Abstract:Linear and miktoarm star-shaped diblock copolymers consisting of single-stranded DNA and poly(N-isopropylacrylamide) (PNIPAAm) with various compositions were synthesized via atom transfer radical polymerization and click chemistry. The temperature-responsive phase transition behavior, micellization, was systematically examined using UV vis spectrometry, high-sensitivity differential scanning calorimetry, dynamic light scattering, and small-angle X-ray scattering. The lower critical solution temperature (LCST) increased, and its enthalpy decreased with decreasing PNIPAAm content The copolymers self-assembled into well-defined nanoparticles having a core composed of PNIPAAm and a coronal layer of DNA above LCST. The particle size and micellar aggregation number of copolymer chains depended on the macromolecular composition and chain architecture. On the other hand, regardless of their factors, the surface area occupied by one DNA strand was found to be almost unchanged. The hybridization of DNA on the nanoparticles with fully complementary one induced the aggregation of the particles in a non-cross-linking configuration. The nanoparticle composed of miktoarm star copolymer showed a quicker DNA-hybridization response in this non-cross-linking aggregation compared with the case of a linear analogue.

Abstract:Poly(N-isopropylacrylamide) (PNIPAAm)-based thermally responsive micelles are of great importance as smart materials for a number of applications such as drug delivery and biosensing, owing to their tunable lower critical solution temperature (LCST). Their design and synthesis in the nanoscale size range have been widely studied, and research interest in their structural and physicchemical properties is continually growing. In this Viewpoint, representative research on the construction of PNIPAAm-based thermally responsive micelles as well as their applications are highlighted and discussed, which would serve as a good start for newcomers in this field and a positive guide for future research.
Synthesis and Characterization of Thermo-Responsive Poly(N-Vinyl-2-Pyrrolidinone)-Block-Poly(N-Isopropylacrylamide) Micelles

Abstract:A novel thermo-responsive diblock copolymer of poly(N-vinyl-2-pyrrolidinone)-block-poly(N-isopropylacrylamide) (PNVP-b-PNIPAM) was synthesized. FT-IR, 1H-NMR and SEC results confirmed the successful synthesis of PNVP-b-PNIPAM diblock copolymer via anionic polymerization. The polymeric micelles formed from PNVP-b-PNIPAM copolymer in aqueous solution were developed and characterized as a potential thermo-responsive and biocompatible drug delivery system. Micellization of the diblock copolymer in aqueous solution was characterized by dynamic laser scattering (DLS), turbidity measurement, tension measurement and transmission electron microscopy (TEM). The thermo-responsive polymeric micelles with the size ranges of 200 to 260 nm and thickness of 30 nm are localized, selected and targeted for drug release, having a great potential in response to external-stimulus such as temperatures from 35 to 39°C. The critical micellization concentration (cmc) of PNVP-b-PNIPAM in aqueous solution is 0.0026 wt% determined by turbidity measurement. The size of micelles determined by DLS increased from 163 to 329 nm with increasing concentration of PNVP-b-PNIPAM from 0.25 to 0.5 wt% in aqueous solution at 40°C, which is determined by DLS.

Abstract:Amphiphilic copolymers are well developed as precursors for the preparation of micellar drug carriers. Poly(N-isopropylacrylamide) (PNIPAAm) is one of the most extensively studied thermo-sensitive polymers that exhibits a lower critical solution temperature (LCST) at around 33 degrees C in aqueous solution. Over the past decade, considerable efforts have been devoted to design and preparation of PNIPAAm-based thermo-sensitive polymeric micelles as delivery vehicles for controlled drug release. Present review highlights the recent developments in this field, and focuses on two categories of PNIPAAm-based copolymer micelles as smart drug delivery systems, i.e. micelles with PNIPAAm as hydrophilic shell-forming segments below the LCST and micelles with PNIPAAm as hydrophobic core-forming segments above the LCST. Both TEM and DLS measurement Suggested that the diameter of micelles was similar to 190 min at 20 degrees C. Diflunisal (DIF)-loaded micelles were prepared by dialysis method. In vitro release test at various temperatures was also performed to Study the effect of temperature on the drug release profiles. Abstract:A novel thermosensitive amphiphilic copolymer comprised of two hydrophobic poly(oleic acid) (POA) segments and one hydrophilic poly(N-isopropylacrylamide) (PNIPAAm) segment was designed and synthesized. The structure of the copolymer was confirmed as Y-shaped by FTIR, 1H NMR, and SEC-MALLS analysis. A cytotoxicity study shows that the P(OA-Y-NIPAAm) copolymer exhibits good biocompatibility. The copolymer may self-assemble into micelles in water, with the hydrophobic POA segments at the cores of micelles and the hydrophilic PNIPAAm segments as the outer shells. The resulting micelles demonstrate temperature sensitivity with a lower critical solution temperature (LCST) of 31.5 degrees C and a critical micelle concentration (CMC) of 12.6 mg L(-1). Transmission electron microscopy (TEM) shows that the micelles exhibit a nanospheric morphology within a narrow size range of approximately 10-30 nm. A study of controlled release reveals that the self-assembled micelles have great potential as drug carriers.

Abstract:To achieve a combination of spatial specificity in a passive manner with a stimuli–response targeting mechanism, a temperature-responsive polymeric micelle was prepared using block copolymers of poly(N-isopropylacrylamide-b-methyl methacrylate) (PNIPAAm-b-PMMA). The critical micelle concentration of amphiphilic block copolymers in aqueous solution was determined by fluorescence spectroscopy using pyrene as a fluorescence probe. Transmission electron microscopy images showed that these nanoparticles were regularly spherical in shape. Micelle size determined by size analysis was around 190nm. The micelles showed reversible dispersion/aggregation in response to temperature cycles through an outer polymer shell lower critical solution temperature (LCST) for PNIPAAm at around 33°C, observed by optical absorbance measurements and dynamic light scattering (DLS). The anti-inflammation drug prednisone acetate was loaded as the model drug in the polymeric nanoparticles. In vitro release behavior of prednisone acetate was investigated, which showed a dramatic thermoresponsive fast/slow switching behavior according to the temperature-responsive structural changes of a micellar shell structure. The reversible and sensitive thermoresponse of this micelle might provide opportunities to construct a novel drug delivery system in conjunction with localized hyperthermia.

Abstract:Thermo-sensitive amphiphilic block copolymers, poly(N-isopropylacrylamide)-block- poly(D,L-lactide) (PNIPAAm-b-PLA), were synthesized through a simple free radical copolymerization route based on a bifunctional initiator, 2,2'-azobis(2-methylpropion amidine) dihydrochloride (AMAD), followed by the ring-opening polymerization of D,L-lactide in the presence of Sn(Oct)2 catalyst. The Chemical structure characterization were conducted by means of 1H NMR, FT-IR, GPC. The amphiphilic PNIPAAm-b-PLA block copolymers could self-assemble into micelles with regularly spherical shape in an aqueous solution, with a TEM diameter range of 46-56 nm, DLS hydrodynamic diameter of 158-199 nm. This behavior depends on the environmental temperature, the hydrophobic interactions among PNIPAAm molecular chains, intermolecular hydrogen bonding between the PNIPAAm chains and water molecules as well as intramolecular hydrogen bonding between -CONH2 groups. The copolymers held a CMC from 5.50 to 6.17 mg L(-1) and LCST from 31.41-32.03 degrees C, being more or less affected by their compositions, PLA or PNIPAAm block length. The as-prepared PNIPAAm-b-PLA block polymers are anticipated to be applied as a potential candidate of drug release carriers.

Abstract:Thermo-responsive amphiphilic poly(methyl methacrylate)-b-poly(N-isopropylacrylamide-co-N-acryloxysuccinimide) (PMMA-b-P(NIPAAm-co-NAS)) block copolymer was synthesized by successive RAFT polymerizations. The uncross-linked micelles were facilely prepared by directly dissolving the block copolymer in an aqueous medium, and the shell cross-linked (SCL) micelles were further fabricated by the addition of ethylenediamine as a di-functional cross-linker into the micellar solution. Optical absorption measurements showed that the LCST of uncross-linked and cross-linked micelles was 31.0°C and 40.8°C, respectively. Transmission electron microscopy (TEM) showed that both uncross-linked and cross-linked micelles exhibited well-defined spherical shape in aqueous phase at room temperature, while the SCL micelles were able to retain the spherical shape with relatively smaller dimension even at 40°C due to the cross-linked structure. In vitro drug release study demonstrated a slower and more sustained drug release behavior from the SCL micelles at high temperature as compared with the release profile of uncross-linked micelles, indicating the great potential of SCL micelles developed herein as novel smart carriers for controlled drug release.

Abstract:A novel thermosensitive amphiphilic ABA triblock poly( methyl methacrylate)-b-poly(N-isopropylacrylamide)-b-poly( methyl methacrylate) copolymer (PMMA-b-PNIPAAm-b-PMMA) comprised of two hydrophobic PMMA segments and one hydrophilic PNIPAAm segment was designed and synthesized. The structure of the copolymer was characterized by FT-IR, (HNMR)-H-1, and GPC analysis. The cytotoxicity study showed that the PMMA-b-PNIPAAm-b-PMMA copolymer exhibited low cytotoxicity. The copolymer was capable of self-assembling into micelles in water and demonstrated temperature sensitivity at around 34.5 degrees C. Transmission electron microscopy (TEM) showed that the micelles exhibit nanosized spherical morphology within a size range of 60 nm with a critical micellar concentration (CMC) at 10 mg l(-1). The drug-loading PMMA-b-PNIPAAm-b-PMMA micelles showed thermosensitive-controlled release which indicates the potential of PMMA-b-PNIPAAm-b-PMMA micelles as drug carriers.

Abstract:A novel thermosensitive amphiphilic copolymer of PNIPAm grafted polyphosphazene (PNIPAm-g-PPP) has been synthesized through a co-substitution reaction of poly(dichlorophosphazene) with amino terminated NIPAm oligomer and glycine ethyl ester (GlyEt). The polymer obtained had a molar ratio of 1:5.25 PNIPAm to GlyEt and had a lower critical solution temperature (LCST) near 30 degreesC. The formation of polymeric micelles was confirmed by fluorescence, dynamic light scattering and transmission electron microscopy measurements.

Abstract:Thermosensitive poly(N-isopropyl acrylamide-co-acrylamide)-block-poly(d.l-lactide) (P(NIPAM-co-AM)-b-PLA) block copolymers were synthesized and characterized in the current study. The block copolymers could self-assemble into nanosized spherical core-shell micelles in aqueous media. Physicochemical properties were examined by surface tension, UV-vis. TEM and DLS measurements. Three water-insoluble drugs. paclitaxel (PTX), 10-hydroxycamptothecine (HCPT) and prednisone, were encapsulated into the copolymer micelles to obtain an optimal micelle-based drug formulation. The experimental results revealed that the copolymer micelles had a cloud point from 43 to 54 C, critical aggregate concentration from ca 7 to 16 mg L-1, and hydrodynamic size from 71 to 81 urn. The PTX-loaded polymer micelle had higher drug loading capacity and entrapment efficiency as well as more excellent thermo-triggered in vitro drug release trait than other two drugs. The MTT cytotoxicity assay indicated that the copolymer micelles exhibited good biocompatibility, and therefore they were potentially feasible as effective target therapy carriers.

Abstract:Tribrachia star-shaped random copolymers with tunable thermosensitive phase transition temperature were designed and synthesized via a simple one-pot ammonolysis reaction approach with trimesic acid as cores. The self-assembly micellization behavior of the copolymers in aqueous solution was examined by surface tension, UV-vis transmittance, transmission electron microscope, and dynamic light scattering measurements, etc. The results indicated that the resultant copolymers formed thermosensitive micelle aggregates through hydrophobic interactions among the isopropyl groups of poly(N-isopropylacrylamide) PNIPAM chains and inter-star association at a polymer concentration above critical aggregation concentrations from 4.06 to 6.55mgL(-1), with a cloud point range from 36.6? to 52.1?, and homogeneously distributed micelle size below 200nm. The arm length and the compositional ratios of the two comonomers had effect on physicochemical properties of the polymer micelle aggregates. Particularly, the cloud point values were enhanced as the (N,N-dimethylacrylamide) DMAM monomer was introduced and reached to 36.6? and 41.0?-44.7? when the mass ratio of NIPAM to DMAM was 90:10 and 80:20, respectively. The thermo-triggered drug release and cytotoxicity were evaluated to confirm the applicability of the random copolymer micelle aggregates as novel drug targeted release carriers.

Abstract:Thermosensitive P(NIPAM-co-NMAM)-g-(PLLA-g-MAAH) graft copolymers were synthesized via a free radical polymerization route using 2,2'-azobisisobutyronitrile(AIBN) as an initiator and 2- aminoethanethiol hydrochloride(AET·HCl) as a chain transfer agent.The structure,molecular weight and polydispersity of the copolymer were characterized by NMR,FTIR and GPC.The morphologies,size and size distribution of the as-synthesized copolymer micelles were investigated by TEM and DLS.The thermosensitive behavior and micellization were examined with UV-vis and surface tension measurements.The experimental results showed that the as-synthesized P(NIPAM-co-NMAM)-g-(PLLA-g-MAAH) copolymers formed sphere-like micelles in aqueous solution,with an average size of approximate 20 nm,a low critical solution temperature of about 42℃,and a critical micelle concentration of ca.28 mg/L.MTT assay indicated that the copolymers had no cytotoxicity or slight toxicity,and could be used as a potential biomedical material.

Abstract:Shell cross-linked (SCL) thermoresponsive hybrid micelles consisting of a cross-linked thermoresponsive hybrid hydrophilic shell and a hydrophobic core domain were synthesized from poly(N-isopropylacrylamide-co-3- (trimethoxysilyl)propyl methacrylate)-b-polymethyl methacrylate (P(NIPAAm-co-MPMA)-b-PMMA) amphiphilic block copolymers. Transmission electron microscopy (TEM) images showed that the SCL micelles formed regularly globular nanoparticles. The SCL micelles showed reversible dispersion/aggregation in response to temperature cycles through an outer polymer shell lower critical solution temperature (LCST) for PNIPAAm at around 33 degrees C, observed by turbidity measurements and dynamic light scattering (DLS). The drug loading and in vitro drug release properties of the SCL micelles bearing a silica-reinforced PNIPAAm shell were further studied, which showed that the SCL micelles exhibited a much improved entrapment efficiency (EE) as well as a slower release rate which allowed the entrapped molecules to be slowly released over a much longer period of time as compared with pure PNIPAAm-b-PMMA micelles.

Abstract:In this paper, novel thermosensitive poly(N-isopropylacrylamide) (PNIPAM) nanocapsules with temperature-tunable diameter and permeability are reported. Firstly, the core-shell composite microparticles were synthesized by precipitation polymerization with isothiocyanate fluorescein (FITC) entrapped SiO2 as core and cross-linked PNIPAM as shell. Then, the SiO2 core was etched by hydrofluoric acid at certain condition and the pre-trapped FITC molecules remained within the inner cavity. The FITC release profile and TEM studies clearly indicate that the release behavior of FITC could be controlled effectively by the external temperature. Above the LCST of PNIPAM (32°C), the dehydrated PNIPAM shell inhibited the release of FITC from the internal cavity while below its LCST, the fluorophore could permeate the swollen shell easily.
Synthesis and Micellization of Thermo- and Ph-Responsive Block Copolymer of Poly(n-Isopropylacrylamide)-block-poly(4-vinylpyridine)

Abstract:A novel double-hydrophilic block copolymer poly(N-isopropylacrylamide)-block-poly(4-vinylpyridine) (PNIPAM-b-P4VP) with low polydispersity which could respond to both temperature and pH stimuli in aqueous solution was synthesized by atom transfer radical polymerization. Micellization of the copolymer in aqueous solution was characterized by dynamic and static laser scattering, 1H NMR and transmission electron microscopy. In aqueous solution, the copolymer existed as unimer at pH 2.8 at 25°C. When the temperature was raised to 50°C at pH 2.8, the copolymer associated into spherical core–shell micelles with the PNIPAM block forming the core and the P4VP block forming the shell. On the other hand, when pH was increased from 2.8 to 6.5 at 25°C, the copolymer associated into spherical core–shell micelles with the core formed by the P4VP block and the shell formed by the PNIPAM block. The process was reversible. The critical aggregation temperature of the block copolymer is 36°C, and the critical aggregation pH value is 4.7.

Abstract:A series of poly(N-isopropylacrylamide)(PNIPAM)-blocked polycarbonates,P(MAC-co-DTC)-b-PNIPAM,were designed as thermosensitive micelles for drug delivery.The amphiphilic block copolymers were synthesized by enzymatic ring-opening copolymerization of HO-PNIPAM,5-methyl-5-allyloxycarbonyl-trimethylene carbonate(MAC) and 5,5-dimethyltrimethylene carbonate(DTC).The novel block copolymers were characterized by1H-NMR and GPC-MALLS analyses.All the amphiphilic copolymers were able to self-assemble into nano-sized micelles in aqueous solutions with hydrophilic PNIPAM shells and hydrophobic polycarbonates cores.The critical micelle concentration(CMC) determined by fluorescence spectroscopy using pyrene as a probe was around 20~60 mg/L.Transmission electron microscopy(TEM) measurements showed that the micelles presented even spherical shape and the diameters of them were about 20~70 nm.The micelles exhibited thermosensitivity at lower critical solution temperature(LCST) of around 38~43℃,which was related to the hydrophilic PNIPAM blocks.In vitro experiments demonstrated that the P(MAC-co-DTC)-b-PNIPAM showed low cytotoxicity.Furthermore,the micelles were loaded with hydrophobic drug of prednisone acetate(PA) and in vitro drug release investigation also displayed thermosensitivity.The results suggested that P(MAC-co-DTC)-b-PNIPAM copolymers would be a promising thermosensitive carrier for drug delivery.
