Abstract:Emotions can be defined as states elicited by rewards or punishments, and indeed the neurology of emotional disorders can be understood in terms of this foundation. The orbitofrontal cortex in humans and other primates is a critical area in emotion processing, determining the value of stimuli and whether they are rewarding or nonrewarding. The cortical processing that occurs before the orbitofrontal cortex primarily involves defining the identity of stimuli, i.e., "what" is present and not reward value. There is evidence that this holds true for taste, visual, somatosensory, and olfactory stimuli. The human medial orbitofrontal cortex is important in processing many different types of reward, and the lateral orbitofrontal cortex in processing nonreward and punishment. Humans with damage to the orbitofrontal cortex have an impaired ability to identify facial and voice expressions of emotions, and impaired subjective experience of emotion. They can have an altered personality and be impulsive because they are impaired at processing failures to receive expected rewards and at processing punishments. In humans, the role of the amygdala in the processing of emotions is reduced because of the great evolutionary development of the orbitofrontal cortex: amygdala damage has much less effect on emotion than does orbitofrontal cortex damage. The orbitofrontal cortex projects reward value information to the anterior cingulate cortex, which is involved in learning those actions required to obtain rewards and avoid punishments. The cingulate cortex thus provides an output route for emotional behavior. In depression, the medial orbitofrontal cortex has decreased connectivity and sensitivity to reward, and the lateral orbitofrontal cortex has increased connectivity and sensitivity to nonreward. The orbitofrontal cortex has major projections to the anterior cingulate cortex, including its subcommissural region, and the anterior cingulate cortex is also implicated in depression.

Abstract:The topics treated in The brain and emotion include the definition, nature, and functions of emotion (Ch. 3); the neural bases of emotion (Ch. 4); reward, punishment, and emotion in brain design (Ch. 10); a theory of consciousness and its application to understanding emotion and pleasure (Ch. 9); and neural networks and emotion-related learning (Appendix). The approach is that emotions can be considered as states elicited by reinforcers (rewards and punishers). This approach helps with understanding the functions of emotion, with classifying different emotions, and in understanding what information-processing systems in the brain are involved in emotion, and how they are involved. The hypothesis is developed that brains are designed around reward- and punishment-evaluation systems, because this is the way that genes can build a complex system that will produce appropriate but flexible behavior to increase fitness (Ch. 10). By specifying goals rather than particular behavioral patterns of responses, genes leave much more open the possible behavioral strategies that might be required to increase fitness. The importance of reward and punishment systems in brain design also provides a basis for understanding the brain mechanisms of motivation, as described in Chapters 2 for appetite and feeding, 5 for brain-stimulation reward, 6 for addiction, 7 for thirst, and 8 for sexual behavior.

Abstract:The orbitofrontal cortex and amygdala are involved in emotion and in motivation, but the relationship between these functions performed by these brain structures is not clear. To address this, a unified theory of emotion and motivation is described in which motivational states are states in which instrumental goal-directed actions are performed to obtain rewards or avoid punishers, and emotional states are states that are elicited when the reward or punisher is or is not received. This greatly simplifies our understanding of emotion and motivation, for the same set of genes and associated brain systems can define the primary or unlearned rewards and punishers such as sweet taste or pain. Recent evidence on the connectivity of human brain systems involved in emotion and motivation indicates that the orbitofrontal cortex is involved in reward value and experienced emotion with outputs to cortical regions including those involved in language, and is a key brain region involved in depression and the associated changes in motivation. The amygdala has weak effective connectivity back to the cortex in humans, and is implicated in brainstem-mediated responses to stimuli such as freezing and autonomic activity, rather than in declarative emotion. The anterior cingulate cortex is involved in learning actions to obtain rewards, and with the orbitofrontal cortex and ventromedial prefrontal cortex in providing the goals for navigation and in reward-related effects on memory consolidation mediated partly via the cholinergic system.

Abstract:This review begins with a brief historical overview of attempts in the first half of the 20th century to discern brain systems that underlie emotion and emotional behavior. These early studies identified the amygdala, hippocampus, and other parts of what was termed the ‘limbic’ system as central parts of the emotional brain. Detailed connectional data on this system began to be obtained in the 1970s and 1980s, as more effective neuroanatomical techniques based on axonal transport became available. In the last 15 years these methods have been applied extensively to the limbic system and prefrontal cortex of monkeys, and much more specific circuits have been defined. In particular, a system has been described that links the medial prefrontal cortex and a few related cortical areas to the amygdala, the ventral striatum and pallidum, the medial thalamus, the hypothalamus, and the periaqueductal gray and other parts of the brainstem. A large body of human data from functional and structural imaging, as well as analysis of lesions and histological material indicates that this system is centrally involved in mood disorders.

Abstract:An approach to depression is described, in which there is over-sensitivity of the lateral orbitofrontal cortex non-reward attractor system, and under-sensitivity of the medial orbitofrontal cortex reward system. Many functional connectivities involving the lateral orbitofrontal cortex are increased in depression, and many functional connectivities involving the medial orbitofrontal cortex are decreased in depression. Involvement of the orbitofrontal cortex in bipolar disorder, autism, attention-deficit hyperactivity disorder, and compulsivity including obsessive-compulsive disorder, is also described.

Abstract:The orbitofrontal cortex represents the reward or affective value of primary reinforcers including taste, touch, texture, and face expression. It learns to associate other stimuli with these to produce representations of the expected reward value for visual, auditory, and abstract stimuli including monetary reward value. The orbitofrontal cortex thus plays a key role in emotion, by representing the reward value of the goals for action. The learning process is stimulus-reinforcer association learning. Negative reward prediction error neurons are related to this affective learning. Activations in the orbitofrontal cortex correlate with the subjective emotional experience of affective stimuli, and damage to the orbitofrontal cortex impairs emotion-related learning, emotional behaviour, and subjective affective state. Top-down attention to affect modulates orbitofrontal cortex representations, and attention to intensity modulates representations in earlier cortical areas that represent the physical properties of stimuli. Top-down word-level cognitive inputs can bias affective representations in the orbitofrontal cortex, providing a mechanism for cognition to influence emotion. Whereas the orbitofrontal cortex provides a representation of reward or affective value on a continuous scale, areas beyond the orbitofrontal cortex such as the medial prefrontal cortex area 10 are involved in binary decision-making when a choice must be made. For this decision-making, the orbitofrontal cortex provides a representation of the value of each specific reward on the same scale, with no conversion to a common currency. Increased activity in a lateral orbitofrontal cortex non-reward area provides a new attractor-related approach to understanding and treating depression. Consistent with the theory, the lateral orbitofrontal cortex has increased functional connectivity in depression, and the medial orbitofrontal cortex, involved in reward, has decreased functional connectivity in depression.

Abstract:It is shown that emotions can usefully be considered as states produced by reinforcing stimuli. The ways in which a wide variety of emotions can be produced, and the functions of emotion, are considered. There is evidence that the amygdala is involved in the formation of stimulus-reinforcement associations, and the orbitofrontal cortex with correcting behavioural responses when these are no longer appropriate because previous reinforcement contingencies change. This evidence comes from the effects of damage to these structures, and from recording the activity of single neurons in these structures in the monkey during the formation and disconnection of stimulus-reinforcement associations. In so far as emotions can be defined as states produced by reinforcing stimuli, then the amygdala and orbitofrontal cortex are seen to be of great importance for emotions, in that they are involved respectively in the elicitation of learned emotional responses, and in the correction or adjustment of these emotional responses as the reinforcing value of environmental stimuli alters. One of the theses advanced is that the changes in emotional behaviour produced by damage to the brain can be analysed and understood by considering how different parts of it function in reinforcement and in the formation and disconnection of stimulus-reinforcement connections. Another thesis is that there is a population of neurons in the amygdala and parts of the temporal lobe visual cortex specialised to respond to faces, and that these neurons may be involved in social and emotional resposes to faces. Some of the outputs of the amygdala and orbitofrontal cortex are directed to the hypothalamus, which not only provides one route for these reinforcing environmental events to produce autonomic responses, but also is implicated in the utilisation of such stimuli in motivational responses, such as feeding and drinking, and in emotional behaviour. Other outputs of the amygdala and orbitofrontal cortex which may enable them to influence behaviour are directed to the striatum, and also back towards some of the cortical regions from which they receive inputs. It is suggested that these latter projections are important in the effects which mood states have on cognitive processing.

Abstract:Abstract The orbitofrontal cortex in primates including humans is the key brain area in emotion, and in the representation of reward value and in non-reward, that is not obtaining an expected reward. Cortical processing before the orbitofrontal cortex is about the identity of stimuli, i.e. ‘what’ is present, and not about reward value. There is evidence that this holds for taste, visual, somatosensory and olfactory stimuli. The human medial orbitofrontal cortex represents many different types of reward, and the lateral orbitofrontal cortex represents non-reward and punishment. Not obtaining an expected reward can lead to sadness, and feeling depressed. The concept is advanced that an important brain region in depression is the orbitofrontal cortex, with depression related to over-responsiveness and over-connectedness of the non-reward-related lateral orbitofrontal cortex, and to under-responsiveness and under-connectivity of the reward-related medial orbitofrontal cortex. Evidence from large-scale voxel-level studies and supported by an activation study is described that provides support for this hypothesis. Increased functional connectivity of the lateral orbitofrontal cortex with brain areas that include the precuneus, posterior cingulate cortex and angular gyrus is found in patients with depression and is reduced towards the levels in controls when treated with medication. Decreased functional connectivity of the medial orbitofrontal cortex with medial temporal lobe areas involved in memory is found in patients with depression. Some treatments for depression may act by reducing activity or connectivity of the lateral orbitofrontal cortex. New treatments that increase the activity or connectivity of the medial orbitofrontal cortex may be useful for depression. These concepts, and that of increased activity in non-reward attractor networks, have potential for advancing our understanding and treatment of depression. The focus is on the orbitofrontal cortex in primates including humans, because of differences of operation of the orbitofrontal cortex, and indeed of reward systems, in rodents. Finally, the hypothesis is developed that the orbitofrontal cortex has a special role in emotion and decision-making in part because as a cortical area it can implement attractor networks useful in maintaining reward and emotional states online, and in decision-making.

Abstract:What can psychopathology and its treatment tell us about cognitive emotional interactions? Standard approaches to interactions between emotion and cognition often adopt a variant of the idea that cognitive processes, subserved by dorsal and lateral cortical circuits, exert control and regulation of ventral, limbic brain areas associated with emotional expression and experience. However, it is becoming clear from studies on depression, post-traumatic stress disorder (PTSD) and obsessive compulsive disorder (OCD), that a binary, opponent theory of cognitive emotion interaction (CEI) and the dorsal-ventral model of neurocircuitry do not fully describe the data. We summarize recent research to suggest that networks of direct and indirect pathways exist by which cognition can regulate pathological emotion, and the inter-relationships of specific nodes within the networks need to be characterized.

Abstract:Preface What produces emotions? Why do we have emotions? How do we have emotions? Why do emotional states feel like something? This book seeks explanations of emotion by considering these questions. One of the distinctive properties of this book is that it develops a conceptual and evolutionary approach (see for example Chapters 2 and 3) to emotion. This approach shows how cognitive states can produce and modulate emotion, and in turn how emotional states can influence cognition. Another distinctive property is that this book links these approaches to studies on the brain, at the level of neuronal neurophysiology, which provides much of the primary data about how the brain operates; but also to neuropsychological studies of patients with brain damage; to functional magnetic resonance imaging (fMRI) (and other neuroimaging) approaches; and to computational neuroscience approaches. The author performs research in all these areas, and this may help the approach to emotion described here to span many levels of investigation. The empirical evidence that is brought to bear is largely from non-human primates and from humans, because of the considerable similarity of their visual and emotional systems associated with the great development of the prefrontal cortex and temporal lobes in primates, and because the overall aim is to understand how emotion is implemented in the human brain, and the disorders that arise after brain damage. To understand how the brain works, including how it functions in emotion, it is necessary to combine different approaches, including neural computation. Neurophysiology at the single neuron level is needed because this is the level at which information is exchanged between the computing elements of the brain. Evidence from the effects of brain damage, including that available from neuropsychology, is needed to help understand what different parts of the system do, and indeed what each part is necessary for. Neuroimaging is useful to indicate where in the human brain different processes take place, and to show which functions can be dissociated from each other. Knowledge of the biophysical and synaptic properties of neurons is essential to understand how the computing elements of the brain work, and therefore what the building blocks of biologically realistic computational models should be. Knowledge of the anatomical and functional architecture of the cortex is needed to show what types of neuronal network actually perform the computation. [Brain Functional Connectivity in Depression During Emotion Processing].

Abstract:Cognitive and emotional dysfunction in depressed patients has been broadly researched. However, the physiological mechanism of the emotional impairment is still obscure. In this study, according to the ethical principles and the procedures of informed consent, we enrolled 16 depression patients and 16 normal subjects in a "learning-judgment" cognitive task. Emotional faces were chosen and used as the visual stimuli, while the subjects' scalp electroencephalographs (EEG) were recorded. We calculated the coherences of the event-related EEG signals between different functional brain regions and used the repeated measure analysis of variance (ANOVA) to analyze the coherences. The results revealed, (1) The depressed patients had a significantly lower inter-hemisphere coherence than did the normal controls at frontal sites across all EEG basic rhythms; (2) The inter-hemispheric coherence of EEG beta activity was significantly correlated with the emotional processing during 1-200 ms after stimulus onset, and the synchronicity with the negative emotion stimulus was higher than that with neutral and positive stimuli; (3) The main effect of race was found at delta and theta bands at about 401-600 ms after stimulus onset. Moreover, the connectivity of inter-hemisphere for own-race stimulus was stronger than that for other-race stimulus.

Abstract:A first tier of cortical processing builds representations of what a stimulus is, i.e. its identity and intensity, independently of reward or emotional value. Structures in this tier include the inferior temporal visual cortex, the insular primary taste cortex, and the pyriform olfactory cortex. A second tier of processing involves two systems, the orbitofrontal cortex and amygdala, in which the reward or emotional value of stimuli is represented. The orbitofrontal cortex becomes more developed and relatively more important than the amygdala in emotion in primates and humans than in rodents. The orbitofrontal cortex projects to the pregenual cingulate cortex, in which there are also affective representations. A third tier of processing is involved in the medial prefrontal cortex in choice decision-making between affective stimuli; in the cingulate cortex in action to reward outcome learning; in the striatum in habit learning; and in the hypothalamus to autonomic and endocrine responses to affective stimuli.
