ANTIOXIDANT ACTIVITY OF BENZYLIDENE-SUBSTITUTED CURCUMIN DERIVATIVES

Abstract:To study the role of extending conjugated structures in antioxidant activity of curcumin and relative derivatives according to relative literature with some modification, different benzylidene substituents (benzylidene, 4-hydroxy benzylidene, 4-methoxy benzylidene and 3-methoxy-4-hydroxy benzylidene) were added into the central active CH2 group of curcumin by the Knoevenagel reaction. Four synthesised benzylidene substituted curcumin derivatives were characterised by H-1 NMR and ESI-MS. The antioxidant activity of the synthesised curcumin derivatives and curcumin was compared by DPPH free radical scavenging, reducing power assay, hemolysis test and H2O2-induced liver homogenate lipid peroxidation experiments. In most experiments (except DPPH free radical scavenging experiment), the curcumin derivatives with benzylidene substituents in the central active CH2 group showed better antioxidant activity than that of curcumin. That was to say, the extending conjugate structure was helpful to improve the antioxidant of curcumin derivatives. However, due to lack of active H-atom in central carbon atom of the heptadienone groups, the antioxidant activity of the synthesised curcumin derivative was poorer than that of curcumin in DPPH free radical scavenging experiment, the mechanism of which involved H-atom donation from the central active CH2 group in the heptadienone link of curcumin.
