Celastrol Induces Apoptosis of HT29 Cells through Inhibiting miR-21 Regulation of Bcl-2 Expression

Abstract:We evaluated the apoptosis induction effects of celastrol in human colorectal cancer cell line HT29 via miRNA-medicated pathway. Microarray analysis was used to profile miRNA expressions in human colorectal adenocarcinoma HT29 cells treated with celastrol. The effect of celastrol on miR-21 expression was assessed by real-time PCR. MiR-21 mimics and inhibitor were transfected to HT29 cells, respectively. Next experiments were performed treating cells concomitantly with miR-21 mimics or inhibitor together with celastrol. Cellular growth inhibition was evaluated by MTT assay, and apoptosis was detected through Tdt-mediated dUTP nick end labeling (TUNEL) assay. Target gene of miR-21 was identified using dual luciferase reporter assay and confirmed with western blot analysis. Significant downregulation of miR-21 expression was seen in celastrol-treated HT29 cells. MiR-21 inhibitor led to a significant decrease of proliferation activity of HT29 cells, whereas miR-21 mimics exerted no significant effect on the proliferation of the transfected cells. The apoptotic cells were increased significantly following transfection of miR-21 inhibitor compared with inhibitor control. Moreover, miR-21 mimics significantly inhibited apoptosis of the cells. Bcl-2 was identified as a target of miR-21 using dual luciferase reporter assay and western blot analysis. In summary, our results demonstrated celastrol induce apoptosis of colorectal cancer cells by inhibiting miR-21 regulation of Bcl-2 expression.
