Abstract:A contrast-enhanced ultrasound-modulated laser feedback imaging system is proposed. Compared with traditional ultrasound modulated optical tomography (UOT), the system exhibits high sensitivity and contrast-to-noise (CNR). The usage of the laser feedback technique decreases the photon consumption in imaging. On the other hand, clinical SonoVue microbubbles are used to enhance the attenuation of light in the ultrasound labeled area inside tissues. In experiments, vessels in phantom are imaged and can be distinguished clearly at a penetration depth of 5.5 cm in reflective mode. Using microbubbles, the contrast-to-noise is increased from 0.78 to 3.73 with the lateral imaging resolution of 0.75 mm. Phantoms of different penetration depths with microbubbles in different concentrations were imaged. The experiment results show that with higher microbubbles concentration and thinner depth comes higher CNR. Although a lot remains to be improved, the proposed method is promising for further development toward practical clinical application.
Mechanism of Contrast-Enhancement in Ultrasound-Modulated Laser Feedback Imaging with Ultrasonic Microbubble Contrast Agent

Abstract:Ultrasound-modulated optical imaging technology is a new type of biological tissue optical detection technology, and sensitive to the change of scattering coefficient and absorption coefficient of biological tissue. This technology is a non-ionizing and non-invasive pathological detection method, which has great potential application in early detection of cancer. However, ultrasound-modulated optical imaging technology is insufficient in signal-to-noise ratio (SNR) and imaging contrast. Frequency-shifted laser feedback technology with microchip laser is a new type of highly sensitive interference technology, whose gain coefficient for weak optical signal can reach 10(6). This technology can greatly improve the SNR of imaging. Combined with the high sensitive laser feedback technology, the ultrasound-modulated laser feedback technology is proposed. The SNR of this technology is better than that of the traditional ultrasound-modulated optical imaging technology. The increase in SNR can achieve greater depth of detection in biological tissue imaging, but there is no significant improvement in imaging contrast. In order to improve the contrast of biological tissue imaging and achieve high resolution imaging of thick biological tissue, we use ultrasound microbubble contrast agent in ultrasound-modulated laser feedback imaging technology. We establish a Monte Carlo photon transport model with microbubbles in order to study the mechanism of contrast enhancement in ultrasound-modulated laser feedback imaging with microbubbles. Finally, we establish an experimental system to verify the correctness of the simulation results. Experimental and simulation results show that in the transparent solution, the ultrasonic microbubble contrast agent can enhance the ultrasound-modulated laser feedback signal and generate harmonic modulation, which can improve the imaging contrast by detecting the enhancement of the fundamental and harmonic signals of the feedback. In the scattering medium, the ultrasonic microbubble contrast agent can significantly attenuate the ultrasound modulated laser feedback signal, and the imaging contrast can be improved by detecting the attenuation of the fundamental and harmonic signal.

Abstract:Theoretical studies on the multi-bubble interaction are crucial for the in-depth understanding of the mechanism behind the applications of ultrasound contrast agents (UCAs) in clinics. A two-dimensional (2D) axisymmetric finite element model (FEM) is developed here to investigate the bubble-bubble interactions for UCAs in a fluidic environment. The effect of the driving frequency and the bubble size on the bubble interaction tendency (viz., bubbles' attraction and repulsion), as well as the influences of bubble shell mechanical parameters (viz., surface tension coefficient and viscosity coefficient) are discussed. Based on FEM simulations, the temporal evolution of the bubbles' radii, the bubble-bubble distance, and the distribution of the velocity field in the surrounding fluid are investigated in detail. The results suggest that for the interacting bubble-bubble couple, the overall translational tendency should be determined by the relationship between the driving frequency and their resonance frequencies. When the driving frequency falls between the resonance frequencies of two bubbles with different sizes, they will repel each other, otherwise they will attract each other. For constant acoustic driving parameters used in this paper, the changing rate of the bubble radius decreases as the viscosity coefficient increases, and increases first then decreases as the bubble shell surface tension coefficient increases, which means that the strength of bubble-bubble interaction could be adjusted by changing the bubble shell visco-elasticity coefficients. The current work should provide a powerful explanation for the accumulation observations in an experiment, and provide a fundamental theoretical support for the applications of UCAs in clinics.

Abstract:The study of micro bubble nonlinear behavior is of great importance in recent years due to the wide applications of micro bubbles in ultrasound diagnosis and treatment. The main problem of the existing models predicting the micro bubble oscillation is that they do not consider the influence of the bubble interaction. Besides, there exists many parameters in the mainstream models, which increases the complexity of the simulation. Giving the complexity and multiple parameters of existing micro bubble models, a new micro bubble model was proposed for lipid-shell Sonovue micro bubble, which decreases the number of the parameters in the model and at the same time considers the influence of the bubbles interaction. Two types of experiments were performed to evaluate the accuracy of the proposed model. One is microscope experiment in which the dynamic behaviors of single Sonovue under different ultrasound exposures were studied. The other evaluation experiment is the in-vivo imaging in which a new mother wavelet was constructed based on the proposed model. The microscope results suggested that the proposed model can simulate the compression-only' behavior of Sonovue accurately. The maximum relative deviation between the experiment and simulation result is smaller than 2.27%. The in-vivo imaging results showed that the contrast-to-tissue ratio (CTR) of imaging has 2.81 dB improvement over that without bubble wavelet transform (BWT) for in vivo rabbit kidney experiment.

Abstract:Ultrasound modulated laser confocal feedback technology is proposed in the imaging inside turbid media. By selecting the detectable signal photons and rejecting the background noise photons in the frequency domain based on the ultrasound modulation, the signal-to-noise ratio (SNR) can be much improved, especially in the turbid media, compared with the traditional imaging without ultrasound modulation. This is a promising method to reach both a larger penetration depth and a better SNR than other optical methods.
A Sensitive Ultrasonic Imaging Method for Targeted Contrast Microbubble Detection

Abstract:We have recently developed a targeted imaging technique for selective and sensitive ultrasound molecular imaging by taking advantage of wideband transient high frequency acoustic emission from ultrasound contrast agents. The imaging modality makes use of a novel multi-frequency co-linear array (two outer 1.4 MHz and one center 5.3 MHz arrays) transducer integrated with the Siemens AntaresSystem. The imaging sequence includes a B-mode imaging pulse sequence in which a short pulse is transmitted with the outer low frequency arrays and received with the inner high frequency array (TLRH: transmit at low frequency and receive at high frequency), followed by a long radiation force pulse to induce immediate bubble adhesion using the center array, and a second B-mode imaging pulse sequence. The RF data obtained from the second B-mode pulse sequence are averaged and then subtracted from the first B-mode sequence. The imaging technique was tested in a targeted imaging phantom, where lipid-shelled biotin microbubbles flow within an avidin coated-cellulose. Results showed that tissue signals were suppressed up to 33 dB and a targeted bubble contrast-to-free bubble signal ratio of up to 23 dB was obtained from the composite sequence imaging.
Bubble-In-Bubble Strategy for High-Quality Ultrasound Imaging with A Structure Coupling Effect

Abstract:We design and synthesize monodisperse multi-layer microbubbles (MLBs) with bubble-in-bubble structure using the polymer-backbone-transition method (PBTM). Besides the tunable layer number and microbubble size, we can accurately control the layer-to-layer space and the thickness of each layer. In comparison to traditional single-layer microbubbles, the MLBs can be used as a novel promising model of contrast agent for high-performance ultrasound imaging. The ultrasound signal of MLBs shows a strong structure coupling effect; as the number of layer increases, both the backscatter signal and lifespan increase significantly. We apply a classical acoustic theory to calculate the ultrasound scattering cross-section for the bubbles. It is found that the MLBs exhibit a larger ultrasound scattering cross-section than single-layer bubbles with the same size under certain experimental conditions. Such theoretical results agree well with the ultrasound experimental results.
Novel Microbubbles for Ultrasonic Imaging Based on Surfactants

Abstract:In this study, Span 60 (sorbitan monostearate) and PEG40S (polyoxyethylene 40 stearate) are employed to generate stabilized microbubbles for use in diagnostic ultrasound. The particle size analysis of the surfactant-coated microbubbles showed that they are appropriately sized for intravenous administration. The dependence on composition in π-A isotherms for mixed monolayers can be considered as evidence of the miscibility, though not necessarily homogeneous, indicating the presence of mutual interactions between the two components. To obtain a kind of novel bi-mode diagnostic agents with both ultrasound and fluorescent imaging properties, we combined microbubbles and CdTe quantum dots (QDs) together via layer by layer assembly (LBL) technique. Through fluorescent spectra and confocal laser scanning microscopy, it was confirmed that CdTe nanoparticles were successfully deposited on the outer surface of the microbubbles. These stabilized microbubbles exhibit a high degree of echogenicity under B-mode scan, regular Doppler and color Doppler. They are useful as contrast agents to provide better contrast resolution for ultrasonic diagnostics, including echocardiography.
Basic physical, acoustic properties, and in vitro ultrasound imaging enhancement of the lipid-coated microbubbles

Abstract:Aim To research on a stable microbubble with good acoustic properties and excellent imaging enhancing effect, and to make it to be a promising agent for the enhancement of ultrasound imaging in the ultrasound diagnosis of cardiovascular system diseases. Methods The morphology, size and zeta potential of lipid-coated microbubbles (LCM), the acoustic properties of backscatter, and the second harmonic scatter of LCM were determined. Furthermore, the relationship between the concentrations and the amplitude values of the second harmonic was investigated by testing the intensities of the second harmonic at different concentrations. The imaging effect of LCM was also studied in vitro. Results The mean diameter of LCM was 3.38 μm with 95% of the bubbles under 5 μm. The scatter signal generated by microbubbles was observed under different concentrations in the bistatic modes while the position of transmitting transducer and receiving transducer was orthogonal. The intensity of the second harmonic scatter fell with the decrease of microbubble concentration. The increase rate of the second harmonic amplitude values generated by the microbubbles versus that by physiological saline was linear with the natural logarithm of bubble concentrations.The LCM could enhance the ultrasound image of thrombus. Conclusion The LCM exhibited good physical state and acoustic properties, which could increase the imaging quality.
High-Speed Ultrasound Imaging of Laser-Induced Cavitation Bubbles

Abstract:While the ultrasound and cavitation based therapies have mushroomed over the years, there is a lack of online monitoring the cavitation bubble dynamics in biological tissue. {\color{black}Here we demonstrate that with ultrasonic plane wave imaging the fast dynamics of single cavitation bubbles can be resolved non-invasively in a tissue mimicking material}. Due to the high contrast of the bubbles, plane wave compounding is not necessary and frame rates of up to 74 kHz can been achieved with a research grade ultrasound scanner. Comparison with simultaneous high-speed imaging demonstrates excellent agreement of both measurement modalities in water and in a tissue mimicking material.

Abstract:Contrast-enhanced ultrasound imaging (CEUS) based on the acoustic nonlinearity of ultrasonic microbubble has received great attention in recent years. Compared with conventional linear ultrasound imaging, nonlinear CEUS can further improve the imaging resolution while overcoming the challenge of clutter filtering. Simulation, acting as an effective tool for research on new mechanisms and technologies of ultrasound imaging, has been a long-term focus of computational acoustics. In the community of biomedical ultrasound, common sound field simulation tools are mainly based on finite element method (FEM), analytical method, k-space pseudospectral method and finite-difference time-domain method (FDTD), which are relatively mature solutions for simulating the nonlinear characteristics of tissue. However, it is still not trivial to simulate nonlinear CEUS by using the prevailing methods, as the nonlinearity of microbubble is often not considered.In this paper, we propose a simulation method of nonlinear CEUS imaging that successfully combines the microbubble nonlinearity and classic k-space pseudospectral method. Specifically, forced oscillation response of the microbubble is computed based on the modified Rayleigh-Plesset equation and such a nonlinear response is further dealt as an additional source for analyzing the nonlinear component propagation and CEUS imaging. To investigate the performance of the proposed method, B-mode images of single microbubble and clustered microbubbles are simulated based on plane wave imaging. The plane wave based CEUS imaging can thus be carried out with different compounding angles and different contrast pulse sequencing (CPS) strategies (pulse inversion, amplitude modulation, pulse inversion & amplitude modulation, and probe element alternation). Different soft-tissue and mechanical parameters of the microbubble can be adjusted by using the proposed nonlinear simulation strategy, thus providing efficient solution for CEUS simulation. Such a method can evaluate the performances of different CPS strategies, and further contribute to the CEUS development.

Abstract:In the medical ultrasound applications, microbubbles are closely tied to the diagnostic/therapeutic uses. For diagnostic applications, their sound scattering properties yield improved imaging, when the microbubbles are used as contrast agents. The harmonics and subharmonics responses from the bubbles assist in distinguishing the acoustic scattering of blood from that of the surrounding tissue. The therapeutic use of microbubbles has recently been the subject of much interest. In a HIFU treatment, the heat generated by the bubble motion contributes an enhanced localized heating effect from the ultrasound. In the lithotripsy of renal calculi, the acoustic cloud cavitation contributes to the comminution of the renal stones. In all these applications, it is essential to understand the microbubbles and bubble cloud dynamics. The bubble motion and bubble cloud behavior are strongly influenced by the internal phenomena of the bubbles, such as thermal diffusion, mist formation, mass diffusion and heat and mass transfer through the bubble wall. In our research, bubble and bubble cloud dynamics in the medical ultrasound field have been examined. In this paper numerical results and the new techniques of medical ultrasound applications with microbubbles, microbubble enhanced HIFU and Cavitation Control Lithotripsy (CCL), are highlighted.

Acoustic Characterization and Enhanced Ultrasound Imaging of Long-Circulating Lipid-Coated Microbubbles.

Abstract:Objectives-A long-circulating lipid-coated ultrasound (US) contrast agent was fabricated to achieve a longer wash-out time and gain more resistance against higher-mechanical index sonication. Systemic physical, acoustic, and in vivo imaging experiments were performed to better understand the underlying mechanism enabling the improvement of contrast agent performance by adjusting the physical and acoustic properties of contrast agent microbubbles. Methods-By simply altering the gas core, a kind of US contrast agent microbubble was synthesized with a similar lipid-coating shell as SonoVue microbubbles (Bracco SpA, Milan, Italy) to achieve a longer wash-out time and higher inertial cavitation threshold. To bridge the structure-performance relationship of the synthesized microbubbles, the imaging performance of the microbubbles was assessed in vivo with SonoVue as a control group. The size distribution and inertial cavitation threshold of the synthesized microbubbles were characterized, and the shell parameters of the microbubbles were determined by acoustic attenuation measurements. All of the measurements were compared with SonoVue microbubbles. Results-The synthesized microbubbles had a spherical shape, a smooth, consistent membrane, and a uniform distribution, with an average diameter of 1.484 m. According to the measured attenuation curve, the synthesized microbubbles resonated at around 2.8 MHz. Although the bubble's shell elasticity (0.20.09 N/m) was comparable with SonoVue, it had relatively greater viscosity and inertial cavitation because of the different gas core. Imaging studies showed that the synthesized microbubbles had a longer circulation time and a better chance of fighting against rapid collapse than SonoVue. Conclusions-Nano/micrometer long-circulating lipid-coated microbubbles could be fabricated by simply altering the core composition of SonoVue microbubbles with a higher-molecular weight gas. The smaller diameter and higher inertial cavitation threshold of the synthesized microbubbles might make it easier to access deep-seated organs and give prolonged imaging enhancement in the liver.
Enhanced visibility through microbubble-induced photoacoustic fluctuation imaging

Abstract:A photoacoustic contrast mechanism is presented based on the photoacoustic fluctuations induced by microbubbles flowing inside a micro vessel filled with a continuous absorber. It is demonstrated that the standard deviation of a homogeneous absorber mixed with microbubbles increases non-linearly as the microbubble concentration and microbubble size is increased. This effect is then utilized to perform photoacoustic fluctuation imaging with increased visibility and contrast of a blood flow phantom.

Abstract:Background Improving imaging quality is a fundamental problem in ultrasound contrast agent imaging (UCAI) research. Plane wave imaging (PWI) has been deemed as a potential method for UCAI due to its’ high frame rate and low mechanical index. High frame rate can improve the temporal resolution of UCAI. Meanwhile, low mechanical index is essential to UCAI since microbubbles can be easily broken under high mechanical index conditions. However, the clinical practice of ultrasound contrast agent plane wave imaging (UCPWI) is still limited by poor imaging quality for lack of transmit focus. The purpose of this study was to propose and validate a new post-processing method that combined with deep learning to improve the imaging quality of UCPWI. The proposed method consists of three stages: (1) first, a deep learning approach based on U-net was trained to differentiate the microbubble and tissue radio frequency (RF) signals; (2) then, to eliminate the remaining tissue RF signals, the bubble approximated wavelet transform (BAWT) combined with maximum eigenvalue threshold was employed. BAWT can enhance the UCA area brightness, and eigenvalue threshold can be set to eliminate the interference areas due to the large difference of maximum eigenvalue between UCA and tissue areas; (3) finally, the accurate microbubble imaging were obtained through eigenspace-based minimum variance (ESBMV). Results The proposed method was validated by both phantom and in vivo rabbit experiment results. Compared with UCPWI based on delay and sum (DAS), the imaging contrast-to-tissue ratio (CTR) and contrast-to-noise ratio (CNR) was improved by 21.3 dB and 10.4 dB in the phantom experiment, and the corresponding improvements were 22.3 dB and 42.8 dB in the rabbit experiment. Conclusions Our method illustrates superior imaging performance and high reproducibility, and thus is promising in improving the contrast image quality and the clinical value of UCPWI.
Real-Time Monitoring and Quantitative Evaluation of Cavitation Bubbles Induced by High Intensity Focused Ultrasound Using B-Mode Imaging

Abstract:A software-based method is proposed to eliminate the flooding interference strips in B-mode images, and to evaluate the cavitation bubbles generated during high intensity focused ultrasound (HIFU) exposures. In vitro tissue phantoms are exposed to 1.12MHz HIFU pulses with a fixed 100 Hz pulse repetition frequency. HIFU-induced cavitation bubbles are detected as hyperechoic regions in B-mode images. The temporal evolution of cavitation bubbles, generated by HIFU pulses with varying driving amplitude and pulse length, is analyzed by measuring the time-varying area of the hyperechoic region. The results show that: first, it is feasible to monitor HIFU-induced cavitation bubble activity in real-time using B-mode imaging; second, more cavitation bubbles can be generated with higher acoustic energy delivered; third, the hyperechoic region is observed to shrink gradually after ceasing the HIFU pulses, which indicates the dissolution of cavitation bubbles. This work will be helpful for developing an effective tool to realize real-time monitoring and quantitative evaluation of HIFU-induced cavitation bubble activity using a current commercialized B-mode machine.
Mass Production of Monodisperse Ultrasound Contrast Microbubbles in Integrated Microfluidic Devices

Abstract:Microbubbles coated by surfactant or polymer have been shown to be the most effective contrast agents for diagnostic ultrasound contrast imaging and targeted molecular imaging probes. The microbubbles produced by flow-focusing units have a narrower size distribution than that of the traditional methods such as sonication or agitation and is nearly monodisperse in size distribution, but with very limited production. This paper investigates the production of monodisperse microbubbles in integrated microfluidic devices in a massive way. The integrated microfluidic devices are composed of three layers: two top layers made from poly(dimethylsiloxane)(PDMS) and one glass slide as the bottom layer. Four and eight FFUs are integrated on a chip sharing the same gas inlet, liquid inlet and outlet channel. Monodisperse microbubbles production efficiency of the three microfluidic devices with one, four and eitht FFUs on a chip are evaluated respectively. It is found that the liquid consumption does not increase linearly with the FFUs number within microfluidic devices while the concentration of the microbubbles increases. The results of this work indicate the potential for mass production of ultrasound contrast agents in large scale integrated microfluidic device for commercial use.
Enhancement of subharmonic emission from encapsulated microbubbles by using a chirp excitation technique.

Abstract:Subharmonic contrast imaging promises to improve ultrasound- imaging quality by taking advantage of an increased contrast to tissue signal. However, acoustic pressures beyond the subharmonic generation threshold using common ultrasound pulses may induce significant contrast microbubble destruction. In this work, a chirp excitation technique is presented to enhance the subharmonic emission from encapsulated microbubbles. Chirp signals with a center frequency of 5 MHz, variable frequency range and duration time are employed to drive microbubbles in numerical simulation and experimental studies. We provide a theoretical evaluation of the chirp excitation pressure threshold and the acoustic pressure dependence of subharmonic based on Church's model and demonstrate that the amplitude and axial resolution of the subharmonic can be optimized by proper selection of the frequency range and time duration of the chirp signal. Measurements are qualitatively in agreement with the simulation. Moreover, we demonstrate that chirp excitation may be able to improve the amplitude of the subharmonic component up to 22 dB over the pulse excitation. The chirp excitation technique could potentially be used for improving the subharmonic contrast imaging quality.

Abstract:A novel method of ultrasound-modulated optical tomography (UOT) detection based on the laser feedback technology is proposed in this Letter. The system has advantages such as a simple structure, high sensitivity, and reflective configuration. Effective penetration depths of up to 9 cm and 5 cm in phantom and biological tissues, respectively, have been demonstrated experimentally. The detection capability is comparable with the state of the art in the transmission mode but with a much lower photon consumption. Although a lot remains to be improved, the proposed method is promising for further development toward practical applications.

Abstract:Objective:Microbubble is a kind of traditional contrast agent for ultrasound imaging and also one of the most important probe carriers for ultrasound molecular imaging.The developing trend of molecular probes on microbubbles is to multimodal,multifunctional and intelligent.Now there have been lots of novel and multifunctional molecular probes developed based on microbubbles in recent years.This review summarized the design and application of different types of ultrasound molecular probes.It includes multimodal molecular probes of various imaging modes,multifunctional molecular probes of integration of diagnosis and therapy,stimulated response ultrasound molecular probes triggered by special pathological conditions and enhanced tissue penetration ultrasound molecular nano-probes.
