Experimental study on tubacin inhibiting the growth of glioma cells

Abstract:Objective To investigate the effects of hi stone deacetylase 6 (HDAC6) selective inhibitor tubacin on glioma cell proliferation,migration and movement,and apoptosis and its mechanism of action.Methods The human glioma U251 cell lines were used as the research objects,a blank control group (CON),a temozolomide (TMZ) positive control group,and a tubacin group were set up.MTT assay was used to detect the cell proliferation activity.Transwell experiment was used to detect the cell migration ability.The high content screening (HCS) technology was used to observe the cell trajectory.Flow cytometry and Western blot were used to detect the apoptosis rate and the related apoptosis protein expression.The DCFH-DA fluorescence staining flow cytometry was used to detect the levels of intracellular reactive oxygen species (ROS).Results The results of MTT showed that the U251 cell reproductive capacity of the tubacin group was significantly lower than that of the CON group (P < 0.05).Compared with the CON group,the cell migration and motility ability was decreased significantly in the tubacin group.There was significant difference (all P < 0.05).The results of flow cytometry showed that the apoptosis rate was significantly increased in the tubacin group (P < 0.05).The results of Western blot showed that the expression levels of Caspase-3 and Bax were increased significantly in the tubacin group compared with the CON group (all P < 0.05),while the expression of Bcl-2 was decreased significantly (P < 0.05).The results of ROS assay found that the ROS level increased significantly in U251 cells after the action of tubacin (P < 0.05),and this effect of tubacin was dose-dependent.Conclusions Tubacin can inhibit glioma cell proliferation,migration,and motility,and promote its apoptosis.Its effect may be produced by inducing the cells to release ROS.
