Abstract:Insulin resistance related diseases such as type 2 diabetes mellitus are spreading rapidly in Asia and are probably closely correlated with catch-up growth caused by rapid elevation of nutrient status. Abstract:Objective To investigate the effects of adult catch-up growth on insulin sensitivity and stress in rats, as well as the probable mechanism of insulin resistance. Methods Male Sprague-Dawley rats were divided into 6 groups:caloric restriction group ( R4, caloric restriction for 4 weeks) and normal controls for 4 weeks ( NC4 ) ; catchup growth group refed with normal chow( RN4, refeeding for 4 weeks after caloric restriction for 4 weeks), catch-up growth group refed with high-fat diet( RH4, refeeding for 4 weeks after caloric restriction for 4 weeks ), normal chow (NC8) or high-fat diet( HF8 ) controls for 8 weeks. The animal model of catch-up growth was devoloped by way of refeeding after caloric restriction as scheduled. The glucose infusion rate( GIR ), 2-deoxyglucose uptake and insulinsitmulated insulin signaling in skeletal muscle during hyperinsulinemic-euglycemic clamp, plasma corticosterone, and 11β-hydroxysteroid dehydrogenase type 1 ( 11β-HSD1 ) mRNA expression level in skeletal muscle were determined.Results After caloric restriction for 4 weeks, plasma corticosterone and 1 1 β-HSD1 mRNA expression in skeletal muscle were significantly higher in R4 group compared with NC4 group( both P＜0. 05 ), but there were no differences in 2-deoxyglucose uptake and Ser473 phosphorylation of Akt in skeletal muscle between two groups. The plasma corticosterone and 11β-HSD1 mRNA expression in skeletal muscle in RN4 group were significantly higher than those in NC8 group, and were higher in RH4 group than those in NC8 and HF8 groups; while the 2-deoxyglucose uptake and insulin-stimulated Ser473 phosphorylation of Akt in skeletal muscle during the clamp in RN4 were remarkably lower than those in NC8 group, and were lower in RH4 than those in NC8, HF8, and RN4 groups (all P ＜ 0. 05 ).Conctusions Catch-up growth rats refed with normal chow or high-fat diet are characterized by significant insulin resistance and stress in the whole body and skeletal muscle. These changes are more evident in catch-up growth rats refed with high-fat diet. The interaction of increased stress and diet might be of utmost importance in the etiology of insulin resistance in catch-up growth animals.
EFFECTS OF CATCH-UP GROWTH AFTER SEMISTARVATION ON INSULIN RESISTANCE IN RATS

Abstract:Objective: To study the effect of catch up growth after semistarvation on insulin resistance in rats. Methods: Wistar rats (n=40) were randomly divided into 5 groups: normal chow group (NC given diet with 18.94% of calories as fat for 12 w); high fat group (HF, given diet with 50.55% of calories as fat for 12 w); food restriction group (FR, 50% of their normal spontaneous food for 4 w); refeeding normal chow group (RN, after 4 w semistarvation, given normal diet for another 8 w ); refeeding high fat diet group (RH, after 4 w semistavation, given high fat diet for 8 w). Changes of body weight, height, and food intake were recorded. At the end of experiment, homeostasis model assessment of insulin resistance (HOMAIR), the rate of abdominal fat (including perirenal fat and epididymal fat) vs weight, plasm protein, blood lipid (including total cholesterol and triglyceride) were detected. Results: There were 82.5% rats getting catch-up growth in refeeding groups. Body weight of both refeeding groups could not catch that of NC group, but the rate of visceral fat vs weight was higher than that of NC group, and this rate of RH group was close to that of HF group. The changes of visceral fat vs body weight were consistent with that of TG levels. All nutritional status except high fat diet could not influence total cholesterol levels. RH and RN group’s HOMAIR were higher than those of NC group and HF group. All groups have similar fastingglucose levels. Conclusion: Catch-up growth can induce insulin resistance, but is not consistent with fat accumulation and hypertriglyceridemia in rats.

Abstract:This study was designed to explore the metabolic changes resulting from catch-up growth in adult (CUGA) induced by varying degrees of nutrition promotion after undernutrition and to confirm whether these changes are transient or not. The CUGA models were developed on rats refed on intakes of normal chow or high-fat diet after a period of caloric restriction. The growth of the rats measured by body weight and length stagnated during caloric restriction and then rapidly accelerated following refeeding. Catch-up growth in adult resulted in an increase in intramuscular and intrahepatic lipid content, visceral fat deposition, and insulin resistance, which is consistent with a transient rise in food efficiency during the early stage of refeeding. In addition, ectopic lipid deposition, visceral fat accumulation, and insulin resistance were more severe in rats refed the high-fat diet than rats refed the normal chow. These findings suggest that CUGA induced by rapid nutrition promotion could result in persistent lipid overaccumulation (increased visceral fat and ectopic lipid deposition) and drastic systemic insulin resistance. The effects of CUGA on metabolic characteristics are dependent on the type of diet that is used for refeeding, especially on the amount of fat intake.
The Characteristics of Changes in Body Composition in Adult Catch-Up Growth Rat and Its Association with System Insulin Sensitivity

Abstract:Objective To observe dynamic changes in body composition in adult catch-up growth (CUGA) rat model and to explore the relationship between body composition and system insulin sensitivity in CUGA.Methods Sprague-Dawley rats were randomly divided into normal chow (NC) and catch-up growth with normal chow (RN) groups.Body composition was measured by dual-energy X-ray absorptiometry,and average glucose infusion rate 60-120 (GIR60-120) in hyperinsulinemic-euglycemic clamp was calculated to evaluate systemic insulin sensitivity.Results After 4-week calorie restriction,percentage of body fat mass (BFM％),percentage of abdominal fat mass (AFM％) and AFM to BFM ratio were lower in RN group than NC group(P＜0.05),while percentage of lean body mass (LBM％) was relative higher in RN group(P＞0.05).After refeeding,BFM％,AFM％,and AFM to BFM ratio in RN group were all increased with the time,whereas LBM％ was gradually decreased.In RN group,GIR60-120 was positively correlated with BFM％,AFM％,and AFM to BFM ratio (P＜0.01),but there was no linear relationship between GIR60-120 and LBM％.Among these factors,AFM to BFM ratio exhibited more significant relationship with GIR60-120 than BFM％ and AFM％.Conclusions The essence of CUGA is a progress that is characterized by a disproportionately higher rate of body fat than lean tissue gain and a redistribution of body fat.This preferential catchup fat,especially the redistribution of body fat,may be a critical determinant for system insulin sensitivity in CUGA.
Insulin Sensitizers Improve the Glp-1 Secretion and the Amount of Intestinal L Cells on High-Fat-Diet-Induced Catch-Up Growth

Abstract:Objective: The aim of this study was to investigate whether insulin resistance can result in impaired glucagon-like peptide (GLP)-1 secretion and to determine whether insulin-sensitizing drugs could improve the secretion of GLP-1 in catch-up growth rats.Methods: Male Sprague Dawley rats were used to establish a catch-up growth model. At the end of weeks 6 and 14, these rats were euthanized to measure energy intake, body weight, plasma triacylglycerol, and nonesterified fatty acid. Fat mass percentage was analyzed by dual-energy x-ray absorptiometry scan. The GLP-1 concentrations were measured by enzyme-linked immunosorbent assay, the glucose infusion rates were measured by hyperinsulinemic-glucose clamp experiment. Quantification of the GLP-1 positive cells in distal ileum was done by immunohistochemical staining method. The L cell line NCI-H716 cells were treated in vitro with palmitate acid, the cells' viability, the carnitine palmitoyl transferase-1, and the insulin signaling pathway were detected.Results: Rats fed a high-fat diet rats had rapidly developed insulin resistance, impaired incretin effect, and a reduction in the number of intestinal L cells. The insulin sensitizers, metformin and pioglitazone, improved insulin resistance and the concentration of circulating GLP-1, increased the relative number of intestinal L cells to a certain degree. In vitro, the NCI-H716 cell viability was decreased and impaired insulin signaling pathway with palmitate acid treatment, metformin treatment could reverse these effects, whereas pioglitazone could not.Conclusions: Insulin resistance caused by a high-fat diet could result in reduced GLP-1 secretion; the insulin sensitizing drugs were able to improve the incretin effect in catch-up growth rats. Selective defect of PI3K-Akt pathway is involved in insulin resistance in small-for-gestational-age rats with catch-up growth

Abstract:AIM:To investigate the changes of insulin postreceptor signal pathway in the rats born small for gestational age(SGA) with catch-up growth(CUG) and to explore the possible mechanism of insulin resistance in CUG-SGA.METHODS: SGA rats born by pregnant rats with dietary restriction were randomly divided into insulin excitation group and control group.Blood glucose and serum insulin level were measured at the 4th week.Insulin resistance was evaluated by the homeostasis model assessment of insulin resistance(HOMA-IR).The β-cell function was evaluated by HOMA-β.The expression of insulin receptor substrate 1(IRS1),protein kinase B(Akt) and extracellular signal-regulated kinase(ERK),and their phosphorylation in gastrocnemius muscle were analyzed by Western blotting using tubulin as an internal control.RESULTS: The levels of serum insulin and HOMA-IR in CUG-SGA group and non-catch-up growth(NCUG)-SGA group were significantly higher than those in appropriate for gestational age(AGA) group(all P<0.01).The levels of serum insulin and HOMA-IR in CUG-SGA group were significantly higher than those in NCUG-SGA group(both P<0.01).The expression of p-IRS1 in CUG-SGA animals under basal condition was not significantly increased after insulin excitation.The expression of p-Akt in SGA group was significantly lower than that in AGA group under the basal condition and insulin excitation state,and that in CUG-SGA group was reduced by 50% as compared with NCUG-SGA group.The expression of p-ERK in CUG-SGA group was significantly higher under the basal condition and was significantly decreased after insulin excitation.A negative correlation between the expression of p-ERK and p-Akt in SGA rats was observed(r=-0.737,P<0.05;r=-0.658,P<0.05).CONCLUSION: SGA individuals all have insulin resistance,especially with CUG.The impairments of insulin receptor PI3K signal pathway in CUG-SGA individuals are more severe than that in NCUG-SGA.The activation of Ras-MAPK-ERK,another insulin signal pathway,brings about growth-enhancing effect on CUG-SGA individuals,which may be the possible postreceptor mechanism of the association of CUG and insulin resistance.
Insulin Resistance and Adiponectin Levels Are Associated with Height Catch-Up Growth in Pre-Pubertal Chinese Individuals Born Small for Gestational Age

Abstract:Background and objective The study was performed to determine whether catch-up growth is associated with the development of insulin resistance and to explore serum endocrine markers associated with the metabolism of adipose tissue in a Chinese population born small for gestational age(SGA) Subjects and methods We recruited 56 children born SGA with catch-up growth and 55 born without catch-up growth, who were further grouped into groups I (with BMI catch-up) and II (without BMI catch-up) respectively, as well as 52 children born appropriate for gestational age (AGA) with normal height. Their serum fasting insulin, fasting glucose, insulin-like growth factor-1, adiponectin, IGFBP-1, triglyceride concentrations, and the homeostasis assessment model for insulin resistance (HOMA-IR) were evaluated. Results (1) The HOMA-IR values in SGA-I with catch-up growth group were significantly higher than those in SGA-II with catch-up growth, SGA-I without catch-up growth and AGA children respectively. (2) The serum adiponectin levels of individuals in the SGA-I without catch-up growth and SGA-II with catch-up growth groups were significantly lower than those from the SGA-II without catch-up growth group. There was no difference in triglyceride or IGFBP-1 levels among the groups. (3) The degree of HOMA-IR was positively correlated with age, current BMI and △height SDS in SGA children. Conclusion The development of insulin resistance and lower levels of adiponectin were closely correlated with higher BMI and the postnatal height catch-up growth in SGA children.
Effect of Catch-Up Growth after Food Restriction on the Entero-Insular Axis in Rats

Abstract:BACKGROUND:Catch-up growth after food restriction (CUGFR) is characterized by a significant change in food intake which could theoretically lead to the change in glucagon-like peptide-1 (GLP-1) secretion that consequently results in altered functions of pancreatic islets.METHODS:Experimental rats were divided into two groups. Rats in CUGFR group were put on food-restriction for 4 weeks, and then allowed full access to food for 0, 2, 4 weeks respectively while rats in the control group were offered ad libitum access to food. Plasma glucose, insulin and GLP-1 level during OGTT were measured in all the rats. Moreover, morphology of intestinal mucosa, number of L cells, beta cell mass, incretin effect and the expression of GLP-1 receptor (GLP-1R) gene in the islets were also determined.RESULTS:The size of pancreatic islets, insulin concentration, plasma GLP-1 concentration, incretin effect, villus height-to-crypt depth ratio and L cells were all significantly decreased in CUGFR group at the end of a 4-week food-restriction period as compared with the controls. Insulin concentration and the villus height-to-crypt depth ratio were increased and finally exceeded the level of the control group over a 4-week catch-up period. Nevertheless, at the conclusion of the study, islet size, L cells number, plasma GLP-1 concentration and incretin effect increased but failed to reach the levels of the controls.CONCLUSION:CUGFR decreases incretin effect and disturbs the entero-insular axis partially by decreasing GLP-1 concentration, which might be responsible for the increased risk of metabolic disorder during CUGFR.
Alteration of FXR Phosphorylation and Sumoylation in Liver in the Development of Adult Catch-Up Growth.

Abstract:Catch-up growth in adult, is increasingly recognized as an important causative factor for the extremely prevalent insulin resistance-related diseases especially in developing countries/territories. We aimed to investigate the alteration of bile acids level, phosphorylation and sumoylation of its interacting protein, bile acid receptor/farnesoid X receptor and their downstream signaling pathway, as well as insulin sensitivity and lipid profile in catch-up growth in adult rats. Male Sprague-Dawley rats were randomly allocated into four groups for two sampling points: caloric restriction group, catch-up growth in adult refed with normal chow and their normal chow controls for four or eight weeks (N4, N8 individually).We found that total serum bile acids and farnesoid X receptor phosphorylation increased without significant changes in farnesoid X receptor sumoylation and its downstream small heterodimer partner expression at the end of caloric restriction stage, while the visceral fat decreased and insulin resistance never occurred in these animals; After refeeding, total serum bile acids, farnesoid X receptor phosphorylation and sumoylation, as well as Cyp7a1, SREBP-1c mRNA levels were higher with significant decrease in small heterodimer partner expression, which is associated fat accumulation, and drastic insulin resistance in whole body and skeletal muscle. Our findings demonstrated that the fat accumulation and insulin resistance are associated with increases of bile acids, alteration of farnesoid X receptor phosphorylation, and sumoylation and its downstream signaling pathway. These changes of bile acids, farnesoid X receptor phosphorylation and sumoylation, as well as their downstream signaling might be of importance in the etiology of fat accumulation and insulin resistance in catch-up growth in adult.
Differences of Glucose Metabolism Between Adipose Tissue and Skeletal Muscle in the Early Stage of Catch-Up Growth in Rats and Its Mechanism

Abstract:Objective To observe the glucose utilization in adipose tissue and skeletal muscle during catch-up growth, and to explore the mechanism of catch-up growth of adipose tissue. Methods Male Wistar rats were randomly divide into normal control group(NC)and catch-up group(RN). Rats in RN group received 50%of food consumed by NC group for 4 weeks, then were re-fed spontaneously as the rats in NC group. In the end of the fifth week(NC1 group and RN1 group)and the sixth week(NC2 group and RN2 group), the experiment was performed. [3]-2-deoxy-glucose was used for detecting the glucose uptake rate. RT-PCR and Western-blot were used for detecting the levels of mRNA and membrane protein of glucose transporter-4(Glut4). Results The glucose uptake rates in adipose tissue and skeletal muscle of RN1 group increased by 189.6%(P<0.01)and reduced by 36.5%(P<0.05)respectively, as compared with NC1 group. After 2 weeks of catch-up growth, the glucose uptake rates in adipose tissue of RN2 group increased by 157.3%(P<0.01)and decreased by 41.5%(P<0.05) in skeletal muscle as compared with NC2 group. However. no significant difference in Glut4 mRNA levels in muscle or in adipose tissue between NC and RN groups were found. The membrane protein of GIut4 after insulin-stimulating in RN1 group and RN2 group reduced by about 46.5%(P<0.01)and 32.1%(P<0.05)in muscle and increased by 116.5%(P<0.01)and 89.9%(P<0.01)in adipose tissue respectively. Conclusion There exists the redistribution of glucose from skeletal muscle to adipose tissue during the early stage of catch-up growth, which results in the catch-up growth of adipose tissue. This change is induced by the tissue-specific alteration of insulin-stimulated Glut4 protein translocation.

Abstract:According to the survey conducted by the Chinese Center for Disease Control and Prevention, the prevalence of type 2 diabetes in China reached 10.9% in 2013, 20-fold increase over the early 1980s. In the recent two decades, the incidence of diabetes in our country has risen up significantly, which is related to catch up growth caused by the rapid promotion of nutrition. It has been revealed that catch up growth can result in thermogenesis suppression, dysfunction of adipose tissue, lipid ectopic deposition in skeletal muscle, liver inflammation, impaired function of intestinal-pancreatic axis and islet β cell injury, leading to visceral fat accumulation and insulin resistance, thus increasing the risk of type 2 diabetes. Therefore, it is of great significance to deeply explore the mechanism of catch up growth and to find effective intervention strategies for the prevention and treatment of type 2 diabetes in China.

Abstract:Objective: To explore the relationship of postnatal catch-up growth and insulin sensitivity. Methods: Fasting glucose, insulin and HOMA value in 44 small for gestational age (SGA) infants and 74 appropriate for gestational age (AGA) infants at age 3 month were studied by a prospective study, and the relations with catch up growth were analyzed in 44 SGA infants. Results: SGA infants had significantly higher fasting insulin concentration and HOMA value than did AGA infants. Adjustment for gestation, there was a negative association between velocity of catch-up growth and insulin concentration and HOMA value in SGA infants ( P < 0. 05). 12 non-catch-up growth in weight and length infants had significantly higher fasting insulin concentration and HOMA value than did they on the third day after birth in SGA group respectively [ insulin : (21. 92 ±14. 25) mU /L vs (11. 08 ±9. 59) mU /L P < 0. 05 ; HOMA : (4. 15 ±2. 96) vs (1. 86 ±1. 71), P < 0. 05 ]. Conclusions: Catch-up growth is closely related with insulin sensitivity in early postnatal life, and poor growth might result in worse insulin sensitivity in SGA infant s during that time.
Involvement of PPARγ/FSP27 in the Pathogenic Mechanism Underlying Insulin Resistance: Tipping the Balance Between Lipogenesis and Fat Storage in Adult Catch-Up Growth Rats

Abstract:Catch-up growth in adult (CUGA) is characterized by visceral fat accumulation, ectopic lipid deposition and insulin resistance (IR). Here, we investigated the determinants of these pathophysiological consequences of CUGA. Rats were divided into different groups: control rats were offered normal chow ad libitum (AL), while experimental rats were put on 4-week caloric restriction (CR) initially, followed by regaining weight-matched normal chow (RN) in the RN group. General characteristics of lipid metabolism, expression level of genes in visceral adipose tissue (VAT), and glucose infusion rate (GIR60–120) by the hyperinsulinemic-euglycemic clamp were examined. After CR, percentage of abdominal fat mass (AFM%) was lower in the RN group than in the AL group but no difference was observed in serum non-esterified fatty acid (NEFA). Expression of fat-specific protein 27 (FSP27) was decreased in the RN group, while the expression of peroxisome proliferator-activated receptors γ (PPARγ), the key lipogenic gene, was increased. After refeeding, AFM% increased over time and serum NEFA persistently elevated in the RN group. Ectopic triglyceride contents were increased whereas insulin sensitivity was impaired. The expression of FSP27 did not follow the increase in the expression of PPARγ. Additionally, we observed a sustained increase in the expression of ATGL and CGI-58 in VAT in the RN group compared with the AL group after CR and refeeding, and a persistent shift-to-the-left of adipocyte size distribution accompanied by enhanced lipogenesis during CUGA. The persistent CR-induced imbalance of lipogenesis/fat storage capacity might be responsible for the CUGA-associated metabolic disorders.

Abstract:Objective To investigate whether liraglutide treatment can prevent catch-up fat in high-fat dietinduced catch-up growth rats,and to discuss the possible underlying mechanisms.Methods Sixty-four SpragueDawley rats were stratified into 4 groups:normal control group,catch-up growth (CUG) group,catch-up growth with liraglutide treatment (CUGL) group,and catch-up growth with liraglutide plus exendin (9-39) treatment (CUGLE) group.All the catch-up growth animals underwent a4 week high-fat diet re-feeding phase after a4 week food restriction phase.Body weight and food intake were measured every day.At the end of food restriction and re-feeding,body composition was measured via Dual energy X-ray absorptiometry.After the rats were sacrificed,body fat distribution and plasma lipid levels were determined.Results By the end of catch-up growth,body weight,energy intake,Lee' s index,fat body mass,visceral adipose tissue/body weight,subcutaneous adipose tissue/body weight,triglyceride,free fatty acid,low density lipoprotein-cholesterol levels were lower in CUGL rats compared with CUG rats (all P＜0.01).There were no significant differences in energy intake,body composition,fat body mass,and blood lipid levels between CUGLE rats and CUG rats,while the weight of CUGLE rats was significantly lower than CUG ones.Conclusions Liraglutide infusion protects rats with high-fat diet induced catch-up growth from catch-up fat mainly via a glucagon-like peptide-1 receptor dependent manner.

Abstract:Insulin resistance, an increasingly prevalent characteristic among children and adolescents with obesity, is now recognized as a significant contributor to the development of type 2 diabetes mellitus (T2DM) and other metabolic diseases in individuals with obesity. Insulin resistance refers to a decrease in the sensitivity of peripheral tissues (primarily skeletal muscle, adipose tissue, and liver) to insulin, which is mainly characterized by impaired glucose uptake and utilization. Although the mechanisms underlying insulin resistance in children with obesity remain incompletely elucidated, several risk factors including lipid metabolism disorders, oxidative stress (OS), mitochondrial dysfunction, inflammation, and genetic factors have been identified as pivotal contributors to the pathogenesis of obesity‐related insulin resistance. In this review, we comprehensively analyze relevant literature and studies to elucidate the underlying mechanisms of insulin resistance in childhood obesity. Additionally, we discuss treatment strategies for pediatric obesity from a perspective centered on improving insulin sensitivity, aiming to provide valuable insights for the prevention and management of pediatric obesity.
