Abstract:Novel divergent domino annulation reactions of sulfur ylides with aryldiazonium tetrafluoroborates have been developed, affording various tetra- and trisubstituted pyrazole derivatives in moderate to good yields. Three molecules of sulfur ylides were applied as C1 synthon to construct the complex products with five new chemical bonds formed in these one-pot reactions.

Abstract:Pyridinium 1,4-zwitterionic thiolates were regarded as powerful and versatile building blocks to prepare nitrogen- and sulfur-containing heterocycles. Herein, we reported a copper-catalyzed formal [4 + 1] annulation of pyridinium 1,4-zwitterionic thiolates and diazo compounds without any additives to access a library of trifunctionalized indolizines in good yields. Besides, isoquinolinium 1,4-zwitterionic thiolates and imidazolium 1,4-zwitterionic thiolates were also applied to this formal [4 + 1] annulation reaction. Of particular note is that various functional groups such as -CO2R, -CO2NR2, -CF3, -CN, and -(O)P(OR)2 could be easily introduced into cycloaddition products indolizines by this strategy.

Abstract:Herein, an efficient, substrate-directed divergent [2+3]/[2+1] annulation of tetra-substituted oxa-dienes and allylic sulfur ylides has been successfully developed. Under precise annulation regulation, a series of functionalized bispirocyclopentane and bispirocyclopropane derivatives were synthesized in a highly stereoselective and economical manner. This protocol offers the advantages of mild conditions, high chemo-, regio- and diastereoselectivity and broad substrate compatibility.

Abstract:A rhodium-catalyzed denitrogenative formal (3 + 3) transannulation of 1,2,3-thiadiazoles with alk-2-enals is achieved, producing 2,3-dihydrothiopyran-4-ones in moderate to excellent yields. An inverse KIE of 0.49 is obtained, suggesting the reversibility of the oxidative addition of thioacyl Rh(i) carbenes to alk-2-enals. The late-stage structural modifications of steroid compounds are realized. Moreover, our studies show that thioacyl carbenes have different reactivities to those of α-oxo and α-imino carbenes, and highlight the importance of heteroatoms in deciding the reactivities of heterovinyl carbenes.

Abstract:Under the catalysis of Pd 2 (dba) 3 . CHCl 3 (5 mol%), dppp (20 mol%), K 2 CO 3 (1.0 eq), the formal [5 + 2] cycloaddition of vinyloxiranes with oxazol‐5‐(4 H )‐ones proceeded readily in THF at room temperature, thus furnishing the desired seven‐ membered lactones in 67‐96% yields. The chemical structure of the title compounds was firmly confirmed by X‐ray diffraction analysis .

Abstract:A diastereoselective and enantioselective formal [4+1] ylide annulation of chiral sulfonium salts with various substituted α‐substituted nitroalkene leading to optically active isoxazoline N‐oxides using CF3SO3− as a key anion has been explored. Abstract:Difluoroenoxysilane, a commonly used difluoroallylating reagent, has attracted considerable attention in recent years. However, its application in the annulation reaction for the construction of fluorinated heterocyclic compounds remains relatively limited. Presented here is the Bronsted acid-catalyzed efficient formal [4 + 2] annulation of difluoroenoxysilanes with alpha-cyano chalcones. The developed protocol demonstrates tolerance to various substituents under mild reaction conditions, providing a reliable approach to construct gem-difluoro-3,4-dihydro-2H-pyrans in good to excellent yields with high diastereoselectivities.

Abstract:We developed a novel method for the synthesis of 2‐substituted‐4,5‐dihydrothiazol‐4‐ols compounds through cyclization of thioamides and 1,4‐dithiane‐2,5‐diol in the presence of Et3N. Stable hydroxy‐substituted thiazole compounds were isolated of substituted thioamides. The reaction conditions are simple, the substrate range is wide, and the target compounds were obtained in up to 82% yield.

Abstract:The synthesis of 4-(trifluoromethyl)-2,3-dihydrothiazoles from alpha-enolic dithioesters and imidoyl sulfoxonium ylides has been achieved under thermal conditions. This transformation is catalyst-free, additive-free, and operationally simple, and it proceeds via a formal insertion of alkenyl S-H generated in situ from dithioesters into ylides followed by an intramolecular annulation. This approach further expands the synthetic application of two versatile sulfur-containing building blocks of fluorinated imidoyl sulfoxonium ylides and alpha-enolic dithioesters.

Abstract:Bromomalonate served as an efficient source of one‐atom synthons to react with benzofuran‐derived azadienes/oxadienes, or pyrazole‐derived oxadienes for the construction of corresponding spirocyclopropane products in the presence of alkaline via 1, 4‐addition induced dearomatization strategy. However, examples of the C1 synthons applied to thiazole‐derived oxadienes have yet to systematically investigated so far. Herein we report an inorganic base promoted spirocyclopropanation reaction between bromomalonate and thiazole‐derived oxadienes for the generation of various spirocyclopropylthiazole derivatives in high yields with excellent diastereoselectivities. A K2CO3 promoted spirocyclopropanation of thiazole‐derived oxadienes with bromomalonate via 1, 4‐addition‐induced dearomatization strategy has been developed. The pathway features a broad substrate scope, operational simplicity and high degree of synthetic efficiency to afford series of functionalized spiro‐cyclopropanation frameworks in 88 %–96 % yields, which enriches the research contents of thiazole‐derived oxadienes involved reactions. All products obtained herein contain two contiguous quaternary centers and their further synthetic applications are also demonstrated.

Abstract:Aiming to develop novel annulation reactions using sulfine intermediates, we have established a regioselective formal [2 + 3] cyclization reaction of α‐keto sulfines generated from the elimination of arylacyl sulfoxides and thioamides. This new strategy allows for the synthesis of thiazole‐5‐thiones directly without catalysts or additives. The approach features a step‐economic, one‐pot characteristics via a tandem sequence of in situα‐keto sulfines generation, regiospecifically nucleophilic attack or [3 + 2] cyclizatoin, dithioate (S=C−S−) skeletons formation, and subsequent intramolecular nucleophilic cyclization. This reaction also represents a novel method for constructing dithioate (S=C−S−) skeletons.

Abstract:A base promoted formal [4+1] cycloaddition of ortho- and para-quinone methides with 3-chlorooxindoles is reported to afford various functionalized 3,2′-tetrahydrofuryl spirooxindoles in high yields under mild conditions. An unexpected reversal in diastereoselectivity was observed from these two types of reactions. Computational studies have provided insight into the origin of diastereoselection, and different configurations of 1,4-addition and 1,6-addition at the oxindole carbanionic center are found to give rise to different diastereomers. The anti-tumor activity of these structurally unique spirooxindole derivatives is reported.

Abstract:A novel cyclization reaction of pyridinium 1,4-zwitterionic thiolates and propiolic acid derivatives mediated by triethylamine is described, which allows the facile synthesis of indolizines under mild reaction conditions. The net transformation involves an acetylide-driven formal [5 + 1] annulation reaction followed by a spontaneous ring-contraction/sulfur extrusion process of transient pyridothiazine intermediates.
