Abstract:Objective To investigate the clinical implication of the decorin expression in cholangiocarcinoma.Methods The decorin expression levels were detected by immunohistochemistry in 44 cases of cholangiocarcinoma and 40 cases of normal bile duct tissues.The relationship between the decorin expression level with sex,age,tumor cell differentiation and lymph node metastasis of patients with cholangiocarcinoma was analyzede.Results Decorin expression was down-regulated in human cholangiocarcinomas cancer tissues.Decorin protein was positive in tumor interstitial tissues and the staining was weaker in cholangiocarcinoma tissues than the adjacent noncancerous tissues.The positive expression rate of decorin in human cholangiocarcinomas cancer tissues and the adjacent normal tissues was 40.91％ and 100.00％,respectively (P ＜ 0.01).The expression of decorin in middle-hight differentiation tumor group (57.14％) was significantly higher than in regional lymph node metastasis group (26.09％,P ＜ 0.05).The expression of decorin was not associated with sex,age and tumor cell differentiation of patients with cholangiocarcinoma.Conclusion The expression of decrorin appears to be tumor specificity to some extent in cholangiocarcinoma.

Abstract:OBJECTIVE:To investigate the value of detecting cytokeratin 19(CK19) and thyroperoxidase (TPO) expression in the diagnosis of thyroid diseases including thyroid carcinoma, multinodular goiter, adenoma and Hashimoto thyroiditis.METHODS:SP immunohistochemistry was used to detect the expressions of CK19 and TPO in paraffin-embedded thyroid tissue specimens obtained from 62 patients with thyroid carcinoma (30 with papillary carcinomas, 22 with follicular variant of papillary carcinomas, and 10 with follicular carcinomas) and 44 with benign thyroid diseases (including 22 with multinodular goiters, 14 with adenoma, and 8 with Hashimoto thyroiditis).RESULTS:CK19 expression was detected in 96.8% of the thyroid carcinomas and in 4.5% of benign thyroid diseases, demonstrating a significant difference in CK19 expression between the two thyroid diseases (P<0.01). TPO expression was found in 100% of benign thyroid disease and in 3.2% of thyroid carcinoma, showing also a significant difference between them (P<0.01).CONCLUSION:CK19 and TPO can be important molecular markers for diagnosing thyroid carcinoma.
The Study on Expression of Human Papillomavirus and Phosphotyrosione and Tyrosine Phosphatase 2 in Condyloma Acuminatum Tissue Remodeling

Abstract:Objective To explore the expression of human papillomavirus (HPV) 6/11,16/18,31/33 group and tyrosine phosphatase 2 (SHP-2) in condyloma acuminatum (CA) tissue remodeling.Methods HPV 6/11,16/18,31/33 group and SHP-2 were detected in skin lesions of 36 patients with condyloma acuminatum, tumor tissues of 13 cases of cervical cancer and normal foreskin tissue of 10 normal controls by in situ hybridization and SP immunohistochemical stain.Results The positive rates of HPV 6/11,16/18,31/33 group in condyloma acuminatum and cervical cancer were 80.5%,38.9%,36.1% and 69.2%,38.4%,53.8% respectively.The results showed that the expression of HPV 6/11,16/18,31/33 group in condyloma acuminatum and cervical cancer were significantly higher than those in normal epidermis (P0.05).The positive expression rates of phosphorylated tyrosine and SHP-2 in condyloma acuminatum and eerviaal cancer were 97.2%,22.2% and 100%,0%,respectively.However the cytoplasm of normal foreskin showed negative expression. Conclusion There is an aberrant activation of phosphorylated tyrosine and SHP-2 in condyloma acuminatum and cervical cancer. SHP-2 may play an important role in the HPV infection.
[Expression of CD10 in Tumor-Associated Fibroblast of Cancerized or Recurrent Colorectal Adenomas].

Abstract:Objective: To investigate the expression of CD10 in tumor-associated fibroblasts (TAF) in colorectal adenomas and its relation to cancerization and recurrence of adenoma. Methods: Tissue samples of low-grade adenoma (n=50), high-grade adenoma (n=50) and colorectal adenocarcinoma (n=50) were collected, and tissue samples at the distal margin of corresponding colorectal lesions were taken as controls. The expression of CD10 in the stromal TAFs, and the expressions of β-catenin, Ki-67, p53 and CyclinD1 in tumor cells were detected by immunohistochemistry (Envision). The correlation of CD10 expression in stromal TAFs with the expressions of β-catenin, Ki-67, p53 and CyclinD1 in tumor cells was analyzed by Spearmen. One hundred samples of low-grade colorectal adenoma were collected, including 57 non-recurrent cases and 43 recurrent cases (16 cases of recurrent adenoma and 27 cases of recurrent adenocarcinoma); the expression of stromal TAF CD10 were determined and compared among groups. Results: There was no TAF in normal colorectal mucosa. The expression rates of TAF CD10 in low-grade adenoma, high-grade adenoma and colorectal adenocarcinoma were 22%, 50% and 78%, respectively (all P<0.05). The expression of Ki-67 and β-catenin in low-grade adenoma, high-grade adenoma, colorectal adenocarcinoma was on a rising trend (all P<0.01). The expression of CyclinD1 in high-grade adenoma was higher than that in colorectal adenocarcinoma and low-grade adenoma (all P>0.05). The expression of p53 in colorectal adenocarcinoma and high-grade adenoma was higher than that in low grade adenoma (all P<0.01). The expression of TAF CD10 was correlated with the expression of p53, Ki-67 and β-catenin-nucleus(r=0.264、0.307、0.320, all P<0.01),but not correlated with CyclinD1 and β-catenin-membrane (r=0.012、-0.073, all P>0.05). The TAF CD10 level was significantly higher in low-grade adenoma with recurrence than that in those without recurrence (P<0.05).The expression of CD10 in recurrent colorectal adenocarcinoma was higher than that in recurrent adenoma (P<0.05). Conclusion: The expression of TAF CD10 is increased gradually in the process of adenoma-cancer, indicating that it may play an important role in the canceration of adenoma. Adenomas with high expression of CD10 TAF are likely to be recurrent and cancerized, and detection of TAF CD10 combined with p53, Ki-67 and β-catenin may be of value in predicting canceration or recurrence of colorectal adenoma.
Decorin impeded the advancement of thyroid papillary carcinoma by thwarting the EGFR/ FER/ SHP2 signaling-induced sustenance of early endosomes

Abstract:Objective: This study explores the inhibition of papillary thyroid carcinoma proliferation by Decorin via the EGFR/SHP2/FER pathway. Method: ology: Thirty-two pairs of papillary thyroid carcinoma tissues and adjacent normal tissues were collected for immunohistochemical analysis. Thyroid cancer cell lines with overexpressed or silenced Decorin were employed in subcutaneous tumor formation experiments in nude mice. Cell membrane proteins were extracted for Western blot and immunofluorescence analyses. Results: Reduced Decorin expression in human papillary thyroid carcinoma was associated with inhibited formation of the EGFR/SHP2/FER complex. Immunohistochemical analysis revealed lower Decorin levels in carcinoma tissues compared to adjacent normal tissues, corroborated by decreased Decorin and PTEN levels in carcinoma as shown by Western Blot. Overexpression of Decorin in mouse models diminished tumor growth, an effect reversed by Decorin silencing and mitigated by FER inhibition. Decorin modulated Rab5-GTP and Rab7-GTP levels, impacting endosome transition and subsequent signaling pathways. Conclusion: Decorin inhibits papillary thyroid carcinoma proliferation by disrupting the EGFR/SHP2/FER pathway and modulating endosomal transport.
Effect of Decorin Gene Overexpression on Migration and Invasion of Cholangiocarcinoma Cells

Abstract:Objective To investigate the effects of decorin (DCN) gene overexpression on the migration and invasion of human cholangiocarcinoma cells.Methods The pEGFP-DCN plasmids and pEGFP-N1 control plasmids were transfected into the human cholangiocarcinoma cell line QBC939 cells.The mRNA and protein expression of decorin and E-cadherin in QBC939 cells transfected with plasmid overexpressing decorin was detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting respectively.Transwell assay was used to observe the migration and invasion.Results Compared to the control plasmids,RT-PCR showed the mRNA expression of decorin in the pEGFP-DCN plasmids was increased by 32.34 fold.Western blotting showed the protein expression of decorin was increased by 2 fold.Transwell assay showed that the migration and invasion capacity of the QBC939 cells transfected with pEGFP-DCN was decreased by 56.9％ and 40.2％ respectively as compared with the control cells.The expression level of E-cadherin was significantly increased after DCN transfection into QBC939 cells as compared with the control cells.Conclusion After the over-expression of DCN gene,the migration and invasion capacity of the QBC939 cells was effectively suppressed,which may be associated with vthe elevated E-cadherin expression.
The Expression of Caspase-10 in Differentiated Thyroid Carcinoma and Association with Its Development and Metastasis

Abstract:Objective To investigate the expression of caspase-10 in differentiated thyroid carcinoma and association with its development and metastasis. Methods Thyroid samples from 37 patients in a period from January 2006 to December 2007, with differentiated thyroid carcinoma were retrospectively analyzed for caspase-10 by immunohistocbemistry(streptavidin-perosidase, S-P), compared to control group of 46 cases with nodtdar goiter. The relationship between the expression of caspase-10 and the clinical pathologic characteristics of thyroid carcinoma were also explored simultaneously. Results caspase-10 were observed as brown or yellow particles located in the cytoplasm or cell membrane of nodular goiter but there were no significant evidence for its positive expression in thyroid carcinoma, caspase-10 expression was markedly down-regulated in differentiated thyroid carcinoma(29.73%,11/37) compared with benign nodules(71.74%,33/46, χ2=14.528, P<0.01). The positive expression in 18 cases with lymph node metastasis(11.11%,2/18) was significantly lower than those in 19 patients without lymph node metastasis(47.37%,9/19; χ2=4.210, P<0.01). There was no significant correlation(P> 0.05) between the expression of caspase-10 and the clinical pathologic characteristics including male, age, TNM stage and pathologic type. Conclusion Down-regulation of caspase-10 may play a critical role in carcinogenesis and development of differentiated thyroid carcinoma.
Reduced decorin expression in the tumor stroma correlates with tumor proliferation and predicts poor prognosis in patients with I–IIIA non-small cell lung cancer

Abstract:Decorin, chiefly synthesized by tumor stroma, is known as a tumor suppressor. However, the clinical and prognostic significance in lung cancer remained unclear. Here, decorin and Ki67 expression was detected by immunohistochemistry (IHC) in I–IIIA non-small cell lung cancer (NSCLC) tissues (n = 264) in comparison to adjacent normal tissues (n = 40). The relationship between the expression of decorin and clinical characteristics, as well as Ki67 index and prognosis, was analyzed. Decorin expression was decreased in both the stroma (P < 0.001) and the tumor cells (P = 0.038) in NSCLC specimens. There was the lowest stromal expression of decorin in patients with G3 adenocarcinoma and higher Ki67 index in the stromal decorin-negative group. The Kaplan–Meier survival analysis demonstrated that lack of decorin in the stroma was correlated with a shorter DFS and OS (P = 0.005 and P = 0.010, respectively), while there was no significant association between decorin expression in the tumor cells and outcome. Multivariate analysis showed that reduced expression of decorin in the stroma was an independent prognostic factor for poor outcome including DFS (HR = 5.685, 95 % CI 0.493–0.933; P = 0.017) and OS (HR = 6.579, 95 % CI 0.484–0.908; P = 0.010). Negative decorin in the stroma combined with high Ki67 index predicted poorer outcomes for I–IIIA NSCLC patients. Our results provide data on decorin expression in both the stroma and cancer cells in NSCLC and reveal that reduced expression of stromal decorin correlates with high Ki67 index and has prognostic significance for poor outcome in I–IIIA NSCLC. Our data suggest that evaluating stromal decorin expression might be useful in assessing the prognosis and malignant potential.
Expression of SRF, E-cadherin and β-catenin in hepatocellular carcinoma and its clinical significances

Abstract:Objective: To investigate the expression of serum response factor(SRF),E-cadherin and βcatenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics of the tumor and prognosis.Methods: Tissue microarray and immunohistochemical study(EnVision method) for SRF,E-cadherin and βcatenin was performed on tissue microarray which consisted of 182 cases of hepatocellular carcinoma,92 cases of adjacent non-neoplastic liver tissues.Results: In the hepatocellular carcinoma group 75.8% tumor samples expressed SRF,and the levels is significantly higher than that in surrounding liver tissue(21.7%,P<0.05).The expression of SRF was positively correlated with the TNM stage,tumor metastasis and recurrence(P<0.05).In the hepatocellular carcinoma group,aberrant expression of E-cadherin and β-catenin was 59.3% and 58.8% respectively,and these levels were higher than those in the adjacent non-neoplastic liver tissue group(30.4% and 26.1% P<0.05).Aberrant expression of E-cadherin andβ-catenin was related to vascular cancer embolus,the TNM stage,tumor metastasis and recurrence(P<0.05).The expression of SRF correlated with aberrant expression of E-cadherin and β-catenin in hepatocellular carcinoma.Conclusion: In hepatocellular carcinoma,the overexpression of SRF and the downregulated expression of E-cadherin and β-catenin may be important mechanisms for promoting the invasion and metastasis of hepatocellular carcinoma and may also play a key role in the occurrence and development of hepatocellular carcinoma.
Expression of Interleukin - 10,E26 Transformation Factor - 1 and Tumor - Associated Macrophages in Cervical Cancer

Abstract:Background Cervical cancer is one of the women malignant tumors with high incidence. Interleukin 10 (IL-10),as a kind of cytokine with immunosuppressive effects,plays an important role in the occurrence and development of cervical cancer. Both tumor - associated macrophages(TAM)and E26 transcription factor 1(Ets1)can promote tumor growth, invasion and migration. Objective To investigate the expression of IL-10,Ets1 and TAM in cervical carcinoma tissues and their correlation with cervical cancer. Methods 72 cervical intraepithelial neoplasia( CIN)patients,76 cervical squamous cell carcinoma(SCC)patients and 40 benign uterine lesion patients underwent total hysterectomy,who were under treatment in Inpatient Department of Tangshan Gongren Hospital from September 2014 to March 2016,were selected into CIN group,SCC group and normal group separately. 3 subjects with cervical tissues in SCC group and 3 with cervical tissues in normal group were selected to make microarray hybridization sequencing by simple random sampling method;the expression levels of IL-10 and Ets1 mRNA in cervical tissues were detected by RT - qPCR method in all patients,the status and expression of TAM and TAM that IL-10 and Ets1 levels expressed by TAM(IL-10 + TAM,Ets1 + TAM)were detected by double immunofluorescence colocalization method,and the relationship between levels of IL-10 + TAM,Ets1 + TAM and clinical data was analyzed. Results Comparing with cervical tissues of normal group,the expression of IL-10 in cervical tissues of SCC group upregulated 3. 4 times,and Ets1 upregulated 2. 9 times(P < 0. 05). Expression levels of IL-10 and Ets1 mRNA in CIN group and SCC group were higher than those in normal group(P < 0. 05);expression levels of IL-10 and Ets1 mRNA in SCC group were higher than those in CIN group (P < 0. 05). The expression level of IL-10 mRNA in CIN group and SCC group was positively correlated with the expression level of Ets1 mRNA(r = 0. 453,P < 0. 01;r = 0. 456,P < 0. 01). TAM increased gradually in cervical tissues in normal group, CIN group and SCC group. The levels of IL-10 + TAM and Ets1 + TAM in CIN group and SCC group were higher than those in normal group(P < 0. 05);the levels of IL-10 + TAM and Ets1 + TAM in SCC group were higher than those in CIN group(P <0. 05). The levels of IL-10 + TAM and Ets1 + TAM in patients of SCC group were not related to age and tumor size(P > 0. 05);the levels of IL-10 + TAM and Ets1 + TAM in patients of SCC group were related to differentiated degree,clinical stage of cervical cancer and lymph node metastasis(P < 0. 05). Conclusion The expression level of IL-10,Ets1 and TAM increases with the progression of cervical lesions,TAM and its secreted IL-10 and Ets1 may be related to cervical cancer and clinical prognosis.
Expression of Vascular Endothelial Growth Factor-C and Its Receptor VEGFR-3 in Cervical Squamous Cell Carcinoma and Its Clinical Significance

Abstract:Objective To investigate the expression of vascular endothelial growth factor-C and its receptor VEGFR-3 in cervical squamous cell carcinomas and its clinical implication.Methods The detection of expression of VEGF-C and its receptor VEGFR-3 was immunohistochemically performed in 10 cases of normal cervical tissue,12 of CIN Ⅰ,12 of CIN Ⅱ,20 of carcinoma in situ and 66 of cervical invasive squamous cell carcinomas. Results The positive expressive rate of VEGF-C was 75.8% in cervical invasive squamous cell carcinomas,while that of VEGF-C expression was 30.0%,41.7%,41.7% and 45.0% in normal cervical tissue,CIN Ⅰ,CIN Ⅱ and carcinoma in situ,respectively.The difference between the invasive squamous cell carcinoma and the normal cervical tissue in VEGF-C expression was statistically significant(P0.01).The expression level of VEGF-C in cervical squamous carcinoma was positively correlated with the positive number of stromal lymphatic vessels stained by VEGFR-3.Enhanced expressions of VEGF-C in cervical squamous cancer cells and stromal VEGFR-3-positive lymphatic proliferation were both positively associated with poor clinical stages and regional lymph node metastasis.Conclusions The enhanced expression of VEGF-C and proliferation of stromal lymphangiogenesis play an important role in the tumoriogenesis and progression of cervical squamous cell carcinoma,and VEGF-C and VEGFR-3 might be valuable markers for predicting the infiltrating potential and lymph node metastasis of cervical squamous cell carcinoma.

Abstract:Objective:To explore the expression and clinical significance of dendritic cell-specific transmembrane protein (DC-STAMP) in papillary thyroid carcinoma (PTC).Methods:A total of 54 cases of papillary thyroid carcinoma in the First Affiliated Hospital of Zhengzhou University were collected. Real-time quantitative polymerase chain reaction (Real-time PCR) and Western blot were used to detect the expression levels of DC-STAMP in papillary thyroid carcinoma tissues and normal thyroid tissues. IHC was used to detect the expression levels of DC-STAMP protein, v-raf murine sarcoma viral oncogene homolog B1 (BRAF) protein and BRAF V600E mutant protein in papillary thyroid carcinoma tissue microarray. The t test was used to analyze the difference in DC-STAMP expression level between papillary thyroid carcinoma tissues and normal thyroid tissues, the χ2 test was used to analyze the correlation between DC-STAMP mRNA and clinicopathological features in papillary thyroid carcinoma, the χ2 test was used to analyze the correlation between DC-STAMP and BRAF V600E mutation. Results:The relative expression value of DC-STAMP mRNA in papillary thyroid carcinoma tissues was significantly higher than those in normal thyroid tissues [increased by (3.354±2.938) times, t=5.886, P<0.05]. Expression level of DC-STAMP mRNA was significantly related to the multifocal tumor ( χ2=4.396, P<0.05). The relative expression value of DC-STAMP protein in papillary thyroid carcinoma tissues was significantly higher than those in normal thyroid tissues. The expression of DC-STAMP in PTC was significantly correlated with BRAF V600E mutation ( χ2=3.962, P<0.05). Conclusion:DC-STAMP was significantly overexpressed in papillary thyroid carcinoma tissues and its expression level was correlated with multifocal tumor. DC-STAMP is significantly associated with BRAF V600E mutation and may play an important role in the invasion and metastasis of papillary thyroid carcinoma.
Protective Role of Decorin in Primary Hepatocellular Carcinoma

Abstract:Hepatocellular carcinoma (HCC) represents one of the most frequent type of primary liver cancers. Decorin, a small leucine-rich proteoglycan of the extracellular matrix, represents a powerful tumor cell growth and migration inhibitor by hindering receptor tyrosine kinases and inducing p21WAF1/CIP1. In this study, first we tested decorin expression in HCCs utilizing in silico data, as well as formalin fixed paraffin embedded tissue samples of HCC in a tissue microarray (TMA). In silico data revealed that DCN/SMA mRNA ratio is decreased in HCC compared to normal tissues and follows the staging of the disease. Among TMA samples, 52% of HCCs were decorin negative, 33% exhibited low, and 15% high decorin levels corroborating in silico results. In addition, applying conditioned media of hepatoma cells inhibited decorin expression in LX2 stellate cells in vitro. These results raise the possibility that decorin acts as a tumor suppressor in liver cancer and that is why its expression decreased in HCCs. To further test the protective role of decorin, the proteoglycan was overexpressed in a mouse model of hepatocarcinogenesis evoked by thioacetamide (TA). After transfection, the excessive proteoglycan amount was mainly detected in hepatocytes around the central veins. Upon TA-induced hepatocarcinogenesis, the highest tumor count was observed in mice with no decorin production. Decorin gene delivery reduced tumor formation, in parallel with decreased pEGFR, increased pIGF1R levels, and with concomitant induction of pAkt (T308) and phopho-p53, suggesting a novel mechanism of action. Our results suggest the idea that decorin can be utilized as an anti-cancer agent.
Identification of potential biomarkers in follicular thyroid carcinoma: bioinformatics and immunohistochemical analyses

Abstract:Abstract Objectives The prevalence of thyroid cancer has shown a progressive rise over time. This study aimed to explore the expression and underlying mechanisms of decorin (DCN) in follicular thyroid carcinoma (FTC), employing bioinformatics analysis and immunohistochemistry techniques. Methods The GSE27155 dataset was obtained from the Gene Expression Omnibus database. Differentially expressed genes (DEGs) and core DEGs were identified through data mining and analysis using the R language and online databases. The expression of core DEGs was validated using The Cancer Genome Atlas database. Additionally, the correlation between DCN and clinicopathological stage, tumor-infiltrating lymphocytes, and hotspot molecules in thyroid cancer was assessed using the Gene Expression Profiling Interactive Analysis and TIMER databases. Immunohistochemical (IHC) analysis was then conducted to verify the differential expression of core DCN in FTC and adjacent tissues. Results We confirmed the downregulation of three DEGs (DCN, GPC3, and PDGFRA). Furthermore, the analysis revealed a significant association between DCN expression and the clinical stage of patients with thyroid cancer (p<0.0001). DCN expression and the infiltration of several immune cells were positively correlated (p 0, p<0.05). Immunohistochemical analyses revealed a significantly lower staining score (3.071 ± 2.493) for DCN protein in cancer tissues than that in adjacent tissues (8.643 ± 2.094) (p<0.0001). Conclusions DCN is underexpressed and contributes to tumor progression in FTC. Thus, DCN serves as a tumor suppressor gene in FTC and a promising therapeutic target.

Abstract:Decorin, a member of the small leucine-rich proteoglycans family, exists and plays multifunctional roles in stromal and epithelial cells. Emerging evidences showed that decorin is dysregulated expression in a wide variety of human tumors and affects a broad biology process of cancer cells, including growth, metastasis, and angiogenesis. Recent studies demonstrated that decorin could affect A549 proliferation though decreasing TGF-β1, cycling D1 expression and increasing P53 and P21 expression. However, limited data are available on the effect of decorin on metastasis of non-small-cell lung cancer (NSCLC) cell lines and how decorin impacts metastasis is still unknown. In this study, we identified decorin mRNA expression through Oncomine database and verified the expression of decorin mRNA and protein in 50 patients who underwent primary surgical resection of a NSCLC in the Department of Thoracic Surgery, Jinling Hospital, Nanjing University School of Medicine, China, between September 2013 and March 2014 by quantitative reverse-transcriptase polymerase chain reaction (qRT-PCR) and Western blot. Also, the correlationship between decorin and the NSCLC patients’ clinical characteristics or survival (www.kmplot.com) was analyzed. Via ectopic expression analyses and Western blot, the roles of decorin in proliferation, metastasis, and the underline mechanism for decorin expression were further explored. We found that decorin was downregulated in NSCLC tissues compared with the adjacent normal lung tissues or normal tissues. Additionally, the expression of decorin was correlated with tumor size, lymph node metastasis, tumor stage, and prognosis. We also showed that overexpression of decorin could inhibit NSCLC cell lines proliferation and metastasis. Through Western blot analysis, we identified that E-cadherin and vascular endothelial growth factor (VEGF) are two key factors responsible for the growth arrest and metastasis inhibition induced by decorin in NSCLC. Our results indicated that decorin plays crucial roles in NSCLC against carcinogenesis and progression. Decorin might be a predictive factor and an attractive therapeutic target for NSCLC patients.
Special Expression of Thy-1 in Different Malignant Tumors

Abstract:To investigate the expression status of Thy-1 in malignant cancerous tissues and to evaluate whether Thy-1 could be a potential tumor marker.Immunohistochemistry method was used to examine Thy-1 expressions in different malignant tumor tissues. We used a monoclonal antibody specific for Thy-1 and the indirect Catalysis Signal Amplification method. With the help of tissue microarray (TMA), we examined expression the status of Thy-1 in 1451 different types of malignant tumor tissues, 144 normal tissues and benign lesions.All the malignant tumor cells were grouped as one group (malignant cells group) and normal or benign tumor tissue cells as another group (normal cells group). Among the entire 1451 malignant cases group, positive Thy-1 expression with diffuse and strong staining was observed in 734 (734/1451; 50.59%) cases. In the benign cases only 6 chronic cervicitis cases showed weak staining for Thy-1. All the normal tissue showed negative staining. One-way ANVOA analysis showed F value between these two groups was 147.229 (P < 0.0001).A significantly stronger expression of Thy-1 in malignant tumors was observed. Special overexpression of Thy-1 in malignant tumors suggests that Thy-1 might be a potential novel tumor marker with respect to cancer progression. Chronic cervicitis has some relationship with cervix carcinoma. When it develops into atypical hyperplasia, it will be a precancerous lesion for cervix carcinoma. In this research we found weak staining for Thy-1 in chronic cervicitis lesion, so Thy-1 may play a crucial role in carcinogenesis for cervix carcinoma. The research about the relationship between Thy-1 expressions in cancer cells and in precancerous lesion will provide some clues to understand the mechanism for carcinogenesis process.

Abstract:Objective To explore the expression of dendritic cells and its clinical significance in patients with different clinical pathological and prognositic of bladder transitional cell carcinoma(BTCC).Methods The dendritic cells were measured in 60 cases of BTCC by IHC using the polyclonal antibody to S-100 protein,compared with 15 normal bladder tissues.The patients were followed-up 3 years.Results There was significant difference in the amounts of DCs between patients with different pathologic grades,clinical stages of BTCC compared with the normal tissue(P<0.01).The amounts of DCs in the recurrent tumours were significantly lower than those non-recurrent group.BTCC's DCs were higher than that of the normal bladder tissue.Conclusion The amounts of DCs in tumour decreased with the grades rising.The DC may be used as a tumour maker in the BTCC,and it is more valuable for prognosis and recurrence of BTCC.

Abstract:目的研究Wnt1和β-catenin在结直肠腺瘤中的表达,及其在结直肠肿瘤发生中的作用.方法用免疫组织化学方法检测61例结直肠腺瘤、20例结直肠癌及其相应正常黏膜石蜡包埋组织标本的Wnt1和β-catenin的表达情况.结果结直肠腺瘤中,随非典型增生程度增高Wnt1表达差异有统计学意义(χ2=16.002,P＜0.05).不同腺瘤组织学类型(χ2=7.509,P＜0.05)、不同非典型增生程度间(χ2=6.666,P＜0.05)β-catenin的表达差异均有统计学意义.Wnt1表达和β-catenin表达呈正相关(P＜0.01,r=0.45).结论 Wnt1和β-catenin在结直肠肿瘤的发生中可能具有重要作用。

Abstract:Introduction: Decoy receptor 3 (DcR3), a newly discovered member of the tumor necrosis factor receptor (TNFR) superfamily, is not only upregulated in cancer cells derived from various cell lineages, but also correlates with the overall survival of certain cancer patients. The objective of the present study was to investigate the expression of DcR3 protein in tissue of gastric precancerous lesions and carcinoma. Material and methods: The expression of DcR3 protein in tissue of gastric carcinoma (GC, n=79), dysplasia (n=45), intestinal metaplasia (IM, n=37) and chronic superficial gastritis (CSG, n=42) was investigated by immunohistochemistry. Results: Expression of DcR3 in GC was significantly higher than that in dysplasia (P<.05); IM (P<.05) and CSG tissue (P<.001), respectively. It was also found that DcR3 expression in well differentiated GC was significantly lower than that in poorly differentiated specimens (P<.05). Moreover, patients in tumor-node-metastasis (TNM) stages I and II showed significantly lower DcR3 expression compared with that in stages III and IV (P<.05). In addition, DcR3 expression in both lymph node metastasis-negative patients and patients without systemic metastasis was significantly decreased in comparison with that in lymph node metastasis-positive patients(P<.05) and patients with systemic metastasis (P<.001). Conclusions: DcR3 is over-expressed in human GC and positively correlated with development and metastases of gastric lesions. The DcR3 gene might serve as an important molecular biological indicator in diagnosing and predicating the clinical outcome in GC patients.

Abstract:Purpose To investigate the expression and clinical significance of DOG-1 and C-erbB-2 in papillary thyroid carcinoma.Methods Immunohistochemical SP method was used to detect the expression of DOG-1 and C-erbB-2 protein in 48 cases of papillary thyroid carcinoma and 30 cases of benign thyroid lesions (15 cases of nodular goiter and 15 cases of Hashimoto's thyroiditis).Results The DOG-1 positive rate of thyroid papillary carcinoma (27.08％,13/48) was significantly higher than that of benign lesions (0,0/30),the difference was significant (P ＜ 0.05).The C-erbB-2 positive rate (39.58％,19/48) was significantly higher than that of benign lesions (3.33％,1/30),the difference was significant (P ＜ 0.05).Positive expression of DOG-1 as well as C-erbB-2 correlated with advanced TNM and presence of lymph node metastasis.The expression of DOG-1 and C-erbB-2 in patients with multifocal carcinomas combined with lymphatic metastasis showed a significant positive correlation(r =0.503,P =0.024).Conclusion The data suggest that DOG-1 and C-erbB-2 contribute to the pathogenesis and progress of thyroid papillary carcinoma.Our results introduce DOG-1 combined with C-erbB-2 as a promising biomarker for recurrence prediction and target therapy.
