Abstract:In vivo vibrational spectroscopic imaging is inhibited by relatively slow spectral acquisition on the second scale and low photon collection efficiency for a highly scattering system. Recently developed multiplex coherent anti-Stokes Raman scattering and stimulated Raman scattering techniques have improved the spectral acquisition time down to microsecond scale. These methods using a spectrometer setting are not suitable for turbid systems in which nearly all photons are scattered. We demonstrate vibrational imaging by spatial frequency multiplexing of incident photons and single photodiode detection of a stimulated Raman spectrum within 60 μs. Compared to the spectrometer setting, our method improved the photon collection efficiency by two orders of magnitude for highly scattering specimens. We demonstrated in vivo imaging of vitamin E distribution on mouse skin and in situ imaging of human breast cancerous tissues. The reported work opens new opportunities for spectroscopic imaging in a surgical room and for development of deep-tissue Raman spectroscopy toward molecular level diagnosis.

Abstract:Label-free vibrational imaging by stimulated Raman scattering (SRS) provides unprecedented insight into real-time chemical distributions. Specifically, SRS in the fingerprint region (400–1800 cm −1 ) can resolve multiple chemicals in a complex bio-environment. However, due to the intrinsic weak Raman cross-sections and the lack of ultrafast spectral acquisition schemes with high spectral fidelity, SRS in the fingerprint region is not viable for studying living cells or large-scale tissue samples. Here, we report a fingerprint spectroscopic SRS platform that acquires a distortion-free SRS spectrum at 10 cm −1 spectral resolution within 20 µs using a polygon scanner. Meanwhile, we significantly improve the signal-to-noise ratio by employing a spatial-spectral residual learning network, reaching a level comparable to that with 100 times integration. Collectively, our system enables high-speed vibrational spectroscopic imaging of multiple biomolecules in samples ranging from a single live microbe to a tissue slice.

Abstract:We report on the synchronization of two commercial picosecond Ti:sapphire lasers with unprecedented low temporal jitter between the pulse trains. Pulse jitter is reduced from a few picoseconds to 20 fs with a stability of several hours. The technology enabling the tight pulse synchronization is reviewed in this article. We demonstrate the usefulness of the synchronization scheme by applying the technique to coherent anti-Stokes Raman scattering (CARS) microscopy. It is shown that CARS images can be acquired with a significant improvement in signal-to-noise ratio. This level of performance brings the fluctuations of the CARS signal down to the fundamental photon shot-noise limit. We present detailed statistical analysis of the pulse jitter and CARS noise along with enhanced CARS vibrational images of polymer beads.

Abstract:Coherent anti‐Stokes Raman scattering performed with a synchronously pumped dual optical parametric oscillator allows to image high wavenumber and cell silent regions simultaneously without motion artefacts. Imaging was performed using an emulsion composed of water, heavy water, and oil, in addition to flowing plastic beads and deuterated HeLa cells. The system is sensitive enough to separate deuterated and regular lipids inside a human cell and allows to effectively perform SHG/THG microscopy due to built‐in group delay dispersion control. Coherent anti‐Stokes Raman scattering microscopy probing two independently selectable vibrational frequencies across the vibrational spectrum is demonstrated. A one‐box femtosecond laser source provides three synchronised 100–200 fs pulses of 77 MHz repetition rate, one at a fixed wavelength of 1025 nm from an Yb oscillator and two independently tuneable ones, over 680–960 nm and 960–1300 nm, respectively, from two optically synchronised parametric oscillators. Combined with spectral focusing, the source allows addressing the fingerprint, cell silent and C–H stretch vibrational regions simultaneously, avoiding the motion artefacts seen in sequential imaging. We demonstrate the microscopy method on water/heavy water/oil emulsions, plastic beads and deuterated lipids inside cells.

Abstract:Stimulated Raman scattering (SRS) microscopy allows label-free chemical imaging based on vibrational spectroscopy. Narrowband excitation with picosecond lasers creates the highest signal levels and enables imaging speeds up to video-rate, but it sacrifices chemical specificity in samples with overlapping bands compared to broadband (multiplex) excitation. We develop a rapidly tunable picosecond optical parametric oscillator with an electro-optical tunable Lyot filter, and demonstrate multicolor SRS microscopy with synchronized line-by-line wavelength tuning to avoid spectral artifacts due to sample movement. We show sensitive imaging of three different kinds of polymer beads and live HeLa cells with moving intracellular lipid droplets.

Abstract:Stimulated Raman Scattering microscopy allows label-free chemical imaging and has enabled exciting applications in biology, material science, and medicine. It provides a major advantage in imaging speed over spontaneous Raman scattering and has improved image contrast and spectral fidelity compared to coherent anti-Stokes Raman. Wider adoption of the technique has, however, been hindered by the need for a costly and environmentally sensitive tunable ultra-fast dual-wavelength source. We present the development of an optimized all-fibre laser system based on the optical synchronization of two picosecond power amplifiers. To circumvent the high-frequency laser noise intrinsic to amplified fibre lasers, we have further developed a high-speed noise cancellation system based on voltage-subtraction autobalanced detection. We demonstrate uncompromised imaging performance of our fibre-laser based stimulated Raman scattering microscope with shot-noise limited sensitivity and an imaging speed up to 1 frame/s.

Abstract:Coherent Raman microscopy provide label-free imaging by interrogating the intrinsic vibration of biomolecules. Nevertheless, trade-off between high chemical-specificity and high imaging-speed currently exists in transition from spectroscopy to spectroscopic imaging when capturing dynamics in complex living systems. Here, we present a novel concept in dual-comb scheme to substantially beat this trade-off and facilitate high-resolution broadband coherent anti-Stokes Raman spectroscopic imaging based on down-converted, automatically varying delay-time in spectral focusing excitation. A rapid measurement of vibrational microspectroscopy on sub-microsecond scale over a spectral span ~700 cm-1 with solid signal-to-noise ratio provides access to well-resolved molecular signatures within the fingerprint region. We demonstrate this high-performance spectroscopic imaging of spatially inhomogeneous distributions of chemical substances as well as the carotenoids accumulation in plant cells. This technique offers an unprecedented route for broadband CARS imaging with hundreds of kHz frame rate using available 1-GHz oscillators.

Abstract:Stimulated Raman scattering (SRS) microscopy is a powerful vibrational imaging technique with high chemical specificity. However, the insufficient tuning range or speed of light sources limits the spectral range of SRS imaging and, hence, the ability to identify molecular species. Here, we present a widely tunable fiber optical parametric oscillator with a tuning range of 1470 cm −1 , which can be synchronized with a Ti:sapphire laser. By using the synchronized light sources, we develop an SRS imaging system that covers the fingerprint and C–H stretching regions, without balanced detection. We validate its broadband imaging capability by visualizing a mixed polymer sample in multiple vibrational modes. We also demonstrate SRS imaging of HeLa cells, showing the applicability of our SRS microscope to biological samples.

Abstract:We present a synchronously pumped fiber optical parametric oscillator for coherent anti-Stokes Raman scattering microscopy. Pulses from a 1 mu m Yb-doped fiber laser are amplified and frequency converted to 779-808 nm through normal dispersion four-wave mixing in a photonic crystal fiber. The idler frequency is resonant in the oscillator cavity, and we find that bandpass filtering the feedback is essential for stable, narrow-bandwidth output. Experimental results agree quite well with numerical simulations of the device. Transform-limited 2 ps pulses with energy up to 4 nJ can be generated at the signal wavelength. The average power is 180 mW, and the relative-intensity noise is much lower than that of a similar parametric amplifier. High-quality coherent Raman images of mouse tissues recorded with this source are presented. (C) 2013 Optical Society of America

Abstract:Coherent anti-Stokes Raman scattering (CARS) spectroscopy with time-delayed ultrashort pulses and a single-pixel photodetector has shown great potential for spectroscopic imaging and transient studies in chemistry and biological research. However, those systems rely on mechanical delay lines or two asynchronous optical combs with inflexible repetition frequencies, technically limiting their acquisition speeds. Here, we demonstrate a hybrid dual-comb CARS system involving a broadband fiber laser and a highly-flexible, frequency-modulated electro-optic comb. We achieve multiplex CARS spectra (2800-3200 cm-1), with a moderate resolution (22 cm-1), at a maximum refresh rate of 1 MHz, limited by the radio-frequency synthesizer we use. Fast spectroscopic CARS imaging is demonstrated for liquid mixtures. Our system enables spectral measurements in the high-wavenumber C-H stretching region at a record speed that is an order of magnitude higher than state-of-the-art systems, which may open up new opportunities for fast chemical sensing and imaging.

Abstract:We present a simple and easily implementable scheme for multiplexed Coherent Anti-Stokes Raman Scattering (CARS) spectroscopy and microscopy using a single femtosecond pulse, shaped with a narrow spectral notch. We show that a tunable spectral notch, shaped by a resonant photonic crystal slab, can serve as a narrowband, optimally time-delayed probe, resolving a broad vibrational spectrum with high spectral resolution in a single-shot measurement. Our single-source, single-beam scheme allows the simple transformation of any multiphoton microscope with adequate bandwidth into a nearly alignment-free CARS microscope.

Abstract:The signal and idler beams from a picosecond, synchronously pumped optical parametric oscillator (OPO) provide the two colors necessary for coherent anti-Stokes Raman scattering (CARS) microscopy. The OPO provides a continuously tunable frequency difference between the two beams over a broad range of Raman shifts (100-3700 cm(-1)) by varying the temperature of a single nonlinear crystal. The near-infrared output (900-1300 nm) allows for deep penetration into thick samples and reduced nonlinear photodamage. Applications of this light source to in vivo cell and ex vivo tissue imaging are demonstrated.

Abstract:We demonstrate a robust, all-fiber, two-wavelength time-lens source for background-free coherent anti-Stokes Raman scattering imaging. The time-lens source generates two picosecond pulse trains simultaneously: one at 1064 nm and the other tunable between 1040 nm and 1075 nm (similar to 400 mW for each wavelength). When synchronized to a mode-locked Ti: Sapphire laser, the two wavelengths are used to obtain on-and off-resonance coherent anti-Stokes Raman scattering images. Real-time subtraction of the nonresonant background in the coherent anti-Stokes Raman scattering image is achieved by the synchronization of the pixel clock and the time-lens source. Background-free coherent anti-Stokes Raman scattering imaging of sebaceous glands in ex vivo mouse tissue is demonstrated. (C) 2016 Optical Society of America
