Abstract:An efficient and practical phosphine-catalyzed vicinal difunctionalization of β-fluoroalkyl α,β-enones with TMSN3 has been developed. Using dppb as the catalyst, the reaction worked efficiently to yield various β-amino α-diazocarbonyl compounds in high yields (up to 94 %). This work marks the first efficient construction of α-diazocarbonyl compounds by phosphine catalysis. Meanwhile, the asymmetric variant induced by the nucleophilic bifunctional phosphine P4 led to various chiral fluoroalkylated β-amino α-diazocarbonyl compounds in high yields and enantioselectivity. NMR and ESI-MS studies support the existence of the key reaction intermediates. In contrast, β-azide carbonyl compounds would be furnished in good yields from β-fluoroalkylated β,β-disubstituted enones.

Abstract:We report a new thiourea-Bronsted acid cooperative catalytic system for the enantioselective cyanosilylation of aldehydes with yields up to 90% and enamioselectivities up to 88%. The addition of an achiral acid was found to be crucial for high asymmetric induction. Mechanistic investigations using a combination of NMR, ESI-MS, and density functional theory computations (including solvent corrections) at the M06/6-31G(d,p) level of theory suggest that the key catalytic species results from the cooperative interaction of bifunctional thioureas and an achiral acid that form well-defined chiral hydrogen-bonding environments.

Abstract:The catalytic potential of carbon nucleophiles has seldom been disclosed due to their reactivity toward carbon electrophiles to form stable carbon-carbon bonds, which are too strong to be cleaved for the expected catalytic cycles. We have developed an efficient catalytic cyanosilylation of carbonyl compounds with an in situ generated alkene/phosphine adduct, which is tuned not to couple with a carbonyl compound or an electron-deficient alkene under the reaction conditions when its nucleophilicity is translated into catalytic activity. In the presence of 3 mol% of methyl acrylate and 3 mol% of triphenylphosphine, a broad range of alkyl, alkenyl, and aryl ketones and aldehydes undergo cyanosilylation reaction with trimethylsilyl cyanide at room temperature to yield structurally diversified cyanohydrin silyl ethers in excellent yields. By using methyl acrylate/triphenylphosphine as a highly effective nucleophilic catalyst system for the cyanosilylation of carbonyl compounds, this study demonstrates a new concept for the development of useful organocatalysis utilizing the nucleophilicity of alkene/phosphine adducts generated in situ.

Abstract:Although recent progress has been made in introducing an SCF3 functionality into a variety of molecules, enantioselective trifluoromethylthiolation remains challenging, especially the -trifluoromethylthiolation of carbonyl compounds. The present study describes a diastereo and enantioselective Cu-catalyzed tandem 1,4-addition/trifluoromethylthiolation of acyclic enones. The tandem reaction enables the asymmetric integration of the -SCF3 group to carbonyl compounds, establishing chiral tertiary -carbon centers and affording -SCF3--substituted carbonyl compounds in 50-92% yields with up to 20:1 dr and 96% ee.

Abstract:We established a strategy for the copper‐catalyzed three‐component 1,4‐phosphinoylcyanation of 1,3‐enynes with diarylphosphine oxides and trimethylsilyl cyanide. The reaction produced various tetrasubstituted phosphinoyl‐ and cyano‐containing allenes with high functional group compatibilities in good yields under mild reaction conditions. A preliminary reaction mechanism involving the formation of phosphinoyl radicals and their regioselective addition to 1,3‐enynes was also proposed.

Abstract:A novel chiral biamide–phosphine multifunctional catalyst has been developed that mediates the asymmetric intermolecular Michael addition of β-carbonyl esters to β-trifluoromethyl enones and 3-aroyl acrylates in the presence of competing methyl acrylate and the inorganic base. This method provides a facile access to structurally diverse trifluoromethyl and quaternary stereogenic centers with excellent enantioselectivity (up to 99% ee) and good diastereoselectivity (up to 13:1 dr). The addition of the inorganic base (K3PO4) does not cause the background racemic reaction and enhances the reactivity by serving as a co-catalyst.

Abstract:Four unprecedented base-catalyzed/mediated nucleophilic additions of TMSCN to alpha-(trifluoromethyl)styrenes and 2-trifluoromethyl enynes were developed. The reaction proceeded smoothly at room temperature under mild and transition-metal-free conditions without affecting the trifluoromethyl group and afforded the corresponding CF3-containing alkyl, alkynyl, and butadienyl nitriles in moderate to excellent yields in a highly regioselective manner, respectively.

Abstract:A copper-catalyzed enantioselective 1,5-cyanotrifluoromethylation of vinylcyclopropanes has been developed using a radical relay strategy. This asymmetric reaction has demonstrated high enantioselective control, broad substrate scope, and mild conditions. Initiated by the in situ generated CF3 radical from Togni’s reagent, this method offers a new solution for remote enantioselective bifunctionalization of alkenes and thus provides a straightforward way for the synthesis of chiral CF3-containing internal alkenylnitriles.

Abstract:A novel copper-catalyzed radical cyanotrifluoromethylation has been achieved through a multicomponent reaction of isocyanides, Togni's reagent, and trimethylsilyl cyanides, affording trifluoroacetimidoyl nitriles in good yields. This reaction demonstrates a unique feature of merging two valuable functional groups-trifluoromethyl (CF) and cyan (CN)-onto the same C atom. The transformation proceeds by the initial addition of the CF radical to isocyanide and the subsequent intermolecular C-CN formation. The products can be successfully transformed to a series of CF-containing amines and imines that may serve in the synthesis of valuable pharmaceuticals and agrochemicals.

Abstract:Trifluoromethylated organic molecules are ubiquitous in pharmaceutics, materials science, and agrochemicals. In the past few decades, great efforts have been devoted to the development of convenient and efficient strategies for the synthesis of these compounds. β-Trifluoromethylated enones are readily available building blocks, which recognize as one of the attractive synthetic feedstock in organic transformations. Thus far, numerous new catalytic systems including novel chiral ligands and organocatalysts have been applied to asymmetric catalytic protocols using β-trifluoromethylated enones and various optically active compounds containing CF3 motif have been synthesized. Hence, this minreview describes the applications of β-trifluoromethylated enones in diverse asymmetric catalytic transformations since 2008, which incorporates the mechanistic aspects, involving the key mechanistic intermediates and the reaction pathways. Finally, the challenges of related fields and future perspectives are also pointed out.
