continuous exposure to adrimycin. MTT assay was used to determine IC50 of K562/ADR cells to adriamycin (ADR), cisplatin (DDP), 5-fluorouracil (5-FU) and vincristin (VCR). The total proteins of K562 and K562/ADR were separated by two-dimensional gel electrophoresis and visualized by silver staining. Proteins with significant expression alterations were selected and their peptide mass fingerprints (PMFs) were obtained by matrix-assisted laser desorption/ionization time of flying mass spectrometry (MALDI-TOF-MS). The PMFs were used to search NCBInr database by AutoMS-Fit software. The results showed that K562/ADR cell demonstrated cross-resistance to other antineoplastic drugs. The IC50 of K562/ADR cells to ADR, DDP, 5-FU, VDR were much higher than those of K562. The proteins differentially expressed in the two cell lines were identified as cell cycle-related proteins, zinc finger protein 165, etc. These proteins are involved in cell cycling and transcription regulation, whose expression alterations may contribute to the multidrug resistant phenotype of K562/ADR cells.

Abstract:Background: Cell adhesion molecule cluster of differentiation 44 (CD44) plays a significant role in cancer cell local invasion, intravasation, migration, and the establishment of metastatic lesions. However, little is known about the underlying mechanism of how CD44 regulates hepatocellular carcinoma (HCC) extrahepatic metastasis (EHM). Methods: The expression of CD44 in HCC tissues and cell lines was detected through western blot and immunohistochemistry (IHC). Through gain- and loss-of-function assays, we examined the oncogenic roles of CD44 in regulating HCC cell growth and metastasis in vitro and in vivo. To identify the potential mechanism, we employed quantitative real-time polymerase chain reaction, and western blot. Results: In this study, CD44 was highly expressed in HCC cells and HCC-patient specimens that exhibited high malignancy potential. The overall survival (OS) was worse and the cumulative recurrence rate was higher in HCC patients with CD44 overexpression than those with low levels of CD44 expression. Our in-vitro and in-vivo experiments showed that CD44 downregulation reduced HCC cell colony formation, migration, and invasion, and I ICC tumor growth and metastasis, and that the pro-metastasis effect of CD44 was mediated by the protein kinase B (AKT)/extracellular signal-regulated kinase (ERK) signaling-chemokine receptor C-X-C chemokine receptor type 4 (CXCR4) axis. The reported capacity of CD44 to induce CXCR4 expression and increase the propensity of tumors to invade and metastasize to distant organs is consistent with the aggressive clinical characteristics of HCCs. Conclusions: CD44 could represent a future therapeutic target for EHM.

Abstract:This study aimed to explore the mechanism of adriamycin resistance in human chronic myelogenous leukemia cells. Proteomic approach was utilized to compare and identify differentially expressed proteins between human chronic myelogenous leukemia K562 cells and their adriamycin-resistant counterparts. The differentially expressed proteins were analyzed by 2-DE (two-dimensional gel electrophoresis), and protein identification were performed on ESI-Q-TOF MS/MS instrument. Out of the 35 differentially expressed proteins between the two cell lines, 29 were identified and grouped into 10 functional classes. Most of identified proteins were related to the categories of metabolism (24%), proteolysis (13%), signal transduction (21%) and calcium ion binding (6%), suggesting that alterations of those biological processes might be involved in adriamycin resistance of K562 cells. We believe this study may provide some clues to a better understanding of the molecular mechanisms underlying adriamycin resistance.
Effects of ADM on Cell Apoptosis and Expression of FAK Mrna in K562

Abstract:Objective To investigate the effects of ADM on proliferation,cell cycle progression,apoptosis and the expression of focal adhesion kinase(FAK) mRNA in human chronic myeloid leukemia (CML) cell line K562, and further to explore the mechanism of regulation of FAK. Methods The CCK8 method was used to detect the effects of ADM on the proliferation of K562 cells; flow cytometry was used to determine the effects of ADM on the cell cycle progression and apoptosis of K562 cells. RT-PCR and Western blot was used to assay the expression of FAK mRNA and FAK protein in K562 cells treated with ADM for 36 hours. Results ADM exhibit a dose- and time-dependent inhibition of K562 proliferation, and the effect was significant (P0.05). ADM could also significantly induce apoptosis of K562 cells (P0.05). ADM arrested K562 cell at S phase. ADM could decrease the expression levels of FAK mRNA and FAK proteins in K562 cells. Conclusions ADM is effective to reduce the expression of FAK mRNA and FAK protein, and to induce cell cycle arrest and apoptosis in K526 cells. The results may provide the research basis for further study of the role of FAK in tumour cell apoptosis.
A novel in-vitro model of the bone marrow microenvironment in AML identifies CD44 and Focal Adhesion Kinase as therapeutic targets to reverse cell adhesion-mediated drug resistance

Abstract:Acute myeloid leukemia (AML) is an aggressive neoplasm. Although most patients respond to induction therapy, they commonly relapse due to recurrent disease in the bone marrow microenvironment (BMME). So, disruption of the BMME, releasing tumour cells into the peripheral circulation, has therapeutic potential. Using both primary donor AML cells and cell lines, we developed an in-vitro co-culture model of the AML BMME. We used this model to identify the most effective agent(s) to block AML cell adherence and reverse adhesion-mediated treatment resistance. We identified anti-CD44 treatment significantly increased the efficacy of cytarabine. However, some AML cells remained adhered, and transcriptional analysis identified focal adhesion kinase (FAK) signalling as a contributing factor; adhered cells showed elevated FAK phosphorylation that was reduced by the FAK inhibitor, defactinib. Importantly, we demonstrated that anti-CD44 and defactinib were highly synergistic at diminishing adhesion of the most primitive CD34high AML cells in primary autologous co-cultures. Taken together, we identified anti-CD44 and defactinib as a promising therapeutic combination to release AML cells from the chemoprotective AML BMME. As anti-CD44 is already available as a recombinant humanised monoclonal antibody, the combination of this agent with defactinib could be rapidly tested in AML clinical trials.

Abstract:Acute myeloid leukemia (AML) has a poor prognosis under the current standard of care. In recent years, venetoclax, a BCL-2 inhibitor, was approved to treat patients who are ineligible for intensive induction chemotherapy. However, complete remission rates with venetoclax-based therapies are hampered by minimal residual disease (MRD) in a proportion of patients, leading to relapse. MRD is a result of leukemic stem cells being retained in bone marrow protective environments; activation of the CXCL12-CXCR4 pathway was shown to be relevant to this process. An important role is also played by cell adhesion molecules such as CD44, which has been shown to be crucial for the development of AML. Here we show that CD44 is involved in CXCL12 promotion of resistance to venetoclax-induced apoptosis in human AML cell lines and AML patient samples, which could be abrogated by CD44 knock down, knockout, or blocking with an anti-CD44 antibody. Split-Venus bimolecular fluorescence complementation showed that CD44 and CXCR4 physically associate at the cell membrane upon CXCL12 induction. In the venetoclax-resistant OCI-AML3 cell line, CXCL12 promoted an increase in the proportion of cells expressing high levels of embryonic stem cell core transcription factors (ESC-TFs: Sox2, Oct4, Nanog) abrogated by CD44 knockdown. This ESC-TF-expressing subpopulation which could be selected by venetoclax treatment, exhibited a basally enhanced resistance to apoptosis and expressed higher levels of CD44. Finally, we developed a novel AML xenograft model in zebrafish, which showed that CD44 knockout sensitizes OCI-AML3 cells to venetoclax treatment in vivo. Our study shows that CD44 is a potential molecular target for sensitizing AML cells to venetoclax-based therapies.
Expression of Caspase-3 and Bcl-2 in leukaemia cell apoptosis and its significance in multiple-drug resistance

Abstract:Objective: To study the regulative role of Caspase-3 on the Bcl-2 protein expression in the process of K562 and K562/AO2 cell apoptosis induced by adriamycin, in order to probe into the roles of Caspase-3 and Bcl-2 in the molecular mechanism of the leukemia multiple-drug resistance. Methods: The terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling method was used to observe the morphological change of the apoptotic cells. Flow cytometry and the enzyme colorimetric activity assay methods were used to examine the rate of cell apoptosis, the Bcl-2 protein expression level and the Caspase-3 activity. Results: 1 The cell apoptosis rate had a time- and dosage-dependent relation with adriamycin in the K562 cell strain. The K562/AO2 cell strain did not have such characteristic. 2 The Caspase-3 activity and the Bcl-2 protein expression level of K562 cell strain without the Caspase-3 inhibitor had a time- and dosage-dependent relation with adriamycin. There was no such relation when the inhibitor is added, but after being affected for 48 hours, the Bcl-2 protein expression level decreased and the activity of Caspase-3 increased. However, the Caspase-3 activity and the Bcl-2 protein expression level of the K562/AO2 cell strain affected by ADR with or without the Caspase-3 inhibitor showed no changes. Conclusion: The change of Caspase-3 can cause the change of the Bcl-2 protein expression level, and they are closely related to apoptosis and have an important effect on multi-drug resistance.
Downregulation of microRNA let-7f mediated the Adriamycin resistance in leukemia cell line

Abstract:Multidrug resistance (MDR) has become the major cause of failure chemotherapy for leukemia and high mortality of leukemia. The study aimed to investigate whether the let-7f mediate the Adriamycin (ADR) resistance of leukemia, and to explore the potential molecular mechanism. Cell proliferation was examined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and the soft agar clone formation assay. Flow cytometry was performed to detected cell cycle and apoptosis. The targeted regulationship was analyzed by dual-luciferase assay. Real-time polymerase chain reaction and Western blot were used to measure the expressions of let-7f, ABCC5, ABCC10, cell cycle-related proteins, and apoptosis-related proteins. The xenograft mouse model was used to conduct the tumor formation assay in vivo. The results demonstrated that the expression of let-7f was lower in multidrug-resistant K562/A02 cell lines compared to that in K562, while ABCC5 and ABCC10 were upregulated. Overexpression of let-7f in K562/A02 cell lines downregulated the ABCC5 and ABCC10 expression, enhanced cell sensitivity to ADR, promoted cell apoptosis, and inhibited cell proliferation. let-7f was proved to negatively regulate ABCC5 and ABCC10. Tumor formation assay further determined that let-7f overexpression increased sensitivity to ADR. Taken together, the let-7f downregulation induced the ADR resistance of leukemia by upregulating ABCC5 and ABCC10 expression. Our study provided a novel perspective to study the mechanism of MDR and a new target for the reversal of MDR.

CD44 inhibition attenuates EGFR signaling and enhances cisplatin sensitivity in human EGFR wild‑type non‑small‑cell lung cancer cells

Abstract:Cluster of differentiation 44 (CD44) as a transmembrane glycoprotein is found to be expressed in non‑small cell lung cancer (NSCLC), is significantly associated with NSLC progression, metastasis and drug resistance. This study aimed to explore whether CD44 inhibition improves the sensitivity of epidermal growth factor receptor (EGFR) wild‑type NSCLC cells to cisplatin and how it affects wild‑type EGFR in NSCLC cells. Small interfering RNA was used to knockdown CD44 expression in EGFR wild‑type NSCLC cell line H460. Results suggested that CD44 downregulation reduced cell growth, promoted G0/G1 cell cycle arrest and induced cell apoptosis in H460 cells and these effects were evidently enhanced when in combination with cisplatin. Deactivation of EGFR signaling pathway including EGFR phosphorylation and its downstream molecules, targets ERK, AKT1 and SRC which were also observed in CD44‑silenced H460 cells with or without EGF stimulation. Furthermore, the CD44 expression level was positively correlated with wild‑type EGFR level in human lung adenocarcinoma tissues and CD44 inhibition significantly accelerated the degradation of EGFR, indicating that enhanced sensitivity of H460 cells to cisplatin by downregulation of CD44 might be due to EGFR degradation. This study demonstrated that suppression of CD44 deactivated EGFR signals in NSCLC cells with wild‑type EGFR, thereby contributing to the inhibition of cell proliferation and the reinforcement of cisplatin sensitivity. It is suggested that downregulation of CD44 could be a novel potential therapeutic strategy for the treatment of EGFR wild‑type NSCLC.
Effects of Anti CD_(44) Antibody on the Cytotoxicity of Leukemic Cells Mediated by Activated T Lymphocyte

Abstract:Objective To study the effect of anti CD 44 antibody alone or combined anti CD 44 with anti CD 2 antibodies on the normal T lymphocyte proliferation and on the cytotoxicity of acute myelogenous leukemic cell mediated by T lymphocyte. Methods The authors pretreated the normal T lymphocyte activated by IL 2 with the anti CD 44 antibody alone, or combined with anti CD 2 antibodies with different concentrations DNA synthesis and the cytotoxicity of leukemic cell line MML 1 mediated by T lymphocyte were respectively detected with [ 3H] TdR incorporation method and double staining flow cytometry. Results Anti CD 44 antibody alone could not induce the proliferation of T lymphocyte activated by IL 2, and showed no influence on the DNA synthesis of T lymphocyte activated by IL 2. However, the combination of anti CD 44 with anti CD 2 antibodies could significantly increase the inhibitory effect of anti CD 2 antibody on the IL 2 activated T lymphocyte proliferation and the DNA synthesis. Anti CD 44 or anti CD 2 antibody alone could inhibit the cytotoxicity of leukemic cell line MML 1 mediated by the IL 2 activated T lymphocyte, and then the PKH2+PI double staining leukemic cells decreased. The combinatory effect of anti CD 44 with anti CD 2 antibodies obviously increased the inhibitory on the cytotoxicity of the activated T lymphocyte. Conclusion The inhibit effect of T lymphocyte proliferation induced by the negative signal transduction resulted from the CD 44 molecule cross ligated with CD 44 antibody might associate with the CD 2 molecule cross ligation. The inhibitory effect of anti CD 44 and anti CD 2 antibody on the cytotoxicity of leukemic cell line MML 1 mediated by the IL 2 activated T lymphocyte might be due to the partial block of the expressions of CD 44 and CD 2 molecules, which might interfere with the Fas FasL ligation.
Cd44/Cellular Prion Protein Interact in Multidrug Resistant Breast Cancer Cells and Correlate with Responses to Neoadjuvant Chemotherapy in Breast Cancer Patients

Abstract:Multidrug resistance (MDR) is one of the most important factors leading to chemotherapeutic failure in patients with breast cancer. The invasive/metastatic ability of MDR cells is strengthened compared with their parental cells. However, the mechanisms underlying MDR have not been fully elucidated. We found that CD44 and the cellular prion protein (PrPc) were both overexpressed in MDR cells (MCF7/Adr and H69AR). Subsequently, we chose the human breast cancer cell line MCF7/Adr, which is resistant to adriamycin, for further research. We discovered that PrPc physically and functionally interacted with CD44. The knockdown of CD44 or PrPc by siRNA in MCF7/Adr cells inhibited cell migration, invasion and proliferation in vitro. However, when the MCF7/Adr cells transfected with CD44 siRNA were incubated with 10 times the peak plasma concentration (PPC) of taxol, their invasive ability was again enhanced. In the breast-carcinoma tissue samples, a significant correlation between the CD44 expression and the PrPc expression was observed in the postneoadjuvant-chemotherapy (NAC) cases. Moreover, in Group 2, which was unresponsive to NAC, the CD44 and PrPc expression levels were significantly increased in the post-NAC cases compared with the pre-NAC cases using the paired-samples t-test. These data indicate that the CD44/PrPc interaction enhances the malignancy of breast cancer cells and affects the responses to neoadjuvant chemotherapy in breast cancer patients. Therefore, blocking the CD44/PrPc interaction may improve outcomes in chemorefractory breast cancer patients.

Abstract:CD44, a non-kinase transmembrane glycoprotein, is overexpressed in several cell types including cancer stem cells and frequently shows alternative spliced variants that are thought to play a role in cancer development and progression. Hyaluronan, the main ligand for CD44, binds to and activates CD44 resulting in activation of cell signaling pathways that induces cell proliferation, increases cell survival, modulates cytoskeletal changes, and enhances cellular motility. The different functional roles of CD44 standard (CD44s) and specific CD44 variant (CD44v) isoforms are not fully understood. CD44v contain additional peptide motifs that can interact with and sequester growth factors and cytokines at the cell surface thereby functioning as coreceptors to facilitate cell signaling. Moreover, CD44v were expressed in metastasized tumors, whereas switching between CD44v and CD44s may play a role in regulating epithelial to mesenchymal transition (EMT) and in the adaptive plasticity of cancer cells. Here, we review current data on the structural and functional properties of CD44, the known roles for CD44 in tumorigencity, the regulation of CD44 expression, and the potential for targeting CD44 for cancer therapy.

Abstract:Chemotherapy remains the standard treatment for acute myeloid leukemia; however, the emergence of drug resistance is a major hurdle in the successful treatment of leukemia. The expression of multidrug resistance-associated protein 4 (MRP4) induces resistance in the adriamycin-resistant acute myeloid leukemia cell line, K562/ADR. The aim of this study was to investigate whether knockdown of MRP4 by lentivirus-mediated siRNA could improve the sensitivity of K562/ADR cells to adriamycin. Five lentivirus-mediated short hairpin RNAs (lv-shRNAs-MRP4) were designed to trigger the gene silencing RNA interference (RNAi) pathway. The efficiency of lentivirus-mediated siRNA infection into K562/ADR cells was determined using fluorescence microscopy to observe lentivirus-mediated GFP expression. MRP4 expression in infected K562/ADR cells was evaluated by real-time PCR and Western blot analysis. The MTS assay was used to measure cell viability and flow cytometry was used to measure apoptosis. The transfection efficiency of K562/ADR cells was over 80 percent. The gene silencing efficacy of lv-shRNA1-MRP4 was superior to the other constructs. Infection of K562/ADR cells with lv-shRNA1-MRP4 led to strong inhibition of MRP4 mRNA and protein expression. Combined treatment with lv-shRNA1-MRP4 and adriamycin decreased cell growth and increased apoptosis compared to treatment with lv-shRNA1-MRP4 or adriamycin alone. These data indicate that in K562/ADR cells MRP4 is involved in drug resistance mechanisms and that lentivirus-mediated knockdown of MRP4 may enhance sensitivity to adriamycin.
