Abstract:Objective:To find out a feasible way to detect the tumor stem-like cells in cervical cancer cells,and to analyze the characteristics of the side population(SP) cells being sorted.Methods:Two different human cervical cancer cell lines(HeLa、SiHa)were stained with Hoechst 33342 and a small fraction of SP cells verified by fluorescence-activated cell sorting(FACS).The expression of BCRP(breast cancer resistance protein) between SP and non-SP cells was compared by Immunocytochemistry.The oncogenicity of the SP and non-SP cells were analyzed by tumor formation in nonobesediabeti-c/severe combined immune-deficient(NOD/SCID) mice.MTS was performed to compare the proliferation and drug sensitivity among SP、 non-SP and HeLa cells.Result:A few of SP cells were identified in HeLa [(1.07±0.32)%] and SiHa [(0.83±0.76)%] cell lines.There was high level expression of BCRP in SP cell compare with non-SP cell.There was no difference for tumor incidence、latency of tumorigenesis and tumor volumes between the SP and non-SP cells sorted from HeLa.However,Tumor latency was shorter and tumor volumes were larger for SP cell sorted from SiHa cells compared with non-SP cells.The growth rate of SP cells was significantly higher than non-SP and HeLa cells after cultured for 7days in vitro.The chemo-resistance result shows the SF(survival fraction) of SP[(86.68±8.78)%] was higher than non-SP[(49.06±6.26)%] and unsorted HeLa[(43.69±4.84)%] in 0.05μmol/L TSA(P<0.05).Conclusion:These SP cells are enriched with stem-like cancer cells characteristics for self-regeneration,high proliferative capacity and chemotherapy resistance.This characterization may provide new insights into novel therapeutic targets for human cervical cancer treatment.
