Abstract:Natural tissues, such as bone, tendon, and muscle, have well-defined hierarchical structures, which are crucial for their biological and mechanical functions. However, mimicking these structural features still remains a great challenge. In this study, we use ice-templated assembly and UV-initiated cryopolymerization to fabricate a novel kind of composite hydrogel which has both aligned macroporous structure at micrometer scale and a nacre-like layered structure at nanoscale. Such hydrogels are macroporous, are thermoresponsive, and exhibit excellent mechanical performance (they are tough and highly stretchable), attractive properties that have a significant impact on the wide applications of composite hydrogels, especially as tissue-engineering scaffolds. The fabrication method in this study including freeze-casting and cryopolymerization can also be applied to other materials, which makes it promising for designing and developing smart and multifunctional composite hydrogels with hierarchical structures.

Abstract:Adaptive hydrogels tailor-made from silk-elastin-like proteins (SELPs) possess excellent biocompatibility and biodegradability with properties that are tunable and responsive to multiple simultaneous external stimuli. To unravel the molecular mechanisms of their physical response to external stimuli in tandem with experiments, here we predict and measure the variation in structural properties as a function of temperature through coarse-grained (CG) modeling of individual and crosslinked SE8Y and S4E8Y molecules, which have ratios of 1:8 and 4:8 of silk to elastin blocks respectively. Extensive structural reshuffling in single SE8Y molecules led to the increased compactness of the structure, whereas S4E8Y molecules did not experience any significant changes as they already adopted very compact structures at low temperatures. Crosslinking of SE8Y molecules at high concentrations impeded their structural transition at high temperatures that drastically reduced the degree of deswelling through extensive suppression of the structural shuffling and the trapping of the molecules in high potential energy states due to inter-molecular constraints. This integrative experimental and computational understanding of the thermal response in single molecules of SELPs and their crosslinked networks should lead to further improvements in the properties of SELP hydrogels through predictive designs and their wider applications in biomaterials and tissue engineering.
Thermo-responsive Gels Based on Supramolecular Assembly of an Amidoamine and Citric Acid.

Abstract:In this work, we report the formation of a novel, aqueous-based thermo-responsive, supramolecular gelling system prepared by a convenient and efficient self-assembly of a long-chain amino-amide and citric acid. To determine the viscosity behavior and to gain insights into the gelation mechanism, a complementary combination of techniques, including Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), dynamic light scattering (DLS), and sinusoidal oscillatory tests, were used. The supramolecular gelator exhibited remarkably reversible sol-gel transitions induced by temperature at 76 °C. At a concentration of 5 wt%, the zero-frequency viscosity of the supramolecular system increased by about four orders of magnitude (from 4.2 to 12 563 Pa s) by changing the temperature from 23 °C to 76 °C. The viscous nature of the supramolecular gel could be preserved up to 90 °C. The synergistic combination of the hydrogen bonding between amino and carboxylic acid groups and the electrostatic interactions arising from the protonation of the amino-group and the deprotonation of carboxylic acid groups enhanced at higher temperatures is presumably responsible for the thermo-responsive behavior. We anticipate that these supramolecular gelators can be beneficial in various applications such as hydrogel scaffolds for regenerative medicine, personal care products and cosmetics, and enhanced oil recovery as viscosity modifiers.

Abstract:A shape-memory hydrogel is a programmable hydrogel material that can store specific shapes and execute functions in response to stimuli. In this report, we developed shape-memory hydrogels by creating double-network polymeric structures using a physically cross-linking elastin-like polypeptide (ELP) and a chemically cross-linking polyacrylamide (PAM). We synthesized the hydrogel matrix by polymerizing the acrylamide mixed in an ELP solution. We exploited the lower critical solution temperature transition of the ELP to enable the hydrogel to hold a new desired shape at an elevated temperature of 55 degrees C. The original shape of the hydrogel can then be recovered by lowering the temperature to 20 degrees C. The shape-memory hydrogels we developed exhibit ultrafast functionality and high repeatability. Taking advantage of the temperature-induced shape-memory capability, we also demonstrate practical functions such as gripping an object and connecting two tubes. Our materials with effective temperature-driven shape-memory functionality will be useful for developing novel materials for biomedical applications in the future.

Abstract:A novel thermoresponsive gelator of (B-co-C)-b-A-b-(B-co-C) topology, comprising a poly(2-(dimethylamino)ethyl methacrylate) (PDMAEMA) weak polyelectrolyte as central block, end-capped by thermosensitive poly(triethylene glycol methyl ether methacrylate/n-butyl methacrylate) [P(TEGMA-co-nBuMA)] random copolymers, was designed and explored in aqueous media. The main target of this design was to control the dynamics of the stickers by temperature as to create an injectable hydrogel that behaves as a weak gel at low temperature and as a strong gel at physiological temperature. Indeed, at low temperatures, the system behaves like a viscoelastic complex fluid (dynamic network), while at higher temperatures, an elastic hydrogel is formed (“frozen” network). The viscosity increases exponentially upon heating, about 5 orders of magnitude from 5 to 45 °C, which is attributed to the exponential increase of the lifetime of the self-assembled stickers. The integration of thermo- and shear responsive properties in the gelator endows the gel with injectability. Moreover, the gel can be rapidly recovered upon cessation of the applied stress at 37 °C, simulating conditions similar to those of injection through a 28-gauge syringe needle. All these hydrogel properties render it a good candidate for potential applications in cell transplantation through injection strategies.

Abstract:Temperature-promoted hydrogel formation was investigated in a mixed solution of sodium cholate and metal ions.Transmission electron microscopy (TEM) was used to characterize the morphology of the microstructures of the hydrogels,and nanofibers were observed.The arrangement of the cholate and metal ions was proposed according to the X-ray diffraction (XRD) measurements.Rheological measurements revealed that the strength of the hydrogels increases with increasing temperature,while the fluorescence of the gels increased as well.Increasing the temperature also increased the gel formation rate.The critical micelle concentration of sodium cholate solution decreased slightly with increasing temperature.On the basis of the experimental results,we propose that the increased hydrophobicity of cholate ions with increasing temperature leads to stronger hydrogen bonding between cholate ions,which accounts for the unique heating-enhanced gelation behavior.

Abstract:Thermal stiffening materials that are naturally soft but adaptively self-strengthen upon heat are intriguing for load-bearing and self-protection applications at elevated temperatures. However, to simultaneously achieve high modulus change amplitude and high mechanical strength at the stiffened state remains challenging. Herein, entropy-mediated polymer-mineral cluster interactions are exploited to afford thermal stiffening hydrogels with a record-high storage modulus enhancement of 13 000 times covering a super wide regime from 1.3 kPa to 17 MPa. Such a dramatic thermal stiffening effect is ascribed to the transition from liquid-liquid to solid-liquid phase separations, and at the molecular level, driven by enhanced polymer-cluster interactions. The hydrogel is further processed into sheath-core fibers and smart fabrics, which demonstrate self-strengthening and self-powered sensing properties by co-weaving another liquid metal fiber as both the joule heater and triboelectric layer.

Abstract:The incorporation of proteins into hydrogel networks has the potential to enhance bioactivity and biocompatibility. In this work, we report on the fabrication of a polymer-protein hydrogel consisting of polymethacrylamide (PMAAm) and bovine serum albumin (BSA). The hydrogel was prepared by in situ polymerization of methacrylamide in the presence of BSA at elevated temperatures. Due to its specific interactions between corresponding functional groups, BSA acts as a cross-linker of polymer chains. Hydrogel with optimized composition and preparation conditions (BSA/methacrylamide ratio and synthesis temperature) demonstrated excellent mechanical properties. Due to the presence of side amide groups in PMAAm, the energy barrier required for heat-induced transformation of globular BSA structures into unfolded linear structures decreased, causing a significant shift in the transition temperature. This transition led to a steep and substantial strengthening of the two-component hydrogel. After compressive and shear deformation, the hydrogel restored damaged structure and demonstrated superior fatigue resistance. Compared to BSA that is globular, it was found that BSA in its unfolded state has a much greater impact on the mechanical properties of the hydrogel.

Injectable Thermogels Based on Block Copolymers of Appropriate Amphiphilicity

Abstract:Some amphiphilic copolymers in water can undergo a reversible sol-gel transition upon heating. If the transition temperature lies between room temperature and body temperature, the aqueous system can be readily mixed with drugs or cells at room temperature, and the mixture is injectable; the injected formulation is physically gelled at body temperature, and the gelation is free of any chemical crosslinking. This affords an amazing biomaterial type, yet many questions are open in light of fundamental research and potential clinical applications. In particular, block copolymers composed of hydrophilic poly(ethylene glycol) (PEG) and some hydrophobic biodegradable polyesters such as poly(lactide-co-glycolide) (PLGA) are of great clinical potential, yet of very unclear physical crosslinking points. This feature article summarizes the corresponding extensive investigations in the authors' group led by Ding at Fudan University for a decade. The Ding group has found the significant effects of the end groups of the copolymer, molar mass dispersity D-M, and other molecular parameters on thermogellability, and revealed the corresponding rules of molecular design, based on their polymer chemistry studies. For instance, with increase in D-M under either given number average or weight average molecule weight, the sol-gel transition temperature for some block copolymer aqueous systems could shift unidirectionally, which indicates that the effect of molecular weight distribution could not be interpreted simply from addition of the effects of molecular weight or D-M affords an independent adjustable parameter. The Ding group has shed light on the mechanism of their thermogelling by putting forward the model of percolated micelle network to describe the internal structure of the physical hydrogel, and extended the range of the thermogellable molecular composition of the copolymers to a large extent by establishing a blend strategy, based on their polymer physics studies. The Ding group have also put forward many strategies for the clinical applications of the thermogels using animal models, including prevention of postoperative tissue adhesion, submucosal cushion for endoscopic submucosal dissection, sustained release carriers of antitumor drugs such as camptothecin derivatives, long-acting formulations of polypeptide drugs such as exenatide in treatment of type II diabetes, and tissue engineering of cartilage. The Ding group has also laid a material basis towards potential products of medical devices and drug carriers including the appropriate way of sterilization and improvement of the handling property of the synthesized polymers. The Ding group suggest some perspectives in the end of this feather article.
Molecularly Engineered Supramolecular Thermoresponsive Hydrogels with Tunable Mechanical and Dynamic Properties

Abstract:Synthetic supramolecular polymers and hydrogels in water are emerging as promising biomaterials due to their modularity and intrinsic dynamics. Here, we introduce temperature sensitivity into the nonfunctionalized benzene-1,3,5-tricarboxamide (BTA-EG4) supramolecular system by incorporating a poly(N-isopropylacrylamide)-functionalized (BTA-PNIPAM) moiety, enabling 3D cell encapsulation applications. The viscous and structural properties in the solution state as well as the mechanical and dynamic features in the gel state of BTA-PNIPAM/BTA-EG4 mixtures were investigated and modulated. In the dilute state (c ∼μM), BTA-PNIPAM acted as a chain capper below the cloud point temperature (Tcp = 24 °C) but served as a cross-linker above Tcp. At higher concentrations (c ∼mM), weak or stiff hydrogels were obtained, depending on the BTA-PNIPAM/BTA-EG4 ratio. The mixture with the highest BTA-PNIPAM ratio was ∼100 times stiffer and ∼10 times less dynamic than BTA-EG4 hydrogel. Facile cell encapsulation in 3D was realized by leveraging the temperature-sensitive sol-gel transition, opening opportunities for utilizing this hydrogel as an extracellular matrix mimic.
Instant Thermal Switching from Soft Hydrogel to Rigid Plastics Inspired by Thermophile Proteins

Abstract:Proteins of thermophiles are thermally stable in a high-temperature environment, adopting a strategy of enhancing the electrostatic interaction in hydrophobic media at high temperature. Herein, inspired by the molecular mechanism of thermally stable proteins, the synthesis of novel polymer materials that undergo ultrarapid, isochoric, and reversible switching from soft hydrogels to rigid plastics at elevated temperature is reported. The materials are developed from versatile, inexpensive, and nontoxic poly(acrylic acid) hydrogels containing calcium acetate. By the cooperative effects of hydrophobic interaction and ionic interaction, the hydrogels undergo significant spinodal decomposition and subsequent rubbery-to-glassy transition when heated to an elevated temperature. As a result, the gels exhibit super-rapid and significant hikes in stiffness, strength, and toughness by up to 1800-, 80-, and 20-folds, respectively, when the temperature is raised from 25 to 70 °C, while the volumes of the gels are almost unchanged. As a potential application, the performance of the materials as athletic protective gear is demonstrated. This work provides a pathway for developing thermally stiffened materials and may significantly broaden the scope of polymer applications.

Abstract:Although micron-sized microgels have become important building blocks in regenerative materials, offering decisive interactions with living matter, their chemical composition mostly significantly varies when their network morphology is tuned. Since cell behavior is simultaneously affected by the physical, chemical, and structural properties of the gel network, microgels with variable morphology but chemical equivalence are of interest. This work describes a new method to produce thermoresponsive microgels with defined mechanical properties, surface morphologies, and volume phase transition temperatures. A wide variety of microgels is synthesized by crosslinking monomers or star polymers at different temperatures using thermally assisted microfluidics. The diversification of microgels with different network structures and morphologies but of chemical equivalence offers a new platform of microgel building blocks with the ability to undergo phase transition at physiological temperatures. The method holds high potential to create soft and dynamic materials while maintaining the chemical composition for a wide variety of applications in biomedicine.

On-Demand Regulation of Dual Thermosensitive Protein Hydrogels

Abstract:Despite considerable progress having been made in thermosensitive protein hydrogels, regulating their thermal transitions remains a challenge due to the intricate molecular structures and interactions of the underlying protein polymers. Here we report a genetic fusion strategy to tune the unique dual thermal transitions of the C-terminal domain (CTD) of spider major ampullate spidroin 1, and explore the regulation mechanism by biophysical characterization and molecular dynamics simulations. We found that the fusion of elastin-like polypeptides (ELPs) tuned the dual transition temperatures of CTD to a physiologically relevant window, by introducing extra hydrogen bonding at low temperatures and hydrophobic interactions at high temperatures. The resulting hydrogels constructed from the fusion proteins were demonstrated to be a promising vehicle for cell preservation and delivery. Microgels with Linear Thermosensitivity in A Wide Temperature Range

Abstract:A newly developed poly(2-methoxyethyl acrylate-co-oligo(ethylene glycol) methyl ether acrylate) (P(MEA-co-OEGA) microgel, synthesized by free radical precipitation copolymerization, displays a linear volume phase transition in water upon heating in a wide range of temperature. Varying cross-linking density in the microgels does not interrupt the linear thermosensitivity. By changing the feeding molar ratio of MEA and OEGA, the linear thermosensitive temperature range can be adjusted, where higher OEGA content results in higher plateau starting temperature. Furthermore, temperature- and pH-sensitive microgels could be achieved by adding another comonomer, acrylic acid (AA). The resulting P(MEA-co-OEGA-co-AA) microgels are ideal candidates to prepare hybrid microgels by in situ formation of silver nanoparticles (AgNPs).

Abstract:Hybrid nanogels that are both thermosensitive and superparamagnetic, and have good biocompatibility are expected to have applications in the biomedical field. In this article, a linearly thermosensitive magnetic microgel was prepared by a radical copolymerization reaction in aqueous dispersion. In this reaction, poly(ethylene glycol) diacrylate was used as a crosslinker, polyvinylpyrrolidone was used as a stabilizer, and 2-methoxyethyl acrylate, poly(ethylene glycol)methyl ether acrylate, and 2-(methacryloyloxy)ethyl acetoacetate were used as copolymer monomers. The thermosensitive magnetic microgel displays a linear volume phase transition in water upon heating over a wide range of temperatures. Transmission electron microscopy, scanning electron microscopy, and dynamic light scattering were used to characterize the morphology and dimensions of the thermosensitive magnetic microgel. This material is expected to be used in magnetically targeted drug delivery systems that require linear drug release.

Abstract:The cellular microenvironment is composed of a dynamic hierarchical fibrillar architecture providing a variety of physical and bioactive signals to the surrounding cells. This dynamicity, although common in biology, is a challenge to control in synthetic matrices. Here, responsive synthetic supramolecular monomers were designed that are able to assemble into hierarchical fibrous structures, combining supramolecular fiber formation via hydrogen bonding interactions, with a temperature-responsive hydrophobic collapse, resulting in cross-linking and hydrogel formation. Therefore, amphiphilic molecules were synthesized, composed of a hydrogen bonding ureido-pyrimidinone (UPy) unit, a hydrophobic alkyl spacer, and a hydrophilic oligo(ethylene glycol) tail. The temperature responsive behavior was introduced by functionalizing these supramolecular amphiphiles with a relatively short poly(N-isopropylacrylamide) (PNIPAM) chain (M n ∼ 2.5 or 5.5 kg/mol). To precisely control the assembly of these monomers, the length of the alkyl spacer between the UPy moiety and PNIPAM was varied in length. A robust sol-gel transition, with the dodecyl UPy-PNIPAM molecule, was obtained, with a network elasticity enhancing over 2000 times upon heating above room temperature. The UPy-PNIPAM compounds with shorter alkyl spacers were already hydrogels at room temperature. The sol-gel transition of the dodecyl UPy-PNIPAM hydrogelator could be tuned by the incorporation of different UPy-functionalized monomers. Furthermore, we demonstrated the suitability of this system for microfluidic cell encapsulation through a convenient temperature sol-gel transition. Our results indicate that this novel thermoresponsive supramolecular system offers a modular platform to study and guide single-cell behavior.

Abstract:Hydrogels are a three-dimensional network material with a high equilibrium water content where chemical, physical, or biomolecular crosslinking systems have been used for the network formation. In this study, we report a thermosensitive cytogel of lactobionic acid/butanoic acid-conjugated poly(ε-l-lysine) (PKLC4). The thermogelation of the aqueous PKLC4 solution (3.5 wt %) was induced by partial dehydration accompanying a random coil-to-β-sheet transition of the polymer. During the sol-to-gel transition, the modulus increased from <0.05 Pa at <10 °C to 1300-1360 Pa at 37 °C. When HepG2 cells were incorporated into the PKLC4 solution, the gel modulus at 37 °C increased to 2300-2670 Pa. Moreover, the gel modulus was significantly affected by the cell type, population of the HepG2 cells, and live/dead states of the HepG2 cells. The cells proliferate better in the biointeractive PKLC4 thermogel than in the bioinert PEG-PA thermogel. To conclude, by combining thermosensitivity and specific binding of the receptor to the substrate, the hydrogel attained a high modulus without delay in gel time. This study provides new insights into hydrogel preparation in that substrate-receptor binding can be utilized as a crosslinking system to control the hydrogel modulus as well as a design principle for three-dimensional cache that improves cytocompatibility for cells.

Abstract:A triblock copolymer, poly(ethylene glycol)-poly(propylene glycol)--poly(ethylene glycol) was end-capped by acryloyl groups using a biodegradable oligolactide as spacer, and such biodegradable amphiphilic macromers could form micelles in water. A nanogel was prepared via polymerizing macromers in a micelle, and a macroscopic physical gelation was found upon heating a concentrated aqueous nanogel suspension. Such a sol-gel transition with a chemically crosslinked nanogel as the building block was thermoreversible. While the hydrogel affords a promising injectable biomaterial; this research reveals new physics of the thermogelling mechanism of amphiphilic block copolymers.
