Abstract:Ganoderma, has been used for clinical applications for thousands of years as a highly-nutritious and significantly effective medicinal herb. The active components and efficacy of Ganoderma are constantly being explored and supplemented every year. In recent years, more and more literature has reported the pharmacological effects of Ganoderma on anti-tumor, liver protection and immunity enhancement, especially on neuroprotection. Numerous research works on the neuroprotective effects of Ganoderma have been documented (e.g., modulation of neurogenesis, amelioration of Alzheimer's disease, therapeutic effect on epilepsy, the protective effect on neural cells in stroke injury, etc.) thus it has drawn increasing attention. However, an integrated and comprehensive review of recent research findings has not been detailed in any great depth. Therefore, the purpose of this review is to summarize and elucidate recent progress of neuroprotective effects of natural Ganoderma and its extracts. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:This literature review provides an up-to-date exploration of the multifaceted attributes of maitake mushrooms (Grifola frondosa), elucidating their bioactive phytochemicals and diverse health advantages, including their substantial role in supporting human health and potential incorporation into the medicinal industry. Carbohydrates and protein are the major constituents contributing to the dry weight of G. frondosa, taking up around 70-80 % and 13-21 %, respectively, with emerging research linking these constituents to various health benefits. By synthesising current research findings, this review emphasises the substantial role of maitake mushrooms in supporting human health and underscores their potential incorporation into the medicinal industry. To further advance our understanding, future research should delve into the mechanisms underlying their health-promoting effects, with a focus on conducting quantitative studies to elucidate physiological pathways and potential drug interactions. Additionally, exploring their integration into functional foods or nutraceuticals through quantitative assessments of bioavailability and efficacy will be crucial for maximising their therapeutic benefits. This review aims to provide comprehensive insights, catalysing further research and innovation in utilising maitake mushrooms for improved well-being and industry advancement.
Extraction, purification, structure and bioactivities of polysaccharides from Grifola frondosa (maitake): a review

Abstract:Grifola frondosa is an edible and medicinal mushroom, and modern pharmacological studies have revealed that it possesses various beneficial activities, which are owing to its complex bioactive components. Among them, polysaccharides are the primary active component of G. frondosa . Although numerous researchers have studied G. frondosa in recent decades, there is currently no review available concerning the polysaccharides of this mushroom. In this paper, we first summarized the methods for extracting and purifying the polysaccharides of G. frondosa polysaccharides. The extraction of polysaccharides directly affects their purity, which in turn affects their bioactivities. Hot water is the common method uesd to extract polysaccharides from this mushroom. Scientists have also developed some new extraction methods, such as cold water extraction, the ammonium sulfate/ethanol aqueous two-phase system (ATPS) and supercritical CO 2 extraction. The extracted crude polysaccharides are usually accompanied by compounds, such as proteins, fats and pigments, which can interfere with the structural identification and subsequent activity analyses. Therefore, further separation and purification of the polysaccharides are necessary. The common purification methods of polysaccharides are ethyl alcohol deposition and column chromatography. Next, we reviewed the molecular weights, monosaccharide compositions, structural characteristics and biological activities of G. frondosa polysaccharides. The polysaccharides had a variety of biological activities, such as anti-tumor, antioxidant, hypoglycemic and hypolipidemic activities among others. This paper will provide useful references for the investigation, production, and application of G. frondosa polysaccharides in functional foods and as therapeutic agents.

Abstract:Background Mushrooms are generally regarded as useful products with multiple applications in the food and health industry. Shiitake (Lentinula edodes) is one of the most cultured and consumed mushrooms around the world. With more complexity in medical science and phenomenon like antibiotic resistance, have led to the search for alternatives for disease treatment and prevention. Shiitake mushroom is nutritionally rich and contains numerous minerals ( Potassium , Manganese, Magnesium , Iron, and Phosphorus) and vitamins (pro-vitamin D 2 , vitamin B 1 , B 2 , B 6 , B 12 , and niacin) which make it a potential source of nutrition. Shiitake mushrooms contain essential macro and micronutrients as well as many bioactive compounds, including polysaccharides, antioxidants, dietary fiber, and ergosterol. Bioactive compounds can be helpful for maintaining the good health of the users and preventing them from diseases. Shiitake mushroom contains phenolics, polysaccharides, sterols, that play role in bodily functions resulting in improved health of the individuals. Scope and approach The review has focused to uncover the therapeutic potential of L.edodes and description of several nutraceutical properties of the shiitake mushroom that can be helpful in preventing and treating diseases. The properties like antimicrobial, antiviral, anticancer, anti-obesity, antidiabetic, antioxidant, etc. are discussed in this review with more focus on the latest update in this context. Key findings and conclusions It has been observed that the shiitake mushroom is one of the useful foods that can confer multiple beneficial effects on the user. Because of presence of many bioactive compounds, it can simultaneously act as an antimicrobial, antiviral, anticancer/antitumor, antidiabetic, antihyperlipidemic, anticholesterol, antioxidative, antiaging, hepatoprotective, and immunomodulatory agent. Nevertheless, the factors like mushroom strain, growth conditions, substrate, extraction method and temperature also influence the outcomes of the studies. Moreover, the results in non-human subjects, such as animals, have shown positive outcomes but further research is required because of varying results. Importantly, the evidence of beneficial effects of Shiitake in humans is scarce and clinical studies are required.

Abstract:Mushrooms are widely used in Asian medicine, both as dietary supplements and as nutraceutical foods. Some principal fungi have recently become popular in Europe for their nutritional and health benefits, and a large number of bioactive compounds have been isolated in several taxa showing antioxidant, anti-inflammatory, antiviral, antitumor activities and other biological properties with great therapeutic application. Separate research has been done separately for the different species. The aim of the present review was to give an overview of the present knowledge regarding bioactive compounds in ten of the most common species, their options in therapeutic use and, for the first time, their possible toxicity or adverse effects. Boletus edulis, Ganoderma lucidum, Grifola frondosa, Hericium erinaceus, Hypsizygus marmoreus, Lentinula edodes, Morchella esculenta, Pleurotus ostreatus, Poria cocos and Trametes versicol have great potential as functional foods and as a source of drugs for pharmaceutical science and medicine. Some adverse effects, however, were found to occur in individual daily consumption and during clinical trials. More research is needed to make a good rational use of these medicinal compounds.

Ganoderma lucidum (Reishi) an edible mushroom; a comprehensive and critical review of its nutritional, cosmeceutical, mycochemical, pharmacological, clinical, and toxicological properties

Abstract:Reishi owes an exceptional value in nutritional, cosmeceutical, and medical treatments; however, none of the studies has provided its future-driven critical assessment. This study documents an up-to-date review (2015-2020, wherever applicable) and provide valuable insights (preclinical and clinical evidence-based) with comprehensive and critical assessments. Various databases 'Google scholar', 'Web of Science', 'ScienceDirect', 'PubMed', 'Springer Link', books, theses, and library resources were used. The taxonomic chaos of G. lucidum and its related species was discussed in detail with solution-oriented emphasis. Reishi contains polysaccharides (α/β-D-glucans), alkaloids, triterpenoids (ganoderic acids, ganoderenic acids, ganoderol, ganoderiol, lucidenic acids), sterols/ergosterol, proteins (LZ-8, LZ-9), nucleosides (adenosine, inosine, uridine), and nucleotides (guanine, adenine). Some active drugs are explored at an optimum level to make them potential drug candidates. The pharmacological potential was observed in diabetes, inflammation, epilepsy, neurodegeneration, cancer, anxiety, sedation, cardiac diseases, depression, hepatic diseases, and immune disorders; however, most of the studies are preclinical with a number of drawbacks. In particular, quality clinical data are intensely needed to support pharmacological activities for human use. The presence of numerous micro-, macro, and trace elements imparts an essential nutritional and cosmeceutical value to Reishi, and various marketed products are available already, but the clinical studies regarding safety and efficacy, interactions with foods/drinks, chronic use, teratogenicity, mutagenicity, and genotoxicity are missing for Reishi. Reishi possesses many valuable pharmacological activities, and the number of patents and clinical trials is increasing for Reishi. Yet, a gap in research exists for Reishi, which is discussed in detail in the forthcoming sections.

Abstract:Maitake (Grifola frondosa) is a medicinal mushroom known for its peculiar biological activities due to the presence of functional components, including dietary fibers and glucans, that can improve human health through the modulation of the gut microbiota. In this paper, a Maitake ethanol/water extract was prepared and characterized through enzymatic and chemical assays. The prebiotic potential of the extract was evaluated by the growth of some probiotic strains and of a selected probiotic consortium. The results revealed the prebiotic properties due to the stimulation of the growth of the probiotic strains, also in consortium, leading to the production of SCFAs, including lactic, succinic, and valeric acid analyzed via GC-MSD. Then, their beneficials effect were employed in evaluating the vitality of three different healthy and tumoral colorectal cell lines (CCD841, CACO-2, and HT-29) and the viability rescue after co-exposure to different stressor agents and the probiotic consortium secondary metabolites. These metabolites exerted positive effects on colorectal cell lines, in particular in protection from reactive oxygen species.

Comprehensive Reviews in Food Science and Food Safety

Abstract:The production of macrofungi (mushrooms) as well as their economic value have been steadily increasing globally. The use of functional foods, dietary supplements, and traditional medicines derived from macrofungi is increasing as they have numerous health benefits as well as abundant nutrients. Macrofungi are diverse with complex and highly varied growth conditions and bioactive constituents, most macrofungal resources have not yet been fully explored and applicated, leading to an urgent need for appropriate strategies to address the problem. Increasing attention has been paid to the macrofungal cultivation and application, in particular, potential prebiotics. Herein, the present review comprehensively summarizes recent progress in the cultivation, newly identified bioactive constituents, and their effects on gut microbiota as well as the potential ways in which they affect human health. Moreover, the macrofungal food development is discussed to improve food nutritional value and change the quality characteristics of food. Finally, the review addresses consumer safety concerns and the prospective genetic manipulation of macrofungi. We hope that this review can provide a comprehensive research reference for ensuring the safety and efficacy, along with maximizing the value and profitability of macrofungi production.
Antioxidative and Immunomodulatory Activities of the Exopolysaccharides from Submerged Culture of Hen of the Woods or Maitake Culinary-Medicinal Mushroom, Grifola frondosa (Agaricomycetes) by Addition of Rhizoma gastrodiae Extract and Its Main Components

Abstract:Grifola frondosa (hen of the woods or maitake) is a famous culinary-medicinal mushroom, and its exopolysaccharides (EPSs) have biological activities with or without supplementation with exogenous additives. In this study, a Rhizoma gastrodiae extract was added to a G. frondosa fermentation system. P-hydroxylbenzaldehyde (HBA), the main product of R. gastrodiae, had the highest utilization rate in the fermentation process (42%). In addition, the EPSs of G. frondosa after addition of R. gastrodiae extract (REPS), of HBA (HEPS), or of a standard solution according to the main component ratio of R. gastrodiae extract (CEPS) were obtained. We then determined the antioxidant and immunomodulatory activities of EPS, REPS, HEPS, and CEPS. Overall, REPS showed the highest antioxidant activities compared with EPS and HEPS (P < 0.05) but similar to that of CEPS (P > 0.05). The half-inhibitory concentration (ED50) values of REPS (< 4 mg/mL) were lower than those of EPS, HEPS, and CEPS. Moreover, REPS was better able to stimulate phagocytosis and nitric oxide production of RAW 264.7 macrophages than were the others, without a significant difference from CEPS (P > 0.05). An interesting and important finding is that a R. gastrodiae extract can increase antioxidant and immunomodulatory activities of EPS preparations from G. frondosa, and the standard solution of the main components of the R. gastrodiae extract may be better for simulating fermentation performed by G. frondosa and biological activities of its major products.

Abstract:Ganoderma lucidum (Reishi) is a popular medicinal mushroom and has been used in oriental medicine because of its promoting effects on health and life expectancy. G. lucidum contains various compounds with a high grade of biological activty, which increase the immunity and show antitumour, antimicrobial, anti-inflammatory, antioxidant and acetylcholinesterase inhibitory activity. Several of these substances belong to the triterpenoids and polysaccharides classes. Proteins, lipids, phenols, sterols, etc. are also present. In the present review, an extensive overview of the presence of antitumour, antimicrobial, antioxidant and antiacetylcholinesterase compounds in G. lucidum extracts will be given, along with an evaluation of their therapeutic effects.

Abstract:In the last few decades, mushrooms have increasingly been used as a source of therapeutic agents or health food supplements. In particular, polysaccharides or polysaccharide-protein complexes derived from mushrooms have received much attention in biomedical research. These bioactive biopolymers are mainly present as glucans with different types of glycosidic linkages, such as (1→3), (1→6)-β-glucans and (1→3)-α-gtucans, while others mostly bind to protein residues as polysaccharide-protein complexes. One of the most promising activities of these polysaccharides is their immunomodulation and anticancer effect. Three antitumor polysaccharides, namely lentinan, schizophyllan, and protein-bound polysaccharide (or polysaccharopeptide), have been isolated from Lentinus edodes (Berk.) Sing., Schizophyllum commune (Fr.: Fr.), and Coriolus versicolor (L.: Fr.) Quel. [= Trametes versicolor (L.: Fr.) Lloyd], respectively, and have become large nutraceutical and pharmaceutical market items, particularly in Japan. The mechanism of their antitumor actions is still not completely understood. But it is widely accepted that antitumor polysaccharides from higher fungi enhance various immune responses in vivo and in vitro, and act as biological response modifiers. Potentiation of the host defense system may result in the activation of many kinds of immune cells that are vitally important for the maintenance of homeostasis. In addition to immunomodulation and antitumor activity, there are new potential applications of mushroom polysaccharides or polysaccharide-protein complexes for pharmaceutical and functional food development. These polysaccharides have been shown to have various potent pharmacological properties, such as antioxidant, free radical scavenging, antiviral, hepatoprotective, antifibrotic, antiinflammatory, antidiabetes, and hypocholesterolemic activities.

Critical review of the phytochemical profiles and health-promoting effects of the edible mushroom Armillaria mellea

Abstract:Research on the nutritional and medicinal properties of wild edible mushrooms has witnessed a significant surge in recent years. Among these mushrooms, Armillaria mellea (AM) stands out due to its abundant biologically active components. The presence of biological compounds in AM, including carbohydrates, sterols, fatty acids, sesquiterpenes, non-hallucinogenic indole compounds and adenosine derivatives, has been demonstrated in previous studies. Notably, specific bioactive substances isolated from AM, such as armillarikin, have exhibited promising anticancer effects. In vitro studies have elucidated the mechanisms behind these effects, further emphasizing the potential of AM in cancer treatment. Consequently, the objective of this study is to provide a comprehensive overview of the phytochemical profiles of AM while thoroughly investigating its therapeutic benefits. Moreover, this research has uncovered novel and effective treatments, including the utilization of ultrasonic disruption extraction in food processing. These findings highlight the potential of AM as a functional food with possible medical applications. By exploring AM's phytochemical composition and therapeutic effects, this study aims to contribute to a deeper understanding of its potential as a valuable natural resource.
Effect of Grifola Frondosa (dicks.:Fr.) S.F.Gray (maitake Mushroom) Aqueous Extract on Nitric Oxide Production Induced by BCG and Cytokines in Rat Hepatocytes

Abstract:: To examine the effect of the Grifola frondosa (maitake mushroom) aqueous extract (MAE) on nitric oxide (NO) production induced by BCG pretreatment in vivo plus cytokines mixture (CM) stimulating conditions in vitro in primary cultured rat hepatocytes. NO accumulations were induced by BCG pretreatment (125 mg kg−1, 2 weeks, i.v.) in vivo or by BCG plus cytokine mixture (CM, including TNF-α, IL-1β, IFN-γ, and LPS) stimulating a in culture medium in vitro. NO production in the cultured supernatant was determined by the Griess reaction. Application of MAE alone did not show marked effects on NO production (P > 0.05) in cultured hepatocytes, even though it was administered after 48 hr and at higher concentrations (800 mg L−1). Under the presence of BCG or/and CM conditions, NO production was significantly increased by MAE at 400 mg L−1 ∼ 800 mg L−1 concentrations (P < 0.05 and P < 0.01, respectively) as the time-dependent manner in culture supernatant. But in similar BCG plus CM stimulating conditions, administration of MAE inhibited NO production at a range of lower concentrations (from 50 mg L−1 to 200 mg L−1, P < 0.05). On the other hand, both aminoguanidine and actinomycin D (ActD), which are inhibitors of selective iNOS enzyme activity and iNOS mRNA transcription, significantly inhibited BCG plus CM-induced NO production in rat hepatocytes (P < 0.05). The results of the present study not only further support the possibility that MAE might be an immunomodulating agent, but also confirm the immune effective mechanism of MAE by regulating NO production both in vivo and in vitro.

Abstract:Abstract The mushrooms have contributed to the development of active ingredients of fundamental importance in the field of pharmaceutical chemistry as well as of important tools in human and animal health, nutrition, and functional food. This review considers studies on the beneficial effects of medicinal mushrooms on the nutrition and health of humans and farm animals. An overview of the chemical structure and composition of mycochemicals is presented in this review with particular reference to phenolic compounds, triterpenoids and sterols, fatty acids and lipids, polysaccharides, proteins, peptides, and lectins. The nutritional value and chemical composition of wild and cultivated mushrooms in Italy is also the subject of this review which also deals with mushrooms as nutraceuticals and the use of mushrooms in functional foods. The nutraceutical benefits of UV irradiation of cultivated species of basidiomycetes to generate high amounts of vitamin D2 is also highlighted and the ability of the muhsrooms to inhibit glycation is analyzed. Finally, attention is paid to studies on bioactivities of some Italian wild and cultivated mushrooms with particular reference to species belonging to the genus Pleurotus . The review highlights the potential of medicinal mushrooms in the production of mycochemicals that represent a source of drugs, nutraceutical, and functional food. Graphic abstract

The active constituents found in mushrooms include polysaccharides, dietary fibers, oligosaccharides, triterpenoids, peptides and proteins, alcohols and phenols, mineral elements (such as zinc, copper, iodine, selenium, and iron), vitamins, and amino acids. The bioactive components found in the G. lucidum mushroom have numerous health properties to treat diseased conditions such as hepatopathy, chronic hepatitis, nephritis, hypertension, hyperlipemia, arthritis, neurasthenia, insomnia, bronchitis, asthma, gastric ulcers, atherosclerosis, leukopenia, diabetes, anorexia, and cancer. In spite of the voluminous literature available, G. lucidum is used mostly as an immune enhancer and a health supplement, not therapeutically. This review discusses the therapeutic potential of G. luidum to attract the scientific community to consider its therapeutic application where it can be worth pursuing.
Regulatory Effects of Ganoderma lucidum, Grifola frondosa, and American ginseng Extract Formulation on Gut Microbiota and Fecal Metabolomics in Mice

Abstract:The bioactivities of Ganoderma lucidum, Grifola frondosa, and American ginseng have been extensively studied and documented. However, the effects of their complexes on the structural properties of intestinal microbiota and fecal metabolism remain unclear. Therefore, this paper aims to present a preliminary study to shed light on this aspect. In this study, an immunocompromised mouse model was induced using cyclophosphamide, and Ganoderma lucidum, Grifola frondosa, and American ginseng extract formulation (referred to as JGGA) were administered via gavage to investigate their modulatory effects on gut microbiota and fecal metabolism in mice. The effects of JGGA on immune enhancement were explored using serum test kits, hematoxylin–eosin staining, 16SrDNA high-throughput sequencing, and UHPLC-QE-MS metabolomics. The findings revealed potential mechanisms underlying the immune-enhancing effects of JGGA. Specifically, JGGA administration resulted in an improved body weight, thymic index, splenic index, carbon scavenging ability, hypersensitivity, and cellular inflammatory factor expression levels in mice. Further analysis demonstrated that JGGA reduced the abundance of Firmicutes, Proteobacteria, and Actinobacteria, while increasing the abundance of Bacteroidetes. Additionally, JGGA modulated the levels of 30 fecal metabolites. These results suggest that the immune enhancement observed with JGGA may be attributed to the targeted modulation of gut microbiota and fecal metabolism, thus promoting increased immunity in the body.

Abstract:The present article reviews the history of mushroom uses in culinary, food and medicinal values; current status and future aspect of mushroom research. Mushrooms contain biologically active polysaccharides, lipid and proteins in fruit bodies, each of them has a distinct role in health as either nutritional value or medicinal elements. Immunostimulating polysaccharides found in mushrooms, are most important for modern medicine. Several of the mushroom biomolecules have undergone phase I, II, and III clinical trials and are used extensively and successfully throughout the world for the treatment of various cancers and other diseases. Medicinal functions played by the mushrooms include antitumor, antibacterial, antioxidant, antiparasitic, antidiabetic, detoxification, cardiovascular, ant hypercholesterolemia, antiviral, antifungal, hepatoprotective, immunomodulating and free radical scavenging. The present review draws attention to nutritional and medicinal importance of mushroom as well as the problems and opportunity in the future development of mushroom research.

Abstract:The present study aims to determine the nutrients and nutraceuticals content and in vitro biological activities of reishi mushroom (Ganoderma lucidum) fruiting bodies. Data of the nutrients composition of Ganoderma lucidum powder indicated that crude fiber and carbohydrates were the most largest compounds (50.19 and 37.33%, respectively) followed by total protein (8.54 %), ash (2.03 %) and crude fat (1.91%.). Also, Ganoderma lucidum powder is rich in different estimated elements (K, P, Ca, Mg, Na, Fe, Zn, Mn, Cu and Se) and vitamins (A, B1,B2, B3, B6, zb9, C and E). Furthermore, nutraceuticals (bioactive compounds) content ofGanoderma lucidumpowder indicated that terpenoids were the most largest compound (2541 mg linalol.100 g-1) followed by polysaccharides (1635 mg starch equivalent. 100 g-1), phenolics (572.67 mg gallic acid equivalent. 100g-1), triterpenoids (495 mg ursolic acid.100 g-1), Flavonoids (279.34 mg catechin.100 g-1), and Lycopene (253.11 µg.100 g-1). The different extracts of Ganoderma lucidumsamples particularly ethanolic and methanolic also recorded several very high biological activities which include antioxidant activity, scavenging of free radicals, inhibition of low density lipoprotein oxidation (anti-atherosclerotic) and as antibacterial and antifungal.Such important biological effects could play important roles in strategies to combat/ prevent/treat many diseases, especially those for which oxidative stress is one of the mechanisms for its occurrence i.e. diabetes, cancer, atherosclerosis, cardiovascular diseases etc. Therefore, the present study recommended like of that Ganoderma lucidumfruiting bodies powder and/or extracts to be included in our daily diets, drinks, food supplementation and pharmacological formulae.
