In Vivo Transfer of Hepatocyte Growth Factor Attenuated Acute Liver Injury in Mice

Abstract:Objective To establish an effective method for hepatocyte growth factor (HGF) gene transfer in vivo and to investigate its effects in acute experimental liver injury.Methods Human HGF cDNA was ligated to pEGFP vector.Recombinant plasmid was transfected into mice with liposome in vivo.The expression of HGF protein and green fluorescent protein (GFP) was observed by fluorescence microscopy.Plasma hHGF concentrations were determined by ELISA.A mouse model of acute hepatic failure was induced by 90% hepatectomy.Plasma alanine aminotransferase (ALT),bilirubin and survival rate were measured.Hepatic PCNA index was assessed by immunohistochemistry.Results The expression of GFP in transfected hepatocytes was observed under fluorescence microscopy.Mice infected with hHGF gene showed a protective effect to acute hepatic failure induced by hepatectomy.Serum ALT and Bil was decreased in mice infected with hHGF gene as compared with control group.The survival rate was remarkably enhanced in the mice infected with hHGF gene.Conclusion Naked DNA injection and transfer with liposome efficiently brings about HGF expression in vivo,which can attenuate acute liver failure.
