Correlation of gene polymorphisms of coagulation factor Ⅶ with its activity and coagulopathy in patients with traumatic brain injury

Abstract:Objective To investigate effects of coagulation factor Ⅶ (CFⅦ) gene polymorphisms on CFⅦ activity (CFⅦa) and coagulopathy in patients with traumatic brain injury.Methods Eighty-four patients with isolated traumatic brain injury emergently admitted between March 2011 and March 2013 were enrolled in the study.Genotyping of-323P0/P10,R353Q,-401G/T,-402G/A,-670A/C,and IVS7 polymorphisms of CFⅦ gene was performed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).Results CFⅦa in different phenotypes of-323P0/P10,R353Q,-401G/T and IVS7 had significant differences (P <0.05),but there was no significant difference in-402G/A or-670A/C (P > 0.05).P10 alleles of-323P0/P10,Q alleles of R353Q,and H7 alleles of IVS7 were risk factors to CFⅦa < 77.5%.Multivariate regression analysis identified Q allele of R353Q (OR =4.401,95% CI:1.275-15.190,P < 0.05) as an independent risk factor of CFⅦa <77.5% and P10 allele of-323P0/P10 (OR =12.323,95% CI:1.445-105.062,P <0.05) as a risk factor independently associated with the presence of coagulopathy.Risk of coagulopathy in patients with P10-H7-Q haplotype was markedly higher than that in patients with other haplotypes (OR =7.624,95% CI:1.325-43.888,P < 0.05).Conclusions Q allele of R353Q is closely associated with the decline of plasma CFⅦa and P10 allele of-323P0/P10 is an independent risk factor to coagulopathy.Risk of coagulopathy in patients with P10-H7-Q haplotype is higher than that in patients with other haplotypes.
