Abstract:Cysteine (Cys) takes part in redox balance in cells as an antioxidant, and imbalance of Cys content in the body can cause a variety of diseases. It is very important to develop a new fluorescent chemosensor to specifically detect Cys intracellular. In this work, a novel NIR fluorescent probe was constructed based on 3-ethyl-1,1,2-trimethyl-1H-benzo[e]indol-3-ium iodide and 5-(4-hydroxyphenyl)furan-2-carbaldehyde. The probe could selectively detect Cys in the presence of homocysteine (Hcy), glutathione (GSH), and other interferences. It also had a number of advantages, including nucleolus-targeting ability, long fluorescence emission wavelength (685 nm), low detection limit (56 nM), and large Stokes shift (172 nm). The probe was employed to enable visualization of Cys in HepG2 cells, and due to its good response in viscous environment, the probe could also locate nucleoli intracellular.

Abstract:Cysteine (Cys) plays key roles in various pathological and physiological processes. Thus, the specific detection of Cys is a great necessity for the related disease diagnosis. Herein, we present a novel fluorescent probe based on a 2-benzothiazolyl-6-quinazolinyl-4-methylphenol moiety as fluorophore and a 2,4-dinitrobenzenesulfonyl moiety as detection functioned group. The probe presented a rapid and specific turn on response to Cys under physiological conditions, which could be convenient observed by the naked eyes. The detection mechanism was elucidated by HRMS, UV/vis absorption, fluorescence and DFT calculation. Furthermore, the probe was applied to monitor the endogenous Cys in living cells and mice.

Abstract:A benzothiazole-based near-infrared (NIR) ratiometric fluorescent probe (HBT-Cys) was developed for discriminating cysteine (Cys) from homocysteine (Hcy) and glutathione (GSH). The probe was designed by masking phenol group in the conjugated benzothiazole derivative with methacrylate group that could be selectively removed by Cys, and therefore an intramolecular charge transfer (ICT) fluorescence was switched on in the NIR region. In the absence of Cys, the probe exhibited a strong blue fluorescence emission at 431 nm, whereas a NIR fluorescence emission at 710 nm was significantly enhanced accompanied by a decrease of emission at 431 nm in the presence of Cys, allowing a ratiometric fluorescence detection of Cys. The fluorescence intensity ratio (I710nm/I431nm) showed a good linear relationship with Cys concentration of 1-40 μM with the detection limit of 0.5 μM. The sensing mechanism was explored based on MS experimental analysis and DFT theoretical calculation. Moreover, the fluorescent probe was successfully used for fluorescence bioimaging of Cys in living cells.

Abstract:Cysteine (Cys), as an important biothiol, plays a major role in many physiological processes like protein synthesis, detoxification and metabolism, and also is closely associated with a variety of diseases; thus the design of novel highly selective and sensitive near-infrared (NIR) fluorescent probes for Cys detection in vivo is of great significance. Herein, we report a selective and sensitive NIR turn-on fluorescent probe (CP-NIR) with large Stokes shift for detecting Cys in vivo. Upon addition of Cys to the solution of the probe, it is absorption wavelength shifts from 550 to 600 nm, accompanying with an obvious enhancement of NIR fluorescence emission centering around 760 nm. This Michael-addition reaction based probe shows a large Stokes shift (160 nm), low detection limit (48 nM), fast response time, and low toxicity. Moreover, this novel NIR probe with good cell permeability was successfully applied to monitoring endogenous Cys in living cells and in a mouse model.

Abstract:In this work, A new near-infrared (NIR) fluorescent probe NIR-L based on dicyanoisophorone was rationally designed, synthesized and characterized for the detection of cysteine (Cys). This probe exhibited highly selective and sensitive response to Cys over other analytes including amino acids, cations and anions under physiological conditions, which can be identified by naked eyes. It was found that the probe showed NIR fluorescence turn-on response at 714 nm toward Cys with a large Stokes shift of 174 nm and a very low detection limit of 28.6 nM. The results of 1H NMR and DFT calculation further confirmed that the addition of Cys triggered a reaction between Cys and NIR-L, leading to the release of a NIR fluorescent dye NIR-OH. Moreover, NIR-L was successfully used to realize the in vivo Cys bioimaging in HeLa cells and mice. Significantly, the sensing system was applied in imaging of Cys in B16-F10 tumor-bearing mice.

Abstract:The near-infrared (NIR) fluorescence sensor for rapid, selective, and sensitive detection of cystenine (Cys) is of great importance in both biological and environmental sciences. Herein, we report a specific probe with turn-on fluorescence property, visible color change with naked-eye, and large wavelength shift on UV spectra for highly selective detection of Cys over homocysteine (Hcy) and glutathione (GSH) in both HEPES buffer (10 mM, pH 7.4) and diluted human serum. The probe based on the conjugate addition-cyclization reaction has a low limit of detection to Cys (0.16 mu M as NIR fluorescence sensor and 0.13 mu M as UV sensor). Kinetic study indicated that the probe has a very rapid response to Cys, owing to the much higher pseudo-first-order reaction constant with Cys (299 M-1 s(-1)) than with Hcy (1.29 M-1 s(-1)) or GSH (0.53 M-1 s(-1)). Upon addition of Cys to a solution of the probe, the color changed from purple to cyan, with the maximum wavelength shifting from 582 to 674 nm in the UV spectrum and a fluorescence emission at 697 nm appearing. It has been successfully applied for determination of Cys in diluted serum and bioimaging of Cys in living cells with low cell toxicity.

Abstract:Cysteine (Cys) plays a crucial role in numerous physiological and pathological processes. Therefore, it is imperative to design a highly selective and sensitive near-infrared (NIR) fluorescent probe to monitor Cys. In this study, we have developed a novel NIR fluorescent probe XA based on Xanthene hybrid tetrahydro-acridine salt dye for specifically tracking of Cys, where a chlorine-substituted tetrahydro-acridine acts as a high Cys-reactive site and water-soluble group. Probe XA exhibits a remarkable "turn-on" NIR emission (830 nm) with an extra-large Stokes shift (305 nm) for monitoring Cys. It also has a high selectivity, rapid response time (6 min) and high sensitivity (LOD as 0.5 μM). We fully characterized and discussed the sensing mechanism of XA toward Cys using HPLC and MS spectrums, as well as quantum theory calculations. Furthermore, the excellent properties of NIR fluorescent detection allow this novel probe to successfully monitor fluctuations of exogenous and endogenous Cys concentration levels in living cells and in vivo.

Abstract:We designed and synthesized a water-soluble near-infrared (NIR) fluorescent probe with the recognition unit of the cyanine-like structure and acrylate group. Through an aromatic ring nucleophilic substitution reaction based on sulfhydryl moiety, an off-on fluorescence response toward cysteine (Cys) was realized. The probe exhibited excellent spectral performance with an emission wavelength of 720nm and a detection limit of 0.20μM. The spectral properties, selectivity and anti-interference performance of the probe were systematically investigated. Density functional theory (DFT) calculations were conducted to clarify the luminescence mechanism of the probe. Furthermore, the probe was successfully applied to the detection of free Cys in human serum and the NIR imaging of endogenous Cys in living cells. Thus, the probe has a promising application prospect in clinical diagnosis and fluorescence imaging.

Abstract:In this work, tetrahydro-acridine salt hybrid coumarin dyes was designed and developed as a Near-infrared (NIR) fluorescent probe CM for the detection and fluorescent bioimaging of Cys/Hcy. The activated chlorine atom in probe facilitates more efficient nucleophilic substitution reaction, resulting in fast responsive time (3 min toward 3 eq. Cys) and high sensitivity toward Cys with detection limit of 46 nM. More importantly, probe displays "Turn -on" near-infrared fluorescence emission (Emmax: 674 nm) with Large Strokes shift (about 200 nm) toward Cys/ Hcy. Furthermore, probe could be used as efficient tool for imaging and detection of Cys/Hcy in mitochondria of living cells and in vivo. This work innovatively proposed a new design strategy for NIR fluorescent probe platform for the detection and fluorescence imaging of bio-thiols.

Abstract:Cysteine (Cys) distribute widely in organisms as the crucial components of proteins, and play important roles in pathophysiological processes of human body. Low level of Cys might induce hepatic injury, edema and growth retardation, while superfluous level of Cys is found to be closely relevant to Alzheimer's and Parkinson's diseases. In this work, a novel near-infrared (NIR) fluorescent probe PFQ-C was developed for highly selective detection of Cys in living cells and mice by utilizing the cyclization removal reaction between acrylate group and Cys. The superior sensitivity (limit of detection, 0.036 μM), NIR emission (655 nm), large Stokes shift (135 nm) and low cytotoxicity of the probe highlight its broad potential for future clinical applications. The response mechanism of the probe towards Cys was clarified by spectroscopy, chromatography and theoretical calculation. In addition, results of fluorescence imaging of cells and mice revealed the good performance of the probe for monitoring the distributions and variations of Cys activity in vivo, which is very useful for the researches on diseases associated with Cys.

Abstract:Hemicyanine dyes are an ideal structure for building near-infrared fluorescent probes due to their excellent emission wavelength properties and biocompatibility in biological imaging field. Developing a near-infrared fluorescent probe capable of detecting cysteine (Cys) was the aim of this study. A novel developed fluorescent probe P showed high selectivity and sensitivity to Cys in the presence of various analytes. The detection limit of P was found to be 0.329 μM. The MTT assay showed that the probe was essentially non-cytotoxic. Furthermore, the probe was successfully used as cysteine imaging in living cells and mice.

Abstract:We designed and synthesized a simple and readily available fluorescent probe 3 for cysteine (Cys) based on naphthalene derivative. The probe is composed of a new class of aggregation-induced emission enhancement (AIEE) active dye 2 based on excited-state intramolecular proton transfer (ESIPT) and an acrylate group as the Cys recognition unit as well as the ESIPT blocking agent, which can be cut off by Cys from the probe in aqueous solution with mild conditions. The probe had great sensitivity and selectivity for the detection of Cys over homocysteine (Hcy) and glutathione (GSH) with a detection limit of 0.05 mu M. Abstract:A novel near-infrared fluorescent probe SWJT-5 based on dicyanoisophorone was synthesized. It achieved the rapid (within 40 s) and discriminative detection of Cys over Hcy and GSH with a large Stokes shift (205 nm). It showed high selectivity and sensitivity for Cys, and had an obvious enhancement of fluorescence emission. The detection limit was 0.43 μM. This probe also had low background interference and little damage to biological samples. Therefore, SWJT-5 had been applied to bioimaging in living cells successfully.

Abstract:Cysteine (Cys) plays a pivotal role in numerous physiological processes and holds significant importance in food analysis. In this study, we designed a novel near-infrared (NIR) fluorescent probe TF for specifically detecting Cys, derived from benzo[e]indole-conjugated tetrahydro-acridine salt. Leveraging the electron-withdrawing properties of the 3,5-bis(trifluoromethyl)benzenethiol group, the probe exhibits a distinctive colorimetric response and a notable enhancement in NIR emission, featuring a substantial Stokes shift of 135 nm, facilitating precise Cys detection. The detection mechanism was elucidated through comprehensive analyses including NMR, MS spectral, and quantum theory calculations. Moreover, the probe demonstrates exceptional attributes such as rapid response (8 min), exceptional selectivity, and sensitivity (with a detection limit as low as 74 nM). The probe's NIR fluorescent response, coupled with its large Stokes shift, enables accurate quantification of Cys in real food samples and facilitates imaging for monitoring both exogenous and endogenous Cys levels in living cells.

Abstract:Ratiometric near-infrared (NIR) fluorescent probes are attractive tools for bioimaging applications. Although many fluorescent probes have been developed for detection of the biologically important cysteine (Cys), ratiometric NIR fluorescent probes for Cys are very rare, particularly those with significant fluorescence enhancement in the NIR region. In this work, a new ratiometric NIR fluorescent probe with significant NIR fluorescent turn-on changes was reported for rapid, colorimetric and ratiometric fluorescent detection of Cys. This probe is able to detect Cys in aqueous solution under mild conditions, with high selectivity and sensitivity for Cys over various analytes including the similar structured homocysteine (Hcy) and glutathione (GSH). The detection limit of this probe for Cys was found to be as low as 0.2 μM, which is far below the normal content of Cys (30–200 μM) in cells. Besides the excellent sensing properties, this probe shows low cytotoxicity and can be conveniently applied to image intracellular Cys in living cells, indicating it is a promising tool for biological applications.

Abstract:A novel red-emitting fluorescent probe (DDNA) for cysteine has been rationally designed and synthesized, which exhibited a low limit of detection to Cys (0.26 mu M) as well as a favorable large stokes shift (lambda(Em)-lambda(Ex) = 128 nm). This novel fluorophore (HDM), which features a large at-conjugation system and typical intramolecular charge transfer (ICT) process, has a long emission wavelength at 631 nm. Besides that, as a turn-on fluorescent probe, it shows high selectivity and sensitivity for Cys over other metal ions and amino acids including the similar structured homocysteine (Hcy) and glutathione (GSH). Finally, the probe DDNA was successfully applied to bioimage intracellular Cys in Hela cells with low cytotoxicity.

Abstract:Two-photon excitable (850 nm) NIR fluorescent turn-on (702 nm) probe TP-NIR was synthesized for selective detection of cysteine (Cys). The probe itself shows is turned on by reaction with Cys in aqueous buffer. In addition, the probe displays greater selectivity for Cys over other thiols, including glutathione (GSH) and homocysteine (Hcy). Moreover, the large Stokes shift, NIR excitation, and NIR emission make this probe suitable for biological imaging.

Abstract:We establish a near-infrared (NIR) ratiometric fluorescent probe Cy-NB for the selective detection of cysteine (Cys) over glutathione (GSH) and homocysteine (Hcy) in mitochondria to indicate oxidative stress. Heptamethine cyanine dye is chosen as the fluorophore of Cy-NB whose emission locates in NIR region. And p-nitrobenzoyl is employed as the fluorescent modulator due to its capability of selective-Cys response. Once triggered by Cys, the uncaged p-nitrobenzoyl rearranges the polymethine π-electron system of the fluorophore, which leads to a remarkable spectrum shifts in absorption and emission profiles. Taking advantage of these spectroscopic properties, we construct a ratiometric fluorescent signal for the detection of Cys with a detection limit of 0.2 µM within 5 min. Our probe Cy-NB can sensitively detect the mitochondrial Cys pool changes under different oxidative stress status in HepG2 cells. We also successfully employ Cy-NB to imaging Cys level changes in living mice. It suggests that mitochondrial Cys can be used as an oxidative stress biomarker with simple potential clinical applications. And our probe Cy-NB is of great potential for further utilizing in exploring the physiological function of Cys in biological systems.

Abstract:Cysteine (Cys) is a crucial biothiol that acts a significant function in food samples and biological systems, including plant roots and living cells. Hence, we developed a novel colorimetric and near-infrared ratiometric fluorescent probe (CT), composed of coumarin and tetrahydroacridine-conjugated indole salt, for the detection of Cys. Upon reaction with Cys, the probe undergoes a specific N-substitution reaction, resulting in a notable colorimetric change and a significant ratiometric fluorescent response in both visible and near-infrared emission channels. These dual-channel ratiometric fluorescence changes are completely independent, enabling the probe to obtain great selectivity, sensitivity, and exceptional detection accuracy. Leveraging these attributes, the probe was employed to provide accurate quantitative analysis of Cys in food samples. Furthermore, confocal imaging demonstrated that the probe could monitor both exogenous and endogenous Cys levels in living cells and track Cys changes in plant roots under heavy metal stress. This work presents a dependable and accurate imaging solution for tracking and identifying Cys of real food, plants, and living cells.
