Effects of small interfering RNA-mediated transthyretin silencing on cellular biological behavior in PC3 cell line of prostate cancer and its mechanism

Abstract:Objective To explore the effect of transthyretin (TTR) on biological behavior of PC3 cells and its mechanism.Methods Three groups were designed,including the blank,the negative control and the experimental.By using the software,negative control RNA and TTR specific small interfering RNA (siRNA) sequences were designed,synthesized in vitro and combined with Lipofectamine2000 into PC3 cells.The expression of TTR was detected by using Western blotting to identify the efficacy of TTR silence.The proliferation,invasion and apoptosis of PC cells were determined by using methyl thiazol tetrazolium (MTT) assay,Transwell assay,and flow cytometry respectively.The level of c-myc was detected by using Western blotting to identify the relationship between c-myc and TTR expression.Results The expression level of TTR was decreased by 88.6% in the siRNA group as compared with the blank group and negative control group.In the siRNA group,the proliferation ability was decreased by 88.7% ; and invasion ability was also depressed to 0.09,significantly lower than that in the blank and negative control groups (0.36 and 0.33 respectively) (P < 0.01).The apoptosis in the siRNA group was up-regulated to 23.5 %,which was significantly different from that in the blank and negative control groups (7.3% and 8.2%,P <0.01).At the same time,the expression of c-myc correlated with the expression of TTR.Conclusion TTR silencing by siRNA can result in the depression of proliferation and invasion of PC3 cells and up-regulatethe apoptosis.TTR is correlated with c-myc signal pathway.
