Anti-Inflammatory Effect of Echinacea purpurea Polysaccharide (EPS) on the Pulmonary Macrophage cell line Raw264.7 in Mice

Abstract:Echinacea purpurea polysaccharide (EPS) has anti-inflammatory and immunomodulatory effects on macrophages and many cytokines involved in immune regulation or inflammatory response. To prove that EPS have inhibiting pneumonia effects on Raw264.7 cells and how these effects work, we used enzyme-linked immunosorbent assay, real-time fluorescence quantitative PCR and western blot assays. After Raw264.7 cells treatment with EPS 12 h, and stimulated by lipopolysaccharide(LPS) for 6 h, the concentrations and the relative mRNA expression level of TNF-alpha, IL-1 beta, IL-6 and IL-8 were increased compared with control group (p < 0.01, p < 0.05), while EPS group could significantly inhibited the increase of proinflammatory cytokines (TNF-alpha, IL-1 beta, IL-6 and IL-8) compared with LPS group (p < 0.05), while it could significantly inhibit the relative mRNA expression of proinflammatory cytokines (TNF-alpha and IL-1 beta). The western blot results showed that EPS promoted the protein levels of NF-kappa B-p65, p38, and inhibited the protein levels of p-ERK (p < 0.01), NF-kappa Bp-p65 (p < 0.05), p-p38 (p < 0.05), I kappa B-alpha, p-I kappa B-alpha, JNK, p-JNK, and ERK compared with LPS group. The effects of inflammatory response by EPS may be related to the effective inhibition the secretion of proinflammatory cytokines, the expression of NF-kappa B-pp65, and the activation of MAPK signaling pathway.
