Novel cognitive enhancer, ZSET1446 promotes neurogenesis in the hippocampal dentate gyrus and improves depressive behavior in the olfactory bulbectomized mice.

Abstract:Neurogenesis persistently occurs in the subgranular zone of the hippocampal dentate gyrus (DG) in adult brain. The impairment of neurogenesis in the DG leads to cognitive dysfunction and depressive behaviors in the olfactory bulbectomized (OBX) mice. OBX mice were orally administered ZSET1446 and investigated depressive behavior using a tail suspension test (TST) and neurogenesis in the DG using 5-bromo-2-deoxyuridine (BrdU) immunoreactivity. The ZSET 1446-treated mice showed a significant decreased immobility time in TST compared with saline-treated group. The chronic treatment with ZSET1446 increased the number of BrdU and NeuN double positive neurons in the DG compared to that in the saline-treated group. Furthermore, ZSET1446 treatment stimulated phosphorylation of ERK and Akt in the DG. These results suggest that treatment with ZSET1446 improves depression behavior with concomitant increased neurogenesis in the DG of the OBX mice.
