Abstract:A new SEK15-derived polyketide compound, strepolyketide D (1), was isolated from salt-lake-derived Streptomyces sp. DBC5, together with two known analogues (2-3). Their structures were elucidated based on spectroscopic analysis of IR, MS, 1 D and 2 D NMR. Compound 2 elicited moderate antioxidation with IC50 value of 39.26 μg/ml. The results of the study revealed that salt-lake actinomycetes of Lake Dabancheng appear to have immense potential as a source of polyketide compounds.

Abstract:Chemical investigation of the EtOAc extract of an insect-associated fungus Letendraea sp. 5XNZ4-2 cultured in Potato Dextrose Broth (1/2 PDB) medium lead to the isolation of three new polyketides, named letendronol D (1), phomopsiketones H-I (2-3). The structures of new compounds were elucidated by the analysis of HRESIMS and NMR spectroscopic data, and the absolute configurations were determined by modified Mosher’s method, ECD calculation and single-crystal X-ray diffraction. Cytotoxicity and antibacterial activities of 1 were assayed and regrettably 1 didn’t display any cytotoxicity and antibacterial activity. 3 was the first phomopsiketone derivative obtaining the lactone.

Abstract:Three novel SEK15-derived polyketides, strepolyketides A-C (1-3), and the known SEK15 (4) were identified from Streptomyces sp. HN2A53. Their structures were established by comprehensive 1D and 2D NMR spectroscopic analysis and HRESIMS data. Compounds 2-3 were the unique dimers of SEK15. Abstract:Marine-derived fungi have been reported to have great potential to produce structurally unique metabolites. Our investigation on secondary metabolites from marine-derived fungi resulted in the isolation of seven new polyketides (phomopsiketones D–G (1–4) and letendronols A–C (5–7)) as well as one known xylarinol (8) in the cultural broth of Letendraea sp. Their structures and absolute configurations were elucidated using a set of spectroscopic and chemical methods, including HRESIMS, NMR, single-crystal X-ray diffraction, ECD calculation, and a modified version of Mosher’s method. Compound 2 showed weak inhibition against nitric oxide production in lipopolysaccaride-activated macrophages with an IC50 value of 86 μM.

Abstract:Three new polyketide metabolites, the twelve-membered macrolides (6R,12S)-6-hydroxy-12-methyloxacyclodoecane-2,5-di one (1), (10S,12S)-10-hydroxy-12-methytoxacyclododecane-2,5-dione (2), and 4,5-dihydroxy-12-methytoxacyclododecan-2-one (3), were isolated from the endophytic fungal strain Cladosporium tenuissimum LR463 of Maytenus hookeri, together with three known compounds, cladospolide A, cladospolide B, and isocladospotide B. Their structures were elucidated by spectroscopic analysis including 1D- and 2D-NMR experiments, and the absolute configurations of 1. and 2 were determined by the Mosher ester method.

Abstract:From the branch tissue of Maytenus hookeri, the endophytic strain Lz531 was isolated, and determined to belong to Streptomyces, according to its 16S rRNA sequence. From the extracts of the fermentation broth of Streptomyces sp. Lz531, two new and four known compounds were isolated. The two new compounds were identified as cyclo(L-PrO-L-Val-L-Val) (1) and 13-methyl-N-(2-phenylethyl)tetradecanamide (2).

Abstract:A series of polyketide-originated metabolites (1-5) were isolated from a marine sponge-derived fungus Mycelia sterilia. Of these, 1-3 were new compounds. Their structures were elucidated by spectroscopic methods as (4R*, 5S*, 6S*, 8S*, 13R*)-1-(2,8-dihydroxy-1,2,6-trimethyl-1,2,6,7,8,8a-hexahydro-naphthalen-1-yl)-3-methoxy-propan-1-one (1), 4,8-dihydroxy-7-(2-hydroxy-ethyl)-6-methoxy-3,4-dihydro-2H-naphthalen-1-one (2) and 1-methyl-naphthalene-2,6-dicarboxylic acid (3). In 1, the proton-proton long-range coupling phenomenon claimed attention and was discussed.

Abstract:Five new polyketides, including two chromones (1–2), two phenyl derivatives (4–5), and a tandyukusin derivative (6), along with five known polyketides (3 and 7–10) were isolated from mangrove endophytic fungus Trichoderma lentiforme ML-P8-2. The planar structures of compounds were elucidated via detailed 1D, 2D NMR, and HR-ESI-MS analysis. ECD spectra, optical rotation values calculation, and alkali hydrolysis were applied in the determination of the absolute configuration of the new compounds. In bioassays, 6 and 9 exhibited promising antifungal activities against Penicillium italicum, with an MIC value of 6.25 μM for both compounds. Moreover, 3 displayed moderate AChE inhibitory activity with an IC50 value of 20.6 ± 0.3 μM.

Abstract:A new polyketide named (1R,2E,4S,5R)-1-[(2R)-5-oxotetrahydrofuran-2-yl]-4,5-dihydroxy-hex-2-en-1-yl(2E)-2-methylbut-2-enoate (1), along with eight known polyketide including one monoterpene (2), three linear furanopolyketides (3-5) and four lovastatin analogues (6-9), was isolated from the endophytic fungal strain Diaporthe sp. SXZ-19 of Camptotheca acuminate. The chemical structures of compounds 1-9 were elucidated on the basis of extensive spectroscopic analyses, including FT-ICR-MS, IR and 1D and 2D NMR experiments. The in vitro cytotoxicity of 1 against the human colon cancer cell line HCT 116 was evaluated but showed no evident activity.

Abstract:Four new polyketides, heterocornol Y (1), stemphyindan (2), pestalospirane C (3), and stemphyspyrane (4), along with five known ones (5–9) were isolated from the endophytic fungus Stemphylium globuliferum 17035 (SG17035) based on the One Strain Many Compounds (OSMAC) strategy allied with an LC-MS approach. These structures were elucidated through extensive spectroscopic analyses, single-crystal X-ray diffraction, and 13C NMR-DP4 analysis. Pestalospirane C (3) and stemphyspyrane (4) featured unprecedented spiroketal skeletons. In addition, the putative biosynthetic logic for compounds 1–4 was proposed. Antibacterial and cytotoxic activities of compounds 1–9 were evaluated. Stemphyspyrane (4) displayed promising antibacterial activity against different pathogens, especially against Staphylococcus aureus, Porphyromonas gingivalis, and methicillin-resistant Staphylococcus aureus (MRSA) with MIC values of 3.125 μM, 6.25 μM, and 12.5 μM, respectively. It is promising as an antibacterial agent for further optimization.

Abstract:From solid cultures of the biologically important endophyte Streptomyces species LR4612, cultivated on Maytenus hookeri, four new and two known compounds were isolated. The new compounds were identified as (2S*,3S*)‐5‐amino‐3‐hydroxy‐5‐oxopentan‐2‐yl 3‐(formylamino)‐2‐hydroxybenzoate (1), N‐[(3R*,4R*)‐3‐amino‐3,4‐dihydro‐4‐methyl‐2,6‐dioxo‐2H,6H‐1,5‐benzodioxocin‐10‐yl]formamide (2), (5β,6α)‐6,11‐dihydroxyeudesmane (3), and 5‐(6,7‐dihydroxy‐6‐methyloctyl)furan‐2(5H)‐one (4); the known compounds were elucidated as sorbicillin (5) and N‐acetyltyramine (6). The structures were established by HR‐ESI‐MS and in‐depth NMR analyses.

Abstract:A new polyketide, 9(E)-3-(5,8-dihydroxynon-3-en-1-yl)-5,6-dihydroxy-2,6-dimethylcyclohex-2-enone (1), and one known compound, 5-chloroisorotiorin (2), were isolated from the mangrove-derived fungus Phomopsis sp. TJM1-5. Their structures were determined by extensive NMR and MS analyses, while the absolute configurations of C-5 and C-6 were established based on experimental and calculated electronic circular dichroism spectra. Compound 2 was isolated from the genus of Phomopsis for the first time. Compounds 1 and 2 were evaluated for their antibacterial activity against five pathogenic bacteria.

Abstract:Three new 6,6-spiroketal polyketides, streptospirodienoic acids A (1), B (2) and streptospirodienoate A (3), were obtained from the fermentation broth of Streptomycetes sp. (no. 0950134). Their structures were constructed by one 6,6-spiroketal core coupled with hexadienoic acid, in which the starter unit might be 2-methylpropionyl-CoA. Their chemical structures were determined by detailed analyses of UV, IR, HRESIMS, 1D, 2D-NMR spectroscopic data and the X-ray crystallography technique. The biosynthetic pathway for 1 and 2 was proposed. Compound 3 showed moderate cytotoxic activity against HCT-116 cells (IC50 = 5.2 mu M), while compounds 1 and 2 showed no activity (IC50 > 10.0 mu M).

Abstract:Three new polyketides 4,6,8-trihydroxy-5-methyl-3,4-dihydronaphthalen-1(2H)-one (1), 5,7-dihydroxy-3-(1-hydroxyethyl)-3,4-dimethylisobenzofuran-1(3H)-one (2) and 1-(4-hydroxy-6-methoxy-1,7-dimethyl-3-oxo-1,3-dihydroisobenzofuran-1-yl) ethyl acetate (3) together with seven known analogues (4-10) were isolated from desert endophytic fungus Paraphoma sp. The structures of these compounds were elucidated by analysis of NMR data. The absolute configuration of (1-3) was established on the basis of CD experiments. The possible biosynthetic pathway of compounds (1-10) was suggested, which implied that these secondary metabolites might be originated from polyketide biosynthesis with different post-modification reactions. Compounds 2, and 5-8 were evaluated for bioactivities against plant pathogen A. solani, whereas none of them displayed any biological effects. In addition, compounds 1, 2 and 5-10 were also tested for cytotoxic activities against three human cancer cell lines (HepG2 cells, MCF-7 cells and Hela cells) without biological effects.

Abstract:Four new polyketide decalin derivatives, penicisteck acids A–D (1–4), and three new nitrogenous compounds (7–9), together with eight previously reported compounds, were isolated from the mangrove endophytic fungus Penicillium steckii SCSIO 41025. Their structures and absolute configurations were characterized by analysis of comprehensive spectroscopic data combined with ECD calculations and modified Marfey's method. Structurally, compounds 1–3 were highly oxygenated decalin derivatives harboring an unusual propanoic acid unit at C‐1. Compound 8 was identified as a rare natural alkaloid‐N‐oxide confirmed by single crystal X‐ray diffraction analysis. A plausible biogenetic pathway of compounds 1–6 is proposed. Compounds 13 and 15 showed significant anti‐bacterial activity against Staphylococcus aureus with the MIC values of 3.9 and 7.8 μg/mL, respectively. Compounds 10 and 12 exhibited potent α‐glucosidase inhibitory activity with the IC50 values of 64.1 and 191.4 μM, respectively.This article is protected by copyright. All rights reserved.

Abstract:Three new polyketides, (2 S )-2,3-dihydro-5,6-dihydroxy-2-methyl-4 H -1-benzopyran-4-one ( 1 ), (2′ R )-2-(2′-hydroxypropyl)-4-methoxyl-1,3-benzenediol ( 2 ), and 4-ethyl-3-hydroxy-6-propenyl-2 H -pyran-2-one ( 3 ) were isolated from the culture broth of Colletotrichum gloeosporioides , an endophytic fungus derived from the mangrove Ceriops tagal . The structures of 1 – 3 were elucidated on the basis of NMR spectra and HR-ESI-MS data. Their absolute configurations were determined by comparing with the experimental and calculated ECD spectrum. Compounds 1 and 3 showed potent antibacterial activities against some of the tested microbes.

Abstract:Mayamycin B, a new antibacterial type II polyketide, together with its known congener mayamycin A, were isolated from Streptomyces sp. 120454. The structure of new compound was elucidated by extensive spectroscopic analysis and comparison with literature data. Sequencing and bioinformatics analysis revealed the biosynthetic gene cluster for mayamycins A and B.

Abstract:Two polyketides were isolated from the mangrove endophytic fungus, Penicillium sp. sk14JW2P, and one derivative with a modified structure. The structures were elucidated by spectroscopic analyses, the structure of the compound 1 was further confirmed by single-crystal X-ray diffraction, and its absolute configuration was determined. Compound 1 and 2 showed acetylcholinesterase (AchE) inhibitory activities with IC50 values of 12±0.3 and 79±2 nM, respectively.

Abstract:A total of seven compounds were isolated from the ISP3 agar cultures of a soil-derived Streptomyces sp. S045 strain. Their structures were determined based on 1D, 2D NMR spectroscopic data, HR ESI mass spectroscopy, X-ray diffraction analysis and comparison with the reported data. The new compounds were identified to be ( S )-4-(1-hydroxyethyl)quinoline-2-carboxamide ( 1 ) and methyl 4-(hydroxymethyl)-2-(4-methylpentyl)-4,5-dihydrofuran-3-carboxylate ( 3 ), respectively. Their anti-bacterial and anti-type III secretion system (T3SS) activities were evaluated.

Abstract:In previous work, a series of bioactive natural products had been isolated from the plant endophytic Streptomyces sp. CS, which was isolated from Maytenus hookeri. To mine new active metabolites, we describe introducing an alien carbamoyltransferase (asm21) gene into the strain CS by conjugal transfer. As a result, three recombinatorial mutants named CS/asm21-1, CS/asm21-2 and CS/asm21-4 were successfully constructed. Three mutants and wild type CS were cultured on solid medium, and the extracts were detected and analyzed by liquid chromatography-mass spectrometry (LC-MS). The LC-MS profiles showed several unknown peaks that were present in the spectra of extracts of the CS/asm21-4 cultured on oatmeal solid medium. Then, three new naphthomycins O-Q (1-3), a new macrolide hookerolide (4) as well as nine known compounds were obtained from the solid cultured medium. Their structures were identified by spectra data. These new compounds showed moderate antimicrobial activities.
