Abstract:Water disinfection of E .coli by high frequency pulsed low electric fields was investigated .The effects of constructional ,electrical and aqueous parameters on disinfection efficiency and energy efficiency were evaluated .Experiments were performed with a 500 mL cylindrical reactor .Average electric intensity of 3‐12 kV/cm was applied and pulsed frequency was up to 20 000 Hz .Optimized structure of high voltage electrode were six equally distributed mesh electrodes with diameter of 40 mm .Both the disinfection efficiency and energy efficiency were enhanced by increasing the electric strength and pulse duration .Water conductivity could influence the pulsed waveforms and single pulse energy ,which changed the disinfection results . Reducing the initial bacterial density made the process more effective and easily realized sterilization .Pulsed frequency did not show any effect on disinfection ,but high frequency could greatly reduce treatment time .When pulsed voltage ,pulsed width ,water conductivity and initial bacteria density were 6 kV ,30 μs ,2 .5 μS/cm and 103‐106 cfu/mL ,respectively ,1‐2 logarithmic reduction were obtained with energy consumption less than 200 J/mL and water temperature lower than 40 ℃ .If initial bacterial density was less than 1 250 cfu/mL ,all the cells could be sterilized ,which was a non‐thermal processing .

Abstract:In this paper, a new multiple-needle-cylinder reactor is designed to study the Escherichia coli inactivation with an unstable pulsed arc discharge in water and investigate the influence of several parameters for promoting its industrial application. The results show that the inactivation rate is mainly determined by energy density. About three orders reduction can be obtained with an initial cell count of 3 × 105 CFU/mL and energy density around 2 J/mL. The inactivation rate can almost reach 100% when the energy density is around 8 J/mL. For reaching the maximum inactivation rate of this system, the parameters including initial water conductivity below 50 μS/cm, initial cell count over 104 CFU/mL, and bubbling flow rate around 3.5 L/min are recommended. The water conductivity, pH value, and concentrations of hydrogen peroxide and ozone are also measured during the treatment to evaluate their influence. The transmission electron microscopy images of E. coli indicate that the intracellular morphology changes after plasma treatment.
Fresh Water Disinfection By Pulsed Low Electric Field

Abstract:In this paper, we describe a pulsed low electric field process for water disinfection. Electric intensity of 0.6-1.7 kV cm(-1) is applied. Experiments are performed with a 1.2 L axis-cylinder reactor. A bipolar pulsed power source with pulsed width of 25 mu s and frequency of 100-3000 Hz is used. Water conductivity of 3-200 mu s cm(-1) is investigated, which can significantly affect pulsed voltage-current waveforms and injected energy. Energy per pulse rises with increased water conductivity. The initial E. Coli density and water conductivity are two major factors influencing the disinfection. No disinfection effect is performed with deionized water of 3 mu s cm(-1). When water conductivity is 25 mu s cm(-1) and bacteria density is 104(-1)06 cfu ml(-1), significant disinfection effect is observed. More than 99% of the cells can be disinfected with an energy density of less than 70 J ml(-1), while water temperature is below 30..

Abstract:To ensure the safety of freshwater which is the priority concern of the general public, it is necessary to inactivate pathogens effectively using innovative methods to disinfect freshwater. A new perspective for disinfection application of slightly acidic electrolyzed water was proposed in this study, namely the disinfection methods of "liquid-liquid". The effects of freshwater disinfection using slightly acidic electrolyzed water by means of continuously dynamic hybrid was evaluated. Under the different conditions of mixture, parameters variation characteristics of slightly acidic electrolyzed water including available chlorine concentration, pH and oxidation-reduction potential showed small fluctuations within a certain range and remained relatively stable. Compared with static mixture, the method of continuously dynamic hybrid had better sterilization effect. Under condition of static mixture, the disinfection effect of slightly acidic electrolyzed water to freshwater followed more sophisticated dynamic model. At different volume ratios, the correlation coefficients (R-2) were all greater than 0.9, which means that the model has good prediction effect. Under condition of dynamic hybrid, the variation of available chlorine concentration kept in a dynamic balance, which is in conformity with the zero-order kinetics model. The disinfection effect of slightly acidic electrolyzed water to freshwater followed first-order kinetic model, which also has better forecasting effect. The increase of temperature within limits contributed to promoting disinfection effect of slightly acidic electrolyzed water. But the disinfection effect became worse along with the increase of turbidity. The Escherichia coli (E. coli) inactivation effect under condition of mixture rendered slightly acidic electrolyzed water a promising disinfectant, which can be applied in freshwater to control natural microbiota. Abstract:Conventional water disinfection methods such as chlorination typically involve the generation of harmful disinfection byproducts and intensive chemical consumption. Emerging electroporation disinfection techniques using nanowire-enhanced local electric fields inactivate microbes by damaging their outer structures without byproduct formation or chemical dosing. However, this physical-based method suffers from a limited inactivation efficiency under high water flux due to an insufficient contact time. Herein, we integrate electrochlorination with nanowire-enhanced electroporation to achieve a synergistic flow through process for efficient water disinfection targeting bacteria and viruses. Electroporation at the cathode induces sub-lethal damages on the microbial outer structures. Subsequently, electrogenerated active chlorine at the anode aggravates these electroporation-induced injuries to the level of lethal damage. This sequential flow-through disinfection system achieves complete disinfection (> 6.0-log) under a very high water flux of 2.4 x 10(4) L/(m(2) h) with an applied voltage of 2.0 V. This disinfection efficiency is 8 times faster than that of electroporation alone. Further, the specific energy consumption for the disinfection by this novel process is extremely low (8 x 10(-4) kW h/m(3)). Our results demonstrate a promising method for rapid and energy-efficient water disinfection by coupling electroporation with electrochlorination to meet vital needs for pathogen elimination.

Abstract:An above water-pulsed nonthermal plasma technique for water disinfection is reported. Experiments are performed with a 2.5 L plasma reactor and Escherichia coli. When its initial number density is less than 10(6) cfu/mL, up to six orders reduction can be achieved within 0.5-2.5 min and at a plasma energy density of less than 2 J/mL. The disinfection performance is always effective by controlling water conductivity of below 1.5 mS/cm, which significantly affects plasma generation. Aqueous hydrogen peroxide, ozone, and nitric acid are continuously accumulated inside the reactor by the processing. Plasma generated UV radiation plays an important role in the cell inactivation. Inactivated cell morphology almost remains the same shape, their intracellular protein like green fluorescent protein, however, is destructed according to fluorescence observation. Abstract:More than 10% of the people in the world still suffer from inadequate access to clean water. Traditional water disinfection methods (e.g., chlorination and ultraviolet radiation) include concerns about the formation of carcinogenic disinfection byproducts (DBPs), pathogen reactivation, and/or excessive energy consumption. Recently, a nanowire-assisted electroporation-disinfection method was introduced as an alternative. Here, we develop a new copper oxide nanowire (CuONW)-modified three-dimensional copper foam electrode using a facile thermal oxidation approach. An electroporation-disinfection cell (EDC) equipped with two such electrodes has achieved superior disinfection performance (>7 log removal and no detectable bacteria in the effluent). The disinfection mechanism of electroporation guarantees an exceedingly low operation voltage (1 V) and level of energy consumption (25 J L(-1)) with a short contact time (7 s). The low operation voltage avoids chlorine generation and thus reduces the potential of DBP formation. Because of irreversible electroporation damage on cell membranes, no regrowth and/or reactivation of bacteria occurs during storage after EDC treatment. Water disinfection using EDCs has great potential for practical applications.

Abstract:Nanowire-assisted electroporation is regarded as an effective point-of-use disinfection technique for drinking water, and its performance under direct voltage (DV, U ) supply is limited by the poor bacterial inactivation on cathode and bacterial adsorption-fouling on anode. Herein, square-wave alternating voltage (SWAV, ± U ) was applied on nanowire electrodes to periodically reverse the cathodic/anodic polarity and their repulsions/attractions with bacterial cells, in turn promoting cell inactivation and fouling resistance. As compared with the gradual decrease in cell inactivation by the biofouling on nanowire anode under DV supply, SWAV supply enabled stable and efficient disinfection with ∼ 2 log higher cell inactivation and ∼ 3 times lower energy consumption (0.32–11.3 Wh/m 3 /log at 1.0–3.0 V) for various G- ( Escherichia coli and Acinetobacter schindleri ) and G+ ( Enterococcus faecalis and Bacillus cereus ) bacteria in deionized water, tap water and lake water. The cell morphology observation, flow cytometry analyses, and quenching experiments of reactive species revealed that electroporation on nanowire interfaces with locally enhanced electric field was the main mechanism for bacterial inactivation under voltages below 3.0 V. It was revealed that reversion of electrophoretic and electrostatic attractions under SWAV supply regulated cell migration, adsorption and desorption on nanowire interface, resulting in cell inactivation and fouling resistance on both nanowire electrodes. This work opens an effective strategy for stable and efficient nanowire-assisted electroporation disinfection.

Abstract:The inactivation of pathogens in liquids has broad applications, ranging from water disinfection to food pasteurization. However, common cell inactivation methods ( e.g ., chlorination, ultraviolet radiation and thermal treatment) have significant drawbacks such as carcinogenic byproduct formation, energy intensiveness and/or nutrient structure destruction. Here, we fabricated a new approach to address these challenges by applying a low-voltage electroporation disinfection cell (EDC) and investigate the critical mechanisms of cell transport to allow high inactivation performance. The EDC prototypes were equipped with two one-dimensional (1D) nanostructure-assisted electrodes that enabled high electric field strength (>107 V m −1 ) near the electrode surface with a low applied voltage (1 V). We have identified that during electroporation disinfection, electrophoresis, dielectrophoresis and hydraulic flow are the three major mechanisms which transport cells into the vicinity of the electrode surface to achieve superior disinfection performance. The EDC treated 70 ml of bacteria sample with an initial cell concentration of 10 7 CFU ml −1 and achieved complete bacteria inactivation (survival rate <0.00001%; no live bacteria detected). Our findings will help to establish a foundation for the future development and implementation of low-voltage electroporation for cell inactivation.
Tuning the Locally Enhanced Electric Field Treatment (LEEFT) between Electrophysical and Electrochemical Mechanisms for Bacteria Inactivation

Abstract:Efficient drinking water disinfection methods are critical for public health. Locally enhanced electric field treatment (LEEFT) is an antimicrobial method that uses sharp structures, like metallic nanowires, to enhance the electric field at tips and cause bacteria inactivation. Electroporation is the originally designed mechanism of LEEFT. Although oxidation is typically undesired due to byproduct generation and electrode corrosion, it can enhance the overall disinfection efficiency. In this work, we conduct an operando investigation of LEEFT, in which we change the electrical parameters to tune the mechanisms between electrophysical electroporation and electrochemical oxidation. Pure electroporation (i.e., without detectable oxidation) could be achieved under a duty cycle of ≤0.1% and a pulse width of ≤2 μs. Applying 2 μs pulses at 7-8 kV/cm and 0.1% duty cycle results in 80-100% bacteria inactivation with pure electroporation. A higher chance of oxidation is found with a higher duty cycle and a longer pulse width, where the antimicrobial efficiency could also be enhanced. For water with a higher conductivity, a higher antimicrobial efficiency can be achieved under the same treatment conditions, and electrochemical reactions could be induced more easily. The findings shown in this work improve the fundamental understanding of LEEFT and help optimize the performance of LEEFT in real applications.

Abstract:Disinfection is an essential process to protect people from waterborne pathogen infections. Conventional water disinfection methods (e.g., chlorination and ultraviolet radiation) have made considerable contributions to water safety, but are accompanied by obvious drawbacks such as carcinogenic byproduct formation, energy intensiveness, and/or regrowth of pathogens. Here, we develop an electroporation-disinfection cell (EDC) that enables fast water disinfection using a polydopamine-protected copper-phosphide-nanowire-modified copper foam (PDA-Cu3PNW-Cu) electrode powered by a high-frequency alternating current (AC; 10(6) Hz). Electroporation relies on a strong electric field to damage microorganism membranes without byproduct formation. The PDA film ensures a robust nanowire structure against the hydraulic detachment and reduces mechanical erosion. High-frequency AC as the power source can effectively eliminate electrochemical corrosion and extend the electrode lifetime. EDCs with PDA-Cu3PNW-Cu electrodes achieved complete disinfection (no live microorganism in the effluent) with a low voltage (peak voltage of 1 V) and a high flux (2 m(3) h(-1) m(-2)). Under such operating conditions, the PDA-Cu3PNW-Cu electrodes enable complete E. coli disinfection (>6.0log removal efficiency) for 15 days. To our knowledge, this is the highest value obtained to date for nanostructure-modified electrodes for low-voltage electroporation: 30 times the previously reported maximum lifetime.

Abstract:Airborne pathogens can widely spread and cause severe diseases in humans, concerning the risks of indoor exposure in hospitals, homes, and malls. Therefore, the inactivation of pathogens in indoor air environments is important. The inactivation of microorganisms in water by nanowire electroporation has been well studied in recent years. But the effectiveness of nanowire-modified electrodes for air disinfection is still unknown. In this study, a disinfection method for the quick inactivation of bacteria in air is reported. The copper meshes were modified with Cu(OH)2 nanowires by electrochemical method. A flow-through device with nanowire-modified electrodes inside was designed to generate, mix, and disinfect bioaerosols. 60-85% inactivation efficiency for bacteria and more than 2-log removal for viruses were achieved with a short air retention time of 0.3 s in high humidity (RH of 100%). Endospores showed a susceptibility to electric disinfection similar to that of the vegetable cells. The inactivation efficiency of bacteria in moderate humidity (RH of 40%) could be significantly improved up to 51% by coating with a high conductive material, poly(3,4-ethylenedioxythiophene):poly(styrene-4-sulfonate) (PEDOT:PSS). The main inactivation mechanism in air was demonstrated as electroporation by detecting membrane integrity. This novel disinfection method may be applied for the disinfection of air-transmitted pathogens in the future.

Abstract:Safe water scarcity occurs mostly in developing regions that also suffer from energy shortages and infrastructure deficiencies. Low-cost and energy-efficient water disinfection methods have the potential to make great impacts on people in these regions. At the present time, most water disinfection methods being promoted to households in developing countries are aqueous chemical-reaction-based or filtration-based. Incorporating nanomaterials into these existing disinfection methods could improve the performance; however, the high cost of material synthesis and recovery as well as fouling and slow treatment speed is still limiting their application. Here, we demonstrate a novel flow device that enables fast water disinfection using one-dimensional copper oxide nanowire (CuONW) assisted electroporation powered by static electricity. Electroporation relies on a strong electric field to break down microorganism membranes and only consumes a very small amount of energy. Static electricity as the power source can be generated by an individual person's motion in a facile and low-cost manner, which ensures its application anywhere in the world. The CuONWs used were synthesized through a scalable one-step air oxidation of low-cost copper mesh. With a single filtration, we achieved complete disinfection of bacteria and viruses in both raw tap and lake water with a high flow rate of 3000 L/(h.m(2)), equivalent to only 1 s of contact time. Copper leaching from the nanowire mesh was minimal.
Nanowire-assisted electroporation via inducing cell destruction for inhibiting formation of VBNC bacteria: Comparison with chlorination

Abstract:The induction of viable but nonculturable (VBNC) bacteria with cellular integrity and low metabolic activity by chemical disinfection causes a significant underestimation of potential microbiological risks in drinking water. Herein, a physical Co3O4 nanowire-assisted electroporation (NW-EP) was developed to induce cell damage via the locally enhanced electric field over nanowire tips, potentially achieving effective inhibition of VBNC cells as compared with chemical chlorination (Cl2). NW-EP enabled over 5-log removal of culturable cell for various G+/G- bacteria under voltage of 1.0 V and hydraulic retention time of 180 s, and with ∼3-6 times lower energy consumption than Cl2. NW-EP also achieved much higher removals (∼84.6 % and 89.5 %) of viable Bacillus cereus (G+) and Acinetobacter schindleri (G-) via generating unrecoverable pores on cell wall and reversible/irreversible pores on cell membrane than Cl2 (∼28.6 % and 41.1 %) with insignificant cell damage. The residual VBNC bacteria with cell wall damage and membrane pore resealing exhibited gradual inactivation by osmotic stress, leading to ∼99.8 % cell inactivation after 24 h storage (∼59.4 % for Cl2). Characterizations of cell membrane integrity and cell morphology revealed that osmotic stress promoted cell membrane damage for the gradual inactivation of VBNC cells during storage. The excellent adaptability of NW-EP for controlling VBNC cells in DI, tap and lake waters suggested its promising application potentials for drinking water, such as design of an external device on household taps.

Abstract:Abstract Water disinfection is a critical step in water and wastewater treatment. The most widely used chlorination suffers from the formation of carcinogenic disinfection by-products (DBPs) while alternative methods (e.g., UV, O 3 , and membrane filtration) are limited by microbial regrowth, no residual disinfectant, and high operation cost. Here, a nanowire-enabled disinfection method, locally enhanced electric field treatment (LEEFT), is introduced with advantages of no chemical addition, no DBP formation, low energy consumption, and efficient microbial inactivation. Attributed to the lightning rod effect, the electric field near the tip area of the nanowires on the electrode is significantly enhanced to inactivate microbes, even though a small external voltage (usually < 5 V) is applied. In this review, after emphasizing the significance of water disinfection, the theory of the LEEFT is explained. Subsequently, the recent development of the LEEFT technology on electrode materials and device configurations are summarized. The disinfection performance is analyzed, with respect to the operating parameters, universality against different microorganisms, electrode durability, and energy consumption. The studies on the inactivation mechanisms during the LEEFT are also reviewed. Lastly, the challenges and future research of LEEFT disinfection are discussed.

Here, we develop polypyrrole nanowire arrays (PPyNWs)-modified electrodes by polymerization of pyrrole on graphite felt for point-of-use water disinfection via low-voltage electroporation. A flow-through mode is specially applied to alleviate diffusion barrier of pyrrole in the porous graphite felt for uniform PPyNWs growth. The flow-through disinfection device using the optimized PPyNWs electrode achieves above 4-log removal for model virus (MS2) and gram-positive/negative bacteria (E. faecalis and E. coli) at applied voltage of 1.0 V and fluxes below 1000 and 2500 L/m<sup>2</sup>/h. Electroporation is recognized as the dominant disinfection mechanism by using square-wave alternating voltage of ±1.0 V to eliminate the electrochemical reactions. In-situ sampling experiments reveal that anode acts as the main disinfection function due to its electric field attraction with negatively charged E. coli cells. The live/dead baclight staining experiments indicate an adsorption-desorption process of E. coli cells on anode, and the adsorption-desorption balance determines the disinfection abilities of PPyNWs anode. Under 1.0 V and 2000 L/m<sup>2</sup>/h, the disinfection device enables above 4-log E. coli removal in tap water within 7-day operation with energy consumption below 20 mJ/L, suggesting its sound application potential for point-of-use water disinfection.

Abstract:Due to the limitations of traditional ultraviolet (UV) in microbial inactivation in water, it is necessary to explore a more suitable and efficient UV disinfection method. In this study, an electron beam excitation multi-wavelength ultraviolet (EBE-MW-UV) system was established and aims to analyze its differential microbial inactivation capabilities in comparison to single-wavelength UV-LEDs in waterborne applications. Furthermore, the inactivation mechanisms of this system on microorganisms were explored. The results showed that EBE-MW-UV had significantly higher inactivation effects on the Escherichia coli , Pseudomonas aeruginosa , Bacillus subtilis and Candida albicans in water compared to UV-LEDs (p<0.05), and the inactivation effect of EBE-MW-UV on Escherichia coli and Pseudomonas aeruginosa at the same UV dose was 3.8 and 1.9 log higher than that of UV-LEDs, respectively, EBE-MW-UV exhibited better inactivation effects on Gram-negative bacteria. Further research found that, under the majority of irradiation doses, neither EBE-MW-UV nor UV-LEDs were significantly affected by the concentration of suspended solids (5 and 20 mg/L) or humic acids (2 and 5 mg/L) in the water. Mechanism analysis revealed that during the disinfection process of EBE-MW-UV, microbial DNA and proteins were initially damaged, which prevented the occurrence of dark repair and led to bacterial inactivation. In addition, UV irradiation led to the production of additional reactive oxygen species (ROS) inside the cells, increasing cell membrane permeability and exacerbating membrane damage. This was accompanied by a decrease in energy metabolism and depletion of ATP, ultimately resulting in microbial inactivation. Therefore, EBE-MW-UV demonstrated more effective disinfection than single-wavelength UV-LEDs, showing great potential. Our research gives new insights into the characteristics of multiple wavelength ultraviolet, and provides scientific basis for the selection of new light sources in the field of ultraviolet disinfection.

Abstract:Water disinfection is conventionally achieved by oxidation or irradiation, which is often associated with a high carbon footprint and the formation of toxic byproducts. Here, we describe a nano-structured material that is highly effective at killing bacteria in water through a hydrodynamic mechanism. The material consists of carbon-coated, sharp Cu(OH)2 nanowires grown on a copper foam substrate. We show that mild water flow (e.g. driven from a storage tank) can efficiently tear up bacteria through a high dispersion force between the nanotip surface and the cell envelope. Bacterial cell rupture is due to tearing of the cell envelope rather than collisions. This mechanism produces rapid inactivation of bacteria in water, and achieved complete disinfection in a 30-day field test. Our approach exploits fluidic energy and does not require additional energy supply, thus offering an efficient and low-cost system that could potentially be incorporated in water treatment processes in wastewater facilities and rural communities.
