Facile Conjugation of BSA onto Bioabsorbable Iron via Mussel Adhesive Protein Inspired Coatings

Abstract:This research concentrated on improving the blood compatibility of the iron stent. Based on the self-polymerization and strong adhesion characteristic of dopamine in alkaline solution, the sheet iron were surface modified via simply immersing them into dopamine aqueous solution. Subsequently, BSA was immobilized onto the resultant membrane by the coupling between BSA and reactive functional groups on polydopamine layer. Field emission scanning electron microscopy (FESEM) and atomic force microscopy (AFM) were employed to investigate the changes in surface morphologies. Attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy were used to determine the chemical compositions of membrane surface. Contact angle was measured to characterize the hydrophilicity. The cell viability was discussed by MTT and the hemocompatibility was researched by Hemolysis test and platelet adhesion experiment. According to the results, the dopamine and BSA are immobilized to the iron surface, and the membranes have good biocompatibility. The most important is that the BSA modified surface has a good blood compatibility which may effectively reduce the occurrence of acute thrombosis.
