Abstract:An umpolung N-alkylation reaction of α-cyclopropyl α-iminothioesters with diethylaluminum chloride or ethylmagnesium bromide affords the corresponding N-ethylated α-aminothioesters in good yields. Subsequent oxidation and reaction of the N-ethylated product with a thiolate or a chloride anion proceed effectively to give the ring-opened products in good yields. In contrast, relatively "hard" nucleophiles did not give the ring-opened products but gave the addition products to the iminium carbon.

Abstract:Alkylation of iminomalonate with Grignard reagents followed by oxidation and allylation gave symmetrical quaternary α-amino diesters in good yields. Subsequent desymmetrization of a diol derivative from these products was conducted via asymmetric carbamylation catalyzed by Cu-Bnbox to give chiral quaternary aminodiol mono-carbamates.

Abstract:A general and transition-metal-free method for the construction of the carbazol-4-amine motif via a vinylogous Michael addition/cyclization/isomerization/elimination reaction of 3-nitroindoles with alkylidene malononitriles has been developed. This novel methodology allows the facile synthesis of a series of di- and trisubstituted carbazol-4-amine derivatives in moderate to good yields. A gram-scale experiment was successfully performed, highlighting the practicability of this method. Moreover, this strategy is also applicable to 3-nitrobenzothiophene, affording the corresponding dibenzo[ b, d]thiophen-1-amine derivatives in moderate yields.

Abstract:The switchable synthesis of 3-aminoindolines and 2'-aminoaryl acetic acids from the same substrates, 3-azido-2-hydroxyindolines, was developed through denitogenative electrophilic amination of Grignard reagents. The key to success is the serendipitous discovery that reaction conditions including solvents and reaction temperature can affect the chemoselectivity. It is notewhorthy that isotope-labeling experiments revealed the occurrence of the aziridine intermediate in the production of 2'-aminoaryl acetic acids.

Abstract:An efficient and general protocol for copper-catalyzed N-arylation of aliphatic amines and amino acids has been developed using aryl iodides under mild conditions (coupling temperature at 25-35 degrees C). For the N-(o-nitrophenyl) amino acid derivatives, subsequent reduction of the nitro group in the presence of tin(II) chloride resulted in 3,4-dihydroquinoxalin-2(1H)-one derivatives in good yields.

Abstract:We report a regioselective, nickel-catalyzed syn-1,2-carboamination of nonconjugated alkenyl carbonyl compounds with O-benzoyl hydroxylamine (N–O) electrophiles and aryl/alkylzinc nucleophiles to afford β- and γ-amino acid derivatives. This method enables preparation of products containing structurally diverse tertiary amine motifs, including heterocycles, and can also be used to form quaternary carbon centers. The reaction takes advantage of a tethered 8-aminoquinoline directing group to control the regiochemical outcome and suppress two-component coupling between the N–O electrophile and organozinc nucleophile.

Abstract:Herein, the construction of 3-indolemalononitriles via base-promoted tandem decarboxylative propargylation/aromatization cascade of 4-ethynyl benzoxazinones with N,N -disubstituted aminomalononitriles has been developed. This protocol features mild reaction conditions, high efficiency and no need of transition metals. Various 3-indolemalononitriles can be obtained in moderate to excellent yields from readily available starting materials. More importantly, this method can be further applied to the synthesis of a wide range of complex indole products.

Abstract:The amide bond is one of nature’s most common functional and structural elements, as the backbones of all natural peptides and proteins are composed of amide bonds. Amides are also present in many therapeutic small molecules. The construction of amide bonds using available methods relies principally on dehydrative approaches, although oxidative and radical-based methods are representative alternatives. In nearly every example, carbon and nitrogen bear electrophilic and nucleophilic character, respectively, during the carbon–nitrogen bond-forming step. Here we show that activation of amines and nitroalkanes with an electrophilic iodine source can lead directly to amide products. Preliminary observations support a mechanism in which the polarities of the two reactants are reversed (German, umpolung) during carbon–nitrogen bond formation relative to traditional approaches. The use of nitroalkanes as acyl anion equivalents provides a conceptually innovative approach to amide and peptide synthesis, and one that might ultimately provide for efficient peptide synthesis that is fully reliant on enantioselective methods.

Abstract:As an alternative metal-free amination, we report a simple and efficient annulation reaction of 1-(3,3,3-trifluoro-2-oxopropyl)pyridin-1-ium bromide and α,β-unsaturated carbonyl compounds with NH4OAc or amines. With the developed protocol, a series of 3-trifluoromethyl aniline derivatives as the major products were obtained in good to excellent yields under Kröhnke pyridine synthesis conditions. The reaction proceeds via a cascade process involving the 1,4-Michael addition of the pyridinium ylide to α,β-unsaturated carbonyl compound, followed by intramolecular addition of carbanion to the keto carbonyl group to form a dienone intermediate.

Abstract:The first catalytic cycloamidination of aminoalkenes with nitriles has been achieved by using rare-earth complexes. This reaction is equivalent to the desired intramolecular hydroamination of alkenylamidines, and allows a new direct access to substituted 2-imidazolines and tetrahydropyrimidines in high yields under operationally simple reaction conditions. Moreover, the methodology is also efficient for synthesis of symmetric and unsymmetric bridged diimidazolines. Compared with the traditional stepwise-mediated synthetic approaches, the present method avoids the use of additives and harsh reaction conditions, and thus leads to a completely different product distribution. Mechanistic data suggest that the reaction involves the initial NH activation by lanthanide complex followed by nitrile insertion into a Ln-N bond to form an amidinate lanthanide intermediate which undergoes the cyclization.

Abstract:A nickel-catalyzed three-component carboamination of unactivated alkenes with organoboronic acids and anthranils has been achieved for the expedient synthesis of δ-aryl and γ-amino acid derivatives. The 8-aminoquinoline (AQ) directing group is crucial for the success of the reaction, and anthranil serves as an arylnitrene precursor in this conversion. This method features mild reaction conditions, good chemo- and regioselectivity, and a broad substrate scope with good functional group tolerance.

Abstract:An efficient approach was developed for the alpha-arylation of imino amides with arylboronic acids. Different substrates were examined for this arylation reaction. For the a-arylation of N-phenylimino amide.. due to the electron-withdrawing properties of the phenyl group, the tautomerization between the amide and the iminol is more difficult. Thus, low reaction rate and low conversion were observed. This phenomenon supported our previously proposed mechanism for the arylation of alpha-amino acid derivatives. Meanwhile, this method provides an alternative approach for the synthesis of alpha-functionalized glycine derivatives.
