Abstract:Near-infrared (NIR) responsive nanoparticles are an important platform for multimodal phototherapy. Importantly, the simultaneous NIR-triggered photodynamic (PDT) and photothermal (PTT) therapy is a powerful approach to increase the antitumor efficiency of phototherapic nanoparticles due to the synergistic effect. Herein, a boron dipyrromethene (BODIPY)-based amphiphilic dye with enhanced electron donor-acceptor-donor (D-A-D) structure (BDP-AP) was designed and synthesized, which could self-assemble into stable nanoparticles (BDP-AP NPs) for the synergistic NIR-triggered PDT/PTT therapy. BDP-AP NPs synchronously generated singlet oxygen (1O2) and achieved preeminent photothermal conversion efficiency (61.42%). The in vitro and in vivo experiments showed that BDP-AP NPs possessed negligible dark cytotoxicity and infusive anticancer performance. BDP-AP NPs provide valuable guidance for the construction of PDT/PTT-synergistic NIR nanoagents to improve the efficiency of photoinduced cancer therapy in the future.
Small-Molecule Porphyrin-Based Organic Nanoparticles with Remarkable Photothermal Conversion Efficiency for in Vivo Photoacoustic Imaging and Photothermal Therapy.

Abstract:Near-infrared (NIR)-absorbing organic nanoparticles (ONPs) are emerging candidates for "one-for-all" theranostic nanomaterials with considerations of safety and formulation in mind. However, facile fabrication methods and improvements in the photothermal conversion efficiency (PCE) and photostability are likely needed before a clinically viable set of candidates emerges. Herein, a new organic compound, [porphyrin-diketopyrrolopyrrole (Por-DPP)] with the donor-acceptor structure was synthesized, where porphyrin was used as a donor unit while diketopyrrolopyrrole was used as an acceptor unit. Por-DPP exhibited efficient absorption extending from visible to NIR regions. After self-assembling into nanoparticles (NPs) (similar to 120 nm), the absorption spectrum of Por-DPP NPs broadened and red-shifted to some extent, relative to that of organic molecules. Furthermore, the architecture of NPs enhanced the acceptor-donor structure, leading to emission quenching and facilitating nonradiative thermal generation. The PCE of Por-DPP NPs was measured and calculated to be 62.5%, higher than most of ONPs. Under 808 nm laser irradiation, the Por-DPP NPs possessed a distinct photothermal therapy (PTT) effect in vitro and can damage cancer cells efficiently in vivo without significant side effects after phototherapy. Thus, the small-molecule porphyrin-based ONPs with high PCE demonstrated promising application in photoacoustic imaging-guided PTT.
D-A-D organic small molecules with AIE effect for fluorescence imaging guided photothermal therapy.

Abstract:Near infrared (NIR) fluorescent organic molecules as fluorescent probes accurately guide photothermal therapy as a potential antitumor method. However, the aggregation and quenching of organic fluorescent molecules and poor tissue permeability greatly limit their therapeutic effect and clinical transformation. In this paper, with a D-A-D structure as the molecular skeleton, cyclopentadithiophene (CPDT) as the donor (D), diketopyrrolopyrrole (DPP) as the acceptor (A), and long-chain isooctane as the shielding unit, organic fluorescent small molecules with a strong absorption band and bright NIR-II emission were synthesized. Then, tetraphenylethylene (TPE) molecules with typical AIE structure characteristics were introduced on both sides of the organic fluorescent small molecules, and an organic small molecular fluorophore (TDA) with AIE characteristics and the photothermal effect was designed. Through a series of experimental characterization techniques, it is proved that TDA NPs have good biocompatibility and tissue permeability, and can accurately locate the tumor location through NIR-II fluorescence imaging to achieve local photothermal treatment of tumors.

Abstract:A schematic representation showing the preparation and application of NCBDT‐4Cl NPs is presented. Phototheranostics with near‐infrared fluorescence and reactive oxygen species generation ability and high photothermal conversion efficiency (PCE) plays a significant role in fluorescence imaging‐guided synergetic photodynamic and photothermal therapy of tumors. Here, a star molecule in organic photovoltaic materials, NCBDT‐4C with an A‐D‐A conjugated structure, was assembled with DSPE‐PEG‐NH2 to prepare water dispersive nanoparticles (NPs). The prepared NCBDT‐4Cl NPs exhibited a maximum NIR absorption peak at 764 nm and a maximum fluorescence peak at 798 nm. These NPs could generate superoxide anion, singlet oxygen (1O2), and heat under 808 nm laser irradiation. The 1O2 generation quantum yield and PCE of the NPs were 37.5% and 53.6%, respectively. The combination of photodynamic and photothermal therapy of cancer was demonstrated in vitro and in vivo. This work presents the advanced application of organic photovoltaic materials in cancer phototherapy.

Abstract:Various phototheranostics have recently been developed for phototherapy. Through proper molecular design, the photochemical and photophysical properties of these phototheranostics can be promoted. Herein, an acceptor-donor-acceptor (A-D-A)-structured dye, BTP-4F-DMO, was synthesized and prepared into water-soluble nanoparticles (NPs). The obtained BTP-4F-DMO NPs had strong absorption from 650 nm to 850 nm and a fluorescence emission peak at ∼900 nm that tailed to ∼1100 nm. The NPs showed a superhigh photothermal conversion efficiency of 90.5% ± 5% and could simultaneously generate • OH and 1 O 2 with a 1 O 2 generation quantum yield of 4.6% under 808 nm laser irradiation. Due to these advanced properties, BTP-4F-DMO NPs can switch the role of autophagy from pro-survival to pro-death, thereby further promoting cancer cell death. These features make BTP-4F-DMO NPs a promising multifunctional phototheranostic agent for NIR-II fluorescence/photoacoustic dual-mode imaging-guided synergetic photodynamic/photothermal therapy. In general, this work provides a strategy for expanding the biomedical applications of organic A-D-A-structured phototheranostics.

Abstract:Organic diradicaloids with unique open‐shell structures and properties have been widely used in organic electronics and spintronics. However, their advantageous optical properties have been explored less in the biomedical field. In this work, the photothermal conversion behaviors of a boron‐containing organic diradicaloid (BOD) are reported. BOD can assemble with 1,2‐distearoyl‐sn‐glycero‐3‐phosphoethanolamine‐poly(ethylene glycol) to form rodlike nanoparticles (BOD NPs). These as‐prepared BOD NPs exhibit high photothermal conversion capability and robust photothermal stability. Notably, they possess morphological superiority, which guarantees the effective photothermal therapy of tumors. This work thus demonstrates the promise of organic diradicaloids as efficient photothermal agents for biomedical applications.

Abstract:Organic nanomaterials have garnered extensive interest in PDT/PTT due to their excellent photo-stability, efficient photothermal and reactive oxygen species generation ability. However, most organic nanomaterials emit very weak in the aggregated state because of the aggregation-caused quenching phenomenon, which limits their application in bioimaging. In this study, a donor-acceptor-donor (D-A-D) type BODIPY-based compound (BODIPY-TPA) was synthesized by connecting triphenylamine (TPA) with BODIPY structure. To further improve the biocompatibility of the compound, a polyethylene glycol chain (PEG) was introduced into the BODIPY core. BODIPY-TPA could transform into the related nanoparticles (BODIPY-TPA NPs) by the self-assembly process. Remarkably, BODIPY-TPA NPs showed strong near-infrared (NIR) fluorescence because of its aggregation-induced emission (AIE) property, which may be ascribed to the twisted structure of TPA. BODIPY-TPA NPs could simultaneously generate singlet oxygen (1O2) and heat under the irradiation of a single laser (635 nm). The photothermal conversion efficiency (PCE) of BODIPY-TPA NPs was calculated to be 20.7%. The subcellular localization experiment indicated the lysosome-targeting properties of BODIPY-TPA NPs in A549 cells. The cytotoxicity of BODIPY-TPA NPs was studied by CCK-8 assay and AM/PI staining on A549 cells. The half maximal inhibitory concentration (IC50) of BODIY-TPA NPs was calculated to be 28.45 μg/mL. This study revealed BODIPY-TPA NPs could potentially be used in bioimaging and photodynamic/photothermal (PDT/PTT) synergistic cancer treatment.

Side-chain Engineering of Organic Photothermal Agents for Boosting Further Red-Shifted Absorption and Higher Photothermal Therapeutic Effect

Abstract:Although organic photothermal agents (PTAs) have been extensively studied in preclinical cancer photothermal therapy (PTT), the internal mechanism, particularly the impact of side chains on photothermal performance, remains inadequately investigated. Herein, we conducted a systematic comparison of the photothermal properties between two organic molecules, namely O-IDTBR with four n-octyl chains and EH-IDTBR with four 2-ethylhexyl chains. With the same conjugated main structure, both O-IDTBR and EH-IDTBR exhibited nearly identical absorption properties (with a peak at 629 nm) in their molecular states. Interestingly, after the formation of nanoparticles (NPs), O-IDTBR NPs with linear alkyl chains exhibit a further red-shifted absorption onset (peak at 711 nm) compared to EH-IDTBR NPs (peak at 662 nm) with branched alkyl chains. Additionally, the photothermal conversion efficiency of O-IDTBR NPs was calculated of 33.7%, which is higher than that of EH-IDTBR NPs (27.7%). This can be attributed to the fact that linear alkyl chains of O-IDTBR NPs promote more intramolecular motions at the aggregated state by extending intermolecular distance and distorting molecular conformation. Therefore, the nonradiative thermal deactivation-induced photothermal property can be further enhanced. Through both in vitro and in vivo experiments, O-IDTBR NPs exhibit effective PTT effect and excellent biocompatibility. This study not only introduces a novel PTA but also opens new avenues for exploring organic optical nano-agents through side-chain engineering to adjust intramolecular motions at the aggregated state.
Diradicaloid-Loaded Polypeptide Nanoparticles for Two-Photon NIR Phototheranostics

Abstract:Stable organic radicals, with unique electronic transitions from the ground state (D0) to the doublet excited state (D1), show promise as high-fluorescence quantum yield dyes. While organic small-molecule photosensitizers (PSs) have advanced for tumor photodynamic therapy (PDT), opportunities exist to enhance their performance and functionality. Herein, we synthesized Thiele's fluorocarbon derivative diradicaloid TFC-I with nearly 100% PLQY and integrated it into amphiphilic polypeptide nanoparticles, P-TI, using a precursor-doping approach. P-TI demonstrated notable features including high photostability, aggregation-induced emission, bright near-infrared fluorescence, substantial quantum yield (37% PLQY), robust near-infrared two-photon absorption (∼400 GM cross section), and superior ROS generation compared to commercial PSs. In vitro and in vivo experiments confirmed that P-TI performed well in mitochondria-targeted PDT, two-photon fluorescence imaging, and biosafety. This work highlights the use of organic stable radicals with precursor-doping for efficient PDT and deep tumor tissue imaging.
NIR Light‐Driving Barrier‐Free Group Rotation in Nanoparticles with an 88.3% Photothermal Conversion Efficiency for Photothermal Therapy

Abstract:Traditional photothermal therapy requires high‐intensity laser excitation for cancer treatments due to the low photothermal conversion efficiency (PCE) of photothermal agents (PTAs). PTAs with ultra‐high PCEs can decrease the required excited light intensity, which allows safe and efficient therapy in deep tissues. In this work, a PTA is synthesized with high PCE of 88.3% based on a BODIPY scaffold, by introducing a CF3 "barrier‐free" rotor on the meso‐position (tfm‐BDP). In both the ground and excited state, the CF3 moiety in tfm‐BDP has no energy barrier to rotation, allowing it to efficiently dissipate absorbed (NIR) photons as heat. Importantly, the barrier‐free rotation of CF3 can be maintained after encapsulating tfm‐BDP into polymeric nanoparticles (NPs). Thus, laser irradiation with safe intensity (0.3 W cm−2, 808 nm) can lead to complete tumor ablation in tumor‐bearing mice after intravenous injection of tfm‐BDP NPs. This strategy of "barrier‐free rotation" provides a new platform for future design of PTT agents for clinical cancer treatment.

Highly Photostable Deep-NIR Chromophore with a Superior Molar Absorptivity for Efficient Photothermal Therapy

Abstract:Organic chromophores, especially those absorbing/emitting in the deep near-infrared region (deep-NIR, i.e., 800 nm and beyond), have prominent potentials in photothermal cancer therapy due to the deep tissue penetration and good biocompatibility. Although many cyanine/DAD-based photothermal agents have been successfully used in clinical and preclinical trials, they are limited to poor photostability and poor molar absorptivity. There is still an imperative demand for robust deep-NIR photothermal agents exhibiting high photostability and high molar absorptivity. We previously invented a bright push–pull fluorochromic scaffold (EC5) absorbing and emitting in the deep-NIR spectral region. In this work, EC5 was rationally tailored to suppress its fluorescence emission and promote its photothermal conversion efficiency. This led to the design and synthesis of EC5PM and EC5DP by changing two or four of its N-alkyl groups of the push–pull headgroups of EC5 into phenyl groups, respectively. The fluorescence intensities of EC5PM and EC5DP are completely quenched due to the TICT effect associated with the N–Ph bonds. In particular, EC5DP maximally absorbs at a longer wavelength of 879 nm with a superior photostability, a high photothermal conversion efficiency of 54.8%, and a superior molar extinction coefficient of 224,000 M–1 cm–1. Upon encapsulation in DSPE-mPEG2000 micelles, the phototoxicity of EC5DP was showcased in 4T1 cells and a remarkable therapeutic effect of tumors was showcased with a 4T1 tumor-bearing mouse model. This work provides a rational strategy to design ultraphotostable and highly deep-NIR absorbing organic chromophores for photothermal therapy.

Inhibiting Radiative Transition-Mediated Multifunctional Polymeric Nanoplatforms for Highly Efficient Tumor Phototherapeutics

Abstract:It is highly desired to explore ideal phototherapeutic nanoplatforms, especially containing satisfactory phototherapeutic agents (PTAs), for potential cancer therapies. Herein, we proposed an effective strategy for designing a highly efficient PTA through inhibiting radiative transition (IRT). Specifically, we developed an ultralow radiative BODIPY derivative (TPA-IBDP) by simply conjugating two triphenylamine units to iodine-substituted BODIPY, which could simultaneously facilitate the nonradiative decay channels of singlet-to-triplet intersystem crossing and intramolecular charge transfer. In comparison to the normal BODIPY compound, TPA-IBDP exhibited an outstanding singlet oxygen yield (31.8-fold) and a higher photothermal conversion efficiency (PCE; over 3-fold), respectively, benefiting from the extended π-conjugated donor-to-accepter (D-A) structure and the heavy atom effect. For tumor phototherapy using TPA-IBDP, TPA-IBDP was conjugated with a H2O2-responsive amphiphilic copolymer POEGMA10-b-[PBMA5-co-(PS-N3)2] to construct a multifunctional phototherapeutic BODIPY-based nanoplatform (PB). PB produced abundant singlet oxygen (1O2) and heat along with negligible fluorescence emission under near-infrared laser irradiation. Additionally, PB could generate a GSH-depletion scavenger (quinone methide, QM) after reacting with the abundant intracellular H2O2 in tumor for the cooperative enhancement of IRT-mediated phototherapy. We envision that this highly efficient multifunctional phototherapeutic nanoplatform cooperated by GSH-depletion could be a valuable paradigm for tumor treatments.

Boron Dipyrromethene-Based Phototheranostics for Near Infrared Fluorescent and Photoacoustic Imaging-Guided Synchronous Photodynamic and Photothermal Therapy of Cancer

Abstract:The regulation of photochemical properties of phototheranostics, especially the absorption, fluorescence, singlet oxygen (1O2) generation, and photothermal conversion efficiency, is a hot research topic. Here, we designed and synthesized four boron dipyrromethene (BODIPY) derivatives with high absorption coefficients and intense fluorescence in the near-infrared (NIR) region. The substituted electron-donating group significantly improved 1O2 generation and fluorescence of BODIPYs, whereas the electron-withdrawing group boosts photothermal conversion. These hydrophobic BODIPYs were further coated with DSPE-PEG-2000 to form water dispersible nanoparticles (NPs). Among these BODIPY NPs, the B-OMe-NPs with methoxyl substituted at the meso-position showed the highest 1O2 generation, a photothermal conversion efficiency of 66.5%, and an NIR fluorescence peak at 809 nm. In vitro and in vivo experiments demonstrated that B-OMe-NPs might be used for NIR fluorescent and photoacoustic imaging-guided photodynamic and photothermal therapy of cancer.
Synthesis of strong electron donating-accepting type organic fluorophore and its polypeptide nanoparticles for NIR-II phototheranostics

Abstract:A novel NIR-II small-molecule D-A type organic fluorophore conjugation of triphenylamine, thiophene, and benzo[c,d] indol groups (TPA-Et) with strong electron-donating and accepting groups has been synthesized. The dye shows a significant Stokes shift for efficient fluorescence in the NIR-II region and high photothermal performance. The TPA-Et was then encapsulated by an amphiphilic copolymer P(OEGMA)20-P(Asp)14, and micelles (P@TP) has been prepared with outstanding NIR-II imaging performance, excellent photothermal conversion efficiency (52.5%) under 808 nm laser irradiation, and good photostability. Fluorescence imaging experiments have consistently shown that P@TP can image tiny blood vessels in mice, enrich effectively in the tumor region, and maintain a relatively stable NIR-II fluorescence signal in the tumor area for a long time up to 60 h. In vivo photothermal therapy has a highly significant anticancer effect without tumor recurrence, demonstrating the apparent advantages of P@TP as a NIR nanotheranostic platform in NIR-II imaging-guided photothermal therapy.
Simple Preparation of Near-infrared-II Organic Small Molecule-based Phototheranostics by Manipulation of the Electron-donating Unit

Abstract:Organic small molecules hold great promise in the field of phototheranostics by virtue of their clear chemical structure, outstanding biocompatibility, and distinguished repeatability. Whereas the synthetic process of them suffers from multi-step and complex reactions. Meanwhile, most of small molecules employed near-infrared-I (MR-I) fluorescence imaging, which could not effectively provide comprehensive tumor information with high resolution. In addition, the reported small molecules showed unsatisfactory singlet oxygen yield and photothermal conversion efficiency, limiting their extensive application in cancer theranostics. Herein, by conjugating diketopyrrolopyrrole (an electron-withdrawing unit) with benzene, aniline, or 1,2-diaminobenzene (electron-donating units), respectively, three novel small molecules DPP-0, DPP-2 and DPP-4 were designed and synthesized by one-step reaction. DPP-0 NPs, DPP-2 NPs and DPP-4 NPs as phototheranostics were further obtained through nanoprecipitation method by self-assembling with Pluronic F127. It was found that with the increase of amino groups, the absorption/emission peaks of NPs were red-shifted. Especially, DPP-4 NPs exhibited strong absorption in the NW-I region, and the maximum emission peak of DPP-4 NPs was located in the near-infrared-II (MR-II) region, suggesting that the optical properties of small molecules can be regulated by changing the electron-donating unit. After irradiation of DPP-4 NPs with a single laser, excellent MR-II fluorescence signal can be obtained for efficient imaging of tumor with high resolution. Moreover, abundant reactive oxygen species (singlet oxygen yield 34.3%) and hyperthermia (photothermal conversion efficiency 40.2%) can also be produced by irradiation of DPP-4 NPs, realizing the combined photodynamic/photothermal therapy guided by MR-II fluorescence imaging.

Abstract:Organic molecules that are active in the near-infrared (NIR) bio-window for photo-to-heat energy-converting theranostics have attracted much attention in cancer therapy. However, current organic NIR-absorbing molecules are still suffering from the inefficient non-radiative decay or serious photobleaching under light irradiation. Herein, we design a convenient strategy to load a new kind of NIR-absorbing nonlinear optical (NLO) chromophore FTC-3f with the biocompatible polymer, methoxy poly(ethylene glycol)-poly(lactideco-glycolide) (mPEG-PLGA). The resulted FTC-3f/PLGA nanoparticles showed excellent stability, high photothermal conversion efficiency (∼51.11%) and good photoacoustic performance. Moreover, in vitro anticancer experiments demonstrated that the FTC-3f/PLGA nanoparticles possess a high PTT (photothermal therapy) efficiency under NIR laser irradiation. This work thus provides an insight into the exploration of versatile organic NIR-absorbing nanoparticles toward future practical PTT application.

Abstract:Photopyroptosis is an emerging research branch of photodynamic therapy (PDT), whereas there remains a lack of molecular structural principles to fabricate photosensitizers for triggering a highly efficient pyroptosis. Herein, a general and rational structural design principle to implement this hypothesis, is proposed. The principle relies on the clamping of cationic moieties (e.g., pyridinium, imidazolium) onto one photosensitive core to facilitate a considerable mitochondrial targeting (both of the inner and the outer membranes) of the molecules, thus maximizing the photogenerated reactive oxygen species (ROS) at the specific site to trigger the gasdermin E-mediated pyroptosis. Through this design, the pyroptotic trigger can be achieved in a minimum of 10 s of irradiation with a substantially low light dosage (0.4 J cm(-2)), compared to relevant work reported (up to 60 J cm(-2)). Moreover, immunotherapy with high tumor inhibition efficiency is realized by applying the synthetic molecules alone. This structural paradigm is valuable for deepening the understanding of PDT (especially the mitochondrial-targeted PDT) from the perspective of pyroptosis, toward the future development of the state-of-the-art form of PDT.
Highly Efficient Multifunctional Organic Photosensitizer with Aggregation-Induced Emission for In Vivo Bioimaging and Photodynamic Therapy

Abstract:Photosensitizers play a critical role in photodynamic therapy (PDT). Multifunctional organic nanoparticles (NPs) that possess bright fluorescence in aggregates, high singlet oxygen (1O2) quantum yield, near-infrared (NIR) absorption and emission, large Stokes shift, two-photon bioimaging, specific organelle targeting, high PDT efficiency, as well as good biocompatibility and photostability are ideal candidate photosensitizers for image-guided PDT. Due to its enhanced fluorescence and high 1O2 generation efficiency in aggregate states, photosensitizers with aggregation-induced emission (AIE) characteristics have attracted increasing interest in PDT. In this study, a new AIE-active Schiff base 5-(((5-(7-(4-(diphenylamino)phenyl)benzo[c][1,2,5]thiadiazol-4-yl)thiophen-2-yl)methylene)amino)-3-methylthiophene-2,4-dicarbonitrile (TBTDC) based on a D-A-π-A skeleton has been designed and synthesized, and it can be readily encapsulated by Pluronic F-127 to form uniform nanoparticles. TBTDC NPs exhibit bright NIR emission at 825 nm with a Stokes shift up to 300 nm, impressive two-photon bioimaging capability with tissue penetration deep into 300 μm, high 1O2 generation quantum yield (0.552), specific targeting to lysosome, as well as good biocompatibility and photostability. Furthermore, TBTDC NPs present remarkable cytotoxicity for tumor cells and suppression of tumor growth in nude mice through reactive oxygen species generation upon white light irradiation. These results reveal that TBTDC NPs have great potential to become excellent candidates for multifunctional organic photosensitizers for two-photon bioimaging and image-guided PDT and are promising in future clinical applications.
A-DA′D-A Structured Organic Phototheranostics for NIR-II Fluorescence/Photoacoustic Imaging-Guided Photothermal and Photodynamic Synergistic Therapy

Abstract:Multimodal imaging-guided combinational phototherapies triggered by a single near-infrared (NIR) laser are highly desirable. However, their development is still a big challenge. Herein, we have developed an "acceptor–donor–acceptor′–donor–acceptor" structured organic phototheranostics (Y16-Pr) with strong light-harvesting ability in the NIR region. After being modified with polyethylene glycol (PEG), the obtained biocompatible nanoparticles (Y16-Pr-PEG NPs) could conduct NIR-II fluorescence imaging (FLI) and photoacoustic imaging (PAI) and perform photothermal therapy (PTT) and photodynamic therapy (PDT) simultaneously. Notably, Y16-Pr-PEG NPs showed an impressive photothermal conversion efficiency (PCE) of 82.4% under 808 nm laser irradiation. The irradiated NPs could also produce hydroxyl radicals (•OH) and singlet oxygen (1O2) for type I and type II PDT, respectively. In vivo and in vitro experiments revealed that the Y16-Pr-PEG NPs significantly inhibit tumor cell growth without apparent toxic side effects under laser irradiation.
