Adhesion and Proliferation of Human Periodontal Ligament Cells on Diseased Cementum Surfaces Conditioned by Minocycline-Hcl

Abstract:Objective To evaluate the effect of 2.5% minocycline-HCL root conditioning on adhesion and proliferation of PDLC on cementum surface.Methods Thirty-four cementum chips were obtained from roots exposed to periodontal pocket.They were from 17 teeth extracted for periodontal disease.Two chips were made from each diseased tooth.Normal cementum chips were obtained from 17 healthy teeth extracted for orthodontic reason.Then root planing was done on all samples.Cementum was confirmed remaining on root chips under microscope.Seventeen diseased cementum chips were conditioned by(2.5%) minocycline-HCl for 2min.The other 17 diseased cementum chips were treated by saline as disease control.And 17 healthy cementum chips were treated by saline as healthy control.Human periodontal ligament cells(PDLC) from healthy teeth were cultured and incubated on the cementum for 16、24 and 72 hours,respectively.The spread of PDLC on root surface was studied by scanning electron microscope(SEM) and the proliferation was quantified using the MTT method.Results The number of PDLC attached to and proliferated on periodontitis cementum surfaces were significantly less than that on healthy cementum surfaces.The PDLC which attached to and proliferated on minocyline-conditioned cementum chips((0.43)±(0.05);(0.68)±(0.07)) was significantly more than that on diseased chips treated by saline((0.25)±(0.03);(0.41)±(0.05))(P(0.01)),but less than that in healthy control((0.54)±(0.07);(0.86)±(0.08))(P(0.01)).Conclusion Adhesion and proliferation of PDLC were suppressed when cultured on cementum chips exposed to periodontal pockets.Root conditioning by minocycline-HCl could improve PDLC attachment and proliferation on root cementum surface affected by periodontitis.
