Abstract:As a global pollution problem, heavy metal contamination poses a serious hazard to soil microorganisms which play an extremely important role in soil chemical cycling and ecological persistence. However, the effects that different levels of heavy metal contamination in soils have on microorganisms and the interactions between them are still unclear. The purpose of this research is to analyze the microbial structure under different levels of heavy metal contamination, find out heavy metal tolerant species under different environmental conditions, then provide useful reference for the bioremediation of contaminated farmland. In this study, 16s rRNA high-throughput sequencing technology was used to investigate the microbial communities in severe level (SL), moderate level (ML), light level (LL) and clean level (CL) of heavy metal contaminated soils, and the relationships between environment variables and microorganisms were analyzed. The results showed that the concentrations of heavy metals and soil physicochemical properties had various impacts on microbial community composition under different heavy metal contamination levels. Most dominant bacteria were in significant negative correlation with Cd in ML region, and significantly correlated with TN and OM in LL region. However, there was no significant correlation between dominant fungi and the physicochemical properties in LL region. And most of the dominant fungi were significantly correlated with the heavy metal concentrations in SL region. The bacterial phyla such as Proteobacteria, Acidobacteria and Bacteroidetes showed more tolerance with heavy metal contamination in SL, ML and LL regions, respectively. Meanwhile, the dominant fungi of Ascomycota, Basidiomycota, Chytridiomycota, Glomeromycota, Zygomycota and Rozellomycota showed stronger correlations with heavy metal contamination in SL and LL regions. These results indicated that some microorganisms had strong tolerance to heavy metal contamination and had certain heavy metals digestion ability, which can create an appropriate soil environment for the growth of food crops.

Abstract:Little is known about the characteristics of arbuscular mycorrhizal fungi (AMF) community in the roots of host plants growing on heavy metal contaminated sites. The objectives of this study were to examine the community structure of AMF associated with the roots of a copper (Cu) tolerant plant—Elsholtzia splendens in a Cu mining area in southeastern Anhui Province, China. Molecular techniques were used to analyze AMF community composition and phylogenetic relationship in E. splendens roots sampled from three Cu mine spoils and two adjacent reference areas. Results obtained showed that root colonization and AMF diversity were very low and negatively correlated with total and extractable Cu concentrations. All the DNA sequences recovered belonged to the genus of Glomus. The principal component analysis (PCA) revealed that the AMF community composition varied remarkably among different sites and was related closely to soil properties, especially Cu concentrations. The distribution pattern of AMF species in various sites suggested the degree of AMF tolerance to Cu contamination. The unique AMF species that presented exclusively in heavily contaminated sites need to be further examined for potential application in phytoremediation of metal contaminated soils.
Comparative transcriptome analysis of ectomycorrhizal fungus Pisolithus albus in response to individual and combined stress of copper and cadmium

Abstract:An ectomycorrhizal fungus Pisolithus albus establishes the natural symbiosis with plant roots on extreme heavy metal (HM)–rich soil and enables their survival in toxic metal concentrations. Understanding P. albus key genes and pathways behind strong metal tolerance is crucial for its successful application in the rehabilitation of metal-contaminated barren lands. Therefore, this study aimed to analyze the whole transcriptome profile of P. albus under individual and combined metal stress of copper (Cu) and cadmium (Cd). At 480 μM Cu and 16 μM Cd toxic concentrations, P. albus has shown growth and survival and accumulated high metal (1.46 μg Cu and 1.13 μg Cd per mg of dry mycelia). The study found a stronger response of P. albus to single-metal stress in high concentration as compared to multi-metal stress in relatively lower concentration. Hence, the intensity of fungal response to HM stress is mainly determined by the metal concentration involved in stress. We have found a total of 11 pathways significantly associated with HM stress, among which amino acid, lipid, and carbohydrate metabolisms were highly affected. The functional enrichment of differentially expressed genes has shown the induced biosynthesis of arginine, melanin, metal chelating agents, membrane phospholipids, fatty acids, folate, pantothenate, ergothioneine, and other antioxidant agents; upregulation of zinc ion uptake, potassium transporters, and lysine degradation; and reduction of phosphatidylcholine degradation, incorrect protein folding, iron uptake, and potassium efflux as the top efficient tolerance mechanisms of P. albus against HM stress. The current study would contribute to understanding fungal HM tolerance and its further utilization in the bioremediation of metal-contaminated abandoned lands. The validation of RNA-sequencing analysis with RT-qPCR of selected genes showed the high credibility of the presented data.

Abstract:Ectomycorrhizal fungi (ECMF) can develop the resistance of host plants to heavy metal stress. However, little is known about the response of ECMF to heavy metal exposure. In this study, the growth and physiological indices of Lepista sordida under Cd and Cu stress were studied. The growth of L. sordida on PDA medium under Cd and Cu stress was observed using scanning electron microscopy (SEM). After the addition of Cd and Cu to the medium, the mycelium started twisting, breaking, sticking together, and even dissolving. In the control group, a good and luxuriant mycelium growth of L. sordida along with the numerous clamp connections was observed. The mycelial biomass decreased with increasing concentrations of heavy metals in a liquid medium. The catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), and ascorbate peroxidase (APX) activities were also investigated, and the results showed that the Cd and Cu treatments caused a significant increase in the antioxidant enzyme activities. The contents of soluble protein, soluble sugar, and free proline in L. sordida were investigated, and it was found that the contents initially increased and then decreased with the increasing concentrations of Cd and Cu. However, the content of malondialdehyde (MDA) increased with the increasing concentrations of Cd and Cu. In conclusion, the present study provides a theoretical basis for the better utilization of Ectomycorrhizal fungal resources for the remediation of soil contaminated with heavy metal.

Abstract:Soil fungi have been widely studied, but the effects of heavy metal contamination at various levels as well as the abundance and diversity of heavy metal tolerant fungi in the contaminated paddy soils are still unknown. The purpose of this study is to analyze the adaptability of fungi at different levels of heavy metal contamination to identify species that have strong adaptability to heavy metals. In this research, the technology of high-throughput sequencing was applied to study fungal communities in severe level (SL), moderate level (ML), light level (LL), and clean level (CL) for soil samples polluted by heavy metal, as well as to analyze the relations between environmental variables and fungal communities. The spearman analysis showed that 6 dominant fungal phyla and 18 dominant fungal genera were significantly correlated with these environmental variables. The α-diversity indexes of the soil fungal community from SL, ML, and CL were, mostly, drastically higher than the LL samples (p < 0.05). Meanwhile, Ascomycota, the main fungal phylum, was spotted to yield a strong tolerance towards heavy metals, especially in ML. The most dominant genera of tolerant fungi in this area, which are Aspergillus, Penicillium, and Fusarium, could absorb and transport the heavy metals with the help of nutrients under certain heavy metal contamination levels. Therefore, this study indicated that some fungi, which have strong biodegradability on heavy metals, can reduce toxicity of heavy metals and create a proper soil environment to grow food crops.

Abstract:s: The experiment was conducted to study heavy metal accumulation, distribution and micro-ambient change of ectomy-corrhizal fungi Boletus edulis in vitro. The study found that the growth modles of ectomycorrhizal fungi were similar in both Cu and Cd stressed environment. Compared with control, low concentration treatments of Cu and Cd enhanced the growth of fungi, but high ones prevented it. Boletus edulis in viro had a strong ability to absorb Cu and Cd, and showed higher heavy metal tolerance. Ecto-mycorrhizal hypha was the main site for heavy metal to accumulate. The concentration of Cu and Cd in the hypha under the highest treatments were 26.6 and 28 times respectively more than control. However, the effect of hyphal exudates on the resistance to heavy metals was relatively small. The accumulated models of two heavy metals were significantly different. Mycelia might alleviate heavy metal damage by changing cultivation environment selectively. This could be one of resistance mechanisms of hypha to heavy metals

Abstract:Ectomycorrhizae have been proved to be able to enhance higher plant to grow in the contaminated area.This paper reviewed achievement of studies on the heavy metal resistance of ectomycorrhizae in the last 20 years,and discussed the positive effect of ectomycorrhizae on the resistance of higher plant to heavy metals attribute to the following mechanisms such as sorption on the external mycelium; exudation of chelating substances and adjustment of fungus slime; filtration and sorption of metals in the hyphal sheath or Hartig net; and hydrophobicity of the hyphal sheath.Some suggestions for future research in this field were put forward based on the mentioned mechanisms.

Abstract:Cadmium (Cd) displays strong toxicity, high mobility, and cannot be degraded, which poses a serious threat to the environment. Cenococcum geophilum (C. geophilum) is one of the most common ectomycorrhizal fungi (ECMF) in the natural environment. In this study, three Cd sensitive and three Cd tolerant strains of C. geophilum were used to analyze the physiological and molecular responses to Cd exposure. The results showed that Cd inhibited the growth of all strains of C. geophilum but had a less toxic effect on the tolerant strains, which may be correlated to a lower content of Cd and higher activity of antioxidant enzymes in the mycelia of tolerant strains. Comparative transcriptomic analysis was used to identify differentially expressed genes (DEGs) of four selected C. geophilum strains after 2 mg/L Cd treatment. The results showed that the defense response of C. geophilum strain to Cd may be closely related to the differential expression of functional genes involved in cell membrane ion transport, macromolecular compound metabolism, and redox pathways. The results were further confirmed by RT-qPCR analysis. Collectively, this study provides useful information for elucidation of the Cd tolerance mechanism of ECMF.

Abstract:s:Two experiments were conducted to investigate the resistance of ectomycorrhizal fungi against heavy metals.In the first experiment,the biomass of Gomphidius viscidus under the intimidation of heavy metals and pH was measured as well as the pH variation under its growing conditions.The results indicated that the pH value had great influence on ectomycorrhizal fungi's resistance against heavy metals.When the pH value was 5.0,the biomass of G.viscidus reached maximum in the control and Cd 50 μmol·L-1 treatment,while the biomass of G.viscidus reached maximum in the treatments of Cu 50 μmol·L-1 and Cu 25 μmol·L-1+Cd 25 μmol·L-1 at the pH value of 4.0.The ectomycorrhizal fungi could adjust its own growing condition,which made itself grow better under unsuitable pH conditions.The ability behaved more significantly under treatments with heavy metals.In the second experiment,the total organic carbon(TOC)exudation of ectomycorrhizas(Formation of Pinus tabulaeformis carr.and Gomphidius viscidus)with or without Cu2+ treatment was measured.The results showed that TOC excreted by ectomycorrhizas after treated with Cu2+ was significant higher than that before treated,indicating that ectomycorrhizal fungi tolerated the toxicity of heavy metals by excreting more organic substances to stabilize heavy metals and decrease their availability.

Abstract:The premise to study on the influence of the heavy metals Cu,Zn and Pb to hypha growth of one mycorrhizal fungi —Leccinum aurantiacum(Bull.) Gray under condition of agar media and standing liquid cultures.We use fungi on EC50 to evaluate the toleration of Leccinum aurantiacum(Bull.) Gray to Cu,Zn and Pb.Through the evaluation of semi-lethal concentration to the toleration of Leccinum aurantiacum(Bull.) Gray found that the toleration to Zn is stronger than Cu and Pb.After culture the reduce of the pH of substance will influence the biomass of Mycorrhizal fungi,This showing that the Mycorrhizal fungi under heavy metal stress can regulate their own pH to ease the pressure of the environment.

Abstract:Fungi possess prominent tolerance and detoxification capacities in highly metal(loid)-polluted systems, yet little is known about their responding behaviors under different contamination conditions. Here, we systematically investigated the structure and function profiles of fungal communities in an abandoned reservoir mainly contaminated by multiple metal(loid)s such as Al, Be, Cd, Co, Cr, and Cu. This abandoned reservoir consisted of three distinct zones, i.e., Zone I with the shortest deprecation time and the highest metal(loid) contamination; Zone II with the medium deprecation time and medium metal(loid) contamination; and Zone III with the longest abandonment time and the lowest metal(loid)contamination. The lowest pH and the highest contents of OM, TN, and TP were also observed for the high-contamination Zone I, followed by the moderate-contamination Zone II and the low-contamination Zone III. Fungal biodiversity was found to be robust and dominated by many endurable genera in Zone I, and notable cooperative relationships among fungal species facilitated their viability and prosperity under severe metal(loid) contaminations. Differently, the lowest biodiversity and fragile co-occurrence network were identified in Zone II. As metal(loid) contaminations reduced from Zone I to Zone III, dominant fungal functions gradually changed from undefined saprotroph guild to parasites or pathogens of plant-animal (i.e. animal pathogen, endophyte, and plant pathogen). Moreover, metal(loid)s combined with physicochemical properties jointly mediated the fungal taxonomic and functional responses to different metal(loid) contamination levels. Overall, this study not only broadens the understanding of taxonomic and functional repertoires of fungal communities under different metal(loid) contaminated conditions, but also highlights the crucial contributions of specific fungi to bioremediation and management in varying metal(loid)-polluted environments.

Abstract:Heavy metal speciation in the rhizospheric soil of Pinus tabulaeformis seedlings was significantly influenced by inoculation of ectomycorrhizal fungi. Compared with CK,the inoculated rhizosphere was much lower in content of exchangeable copper and cadmium, but much higher in content of organically bonded forms, showing a remarkable tendency of loosely associated forms transforming into tightly associated forms. Ectomycorrhizae evidently decreased bioavailability of heavy metal in the rhizosphere, and alleviated the damage of heavy metal to its host plant.

Abstract:Filamentous fungi‐copper (Cu) interactions are very important in the formation of natural ecosystems and the bioremediation of heavy metal pollution. However, important issues at the proteome level remain unclear. We compared six proteomes from Cu‐resistant wild‐type ( WT ) Penicillium janthinellum strain GXCR and a Cu‐sensitive mutant ( EC ‐6) under 0, 0.5, and 3 mmol/L Cu treatments using i TRAQ . A total of 495 known proteins were identified, and the following conclusions were drawn from the results: Cu tolerance depends on ATP generation and supply, which is relevant to glycolysis pathway activity; oxidative phosphorylation, the TCA cycle, gluconeogenesis, fatty acid synthesis, and metabolism are also affected by Cu; high Cu sensitivity is primarily due to an ATP energy deficit; among ATP generation pathways, Cu‐sensitive and Cu‐insensitive metabolic steps exist; gluconeogenesis pathway is crucial to the survival of fungi in Cu‐containing and sugar‐scarce environments; fungi change their proteomes via two routes (from ATP , ATP ‐dependent RNA helicases ( ADRH s), and ribosome biogenesis to proteasomes and from ATP , ADRH s to spliceosomes and/or stress‐adapted RNA degradosomes) to cope with changes in Cu concentrations; and unique routes exist through which fungi respond to high environmental Cu. Further, a general diagram of Cu‐responsive paths and a model theory of high Cu are proposed at the proteome level. Our work not only provides the potential protein biomarkers that indicate Cu pollution and targets metabolic steps for engineering Cu‐tolerant fungi during bioremediation but also presents clues for further insight into the heavy metal tolerance mechanisms of other eukaryotes.
The effects of ectomycorrhizal fungi on heavy metals' transport in Pinus massoniana and bacteria community in rhizosphere soil in mine tailing area

Abstract:Pinus massoniana is one of the potential tree species of afforestation in barren mine tailing area, and ectomycorrhizal fungi contributes remarkably to its survival. However, how ectomycorrhizal fungi interacts with Pinus massoniana under heavy metals' stress is unknown. Two systems (Pinus massoniana inoculated with and without ectomycorrhizal fungi) were designed, and each system contained rhizosphere and non-rhizosphere, while bulk soil was sampled as control. The results showed that treatment of ectomycorrhizal fungi inoculation could obviously improved the absorption of soil moisture, total carbon/total nitrogen and nutrients, while reduced the bulk density and heavy metals of soil when compared with control (p＜0.05). The heavy metals accumulating in plants' roots with ectomycorrhizal fungi were greater than that without ectomycorrhizal fungi. Conversely, they were lower in shoots with ectomycorrhizal fungi. The bacterial community were affiliated with 23 bacterial phyla, 70 classes, 115 orders, 201 families, and 363 genera. Constrained Principal Coordinate Analysis and redundancy analysis demonstrated that bacterial communities structure in the soil inoculated with or without ectomycorrhizal fungi and bulk soil were distinguishable, but no difference between rhizosphere and non-rhizosphere. The LEfSe analysis showed Acidobacteria, Actinobacteria, and Proteobacteria were the dominant phyla that contributed to the difference among treatments.
Tolerance and Physiological Responses to DDT of Four Ectomycorrhizal Fungi

Abstract:Growth effect,tolerance and physiological responses of four species of ectomycorrhizal fungi,i.e.Boletus edulis,Gomphidius viscidus,Laccaria bicolor and Leccinum scabrum,growing in the media with different mass concentrations of DDT were studied in vitro.The results show that no impact was observed from DDT treatment on the growth mode of the mycelia,but the growth rate and biomass increment were somewhat restrained.No significant impact was measured on growth of Gomphidius viscidus and Laccaria bicolor under the treatment of 5.0 mg/L DDT and on growth of Boletus edulis under the treatments of 5.0 mg/L and 25.0 mg/L DDT.Under the treatments of DDT at higher levels,activities of polyphenol oxidases,laccase and peroxidase of Gomphidius viscidus were significantly enhanced,and pH in liquid media of it declined to a higher extent.It is concluded that Boletus edulis and Gomphidius viscidus reacted to DDT stress through different mechanisms and both had the potential capabilities of degrading DDT.
[Effect of Ectomycorrhizae on Heavy Metals Sequestration by Thermostable Protein in Rhizosphere of Pinus Tabulaeformis under Cu and Cd Stress].

Abstract:Copper and cadmium in soil is a severe soil pollution problem in China. It is effective to remediate the soil by sequestrating Cu and Cd with tolerant plant and microorganism. Ectomycorrhizae could exude a large number of organic matter to reduce the biological effectiveness of heavy metals. Therefore, under Cu or Cd stress, thermostable protein in rhizosphere exuded by roots of pine seedling (Pinus tabulaeformis) associated with ectomycorrhizal fungi (Xerocomus chrysenteron) can have the potential of sequestration for heavy metals. The results illustrated that the association didn't impact on exudation of thermostable protein but treated with different concentrations of Cu, the total thermostable protein (TTP) and easily extracted thermostable protein (EETP) in mycorrhizal rhizosphere increased by 2.64 to 11.79 times compared with non-mycorrhizal one. While treated with Cd, it was 1.49 to 7.56 times. Further analysis of metal content in rhizosphere showed that association significantly increased the Cu sequestration in mycorrhizal rhizosphere where relative content of Cu was 1.81 to 2.75 times higher than those in non-mycorrhizal rhizosphere while most of Cu was sequestrated by thermostable protein, 4.19 to 43.00 times higher in protein than in root cell. Meanwhile results of Cd treatments showed a similar trend with Cu treatments. That indicated that under excessive heavy metals stress, ectomycorrhizal association facilitated the exudation of thermostable protein so as to extend the capacity of sequestration for excessive heavy metals and mitigate the phytotoxicity from heavy metals polluted soil.

Abstract:Ectomycorrhizal fungi have huge potential value, both nutritionally and economically, but most of them cannot be cultivated artificially. To better understand the influence of abiotic and biotic factors upon the growth of ectomycorrhizal fungi, mycosphere soil and bulk soil of five ectomycorrhizal fungi (Calvatia candida, Russula brevipes, Leucopaxillus laterarius, Leucopaxillus giganteus, and Lepista panaeola) were used as research objects for this study. Illumina MiSeq sequencing technology was used to analyze the community structure of the mycosphere and bulk soil bacteria of the five ectomycorrhizal fungi, and a comprehensive analysis was conducted based on soil physicochemical properties. Our results show that the mycosphere soil bacteria of the five ectomycorrhizal fungi are slightly different. Escherichia, Usitatibacter, and Bradyrhizobium are potential mycorrhizal-helper bacteria of distinct ectomycorrhizal fungi. Soil water content, soil pH, and available potassium are the main factors shaping the soil bacterial community of the studied ectomycorrhizal fungi. Moreover, from the KEGG functional prediction and LEfSe analysis, there are significant functional differences not only between the mycosphere soil and bulk soil. ‘Biosynthesis of terpenoidsand steroids’, ‘alpha-Linolenic acid metabolism’, ‘Longevity regulating pathway-multiple species’, ‘D-Arginine and D-ornithine metabolism’, ‘Nitrotoluene degradation’ and other functions were significantly different in mycosphere soil. These findings have pivotal implications for the sustainable utilization of ectomycorrhizal fungi, the expansion of edible fungus cultivation in forest environments, and the enhancement of derived economic benefits.

Abstract:Copper (Cu) contamination threatens the stability of soil ecosystems. As important moderators of biochemical processes and soil remediation, the fungal community in contaminated soils has attracted much research interest. In this study, soil fungal diversity and community composition under long-term Cu contamination were investigated based on high-throughput sequencing. The co-occurrence networks were also constructed to display the co-occurrence patterns of the soil fungal community. The results showed that the richness and Chao1 index both significantly increased at 50 mg kg<sup>-1</sup> Cu and then significantly decreased at 1600 and 3200 mg kg<sup>-1</sup> Cu. Soil fungal diversity was significantly and positively correlated with plant dry weight. Specific tolerant taxa under different Cu contamination gradients were illustrated by linear discriminant analysis effect size (LEfSe). Soil Cu concentration and shoot dry weight were the strongest driving factors influencing fungal composition. The relative abundance of arbuscular mycorrhizal fungi increased first and then declined along with elevating Cu concentrations via FUNGuild analysis. The interactions among fungi were enhanced under light and moderate Cu contamination but weakened under heavy Cu contamination by random matrix theory (RMT)-based molecular ecological network analysis. Penicillium, identified as a keystone taxon in Cu-contaminated soils, had the function of removing heavy metals and detoxification, which might be vital to trigger the resistance of the fungal community to Cu contamination. The results may facilitate the identification of Cu pollution indicators and the development of in situ bioremediation technology for contaminated cultivated fields.

Abstract:Background and aims Microbial properties are often used to assess the recovery of soil health during phytoremediation. A field survey was conducted to test the effects of different plant metallophytes (excluder and hyperaccumulator) on soil microbial characteristics. Methods Microbial properties in the rhizosphere of four metallophytes ( Sedum alfredii , Rubus hunanensis, Lysimachia christinae and Clinopodium gracile ) growing naturally on highly Cd-, Zn- and Pb-contaminated soils were investigated. Microbial biomass carbon, basal respiration, enzyme activities and phospholipid fatty acids were analyzed to study microbial community function and composition. Results The total microbial biomass, bacterial, actinomycete, fungal, and protozoan PLFAs, basal respiration and enzyme activities in the rhizosphere of metallophytes were significantly higher than in bulk soil. Moreover, the largest increases were found in the rhizosphere of the hyperaccumulator S. alfredii . The microbial community composition in the rhizosphere of S. alfredii was significantly different from the other 3 plants. Redundancy analysis showed that soil physico-chemical properties such as metal concentrations, labile carbon and pH explained most of the variation in microbial community biomass, activity and structure. Conclusions Metallophytes using different strategies to adapt to metal-rich soils induced different effects on soil microbial properties, which were also influenced by physico-chemical characteristics.

Abstract:Endophytes play an important role in helping plants resist heavy metal stress. However, little is known about the effects of different heavy metals on the diversity and composition of endophyte communities. In this study, we used 16S and ITS amplicon sequencing to reveal the structure and function of endophytes in Symphytum officinale under different heavy metal stressors. The results showed that the endophytic fungal diversity decreased compared with the control under the different heavy metals stressors, while the diversity of endophytic bacteria showed an increasing trend. The biomarker analysis indicated that Zn and Pb stress led to obvious branches. Specific OTUs analysis showed that there were 1224, 597, and 1004 OTUs specific under Zn, Pb, and Cd stress in the bacterial community and 135, 81, and 110 OTUs specific under Zn, Pb, and Cd stress in the fungal community. The co-occurrence network showed changes in microbial interactions under heavy metal contamination conditions, suggesting that endophytic bacteria play an important role in the resistance of host plants. The Spearman analysis showed that the correlation between endophytic bacteria and endophytic fungi in relation to heavy metal transport exhibited variations. Our results expand the knowledge of the relationships of plant–microbe interactions and offer pivotal information to reveal the role of endophytes under different heavy metal stress conditions.
