MATE transporter gene of Zea mays regulates plant development and hypocotyl elongation

Abstract:[Objective] This study explored the function of multidrug and toxic compound extrusion (MATE) family gene ZmMATE884 (GRMZM2G423884) in maize during plant development.[Method] In this paper,the MATE protein family gene ZmMATE884 was cloned from the whole genomic DNA of maize,and the gene expression pattern was analyzed by quantitative RT-PCR.The possible transmembrane domain of ZmMATE884 protein was predicted by protein homology comparison.Transient expression vectors of ZmMATE884 C-terminal fused with fluorescent protein were constructed for the transformation of Arabidopsis mesophyll protoplasts to analyze subcellular localization.Then,the ZmMATE884 overexpression transgenic Arabidopsis lines were constructed by Agrobacterium tumefaciens infection technique.Arabidopsis transgenic overexpression lines and wild-type seeds were incubated on 1/2 MS solid medium with and without 1 % sucrose for 6 days in the dark or 7 days in the light to observe the hypocotyl elongation.[Result] ZmMATE884 was highly expressed in roots,base leaf,tip leaf and embryos.ZmMATE884 had 12 transmembrane domains,and it was partially localized in golgi and endosomes.The transgenic plants showed faster rosette leaves initiation and earlier flowering.The hypocotyls of the ZmMATE884 gene overexpression transgenic Arabidopsis lines were significantly shorter than that of wild-type.[Conclusion]ZmMATE884 involved in the regulation of plant development and elevated ZmMATE884 expression levels resulted in accelerated plant growth and earlier flowering.In addition,ZmMATE884 was a negative regulator of hypocotyl elongation.
