Abstract:Background: The ketogenic diet (the KD) is an effective treatment for intractable epilepsy, especially in the paediatric population, and a growing number of studies have shown the neuroprotective role of the KD. However, few studies focused on the neuroprotective effects of the KD in traumatic brain injury (TBI). The present study aimed to investigate the effects of the KD on TBI. Methods and procedures: Male Sprague-Dawley rats (n = 60) were randomly divided into four groups according to the diet fed (the KD vs normal diet) and whether brain was injured or not. TBI was produced using Feeney weight drop model. Brain oedema was estimated by wet/dry weight ratio; Bax and Bcl-2 mRNA levels were determined by RealTime-PCR; Bax and Bcl-2 protein levels were detected by Western blot. Furthermore, cellular apoptosis in the penumbra area was examined using terminal deoxynucleotidyl transferase-mediated dUTP nick end labelling (TUNEL) method. Main outcomes and results: The results indicated that both Bax mRNA and protein levels were significantly elevated 72 hours after TBI and decreased by KD administration. Neither TBI nor the KD affected Bcl-2 mRNA and protein levels. KD administration also reduced brain oedema and cellular apoptosis. Conclusion: These results suggest that the KD might be a useful treatment for children suffering from the consequences of TBI.

Abstract:Ketone bodies have been shown to be favorable alternative metabolic substrates and are protective under neuropathologies. At the same time, cytochrome c release has been reported following traumatic brain injury (TBI) and precipitates apoptosis via the mitochondrial pathway. The present study investigated the effects of a ketogenic diet (KD) on TBI. TBI was produced using the Feeney weight-drop model and the animals were fed either normal diet (ND) or KD. Brain edema was estimated by wet/dry weight ratio; cytochrome c was detected by Western blotting; cellular apoptosis in the penumbra area was examined by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) and active caspase-3 immunohistochemical staining. The results show that brain edema, cytochrome c release, and cellular apoptosis were induced after TBI and that KD reduced these changes dramatically. These findings suggest that KD has potential therapeutic benefit in TBI.
Neuroprotective Effects of (-)-epicatechin on Traumatic Brain Injury Mice by Nuclear Factor E2-related Factor 2 Pathway

Abstract:Objective To investigate the neuroprotective effect of (-)-epicatechin (EC) on traumatic brain injury (TBI) mice.Methods TBI mouse model was prepared by modified weight drop method.Animals were randomly divided into Sham group,TBI + Veh group and TBI + EC group.At 3 days after TBI,Western blotting was used to detect the expression of Nrf2,SOD1 and NQO1.On days 7 and 28 post TBI,Neurologic deficit score (NDS) and wire hanging test were used to detect the motor function.Tail suspension test and forced swim test were used to evaluate the depressive status of mice after TBI on day 28.Results (1) Compared with Veh group,the expression of Nrf2 in nucles was obviously higher (1.354 ±0.432 vs.0.847 ±0.345) in EC treated group.The expression of SOD1 and NQO1 (1.128 ± 0.326) and (1.325 ± 0.427),P ＜ 0.05) was significantly increased,compared with them in the Veh group [(0.855 ± 0.365) and (1.025 ± 0.317)].NDS showed that EC treatment group on 7 d and 28d scores of motor function [(7.18 ± 1.26) and (3.25 ± 0.91)] significantly improved,compared with the Veh Group [(9.75 ± 1.16) and (6.63 ± 1.06),P ＜ 0.05].In the wire hanging test,the mice in EC group have more latency time (22.48 ±2.26) s and (45.35 ±2.92) s] compared to it in the Veh group [(12.23±2.11) sand (22.53±2.32) s,P＜0.05] on day 7 and 28 post TBI.(3) On day 28,in the tail suspension test and force swim test,the immobility time in the EC treated group [(202.88 ± 12.56) and (101.98 ± 22.43)] is shorter than it in the vehicle treated group [(244.48 ± 22.76) and (145.39 ± 19.86),P ＜ 0.05].(4) On day 28 post injury,compared with the Veh group,the lesion volume is smaller in the EC treatd group [(8.95 ± 1.59) mm3 vs.(11.52 ± 1.17) mm3,P ＜0.01].Conclusion EC protects the brain after TBI by NRf2 pathway.
Ketogenic Diet Modulates Neuroinflammation via Metabolites from Lactobacillus reuteri After Repetitive Mild Traumatic Brain Injury in Adolescent Mice

Abstract:Repetitive mild traumatic brain injury (rmTBI) is associated with a range of neural changes which is characterized by axonal injury and neuroinflammation. Ketogenic diet (KD) is regarded as a potential therapy for facilitating recovery after moderate-severe traumatic brain injury (TBI). However, its effect on rmTBI has not been fully studied. In this study, we evaluated the anti-neuroinflammation effects of KD after rmTBI in adolescent mice and explored the potential mechanisms. Experimentally, specific pathogen-free (SPF) adolescent male C57BL/6 mice received a sham surgery or repetitive mild controlled cortical impacts consecutively for 7 days. The uninjured mice received the standard diet, and the mice with rmTBI were fed either the standard diet or KD for 7 days. One week later, all mice were subjected to behavioral tests and experimental analysis. Results suggest that KD significantly increased blood beta-hydroxybutyrate (β-HB) levels and improved neurological function. KD also reduced white matter damage, microgliosis, and astrogliosis induced by rmTBI. Aryl hydrocarbon receptor (AHR) signaling pathway, which was mediated by indole-3-acetic acid (3-IAA) from Lactobacillus reuteri (L. reuteri) in gut and activated in microglia and astrocytes after rmTBI, was inhibited by KD. The expression level of the toll-like receptor 4 (TLR4)/myeloid differentiation primary response 88 (MyD88) in inflammatory cells, which mediates the NF-κB pathway, was also attenuated by KD. Taken together, our results indicated that KD can promote recovery following rmTBI in adolescent mice. KD may modulate neuroinflammation by altering L. reuteri in gut and its metabolites. The inhibition of indole/AHR pathway and the downregulation of TLR4/MyD88 may play a role in the beneficial effect of KD against neuroinflammation in rmTBI mice.
Neuroprotective Effect of Erythropoiet in Treatment of Traumatic Brain Injury in Rats

Abstract:Objective To evaluate the neuroprotective effects of erythropoiet (EPO) for traumatic brain injury (TBI) in rats and investigate the possible role of endoplasmic reticullum stress response and Caspase-12-induced apoptosis.Methods According to the random number table,140 male Sprague-Dawley rats were divided into sham injury group,sham injury plus EPO group,TBI group and TBI plus EPO group,with 35 rats per group.TBI was induced by a fluid percussion device.EPO (5 000 U/kg in saline) was administered intraperitoneally at 6 hours after injury.The rate of TUNEL positive cells in injured cortex were measured to evaluate cell apoptosis status.Neurological function was assessed at days 1,4,7,21,28 and 35 after intervention using a modified neurological severity score (mNSS).At 24 hours after injury,the expressions of Caspase-12 in injured cortex and C/EBP-homologous protein (CHOP) which was the symbol of ERS response were measured by Western blot and immunofluorescent staining to assess the changes of ERS response after TBI and EPO treatment.Results TUNEL-positive staining cell density was significantly increased by (30.3 ± 2.3) ％ in the injured cortex 24 hours after injury (P ＜ 0.01).Compared with TBI group,TBI plus EPO group had a significant decrease of the positive rate of TUNEL cells [(14.6 ± 1.5) ％] (P ＜ 0.01).Compared with TBI group,mNSS score significantly was decreased in TBI plus EPO group at 7-35 days after injury (P ＜ 0.05).At 24 hours after injury,the results of Western blot showed that the expression levels of Caspase-12 and CHOP in the injured cortex in TBI group were higher than those in sham group,but that in TBI plus EPO group was lower than those in TBI group (P ＜ 0.01).At 24 hours after injury,the results of immunofluorescent staining showed the rates of Caspase-12 and CHOP positive cells in the injured cortex in TBI group were higher than sham group (P ＜ 0.01).But the rates of Caspase-12 and CHOP positive cells in TBI plus EPO group was lower than that in TBI Group (P ＜ 0.01).Conclusion Exogenous EPO has significant neuroprotective effects on TBI rats.EPO may exert its neuroproective effects through suppression of ERS response and inhibition of Caspase-12-induced apoptosis.
The Effects of Ketogenic Diet on Seizure and the Pathology of Hippocampus CA3 Area of Immature Rats

Abstract:Objective To explore the effects of ketogenic diet (KD) on the PTZ-induced seizure threshold and the pathological changes of hippocampus in immature rats. Methods To determinate the PTZ-induced seizure threshold of KD and the control groups by tail-vein infusion of PTZ and the seizure scoring as well as the pathological changes of hippocampus CA3 area of the two groups. Results The KD group showed higher PTZ-induced seizure threshold than the control group( P 0.01),but there was no difference of seizure intensity between them( P 0.05).The counts of normal neuron cells in hippocampus slides CA3 area of KD group are higher than that of the control group. Conclusions KD can elevate PTZ-induced seizure threshold in immature rats but does not change the seizure intensity.KD has showed the seizure protection effect in rats.

Abstract:Dietary modification would be the most translatable, cost-efficient, and, likely, the safest approach available that can reduce the reliance on pharmaceutical treatments for treating acute or chronic neurological disorders. A wide variety of evidence suggests that the ketogenic diet (KD) could have beneficial effects in acute traumatic events, such as spinal cord injury and traumatic brain injury. Review of existing human and animal studies revealed that KD can improve motor neuro-recovery, gray matter sparing, pain thresholds, and neuroinflammation and decrease depression. Although the exact mechanism by which the KD provides neuroprotection is not fully understood, its effects on cellular energetics, mitochondria function and inflammation are likely to have a role.
Ketogenic diet attenuates post-cardiac arrest brain injury by upregulation of pentose phosphate pathway–mediated antioxidant defense in a mouse model of cardiac arrest

Abstract:ObjectiveThe aim of this study was to investigate the effect of the ketogenic diet (KD) on post-cardiac arrest brain injury in a mouse model of cardiac arrest and cardiopulmonary resuscitation.MethodsMice were fed a KD for 4 wk and then subjected to cardiac arrest and cardiopulmonary resuscitation. The HT22 cells after β-hydroxybutyrate (β-OHB) treatment were exposed to oxygen-glucose deprivation/reoxygenation. Survival and neurologic function were measured after return of spontaneous circulation. Positron emission tomography/computed tomography scanning, 13C-magnetic resonance spectroscopy analysis, and seahorse assay were performed to explore the mechanism underlying the phenotype.ResultsResults of this study demonstrated that KD improved neurologic function and reduced apoptotic neurons in cardiac arrest and cardiopulmonary resuscitation mice. With no alteration of glucose uptake, KD suppressed glucose oxidation in mouse brain. Consistently, the glycolytic capacity of the HT22 cells was also downregulated by β-OHB treatment. Moreover, KD increased nicotinamide adenine dinucleotide phosphate/oxidized nicotinamide adenine dinucleotide phosphate and reduced glutathione/oxidized glutathione couples and reduced reactive oxygen species in the brain, probably due to activation of glucose-6-phosphate dehydrogenase, the rate-limiting enzyme in the pentose phosphate pathway. Pharmacologic inhibition of pentose phosphate pathway totally abolished the influence of β-OHB on glycolysis, and post–oxygen-glucose deprivation/reoxygenation cell viability and reactive oxygen species production in HT22 cells.ConclusionKD improved survival and attenuated post-cardiac arrest brain injury, which was mediated by upregulation of pentose phosphate pathway and related antioxidant defense system.

Abstract:Ketogenic diet (KD) is a classical nonpharmacological therapy that has recently been shown to benefit cerebral ischemia, but the mechanism remains unclear. This study investigated the neuroprotective effects of KD pretreatment and β-hydroxybutyrate (BHB, bioactive product of KD) post-treatment in a mouse model of temporary middle cerebral artery occlusion (tMCAO). Neurological function, infarct volume, as well as inflammatory reactions are evaluated 24 hours after ischemia. Results showed that both KD pretreatment or BHB post-treatment improved the Bederson score and Grip test score, reduced infarct volume and the extravasation of IgG, suppressed the over-activation of microglia, and modulated the expression of cytokines. Mechanically, we found that both KD pretreatment or BHB post-treatment significantly stimulated the expression of interleukin-1 receptor-associated kinase M (IRAKM) and then inhibited the nuclear translocation of NF-κB. IRAKM deletion ( Irakm −/− ) exacerbated tMCAO-induced neurovascular injuries, and aggravated neuroinflammatory response. Moreover, KD pretreatment or BHB post-treatment lost their neuroprotection in the tMCAO-treated Irakm −/− mice. Our results support that KD pretreatment and BHB post-treatment alleviate ischemic brain injury in mice, possibly via an IRAKM-dependent way.

Ketogenic Diet Attenuates Neuronal Injury Via Autophagy and Mitochondrial Pathways in Pentylenetetrazol-Kindled Seizures.

Abstract:Autophagy alterations have been observed in a variety of neurological disorders, however, very few studies have focused on autophagy alterations in epilepsy. The ketogenic diet (KD) likely ameliorates neuronal loss in several seizure models. However, whether this neuroprotective function occurs via starvation-induced autophagy and its prevalence in chronic kindled seizures remains unknown. The aim of this study was to determine the role of autophagy following seizure under KD, and the potential mechanism involved. Pentylenetetrazol (PTZ)-kindled rats, which were fed a Normal diet (ND) or KD, were pretreated with intraventricular infusions of saline, autophagy inducer rapamycin (RAP), or inhibitor 3-methyladenine (3-MA). KD alleviated seizure severity, decreased the number of Fluoro-jade B (FJB)-positive cells in the hippocampus of kindled rats. These effects were abolished by 3-MA pretreatment. RAP pretreatment did not affect seizure severity, but decreased the number of FJB-positive cells in ND group. KD decreased the percentage of damaged mitochondria in kindled group. Hippocampal Beclin-1 was increased by KD in vehicle group. The autophagy proteins Atg5, Beclin-1 and the ratio of microtubule-associated protein 1 light chain 3 (LC3) II to LC3-I in kindled KD-fed rats were higher, and the autophagy substrate P62 was lower than those in the kindled ND-fed rats, indicating an increase in autophagy following KD. Pretreatment with RAP increased the level of LC3-II/LC3-I, and pretreatment with 3-MA increased the level of P62 in KD-fed rats. To further clarify the mechanism of autophagy protection, the levels of key mitochondria related molecules were examed. The results showed that mitochondrial cytochrome c was up-regulated, cytosolic cytochrome c and the downstream cleaved caspase-3 was down-regulated in KD-fed rats, indicating a decrease in mitochondrial apoptosis. Taken together, our results indicated that KD activates autophagic pathways and reduces brain injury during PTZ-kindled seizures. The neuroprotective effect of KD is likely exerted via a reduction of mitochondrial cytochrome c release.
Ketogenic Diet Improves Brain Ischemic Tolerance and Inhibits NLRP3 Inflammasome Activation by Preventing Drp1-Mediated Mitochondrial Fission and Endoplasmic Reticulum Stress.

Abstract:Background: Neuroprotective effects of ketogenic diets (KD) have been reported in stroke models, and nucleotide-binding domain (NOD)-like receptor protein 3 (NLRP3) inflammasome has also been implicated in the pathogenesis of stroke. This study aimed to investigate the effects of KD on NLRP3 inflammasome and explore the potential molecular mechanisms. Methods: In in vivo study, mice were fed with KD for 3 weeks and then subjected to middle cerebral artery occlusion/reperfusion (MCAO/R)-injury. In in vitro study, SH-SY-5Y cells were treated with beta-hydroxybutyrate (BHB) followed by oxygen-glucose deprivation/reoxygenation (OGD/R). NLRP3 inflammasome activation and related regulatory mechanisms were evaluated. Results: Mice fed with KD had increased tolerance to MCAO/R. KD inhibited endoplasmic reticulum (ER) stress and suppressed TXNIP/NLRP3 inflammasome activation in the brain. The in vitro study showed BHB (10 mM) prevented the mitochondrial translocation of dynamin-related protein 1 (Drp1) to inhibit mitochondrial fission. Furthermore, BHB decreased reactive oxygen species (ROS) generation, inhibited ROS-NLRP3 pathway in OGD/R-treated cells, and suppressed ER stress-induced NLRP3 inflammasome activation. Conclusions: KD may suppress ER stress and protect mitochondrial integrity by suppressing the mitochondrial translocation of Drp1 to inhibit NLRP3 inflammasome activation, thus exerting neuroprotective effects. Our findings provide evidence for the potential application of KD in the prevention of ischemic stroke.

Abstract:Objective To investigate the effect of ketogenic diet on hippocampus injury in pentylenetetrazol-kindled seizure and its relation to mammalian target of rapamycine (mTOR) pathway in rats.Methods The PTZ-kindled seizure model was induced by intra-ventricular infusion of 35mg/kg pentylenetetrazol (PTZ) in male Sprague-Dawley (SD) rats.Forty four rats,in which the seizure was fully kindled,were divided into 4 groups with 11 rats in each group.Rats in ND+PTZ and ND+PTZ+RAP groups were fed with normal diet,while those in KD+PTZ and KD+PTZ+RAP were fed with ketogenic diet.The mTOR pathway inhibitor rapamycin(RAP) was given to rats in ND+PTZ+RAP and KD+PTZ+RAP groups by intra-ventricular infusion 30min before PTZ injection.The seizure stages and the neuron in hippocampal CA1,CA3 and DG regions were counted.And the expressions of pS6,S6,pAkt,Akt,pAMPK and AMPK were detected by Western blot.Results The seizure stage of the kindled rats in the KD+PTZ group was lower than that of ND+PTZ group significantly (P＜0.05).The stages of KD+PTZ+RAP group were lower than those of ND+PTZ and ND+PTZ+RAP groups significantly (P＜0.05).The density of neuron in hippocampal CA1 subarea in KD+PTZ group was higher than ND+PTZ group significantly (P＜0.05),and the density of KD+PTZ+RAP was higher than ND+PTZ+RAP group (P＜0.05).Additionally,the density of ND+PTZ+RAP was higher than ND+PTZ (P＜0.05).Western blot result shows that the expression of phosphor-S6 to total S6(pS6/S6) and phosphor-Akt to total Akt(pAkt/Akt) was increased in ND+PTZ group compared to control (P＜0.05).The expression levels of phosphor-AMPK to total AMPK (pAMPK/AMPK) in all kindled groups were higher than those of non-kindled control groups (P＜0.05).Compared to the ND+PTZ group,the expression levels of pS6/S6 and pAkt/Akt were decreased (P＜0.05),while the pAMPK/AMPK was increased in KD+PTZ group (P＜0.05).Compared to the KD+PTZ group,the expression of pS6/S6 and pAkt/Akt in KD+PTZ+RAP group were decreased (P＜0.05).Similarly,compared to the ND+PTZ group,the expression of pS6/S6 and pAkt/Akt in ND+PTZ+RAP group was also significantly decreased (P＜0.05).Moreover,compared to ND+PTZ+RAP,the expression level of pS6/S6 in KD+PTZ+RAP group was decreased significantly (P＜0.05),while the expression level of pAMPK/AMPK was increased (P＜0.05).Conclusion Our study indicated that ketogenic diet has the anti-seizure and neuronal protective effect in PTZ kindled rat model,which may be related to the mTOR pathway suppression.Moreover,the mTOR pathway is modulated by both the inhibition of Akt and activation of AMPK pathway.

Abstract:Objective: To investigate the effects of ketogenic metabolism on macrophage polarization, inflammation inhibition, and function recovery after acute spinal cord injury (SCI) in rats.Methods: Sixty-four adult male Sprague–Dawley rats were randomly and equally divided into sham, standard diet (SD), ketone diet (KD), and 1, 3-butanediol (BD) groups. All animals underwent C5 unilateral laminectomy, whereas the SD, KD, and BD groups underwent C5 spinal cord hemi-contusion. The impact rod with a diameter of 1.5 mm was aligned 22.5° to the left and 1.4 mm to the midline, and then triggered to deliver a set displacement of 1.5 mm at a speed of 100 mm/s. The gene expression of inflammatory factors as well as the protein expression of inducible nitric oxide synthase, arginase-1, and inflammatory factors were measured at 1 week post-injury. Serum ketone and behavior were evaluated every second week for 12 weeks. Then, histological analyses of the gray and white matter at the epicenter were conducted at 12 weeks post-injury.Results: The serum ketone levels of the KD and BD groups were significantly increased when compared with the SD group. The gene and protein expression of TNF-α and IL-1β tended to increase after the SCI, but were inhibited in the KD and BD groups. The protein expression of inducible nitric oxide synthase, marker of M1 macrophage, was inhibited in the KD and BD groups; on the other hand, the expression of arginase-1, marker of M2 macrophage, was boosted in the KD and BD groups. The usage of the ipsilateral forelimb was higher in the KD group than in the SD group. The hemi-contusive injury resulted in an obvious ipsilateral lesion area at the epicenter, and there was no significant difference between groups regarding the lesion size. However, the spared gray matter area was significantly greater in the KD group than in the SD and BD groups.Conclusion: The present study suggests that ketogenic metabolism promotes macrophage polarization to M2, inhibits an inflammatory response, and alleviates the loss of gray matter after SCI. A higher ketone level, such as that induced by the ketogenic diet, seems to benefit function recovery after SCI.

Abstract:The present study aimed to investigate the protective effects of docosahexaenoic acid (DHA) on traumatic brain injury (TBI) in rats. A model of TBI was induced by lateral fluid percussion injury in adult rats and rats were randomly divided into the TBI-model group, TBI-low DHA group and TBI-high DHA group, while other healthy rats were assigned to the sham-operated group. Motor recovery was tested with beam-walking trials at 2, 7 and 15 days post-TBI. Cognitive recovery was tested with Morris water maze trials at 15 days post-TBI. The expression levels of caspase-3, B-cell lymphoma 2 (Bcl-2) and Bcl-2-associated X protein (Bax) were measured by western blotting. DHA protected against motor deficits induced by TBI in beam walking tests. All TBI-model groups had longer escape latency and swimming distances than the sham groups. Compared with the TBI-low DHA group, the TBI-high DHA group demonstrated shorter escape latency and swimming distances. DHA inhibited the expression of caspase-3 and the inhibition effect was more obvious at a high dosage. Furthermore, DHA dose-dependently rescued neurons by upregulating the Bcl-2:Bax ratio. DHA supplementation was a viable strategy to mitigate injury from TBI.

Abstract:The ketogenic diet (KD) controls seizure and improves cognition in patients with drug refractory epilepsy. However, few experimental models have shown this neuroprotective effect on cognition. In this study, we investigated the cognitive protective effects of KD in pentylenetetrazol (PTZ)-kindled rats. We used two relatively low-stress behavioral assessment methods, the novel object recognition (NOR) task and the novel placement recognition (NPR) task, to reveal impairment in item and spatial memory, respectively. We used the Morris water maze (MWM) test for comparisons amongst memory assessment methods. The KD group had a slower body weight gain and shorter bregma-lambda length than the control normal diet (ND) group. KD did not increase anxiety or decrease motor activities in an open-field test. KD attenuated the decrease in exploration ratio both in NOR and NPR tasks in kindled rats. Compared to the kindled ND rats, kindled KD rats stayed longer in target quarter during the probe trial testing of MWM. However, there were no differences in memory acquisition based on the MWM test results. In conclusion, KD attenuated the spatial and item memory impairment in PTZ-induced seizures.
Neuroprotective mechanisms of OXCT1 via the SIRT3-SOD2 pathway after traumatic brain injury

Abstract:Background Ketones are not only utilized to produce energy but also play a neuroprotective role in many neurodegenerative diseases. However, whether this process has an impact on secondary brain damage after traumatic brain injury (TBI) remains unknown. OXCT1 (3-Oxoacid CoA-Transferase 1) is the rate-limiting enzyme in the intra-neuronal utilization of ketones. In this study, we investigated whether reduced expression of OXCT1 after TBI could impact neuroprotective mechanisms and exacerbate neurological dysfunction. Materials and methods Experimental TBI was induced by a modified version of the weight drop model, it is a model of severe head trauma. Expression of OXCT1 in the injured hippocampus of mice was measured at different time points using immunoblotting assays. The release of abnormal mitochondrial cytochrome c from neurons of the mouse injured lateral hippocampus was measured 1 week after TBI using immunoblotting assays. Neuronal death was assessed by Nissl staining and the level of reactive oxygen species (ROS) within the neurons of the injured lateral hippocampus was assessed by Dihydroethidium staining. Results OXCT1 was overexpressed in hippocampal neurons by injection of adeno-associated virus into the lateral ventricle. OXCT1 expression levels decreased significantly 1 week post-TBI. After comparing the data obtained from different groups of mice, OXCT1 was found to significantly increase the expression of SIRT3 and reduce the proportion of acetylated SOD2, thus decreasing the production of ROS in the injured hippocampal neurons, reducing neuronal death, and improving cognitive function. Conclusions OXCT1 has a critical previously unappreciated protective role in neurological impairment following TBI via the SIR3-SOD2 pathway. These findings highlight the potential of OXCT1 as a simple treatment for patients with TBI.

Abstract:Purpose: To investigate the effects of abietic acid (AA) on traumatic brain injury (TBI) in a rat model, and the underlying mechanisms of action. Methods: Twenty male Sprague-Dawley (SD) rats were randomly divided into four groups of 5 animals each: Sham, TBI, TBI + AA (14 mg/kg), and TBI + AA (28 mg/kg). Controlled cortical impact (CCI) model was used to induce TBI in rats. Western blot was used to determine the protein levels of Claudin 3, Occludin, ZO-1, Bax, Bcl-2, cleaved-caspase3, p65 NF-κB, p-p65 NF-κB, ERK, p-ERK, JNK, p-JNK, p38 and p-p38, while the expressions of TNF-α, IL-6 and IL-1β were determined using the applicable assay kits. Neurological deficit was assessed based on mNSS and brain water content. Results: Treatment with AA significantly reduced TBI-induced blood-brain barrier (BBB) damage, as well as apoptosis and neuroinflammation in a concentration-dependent manner (p < 0.001). The neuroprotective effect of AA was associated with the inhibition of the MAPK signaling pathways and subsequent suppression of NF-κB (p < 0.001). Conclusion: Abietic acid serves as a potential novel candidate for the treatment of TBI, and its anti-inflammatory and anti-apoptotic effects are related to the suppression of MAPK signaling pathways. Therefore, abietic acid may serve as a novel candidate for the treatment of TBI in humans.

Abstract:Traumatic brain injury (TBI) is the primary cause of child mortality and disability worldwide. It can result in severe complications that significantly impact children's quality of life, including post‐traumatic epilepsy (PTE). An increasing number of studies suggest that TBI‐induced oxidative stress and neuroinflammatory sequelae (especially inflammation in the hippocampus region) may lead to the development of PTE. Due to the blood‐brain barrier (BBB), typical systemic pharmacological therapy for TBI cannot deliver berberine (BBR) to the targeted location in the early stages of the injury, although BBR has strong anti‐inflammatory properties. To break through this limitation, we developed a microenvironment‐responsive Gelatin methacrylate hydrogel (GM/PB) to deliver BBR for regulating neuroinflammatory reactions and removing ROS in the brain trauma microenvironment through poly(propylene sulfide)60 (PPS60). In situ injection of the GM/PB hydrogel efficiently bypassed the BBB and was administered directly to the surface of brain tissue. In post‐traumatic brain injury models, GM/PB has the potential to mitigate oxidative stress and neuroinflammatory responses, facilitate functional recovery, and lessen seizing. Abstract:Abstract To maintain its functional abilities, the mature brain obtains energy from glucose produced in carbohydrate metabolism. When carbohydrates are eliminated from the diet, the energy comes from the oxidation of fatty acids. In this metabolic state called ketosis, ketone bodies are formed: β-hydroxybutyric acid (bHb), acetone, and acetoacetate as alternative source of energy passing through the blood–brain barrier easily. The ketosis state can be achieved through various strategies like caloric restriction, supplementation with medium-chain triglycerides, intense physical training, or ketogenic diet (KD). Using KD, drug-resistant epilepsy has been successfully treated in children and adults. It can also exert neuroprotective influences in cases of brain damage, glioblastoma multiforme, and Alzheimer's or Parkinson's diseases. Although many possible mechanisms of KD activity have been proposed, newer hypotheses appear with the research progress, mostly characterizing the brain under pathological but not normal conditions. Since different pathological conditions may affect the mechanism of KD action differently, additional research on the normal brain appears reasonable. For this purpose, young adult rats were treated with 4-month-lasting KD. Then, MRI structural measurements, spectroscopy, and tractography were performed. The procedures revealed significant increases in the concentration of glutamine, glutamate, glutathione and NAA, accompanied by changes in the pattern of neuronal connections of the striatum and hippocampal formation. This implies a possible involvement of these structures in the functional changes occurring in the brain after KD application. Thus, the investigations on the normal brain add important details concerning mechanisms underlying KD effects without their possible modification by a pathological status.

Abstract:Objective Oxidative stress is a key pathophysiological response to traumatic brain injury (TBI). The Nrf2-ARE signal pathway has been found to have an antioxidative effect. This study aimed to verify the protective effect of Nrf2 inducer t-BHQ on traumatic brain injury in rats. Methods Fifty-four SD rats were equally randomized to three groups: control, TBI and medication. The rats in the medication group were intraperitoneally injected with t-BHQ 24 hours before brain trauma, and TBI was induced by Feeney's free weight drop method. Water content of the local brain tissue was measured, and apoptotic cells were detected by TUNEL 24 hours after brain trauma. Results Pretreatment with t-BHQ significantly decreased the local content of water and the apoptosis of the injured neurons. Conclusion Pretreatment with t-BHQ can effectively protect the damaged brain tissues after traumatic brain injury.
