Neuroprotective Effects in PC12 Cells by Artemether, a Clinical Antimalarial Agent

Abstract:Artemether, a clinical antimalarial agent, has been shown for the first time with neuroprotective effects against sodium nitroprusside (SNP)-induced injuries in PC12 cells. It showed low cytotoxicity to PC12 cells at a concentration up to 30 mu M. Artemether significantly decreased intracellular reactive oxygen species (ROS) and cell apoptosis, preventing the declined of mitochondrial membrane potential induced by SNP. It up-regulated the phosphorylations of extracellular signal-regulated kinase (ERK1/2) and p38 mitogen-activated protein kinase (MAPK) in time-and dose-dependent manner. Consistent with the activation of two MAPKs, neuroprotective effects induced by artemether were inhibited by either the ERK specific inhibitor PD98059 or the p38 specific inhibitor PD163916, respectively. All the current data, together with results from other groups, indicate that artemether is a potential candidate to treat neurodegenerative diseases.
