Relationship of high mobility group boxl and inflammation

Abstract:The early studies of high mobility group box1 (HMGB1) focused on its role as a nonhistone DNA-binding protein. HMGB1 recently has been identified as a proinflammatory cytokine with large proinflammatory activity. It is now known that HMGB1 is secreted by activated monocytes, macrophages,mature dentritic cells(DC) and natural killer(NK) cells,also passively released by necrotic or damaged cells,and then it triggers immune inflammation response. HMGB1 itself has cytotoxicity, mice administrated HMGB1 systemically will die in a dose-dependent manner; macrophages and neutrophiles exposed with HMGB1 enhance the production of proinflammatory cytokines such as TNF and IL-1β. Anti-HMGB1 therapy confers significant protection against lethal endotoxaemia and sepsis, latently widens the therapeutic window of severe infections in clinic. In addition, HMGB1 also contributes to neurite outgrowth and the development of turnout.
