Novel Lipophilic Jatrorrhizine: Synthesis, Antimicrobial, and Cytotoxic Activity Evaluation

Abstract:By introducing n-octyl group to C-13 and long carbon-chain alkyl groups to C-3 of jatrorrhizine, long carbon chain acyl groups to C-3 of jatrorrhizine, a series of novel lipophilic jatrorrhizines were synthesized and their structures were characterized by H-1 NMR, C-13 NMR and MS spectroscopy. All target compounds have been screened for their antimicrobial and cytotoxic activities in vitro in order to discuss their structure-activity relationships. The results indicated that most of these derivatives exhibited higher antimicrobial activities than that of jatrorrhizine with low MIC values ranging from 0.25 to 128 mu g/mL, which were comparable to or even better than those of the reference drugs jatrorrhizine and erythromycin, especially against Gram-positive bacteria and fungi. In addtion, in cytotoxic activity evaluation, most of these derivatives gave the most potent inhibiton on two hunan cancer cells (Hela and NCI-H460) than that of jatrorrhizine, in all cases the 3-O-lauroyljatrorrhizine and 13-octyloxy-3-hexanejatrorrhizine displayed the highest cytotoxic activity in all compounds, with IC50 values of 0.25 mu M for Hela and 0.37 mu M for NCI-H460, respectively. The cytotoxic activity of all compounds were stronger than that of jatrorrhizine. In our study, the antimicrobial and cytotoxic activities of compounds performance consistency. Too long or too short carbon chain substituents decreased activity. These results suggested that the presence of lipophilic substituents with moderate size might be crucial for the optimal antimicrobial and cytotoxic activities.
